1.Effects of bisdemethoxycurcumin promoting neuronal differentiation of neuroblastoma cells in mice and its mechanism

OBJECTIVE To study the effects of the curcumin derivative bisdemethoxycurcumin (BC) promoting neuronal differentiation of neuroblastoma cells Neuro-2a (N2a) in mice and its mechanism. METHODS The effects of BC (1, 2, 4, 6, 8, 10 μmol/L) on the viability of N2a cells were detected by MTT assay to determine the concentration range of drug treatment. The control group, retinoic acid (RA) group (10 μmol/L) and BC groups (1, 2 and 4 μmol/L) were set up, and the length of neural protrusions of the differentiated cells was measured and the cell differentiation rate was calculated after 48 h and 72 h of culture. Compared with 0 min group, Western blot was used to detect the phosphorylation levels of protein kinase B (Akt), extracellular- signal regulated kinase 1/2 (ERK1/2), and p38 mitogen-activated protein kinase (p38) proteins in cells treated by 4 μmol/L BC for 5, 15, 30, 60, 120 min. After intervention with inhibitors LY294002 (LY) and PD98059 (PD), the effects of BC on Akt and ERK1/2 protein phosphorylation levels and promoting neural differentiation were further validated. RESULTS According to the MTT experiment, the BC concentrations for subsequent induction of cell differentiation were determined to be 1, 2, and 4 μmol/L. After 48 hours of differentiation, compared with the control group, the cell differentiation rate in RA group and BC 1, 2 and 4 μmol/L groups, the length of cellular neural processes wjxhhxx413@163.com in the BC 4 μmol/L group significantly increased (P<0.05 or P<0.01);after inducing differentiation of BC for 72 hours,compared with the control group, the cell differentiation rate and the length of cellular neural processes in the RA group, the cell differentiation rate in the BC 4 μmol/L group, and the length of cellular neural processes in the BC 2 μmol/L group all significantly increased (P<0.05 or P<0.01).Compared with the 0 min group, the phosphorylation levels of Akt, ERK1/2, and p38 proteins in cells of the 5, 15, 30, 60 and 120 min groups increased to varying degrees after treated by 4 μmol/L BC, and some differences were statistically significant (P<0.05 or P<0.01). After adding the inhibitor LY/PD, compared with the BC group, the phosphorylation level of ERK1/2 protein in the PD+BC group cells were significantly reduced (P<0.01), and the cell differentiation rates in the LY group, LY+BC group, PD group, and PD+BC group was significantly reduced (P<0.01). CONCLUSIONS BC promotes N2a cell differentiation mainly by increasing cell differentiation rate and neural protrusion length. The mechanism may be related to the activation of mitogen-activated protein kinase/ ERK and PI3K/Akt signaling pathways.
2.Relationship among physical activity,mild depressive symptoms and frontal alpha power asymmetry in college students

Objective:To investigate the correlation among physical activity,mild depressive symptoms and frontal alpha power asymmetry in college students.Methods:Seventy college students with mild depressive symp-toms who conformed to the standard of the Self-Rating Scale for Depression(SDS)of 53-62 and 70 normal col-lege students were recruited.The frontal alpha power was measured under quiet and closed-eye state,and the total physical activity(PA)was assessed with the International Physical Activity Questionnaire.Results:The college students with mild depressive symptoms had lower Total PA scores,right frontal alpha power and frontal alpha a-symmetry(FAA)than the normal controls(P<0.001).In college students with mild depressive symptoms,the to-tal PA scores(r=-0.29,P<0.05)and FAA(r=-0.41,P<0.001)were negatively correlated with SDS scores,and the total PA scores were positively correlated with FAA(r=0.34,P<0.01).Conclusion:The college students with mild depressive symptoms may have reduced physical activity and asymmetric right lateralization of frontal alpha power.There is a correlation among depressive symptoms,physical activity and frontal alpha power a-symmetry in college students with mild depressive symptoms.
3.Preparation of self-assembling peptide-based hydrogels and its application in traumatic hemorrhage of hepatic parenchymal

Objective To prepare FLIVIGSII peptide(FI peptide)and investigate its physicochemical properties and hemostatic effect in vivo and in vitro.Methods The self-assembling peptide-based hydrogels were prepared by the FI peptide mixed with water.After gross observation for the hydrogel state of the FI peptide,scanning electron microscopy(SEM)and transmission electron microscopy(TEM)were used for its microstructure,and dynamic light scattering(DLS)was performed for its size.The hemostatic effect of FI peptide after being mixed with blood samples treated with 3.8%sodium citrate was observed,and the microstructure of the blood clot was observed with SEM.CCK-8 assay and hemolysis assay were performed to verify its biocompatibility.After a rat model of hepatic parenchymal perforation and hemorrhage was established,15 female SD rats(6~8 weeks old,weighing 150 g)were randomly divided into control group,FI peptide group and fibrin sealant group.The hemostatic effect of FI peptide and prognosis was observed and analyzed after treatment in each group,and the hemostatic mechanism was also investigated.Results FI peptides were successfully prepared,and it could rapidly self-assemble into a nanofiber network hydrogel in water,and further cause formation of blood clots.SEM showed that FI peptides self-assembled to form fibrous hydrogels after mixing with water.TEM results verified that the FI peptide formed into nanofibers in a diameter of 13.70±2.31 nm after gelatinization in water,and DLS results verified that the FI peptide formed polydisperse and multi-size nanofibers in water(in a range of 148.2~208.0 nm or 575.0~807.0 nm).The fibrous hydrogel formed by the FI peptide mixed with the blood could envelop the red blood cells,thus form a physical hemostatic barrier to achieve blood clotting in seconds.FI peptide hydrogel had no cytotoxicity to normal hepatocytes(L-O2 cells)and did not cause hemolysis of red blood cells.In in vivo experiment,FI peptide quickly formed nanofiber hydrogel when in contact with blood,thus formed physical hemostasis barrier to achieve hemostasis within a few seconds(hemostasis time<5 s).Conclusion The FI peptide exhibits a rapid and efficient hemostatic effect,indicating a promising clinical application in the hemostasia of hepatic parenchymal traumatic hemorrhage.
4.Analysis of the risk factors of persistent inflammation-immunosuppression-catabolism syndrome in patients with extensive burns

Objective:To investigate the risk factors and treatment outcome of persistent inflammation-immunosuppression-catabolism syndrome (PICS) in patients with extensive burns.Methods:A retrospective case series study was conducted. From January 2017 to December 2021, 220 patients with extensive burns who were admitted to Guangzhou Red Cross Hospital of Jinan University met the inclusion criteria, including 168 males and 52 females, aged 18-84 (43±14) years. According to the occurrence of PICS, the patients were divided into PICS group (84 patients) and non-PICS group (136 patients). The general data such as sex, age, complication of underlying diseases and acute physiology and chronic health evaluation Ⅱ (APACHE Ⅱ) score on admission, sepsis-related organ failure evaluation (SOFA) scores on admission and 14 days post admission, and proportion of patients with mechanical ventilation over 48 h during treatment, special conditions such as total burn area, full-thickness burn area, proportion of patients admitted within 48 h post injury, and exposed deep wound area at the 30 th day post injury, outcome indicators such as hospitalization day, total cost of hospital stay, number of surgeries, and death of patients in the 2 groups were collected and analyzed. Data were statistically analyzed with independent sample t test, Mann-Whitney U test, and chi-square test. The multivariate logistic regression analysis was performed on the indicators with statistically significant differences between the two groups except for outcome indicators, and the independent risk factors influencing secondary PICS in patients with extensive burns were screened. Results:The APACHE Ⅱ and SOFA scores on admission, and proportion of patients with mechanical ventilation over 48 h during treatment of patients in PICS group were significantly higher than those in non-PICS group ( t=6.78, Z=-4.75, χ2=4.74, respectively, P<0.05). There were no statistically significant differences in the rest of general data of patients between the two groups ( P>0.05). The total burn area, full-thickness burn area, and exposed deep wound area at the 30 th day post injury in PICS group were significantly greater than those in non-PICS group ( t=6.29, Z=-7.25, Z=-8.73, P<0.05), the exposed deep wound areas at the 30 th day post injury in PICS group and non-PICS group were respectively 25% (15%, 35%) total body surface area (TBSA) and 8% (0, 13%) TBSA, while the proportion of patients admitted within 48 h post injury was significantly lower than that in non-PICS group ( χ2=6.13, P<0.05). The hospitalization day, total cost of hospital stay, and number of surgeries of patients in PICS group were significantly higher than those in non-PICS group (with Z values of -7.12, -8.48, and -6.87, respectively, P<0.05), while the deaths of patients in the 2 groups were similar ( P>0.05). The APACHE Ⅱ score on admission and exposed deep wound area at the 30 th day post injury both were the independent risk factors for PICS in patients with extensive burns (with odds ratios of 1.15 and 1.07, 95% confidence intervals of 1.06-1.25 and 1.05-1.10, respectively, P<0.05). Conclusions:The APACHE Ⅱ score on admission and exposed deep wound area at the 30 th day post injury are the independent risk factors for PICS in patients with extensive burns. The patients with secondary PICS had good prognosis with more surgical intervention and hospitalization day, and higher total cost of hospital stay.
5.Improvement of a mouse model of valproic acid-induced autism.

OBJECTIVE:
To establish an improved mouse model of valproic acid (VPA)-induced autism that better mimics human autism.
METHODS:
We established mouse models of autism in female C57 mice by intraperitoneal injection of sodium valproate either at a single dose (600 mg/kg) on day 12.5 after conception (conventional group) or in two doses of 300 mg/kg each on days 10 and 12 after conception (modified group), and the control mice were injected with saline only on day 12.5. The responses of the mice to VPA injection, the uterus, mortality rate, and abortion rate were compared among the 3 groups. The morphology and development of the offspring mice were assessed, and their behavioral ontogeny was evaluated using 3- chambered social test, social test, juvenil play test, and open field test.
RESULTS:
The mortality and abortion rates were significantly lower in the modified model group than in the conventional group ( < 0.01). Compared with those in the control group, the offspring mice in both the conventional group and the modified group showed developmental disorders ( < 0.05). The mortality rate of the newborn mice was significantly lower in the modified group than in the conventional group with a rate of curvy tail of up to 100% ( < 0.001). The offspring mice in both the modified group and conventional group exhibited autism-like behavioral abnormalities, including social disorder and repetitive stereotyped behavior ( < 0.05).
CONCLUSIONS
The mouse model of autism established using the modified method better mimics human autism with reduced mortality and abortion rates of the pregnant mice and also decreased mortality rate of the newborn mice.

6.Acupuncture combined with ganglioside and mecobalamine injections can relieve the symptoms of idiopathic facial palsy

Objective To observe the clinical effect of acupuncture combined with ganglioside and mecobalamine injections on idiopathic facial palsy (IFP).Methods A total of 120 IFP patients were divided into a ganglioside group,a mecobalamine group and a consociation group,each of 40.All were given routine drug and acupuncture treatment,while the ganglioside and mecoblamine groups were additionally provided with the corresponding injections of ganglioside and mecobalamine,respectively.The treatment was administered once daily for a total of 14 days.All the groups were evaluated using the House-Brackmann facial nerve grading system before and after the interventions of 14 days ended.The compound muscle action potential (CMAP) wave amplitude and R1 incubation,the motor nerve conduction velocity (MCV) and the sensory nerve conduction velocity (SCV) of the affected facial nerves were observed and compared before and after the 14 days of treatment.Results The total effectiveness rate of the consociation group (95.0%) was significantly higher than that of the ganglioside (90.0%) or the mecobalamine group (87.5%).After the treatment the CMAP wave amplitude had improved significantly and the R1 latency had shortened significantly in the consociation group compared to the other 2 groups.The average MCV and SCV of the consociation group had also improved significantly compared with the other two groups.Conclusion Acupuncture combined with ganglioside and mecobalamine injections is an effective therapy for idiopathic facial palsy.It can improve the patient's clinical symptoms,shorten the CMAP wave amplitude and R1 latency on the affected side and improve MCV and SCV.
7.Effects of sulfur lamp, heat radiation lamp, fluorescent lampand LED lamp lighting on reproductive system in depressive male rats

Objective To investigate the effects of different lighting(sulfur lamp, heat radiation lamp, fluorescent lamp and LED lamp) on reproductive system in depressive male rats.Methods CUMS depressive model of rats was established through the stimulations of chronic unpredictable mild stress(CUMS), then they were randomly divided into 5 groups:depressive model group, sulfur lamp group, heat radiation lamp group, fluorescent lamp group and LED lamp group.After 45 d of continuous illumination, serum was collected and reproductive organs were removed from rats, then the organ index of testicle,epididymis,seminal vesicle and kidney was calculated, the concentrations of testosterone (T), estradiol (E2), prolactin (PRL) and progesterone (PROG)in the serum were detected by ELISA, the histomorphological lesion of testicle was observed under microscope with hematoxylin-eosin (HE) staining, the expression of melatonin receptors (Mel 1a), 3β-HSD and P450scc of testicle were detected by Western blot.Results The organ index of testicle, epididymis and seminal vesicle, the concentration of T, E2,PRLand PROG, and the expression of Mel 1a, 3β-HSD and P450scc in the depressive rats were significantly lower than that of the controls(P<0.05), and the seminiferous tubules of the testis of depressive rats were atrophied.However, the level of T, E2, PRL and PROG were increased,the cell morphology of the testicle was improved, and the expression of Mel 1a, 3β-HSD and and P450scc were upregulated after the sulfur lamp lighting in the depressive male rats compared with depressive model group (P<0.05).Conclusions The testis structure and functions of depressive male rats are improved by sulfur lamp lighting.
8.Disease characteristics of a group of military men in Chinese PLA Air Force between 2009 and 2013

Objective To analyze the characteristics and changes in the disease spectrum of Air Force(AF) servicemen in order to provide reference for the determinatron of the overall level of health of AF and priorities of disease prevention and control.Methods The data of outpatient and inpatient diseases between 2009 and 2013 in AF was collected and analyzed statistically according to the International Classification of Diseases-9(ICD-9),and statistical description was performed with SPSS18.0.Results Between 2009 and 2013,unit B had the highest number of outpatients and inpatients,accounting for 47.74% and 39.64% respectively.The upper respiratory tract infections,gastroenteritis/gastritis,chronic low back pain,skin diseases,nasal sinusitis,training injury,trauma,hemorrhoids,varicocele and stone diseases were the common and frequently occurring diseases in AF,which were the top ten in the disease spectrum.Among them,the incidence and admission rate of skin diseases kept declining.However,the incidence of gastroenteritis/gastritis in unit A,the incidence and admission rate of chronic low back pain in unit B and D,the incidence of injury in unit D,as well as the incidence of neurological headache and hypertension in unit C and D were higher than in other units.Conclusion Respiratory system disease,and occupation-related diseases such as low back pain,training injuries,trauma,skin disease and digestive system disease are the focus of health care in AF.Health education and health promotion for these diseases should be improved,and health intervention for key populations should be enhanced.
9.Inhibitory Effect of Asiaticoside on Bleomycin-induced Skin Cicatrization in Mice

Objective To evaluate the inhibitory effect of asiaticoside on bleomycin-induced skin cicatrization. Methods Thirty male C57BL/6 mice were randomly divided into three groups:negative control group,model control group,and asiaticoside group,ten in each group.In model control group and asiaticoside group,1 mg·mL-1bleomycin was subcutaneously injected into the dorsal skin of mice every day;4 h later,1 mL 0.9% sodium chloride solution 1 mL asiaticoside(20 mg·mL-1) was injected into the lesion skin in the model control group and the asiaticoside group,respectively.In the negative control group, the same volume of 0.9% sodium chloride solution was subcutaneously injected into the dorsal skin of the mice at the two time points every day.After 21 days,skin specimens were harvested to observe the histomorphology and detect myofibroblast proliferation and expression of inflammatory factors. Results The skin scar was significantly attenuated in the asiaticoside group as compared with the model control group,and the dermal thickness measured exhibited a gradual decrease in asiaticoside group.The expression of α-antismooth muscle antisbidy and infiltration of inflammatory cells were significantly lower in the asiaticoside group than in the model control group. Conclusion Asiaticoside inhibits the development of skin scar of mice by regulating proliferation and differentiation of myofibroblasts and down-regulating inflammatory cells.
10.Effect of Fasudil Combined with Fibrinogenase for Injection on 30 Cases of Sudden Sensorineural Hearing Loss of the Old People

Objective To observe the clinical efficacy of fasudil combined with fibrinogenase for injection on sudden sensorineural hearing loss of old people. Methods Ninety old patients with sudden sensorineural hearing loss were randomly divided into fasudil group, fibrinogenase group and combined group (n=30 in each group).After the treatment, improvement of deafness and tinnitus were compared among the three groups. Hemorrheology, coagulation function, blood routine examination, liver and kidney function profile, and blood lipid were compared before and after the treatment. Results The total effective rate of deafness was significantly higher in the combined group ( 93. 3%) than in the fasudil group ( 90. 0%) and the fibrinogenase group (86.7%) (both P<0.05).The total effective rate of tinnitus was significantly higher in the combined group (93.3%) than in the fasudil group (83.3%) and the fibrinogenase group (80.0%) (both P<0.05).After the treatment, blood viscosity and coagulation function were significantly improved in the three groups (all P<0.05).As compared with the fasudil group and the fibrinogenase group, blood viscosity was significantly improved in the combined group (both P<0.05). Conclusion Fasudil combined with the fibrinogenase for injection for sudden sensorineural hearing loss of the old people has preferable curative efficacy, and can effectively improve clinical symptom, blood viscosity and coagulation function of sudden sensorineural hearing loss.
