Objective To study the protective effect of lipoxin A4(LXA4)against sepsis-induced acute kidney in-jury(SAKI)in rats and its effect on the expression of toll-like receptor 4(TLR4),myeloid differentiation factor88(MyD88),nuclear factor kappa B(NF-κB)in the kidney. Methods Forty male specific pathogen-free C57BL/6J mice were randomly divided into SAKI group,SAKI+LXA4 group, sham procedure group, sham procedure+LXA4 group. The mice in SAKI group and SAKI+LXA4 group were given cecum ligation and puncture(CLP)to establish SAKI animal models. The mice in SAKI+LXA4 group and sham procedure+LXA4 group were given LXA4(40 ng/kg)with intraperitoneal injection 30 mins after CLP.All mice were sacrificed at the 24th hour after CLP to collect serum, urine and kidney tissues. The enzyme linked immunosorbent assay(ELISA)was used to test the serum creat-inine(Scr),blood urea nitrogen(Bun),interleukin-1 β(IL-1β),IL-6,tumor necrosis factor-α(TNF-α)and urine neutrophil gelatinase-associated lipocalin(NGAL),kidney injury molecule 1(KIM-1)levels of mice. The pathologi-cal changes were examined through hematoxylin-eosin(HE)staining and Periodic Acid-Schiff(PAS)staining.And the mRNA levels of TLR4,MyD88,NF-κB p65 were detected through real-time PCR(RT-PCR);the expression lev-els of TLR4,MyD88,NF-κB p65,phospho-NF-KB p65(p-NF-KB p65)were detected through immunohistochemistry(IHC)and Western blot assay. Results ELISA showed that the values of Scr,Bun,IL-1 β,IL-6,TNF-α,NGAL,KIM-1 in SAKI group were higher than those in SAKI+LXA4 group(P<0.05),and there were no significant differences in Scr,Bun,IL-1 β,IL-6,TNF-α,NGAL,KIM-1 between sham procedure group and sham procedure+LXA4 group.HE and PAS staining showed that SAKI group had severer pathological changes than SAKI+LXA4 group in the kidney structure(P<0.05),while pathological structures of the kidney were normal in sham proce-dure group and sham procedure+LXA4 group.RT-PCR showed that the mRNA levels of TLR4,MyD88,N F-κB p65 in SAKI group,SAKI+LXA4 group were higher than sham procedure group and sham procedure+LXA4 group(P<0.05);the mRNA levels of TLR4,MyD88,NF-κB p65 in SAKI group were higher than SAKI+LXA4 group(P<0.05);The results of IHC,Western blot assay were as follows:The expression levels of TLR4,MyD88,NF-κB p65,p-NF-κB p65 in SAKI group,SAKI+LXA4 group were higher than sham procedure group and sham procedure+LXA4 group(P<0.05);the expression levels of TLR4,MyD88,NF-κB p65,p-NF-κB p65 in SAKI group were higher than SAKI+LXA4 group(P<0.05).Conclusion TLR4/MyD88/NF-KB pathway plays an important role in the occurrence and development of SAKI,and LXA4 may reduce SAKI by inhibiting TLR4/MyD88/NF-κB sig-naling pathway.
2.Preliminary study on the distributions of dose differences and their correlations with tumor types in independent verification
Chinese Journal of Medical Physics 2024;41(3):289-293
Objective To explore the distributions of dose differences and their correlations with tumor types in independent three-dimensional dose verification for intensity-modulated radiotherapy(IMRT)plan, and to establish prediction models. Methods The fixed field IMRT plans of 180 patients with head and neck, chest, and abdominal tumors were collected.Independent sample t-test was used to analyze the consistency between the initial dose calculated in treatment planning system and the verified dose for evaluating the feasibility of independent validation. The distributions of planning target volume(PTV)dose differences among different tumor types were analyzed. The correlations of PTV dose differences with conformity index(CI)and homogeneity index were analyzed using correlation analysis and multiple linear regression method. Results The PTV dose differences for head and neck, chest, and abdominal tumors were within±1.2%,and the average 3%/3 mm gamma passing rate between the initial dose and the verified dose was higher than 99.5%,indicating good consistency in dose distribution between independent validation and treatment planning system. The distributions of PTV dose differences were different in head and neck, chest, and abdominal plans, and were significantly correlated with CI which could be used to preliminarily determine the PTV dose differences in independent validation. The multiple regression equation based on CI and homogeneity index could be used to estimate the dose verification differences of IMRT plan. Conclusion Independent verification can quickly achieve pre-treatment validation for radiotherapy plans, improving the efficiency of quality control of radiotherapy plans. The estimation based on dose differences provides guidance for optimizing radiotherapy plans.

Abstract
In recent years, the overall incidence of scoliosis in children and adolescents in China is on the rise, and it has become the third largest "killer" affecting the health of children and adolescents in China after obesity and myopia. The dysfunction caused by spinal deformity has no obvious pathological manifestations or clinical symptoms in children and adolescents (except for severe scoliosis). It is easy to be ignored in the early stage, but if it is not intervened in time, this will cause serious damage to the long term prognosis and quality of life of the patients. Therefore, through early scoliosis screening, we can effectively monitor the spinal health status of children and adolescents in China, ensure timely diagnosis of the disease, timely intervention (brace and other conservative treatment), and play the role of early prevention. Avoid further deterioration of scoliosis and surgical intervention. This article will focus on the research progress of screening and conservative treatment of scoliosis.

Objective:To study the association between preoperative hemoglobin amount and incidence of lower limb deep vein thrombosis (DVT) in patients with lower limb fracture. Methods:A retrospective study was performed of the 2, 482 patients with lower limb fracture who had been treated at Department of Orthopaedics Trauma, Honghui Hospital Affiliated to Xi'an Jiaotong University from July 2014 to August 2019. They were 1, 174 males and 1, 308 females with an age of (60.6±19.3) years. Recorded were the patients' age, gender, injury time, hemoglobin amount, D-dimer measurement, combined medical conditions, time and results of ultrasound vein examination on both lower extremities. According to the ultrasound results, the patients were divided into a thrombosis group and a thrombosis-free group. The 2 groups were compared in hemoglobin amount. Logistic regression was used to analyze the relationship between preoperative hemoglobin amount and incidence of lower limb DVT. The patients were divided into 5 groups according to the quintile of hemoglobin amount; the incidences of thrombosis were compared between the 5 groups. Results:The total incidence of DVT in this cohort was 29.53%(733/2, 482). The hemoglobin amount in the thrombosis group was (116.57±19.24) g/L, significantly lower than that in the thrombosis-free group (124.76±19.79) g/L ( P<0.05). The preoperative hemoglobin amount was a risk factor for incidence of DVT after a lower limb fracture ( OR=0.985, 95% CI: 0.980 to 0.990, P<0.001). As the quintile level of hemoglobin increased, the incidence of DVT showed a downward trend. In comparison of the group with the highest DVT incidence (40.58%) and the group with the lowest DVT incidence (17.27%), the risk increased by 2.386 times (95% CI: 1.718 to 3.315). Conclusions:The preoperative hemoglobin amount can affect the DVT incidence after a lower limb fracture, and a low hemoglobin amount may more likely lead to lower limb DVT.
