1.Correlation analysis of AT1-AA,serum uric acid and blood urea nitrogen with hypertensive disorders complicating pregnancy

		                        			Objective To analyze the relationship between angiotensin type Ⅱ 1 receptor autoantibodies (AT1-AA),blood uric acid and urea nitrogen with hypertensive disorder complicating pregnancy (HDCP). Methods A total of 125 patients with HDCP diagnosed in the obstetrics department of Tianjin Municipal Bao-di District People's Hospital from August 2021 to February 2023 were randomly selected as the HDCP group, and 55 healthy pregnant women admitted to this hospital during the same period were selected as the control group.The HDCP group was divided into the gestational hypertension subgroup (n=77) and preeclampsia group (n=48) according to the degree of disease progression.The levels of serum uric acid,blood urea nitro-gen and AT1-AA were detected and compared between the HDCP group and control group and between the gestational hypertension subgroup and preeclampsia subgroup.Results There were no statistically significant differences in the age,gestational time,D-dimer,serum calcium and serum creatinine between the control group and the HDCP group (P>0.05).The levels of peripheral blood AT1-AA,serum uric acid and blood u-rea nitrogen in the HDCP group were higher than those in the control group,and the differences were statisti-cally significant (P<0.05).The levels of peripheral blood AT1-AA,serum uric acid and blood urea nitrogen in the preeclampsia subgroup were significantly higher than those in the gestational hypertension subgroup, and the differences were statistically significant (P<0.05).The Spearman rank correlation coefficient analysis showed that the levels of peripheral blood AT1-AA (r=0.301),serum uric acid (r=0.380) and blood urea nitrogen (r=0.257) levels were positively correlated with the severity degree of HDCP (P<0.01).The area under the curve (AUC) of AT1-AA,serum uric acid and blood urea nitrogen detection alone and combined de-tection in the diagnosis of HDCP were 0.680,0.714,0.647 and 0.725,and the corresponding Youden coeffi-cients were 0.368,0.380,0.306 and 0.439,indicating that the value of combined testing was higher than that of single item detection in the diagnosis of HDCP.Conclusion AT1-AA,serum uric acid and blood urea nitro-gen are correlated with the occurrence and development of HDCP,and the 3-item combined detection has high-er value.

		                        			Ferroptosis is a form of iron-dependent cell death caused by a disorder of iron lipid metabolism.Its mechanism mainly includes lipid metabolism and iron metabolism,which are complicated and strictly regulated by various metabolic and signaling pathways.Current studies have shown that ferroptosis is associated with cancer,atherosclerosis,and neurological diseases.Traditional Chinese medicine(TCM)is characterized by various active ingredients,relatively high safety,and lower treatment costs,which possess unique advantages in clinical applications.It has been widely used in the treatment of various diseases.Taking the regulation of ferroptosis by traditional Chinese medicine as the entry point may be a new direction for the future prevention and treatment of various diseases.In this review,we discussed the mechanism of iron lipid metabolic disorders based on ferroptosis and the research progress of traditional Chinese medicine intervention to provide reference and treatment strategies for the prevention and treatment of related diseases.
		                        		
		                        		
		                        		
		                        	
3.Attitudes of mothers of very low birth weight infants to breast milk donation in neonatal intensive care unit and its influencing factors

		                        			Objective:To investigate the willingness and influencing factors of mothers of very low birth weight infants (VLBWIs) in the neonatal intensive care unit (NICU) to donate breast milk or accept donated breast milk.Methods:Using the convenient sampling method, a total of 124 mothers of VLBWIs who were admitted to NICU of Children's Hospital of Fudan University from March to July 2019 were selected as investigation objects. The self-designed questionnaire was used to assess the willingness to donate breast milk, the willingness to accept breast milk donation and knowledge of breast milk donation. The survey was conducted by downloading the questionnaires through mobile terminals to analyze attitudes of mothers of VLBWIs towards breast milk donation and its influencing factors.Results:Among 124 mothers of VLBWIs, 51.6% (64/124) were reluctant to donate breast milk, mainly because they feared that the amount of breast milk after donation would affect the feeding of their children. 43.5% (54/124) did not want their children to receive donated breast milk, mainly because of doubts about the safety of donated breast milk. Adequate lactation was the main factor affecting the willingness to donate breast milk ( P<0.05) . Conclusions:It is necessary to strengthen the publicity and education on the safety of breastfeeding and breastmilk donation and improve the cognition of breastmilk donation.

		                        			Total 732 subjects aged 30-60 years undergoing health check-up at Beijing Hospital Medical Examination Center in 2009,who had no history of non-alcoholic fatty liver disease (NAFLD) were recruited in the study.According to the quartile of hemoglobin (HGB) level,the subjects were divided into 4 groups:Q1:HGB ≤ 131 g/L (n =192),Q2:HGB > 131 g/L and ≤ 140 g/L (n =178),Q3:HGB > 140 g/L and ≤152 g/L (n =184),Q4:HGB > 152 g/L (n =178).All participants were followed up for 4 years,the prevalence rates of NAFLD in groups Q1,Q2,Q3 and Q4 were 8.3% (16/192),17.4% (31/178),23.4% (43/184) and 25.3% (45/178),respectively (P <0.05).Logistic regression showed that the rates of NAFLD in groups Q2,Q3 and Q4 were 2.32 (1.22-4.41),3.36 (1.81-6.21) and 3.72(2.02-6.87) times higher as group Q1 (P < 0.05).Multiple logistic regression analysis showed that the hemoglobin level,TG and BMI were the independent risk factors of NAFLD.
		                        		
		                        		
		                        		
		                        	
6.Evaluating the applied effectiveness of Chinese diabetes risk score in physical examination of elderly population

		                        			Objective To analyze the effect of Chinese diabetes risk score in health checkup of elderly population and to explore the risk factors of abnormal glucose metabolism in the elderly patients.Methods Chinese diabetes risk score(C-DRs)screening,glucose tolerance test(OGTT),blood biochemical parameters and history collection were performed in 1 181 elderly people participating the health checkup.The area under the ROC curve(AUC)was used to evaluate the accuracy of the screening method.The effect of different cumulative C-DRs on screening target population was reflected by the Gordon index.Multivariate logistic regression analysis was used to analyze relevant risk factors for the glucose metabolic abnormalities.Results The AUC of screening for diabetes was 0.749(95%CI:0.715-0.782),and the best cut-point value was 32.5 points.The sensitivity was 86.50%,the specificity was 60.84%,and the Gordon index was 0.47(P=0.000).The AUC of screening for the pre-diabetes was 0.760(95%CI:0.733-0.787),and the best cut-point was 33.5 points.The sensitivity was 70.89%,the specificity was 68.72%,and the Gordon index was 0.40(P=0.000).The AUC of screening for MS was 0.797(95% CI:0.772-0.823),and the best cut-point value was 32.5 points.The sensitivity was 83.62%,the specificity was 64.90%,and the Gordon index was 0.49(P=0.000).The AUC of screening for insulin resistance was 0.609(95%CI:0.645-0.734),and the best cut-point value was 30.5 points.The sensitivity was 81.25%,the specificity was 44.81%,and the Gordon index was 0.26(P=0.000).Multiple logistic regression analysis showed that age over 80 years,abdominal obesity(waist circumference,male ≥ 90 cm,female ≥ 85 cm),hypertension,hypertriglyceridemia,family history of diabetes were risk factors for abnormality of glucose metabolism in the elderly.The odd ratio values of the above were 1.557,1.543,1.495,1.569,1.625,1.715(all P<0.05).Conclusions Chinese diabetes risk score may be used to screen for diabetes,metabolic syndrome and insulin resistance in the elderly population.Old age,abdominal obesity,hypertension,hypertriglyceridemia and family history of diabetes are independent risk factors for abnormal glucose metabolism in the elderly population.
		                        		
		                        		
		                        		
		                        	
7.Effects of apigenin on self-renewal and uPAR expression in NCI-H446 cell line.

		                        			To investigate the effect of apigenin on self-renewal for sphere-forming cells in human small cell lung cancer cell line NCI-H446 and the underlying mechanisms.
 Methods: Sphere-forming cells from NCI-H446 cell line were cultured in stem cell-conditioned culture medium with ultra-low attachment surface plates. The rate of sphere-forming cells in the second passage sphere-forming cells was used to evaluate the inhibitory effects of apigenin on the self-renewal for sphere-forming cells. The protein level of urokinase-type plasminogen activator receptor (uPAR) in spheroids was analyzed by Western blot.
 Results: Apigenin signifcantly inhibited the self-renewal of the second passage sphere-forming cells [0, 5.0, 10.0, 20.0 μmol/L apigenin: (18.2±1.9)%, (13.6±1.7)%, (10.6±1.6)%, (6.9±1.3)%, respectively] and down-regulated uPAR expression in a concentration-dependent manner (P<0.05).
 Conclusion: Apigenin inhibits the self-renewal capacity of sphere-forming cells in NCI-H446 cells, which may be associated with down-regulation of uPAR expression.

8.Differentiation of SOX-9 and GDF-5 co-transfected bone marrow mesenchymal stem cells into nucleus pulposus cells

		                        			BACKGROUND:Transplantation of mesenchymal stem cels to prevent and treat degeneration of the intervertebral disc is a feasible method. Mesenchymal stem cels co-transfected by SRY-related high mobility group-box gene 9 (SOX-9) and growth differentiation factor-5 (GDF-5) can differentiate into nucleus pulposus cels, in order to obtain greater effect of induction and proliferation of nucleus pulposus cels. OBJECTIVE:To investigate the effect of SOX-9 and GDF-5 co-transfection on the differentiation of rabbit bone marrow mesenchymal stem cels into nucleus pulposus cels. METHODS: We separated and cultured bone marrow mesenchymal stem cels from the bone marrow of rabbit aged 4 months. Passage 3 cels were divided into five groups andin vitro induced to differentiate into nucleus pulposus cels: non-transfected group, empty vector transfection group, SOX-9 transfection group, GDF-5 transfection group, SOX-9 and GDF-5 co-transfection group. At 14 days after transfection, RT-PCR was employed to assay SOX-9, GDF-5 and colagen type II mRNA expressions in bone marrow mesenchymal stem cels. The marker of nucleus pulposus cels-KRT19 expression was also detected by immunohistochemical staining. RESULTS AND CONCLUSION:In the co-transfection group, the mRNA expressions of SOX-9, GDF-5, and colagen type II were significantly higher than those in the SOX-9 transfection group, GDF-5 transfection group, and both these two groups, respectively (P < 0.05). Cels were positive for KRT19 in the SOX-9 and GDF-5 groups, and strongly positive for KRT19 in the co-transfection group. These findings indicate that double gene-transfected bone marrow mesenchymal stem cels are better than single gene-transfected cels with regard to differentiation into nucleus pulposus cels and secretion of extracelular matrix.
		                        		
		                        		
		                        		
		                        	
9.Study on relationship of quercetin on cervical carcinoma in nude mice model of tumor growth and the expression of heparanase

		                        			Objective To explore effect of quercetin on growth of cervical cancer,and its relationship to expression heparanase, and to investigate the possible mechanism of the inhibition effect of quercetin on cervical cancer .Methods HeLa cells were injected into nude mouse to establish the model of cervical cancer.30 nude mice with palpable tumors were randomly assigned to five groups:25,50,100 mg/kg quercetin groups,PI-88 positive group and blank control group.Transmission electron microscope was used to observe the ultrastructural changes of the vascular endothelial cells in each group.The expression of heparanase protein was detected by Western blot.Used blank control group for grey reference, relative grey value of each group was calculated for the further statistical analysis.Results 50,100 mg/kg quercetin groups and group of PI -88 made the volume of tumor tissues smaller.In 50 mg/kg quercetin group, swelling of VEC and mitochondria in tumor tissues was observed.In 100 mg/kg quercetin group, the disappearance of membraneaceous structures was found.In PI-88 positive group, the atresia of lumen, vacuole-shaped smooth endoplasmic reticulum and a dead cell were observed.50 and 100 mg/kg quercetin groups decreased the heparanase protein levels significantly(P<0.05).Furthermore, PI-88 positive group could decrease the heparanase protein levels significantly(P<0.05), compared to 50 and 100 mg/kg quercetin groups.However, the difference between the 25 mg/kg quercetin group and blank control group showed no statistical significance.Conclusion The growth of cervical cancer could be inhibited by 50, 100 mg/kg quercetin.Meanwhile, the ultrastructural changes of tumor endothelial cells could be induced by 50,100 mg/kg quercetin.These changes may relate with the decrease of heparanase protein level .
		                        		
		                        		
		                        		
		                        	
10.Effect of Gambogic acid on proliferation and invasion in colon cancer LoVo cell line

		                        			Objective To investigate the effects of gambogic acid on proliferation,invasion and protein expression of MMP-2 in LoVo human colon cancer cell line.Methods The CCK-8 assay was used to determine the anti-proliferation ability.Transwell chamber assay was used to study the invasion of LoVo colon cancer cells.Western Blot was applied to examine the protein expression of MMP-2.Results CCK-8 assay showed that after cells treated with 1,2,4 μmol/L GA for 24 h,48 h the inhibition rates were(0.16±0.11)%,(0.24±0.08)%,(0.58±0.01)%,(0.67±0.03)%,(0.79±0.01)% and (0.27±0.05)%,(0.69±0.09)%,(0.85±0.01)%,(0.87±0.01)%,(0.89±0.01)%,and there had statistically significant differences between control group (0 μmo/L) with experimental group (P<0.05).Gambogic acid can effectively inhibit LoVo cells invasion at a low-dose concentration,which the numbers of cells that digested the matrigel in control group (0 μmo/L)was 120.50± 8.69 and 69.83 ±4.75 in experimental group (1 μnol/L),P< 0.05).Western Blot revealed that Gambogic acid can down-regulate the expression of MMP-2,which the levels of down-regulating the expression of MMP-2 after treated with 1,2,4 umol/L GA were 48.67%,74.72%and 82.70% (P<0.05) Conclusion Gambogic acid can effectively inhibit the growth and invasion of the LoVo cells,which may be related with down-regulating the expression of MMP-2.
