1.Effects of Gouteng Jiangya Jieyu Prescription on Learning and Memory Ability and Hippocampal Autophagy-Related Protein Expression in Rats with Hypertension Complicated with Depression

Objective To study the effects of Gouteng Jiangya Jieyu Prescription(Uncariae Ramulus cum Uncis,Gastrodiae Rhizoma,Pheretima,Puerariae Lobatae Radix,Salviae Miltiorrhizae Radix et Rhizoma,etc.)on learning and memory ability,hippocampal inflammatory response and autophagy-related protein expression in rats with hypertension complicated with depression(HD).Methods Thirty spontaneously hypertensive rats(SHR)were randomly divided into model group,positive control group(Levamlodipine Besylate 0.45 mg·kg-1+Fluoxetine Hydrochloride 1.80 mg·kg-1)and Gouteng Jiangya Jieyu Prescription high-,medium-and low-dose groups(25.38,12.69,6.34 g·kg-1).Another 6 SD rats were used as blank control group.The SHR rats were intervened by chronic mild unpredictable stress combined with solitary rearing to replicate the HD rat model.At the same time,intragastric administration was given once a day for 6 weeks.The systolic blood pressure and diastolic blood pressure of rat tail artery were measured by non-invasive sphygmomanometer.The learning and memory ability of rats was detected by Morris water maze test.The ultrastructure of hippocampal neurons was observed by transmission electron microscope.The contents of interleukin-1β(IL-1β),IL-18 and IL-10 in hippocampus were detected by ELISA.The expression of autophagy-related proteins Beclin1 and Bcl-2 in hippocampus was detected by immunohistochemistry.The expression of autophagy-related proteins LC3Ⅰ and LC3Ⅱ in hippocampus was detected by Western Blot.Results Compared with the blank control group,the SBP and DBP of the rats in the model group were significantly increased from week 1-6(P<0.01).The escape latency was significantly prolonged on the third and fourth day(P<0.01).The first time of crossing the platform was significantly prolonged(P<0.01),the times of crossing the platform area was significantly reduced(P<0.05),and the retention time of the platform area was significantly shortened(P<0.01).The neuronal cell body was obviously swollen,the ridge was destroyed,the nucleus was shrunk,and a large number of autophagosomes appeared;the contents of IL-1β and IL-18 in hippocampus were significantly increased(P<0.01).The ratio of LC3Ⅱ/LC3Ⅰ protein expression and the expression of Beclin1 protein in hippocampus were significantly up-regulated(P<0.05,P<0.01),and the expression of Bcl-2 protein was significantly down-regulated(P<0.01).Compared with the model group,the SBP of rats in the low-dose group of Gouteng Jiangya Jieyu Prescription was significantly decreased at the weeks 1,3,4,5,6(P<0.01),and the DBP was significantly decreased at weeks 1,3,4,5(P<0.05,P<0.01).The SBP of the rats in the medium-dose group of Gouteng Jiangya Jieyu Prescription was significantly decreased at weeks 1,5,6(P<0.01),and the DBP was significantly decreased at week 4(P<0.05).The SBP of rats in the high-dose group of Gouteng Jiangya Jieyu Prescription was significantly decreased in the first week(P<0.01).The escape latency of rats in the high-and medium-dose groups of Gouteng Jiangya Jieyu Prescription was significantly shortened on the third day(P<0.05),and the escape latency of rats in the high-and low-dose groups of Gouteng Jiangya Jieyu Prescription was significantly shortened on the fourth day(P<0.05).The first crossing platform time of rats in the high-,medium-and low-dose groups of Gouteng Jiangya Jieyu Prescription was significantly shortened(P<0.01).The times of rats crossing the platform area in the medium-and low-dose groups of Gouteng Jiangya Jieyu Prescription were significantly increased(P<0.05),and the retention time in the platform area was significantly prolonged(P<0.05).In the administration group,the degree of hippocampal neuron damage was reduced,the nuclear shrinkage was significantly improved,and the autophagosomes were reduced.The contents of pro-inflammatory factors IL-1β and IL-18 in the hippocampus of rats in the high-and medium-dose groups of Gouteng Jiangya Jieyu Prescription were significantly decreased(P<0.05,P<0.01).The content of anti-inflammatory factor IL-10 in the hippocampus of rats in the high-dose group of Gouteng Jiangya Jieyu Prescription was significantly increased(P<0.01).The protein expression ratio of LC3Ⅱ/LC3Ⅰ in hippocampus of high-,medium-and low-dose groups of Gouteng Jiangya Jieyu Prescription was significantly down-regulated(P<0.01),and the expression of Bcl-2 protein was significantly up-regulated(P<0.01).The expression of Beclin1 protein in the hippocampus of the high-and medium-dose groups of Gouteng Jiangya Jieyu Prescription was significantly down-regulated(P<0.05,P<0.01).Conclusion Gouteng Jiangya Jieyu Prescription can reduce the tail arterial pressure of HD rats,improve their learning and memory ability,and alleviate hippocampal neuronal damage.The mechanism may be related to reducing the release of inflammatory factors,increasing the level of anti-inflammatory factors,and regulating the expression of hippocampal autophagy-related proteins LC3Ⅱ/LC3Ⅰ,Beclin1 and Bcl-2.
2.Effect of Artemisinin and Its Derivatives in Treatment of Nervous System Diseases: A Review

With the rapid development of social economy, the number of patients with nervous system diseases has increased, and the incidence of the population has a trend of younger, which has a serious impact on life health and social economy. Artemisinin is an active antimalarial component extracted and isolated from Artemisia annua, a Chinese medicinal material. Artemisinin and its derivatives, in addition to the antimalarial effect, also have anti-parasitic, anti-fungal, anti-viral, hypoglycemic, hypolipidemic, anti-tumor, and anti-inflammatory effects, showing a wide range of pharmacological activities. In the past five years, research on the new pharmacological effects of artemisinin and its derivatives has been deepening, and the efficacy of artemisinin and its derivatives in nervous system diseases has attracted much attention, including anti-neuroinflammation, anti-oxidative stress, maintaining the stability of the blood-brain barrier, regulating the release of neurotransmitters, repairing neuronal damage, and promoting neuronal regeneration. These pharmacological effects indicate that artemisinin and its derivatives are potentially capable of neuroprotection. By sorting out literature on the pharmacological activity of artemisinin and its derivatives in nervous system during 2019-2024, this paper systematically summarized the protective effects of artemisinin and its derivatives against nervous system diseases such as stroke, neurodegenerative diseases, neuroimmunological diseases, neuralgia, and nervous system tumors. This review is expected to provide clues and evidence for new indication expansion of artemisinin drugs, innovative drug development, and clinical treatment of nervous system diseases.
3.Mechanisms of Outdoor Air Pollution Affecting Cardiovascular Diseases: A Review

The impact of air pollution on human health has always been a research hotspot in the global health field. Outdoor air pollutants composed of multiple components can enter the human body through various pathways. Cardiovascular diseases are a group of diseases caused by outdoor air pollutants. Studies have shown that the incidence of cardiovascular diseases, including hypertension, arrhythmia, and heart failure, is significantly increased among people exposed to air pollution environments. Air pollutants such as fine particulate matter, nitrogen dioxide, ozone, carbon monoxide, and sulfur dioxide are closely related to the occurrence of cardiovascular diseases, and short-term and long-term exposure causes different cardiovascular risks. By reviewing the relevant research reports from 2019 to 2024, this article summarizes the epidemiological evidence of cardiovascular diseases caused by different air pollutants. It generalizes the pathways through which air pollutants accelerate the progression of cardiovascular diseases. These pathways include oxidative stress, inflammatory response, thrombosis, extracellular vesicle release, endoplasmic reticulum stress, apoptosis, endothelial dysfunction, autonomic nervous system imbalance, and their interactions. Based on the different mechanisms of air pollution on cardiovascular diseases, the article analyzes the main progress in drug intervention and summarizes the roles of various active ingredients and compound prescriptions of traditional Chinese medicine in treating air pollution-related cardiovascular diseases, providing reference for the research on the mechanisms and drug interventions of air pollution-related cardiovascular diseases.
4.Optimized CRISPR/Cas9 System in Brain Science and Application Prospects in Field of Traditional Chinese Medicine

Clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR associated nuclease 9 (CRISPR/Cas9) is a self-defense system found in bacteria and archaea that enables targeted gene editing based on the principle. Due to its universality, efficiency, and simplicity, CRISPR/Cas9 has been applied in the pathological mechanism and prevention and treatment of diseases in many fields. Cerebrovascular diseases and central nervous system diseases seriously endanger human health. Stroke is related to genetics, unhealthy living habits, chronic diseases, and other factors. The brain tissue structure is complex and the cell types are diverse. It is difficult for a universal gene editing platform to study target genes safely, specifically, and efficiently. Scholars have continuously improved and optimized gene editing technology, explored the potential and research methods of gene editing technology, and promoted the research process of brain science. After a brief introduction to the mechanism of CRISPR/Cas9, this paper mainly summarized the optimization of the system in the fields of cerebral science including delivery methods, adeno-associated virus assembly, and new nanomaterials. Its application in cerebrovascular research including vascular homeostasis, microglial homeostasis, angiogenesis, blood-brain barrier, and drug screening was also summarized. Finally, this paper prospected the development of CRISPR/Cas9 in traditional Chinese medicine, hoping to provide references for related research design.
5.Protective Effect of Shenlian Prescription on Acute Lung Injury Induced by Particulate Matter Exposure in Rats

ObjectiveTo observe the protective effect of Shenlian prescription on acute lung injury induced by particulate matter (PM) exposure in rats and explore the mechanism. MethodFifty male SD rats were randomly divided into the control group, model group, Shenlian low-dose group (4.32 g·kg-1), Shenlian high-dose group (8.64 g·kg-1), and roflumilast group (3.46 mg·kg-1), with 10 in each group. Pre-administration with drugs by gavage was performed for one week. On the 8th and 11th days, the control group was instilled with normal saline in the trachea and the other groups with PM suspension to establish a rat model of acute lung injury induced by PM exposure. After modeling, drugs were given continuously until the end of the experiment. Forty-eight hours after the last exposure, the lung function of rats was detected. Then the rats were sacrificed and the lung morphological changes and pathological changes by hematoxylin-eosin (HE) staining were observed. CD68 expression in lung was detected by immunohistochemistry, and the levels of lung injury markers surfactant protein A (SP-A) and Clara cell protein16 (CC16) in serum were detected by enzyme-linked immunosorbent assay (ELISA). The mRNA expression of interleukin-1α (IL-1α), IL-6, IL-18, and monocyte chemoattractant protein-1 (MCP-1) in lung tissue was measured by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). ResultCompared with those in the control group, the rats in the model group had decreased lung function and obvious structural damage of lung tissue, PM deposition, and infiltration of CD68 positive cells. The expressions of IL-1α, IL-6, IL-18, and MCP-1 in lung tissue were increased (P<0.01). Compared with the model group, Shenlian prescription low and high doses restored the rats' lung function injury(P<0.05,P<0.01), improved lung morphological and pathological structure, and reduced PM deposition. Infiltration of CD68 positive cells in lung was not significantly decreased. The levels of inflammatory factors IL-1α, IL -6, IL-18, and MCP-1 in lung were lowered (P<0.01). ConclusionShenlian prescription could protect the rats' lung injury caused by PM exposure, improve lung morphology, and reduce PM deposition and inflammatory factor expression.
6.Analysis on Difference of Medicinal Properties Between Atractylodes lancea and A. chinensis Based on Correlation of Energy Metabolism and Dryness

ObjectiveTo explore the correlation between dryness and energy metabolism of Atractylodis Rhizoma, and to analyze the difference of medicinal properties between Atractylodes lancea and A. chinensis. MethodA total of 110 healthy SD rats were randomly divided into 11 groups, including normal group, volatile oil of A. lancea 1-5 group (S1-S5 group, doses of 447, 473, 442, 489, 496 mg·kg-1) and volatile oil of A. chinensis 1-5 group (N1-N5 group, doses of 197, 118, 281, 222, 185 mg·kg-1), the administration volume was 0.01 mL·g-1 with intragastric administration for 21 days. Dryness effect of A. lancea and A. chinensis on rats was evaluated by comparing the body weight, drinking water volume, urine volume, whole blood viscosity and pathological sections of submandibular gland stained with hematoxylin-eosin (HE). The expression of aquaporin 2 (AQP2) in rat kidney was measured by immunohistochemistry, the mRNA expressions of cytochrome C oxidase subunit 7A2 (COX7A2) and succinate dehydrogenase (SDH) complex subunit D (SDHD) in liver tissue were detected by real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). The contents of SDH, lactate dehydrogenase (LDH) and sodium ion-potassium ion-adenosine triphosphatase (Na+-K+-ATPase) in rat plasma were determined by colorimetry. The quality of A. lancea and A. chinensis was evaluated by coefficient of variation method, and Pearson correlation coefficient was used to analyze the correlation between dryness and energy metabolism. ResultCompared with the normal group, the amounts of drinking water and urine in volatile oil of A. lancea group and volatile oil of A. chinensis group increased, and the submandibular gland acini atrophied, the whole blood viscosity of rats in the volatile oil of A. lancea group increased significantly (P<0.01), the expression levels of COX7A2 and SDHD mRNA, the activities of SDH, LDH and Na+-K+-ATPase increased significantly (P<0.01), and the expression of AQP2 in kidney decreased significantly (P<0.01). Compared with the normal group, the expression level of COX7A2 mRNA, SDH activity and whole blood viscosity in the volatile oil of A. chinensis group increased (P<0.05, P<0.01), the AQP2 and SDH mRNA expression levels, LDH and Na+-K+-ATPase activities had no significant difference. The comprehensive score analysis of each index showed that the effect of volatile oil of A. lancea on dryness and energy metabolism was stronger than that of volatile oil of A. chinensis, and there was a positive correlation between dryness index and energy metabolism index. ConclusionThe two indexes show that medicinal properties of A. lancea is stronger than that of A. chinensis, and energy metabolism is closely related to the dryness of Atractylodis Rhizoma. It is suggested that it is reasonable to evaluate the dryness effect of Atractylodis Rhizoma from the perspective of energy metabolism, which can further enrich the evaluation indexes of medicinal properties.
7.Relationship between fundamental movement skills and physical fitness in children

Objective:
To investigate the relationship between fundamental movement skills(FMS) and physical-related fitness(PF) in 5-6 aged children in Beijing and to provide the corresponding theoretical basis for the further development of the scien tific research work of children sports activities.
Methods:
M-ABC-2(Movement Assessment Battery for Children-2)was used to assess FMS of the children aged 5-6 in 3 kindergartens of Beijing; “National Physical Fitness test Standards Manual-Preschool Part” was used as the standard for assessment of children’s PF; Canonical Correlation Analysis was used to analyze the relationship between FMS and PF.
Results:
A total of 107 children’s complete data were obtained. The overall development level of 5-6 yearold children’s physical fitness could positively predict their the manual dexterity (B=0.24, P<0.05); In the test of individual PF, the muscle strength and balance ability exhibited by the tennis throwing and walking balance wood were also strongly correlated with the manual dexterity (r=0.43, P<0.01), and the standing long jump and the continuous jump of both feet also showed a positive correlation with the manual dexterity (r=0.35, P<0.05).
Conclusion
There is a certain relationship between the PF and FMS of children aged 5-6. The development of coordination and stability will play a positive role in promoting the development of their FMS. At the same time, parents and teachers should pay attention to the development of children's fine motor skills to promote their overall healthy growth.
8.The analysis of myofascial trigger points in the infraspinatus muscle misdiagnosed as cervical spondylotic radiculopathy

Objective:To analyze the cause of myofascial trigger points(MTrPs)in the infraspinatus muscle misdiagnosed as cervical spondylotic radiculopathy(CSR).Methods:Clinical data of patients with MTrPs in the infraspinatus muscle misdiagnosed as CSR were collected in Peking University Third Hospital from October 2016 to September 2019.Results:A total of 28 cases with MTrPs in the infraspinatus muscle were misdiagnosis as CSR.All patients received CT or magnetic resonance imaging evaluation of cervical spine, while none of them received physical examination in posterior shoulder area before confirming MTrPs in the infraspinatus muscle.The pain range was mainly located in the radial side of the shoulder, upper arm and forearm(89.3%)and was mainly unilateral(89.3%). The MTrPs in the supraspinatus muscle were mostly 2~3 points(67.8%). The visal analogue scale(VAS) score was significantly decreased after dry acupuncture as compared with pre-treatment[(2.7±1.4) vs.(6.1±1.3), P<0.05], and the overall treatment efficiency defined as VAS score decrement by more than 50%, was 82.1%. Conclusions:The neglect of medical history inquire and physical examination, and over-reliance on imaging findings are the important factors for misdiagnosis of MTrPs in the infraspinatus muscle as CSR.
9.Initial experience of using improved motion sensitized driven equilibrium prepared balanced steady-state free precession

Objective To assess the application value of NCE-MRA using iMSDE prepared bSSFP sequence in lower limb in patients with diabetes. Methods This prospective study included a total of 35 patients with type II diabetes who underwent CE-MRA on the 1.5T MR scanner after the NCE-MRA. The obtained MIP images were independently rated by two radiologist with a four score table and using CE-MRA as a reference standard to evaluate the diagnostic accuracy of NCE-MRA for the narrowed arteries. The difference of the percent age of diagnostic arterial segments between NCE-MRA and CE-MRA on diabetic patients was evaluated by the χ2 test. Results Compared with CE-MRA, the diagnostic-value arterial segment ratio of pelvic arteries on NCE-MRA is decreased significantly. The sensitivity, specificity, positive predictive value, negative predictive value, and accuracy of two readers in NCE-MRA were 95%/87%, 96%/95%, 57%/72%, 99%/99%, and 96%/95%(k=0.76), respectively. Conclusions The NCE-MRA using iMSDE prepared bSSFP sequence is capable of depicting vascu-lar lesions for the lower extremities in diabetic patients with the advantages of contrast agent free, short scan times and good diagnostic value in the thigh and calves.
10.Comparative Study on the Content of Microelement Cu, Zn and Mn in Eye Tissue of Rats Treated by Rhi-zoma Corydalis before and after Vinegar Processing

Objective:To study the influence of Rhizoma Corydalis before and after vinegar processing on the microelement content in the eye tissues of rats to verify the vinegar processing leading the herb to liver meridian. Methods: The rats were divided into the crude Corydalis group, vinegar processing group and negative group. The content of microelement Cu, Zn and Mn in rat ocular tissues was determined and compared after the administration. Results:Corydalis could significantly increase the content of Cu and Zn in fun-dus blood and that of Zn and Mn in the eyeball vitreous, and Rhizoma Corydalis after vinegar processing showed more obvious effect. Conclusion:Rhizoma Corydalis with vinegar processing can improve the distribution of microelements in eye tissues, and because liver opens at eyes, the results provide evidence for Corydalis after vinegar processing leading to liver meridian.
