BACKGROUND:Traumatic spinal cord injury primarily relies on scale assessment and imaging examinations in clinical practice.However,there are limitations in predicting the prognosis of the injury.Therefore,the use of metabolomics technology for biomarker screening is significant for estimating the extent of damage,injury and recovery,as well as developing new therapies. OBJECTIVE:To characterize the metabolic features of patients with traumatic spinal cord injury using metabolomics technology and explore potential biomarkers and disrupted metabolic pathways. METHODS:Serum and urine samples were collected from 20 patients with traumatic spinal cord injury(observation group)and 10 healthy subjects(control group).Metabolites were analyzed and multivariate statistical analysis was then performed for data processing to screen differential metabolites.Metabolic pathway enrichment was performed using MetaboAnalyst software.Logistic regression was applied to construct a biomarker combination model,and its relationship with the American Spinal Injury Association grading was analyzed. RESULTS AND CONCLUSION:Significant differences in 160 and 73 metabolites were detected in the serum and urine samples of the two groups,respectively.Pathway enrichment analysis showed evident disturbances in lipid metabolism after traumatic spinal cord injury,including sphingolipid,arachidonic acid,α-linolenic acid,and arachidonic acid metabolism,as well as glycerophospholipid and inositol phosphate biosynthesis.The combination of two identified biomarkers,telmisartan and quercetin glycoside,showed a correlation with the American Spinal Injury Association grading in both serum and urine levels.Thus,metabolomics technology provides assistance in further understanding the pathological mechanisms of traumatic spinal cord injury and screening therapeutic targets.The identified metabolic biomarker combination may serve as a reference for assessing the severity of traumatic spinal cord injury.

Objective To investigate the effect of astragalus polysaccharides injection (APS) on autophagy of human nasopharyngeal cancer CNE-2 cells.Methods The inhibitory effect of APS on proliferation of CNE-2 cells was measured by CCK8 assay.Morphological changes of autophagy of APS was detected by acridine orange (AO) staining.Transmission electron microscopy was perform to observe the morphological alterations in the autophagic cells.The expression of Beclin-1 protein was detected by Western blot assay.Results APS markedly inhibited cell growth in a dose-and time-dependent manner.Increases of the number of large vacuoles and double layered membrane structure were observed by transmission electron microscopy.Results of AO staining revealed more bright red cytoplasm or nucleus in the ceradime group,which proved the existence of acidic vesicular organelles.Western blotting showed that Beclin-1 protein level was up-regulated.Conclusion APS may induce autophagy of human nasopharyngeal cancer CNE-2 cells via up-regulating the expression of apoptosis-related gene of beclin1 and inhibit the proliferation of CNE-2 cells.

Objective To evaluate the toxicity and efficacy of thermotherapy used with a streptococcus A preparation (OK-432) and cisplatin for treating malignant pleural effusion (MPE),and to assess patients' tolerance.Methods A total of 158 MPE patients were randomized into experimental and control groups.Closed drainage of the thoracic cavity was conducted with all the patients using a central venous catheter,and systematic chemotherapy was administered individually on the basis of each patient's condition.Patients in the control group were treated with intrapleural administration of OK-432 (0.5 mg) and cisplatin (40-60 mg) weekly for a maximum of 4 weeks,while those in the experimental group were given 60min of high frequency electrical thermotherapy 30-60 min after the administration of the drugs.The thermotherapy was at 40-43.5 ℃ and administered twice a week.Efficacy,quality of life and toxicity were compared between the two groups.Results The overall response rate in the experimental and control groups was 90.6% and 79.5% respectively;and grade I-II toxicity was similar.No grade Ⅲ or more serious toxicity was observed.The life quality scores in both groups were significantly improved after treatment.Conclusions Thermotherapy combined with intrapleural OK-432 and cisplatin can improve the effectiveness of therapy and life quality in MPE patients without increased toxicity.

BACKGROUND: The surgical resection rate of pdmary hepatic carcinoma is low, thus, the local treatment arose more attention. Microwave coagulation therapy can inactivate rather than kill the hepatic carcinoma ceils. Slow-release chemotherapy has been used in treating primary hepatic carcinoma because it can form local high concentrations and last for a long time. OBJECTIVE: To observe the therapeutic effect of epirubicin-loaded chitosan microspheres combined with microwave coagulation on treating hepatocellular carcinoma in mica. METHODS: Epirubicin-loaded chitosan microspheres were prepared by using emulsion-chemical cross linking technique. The surface morphology and particles size of chitosan microspheres were observed by scanning electron microscope. Ultraviolet speotrophotometer was used to analyze the entrapment efficiency, entrapment efficiency and cumulative release rates of epirubicin-loaded chitosan microspheres. Totally 24 mice with transplanted subcutaneous H22 HCC were divided into 4 groups, which were respectively treated by microwave coagulation therapy, intratumoral injected with physiological saline after microwave coagulation therapy, intratumoral injected with epirubicin after microwave coagulation therapy, intratumoral injected with epirubicin-ioaded chitosan microspheres after microwave coagulation therapy. The tumor inhibitory rate was calculated. RESULTS AND CONCLUSION: The average diameter of chitosan microsphere was 105 μm, with uniformed particle diameter. The ratio of drug loading was 11% and the entrapment was 80%. The in vitro drug cumulative release rate was 84% after 2 weeks. The tumor inhibitory rates of the microwave coagulation combined with physiological saline, epirubicin, and drug carried microspheres groups were 8%, 20%, and 47%. It suggested that chitosan microsphere is a safe and effective slow-release dosage form, which exhibits strong anti-tumor effect when combined with microwave treatment.

Objective To investigate the apoptosis-inducing effect of L-gossypol on human nasopharyngesl carcinoma cell line CNE2 and its possible mechanism.Methods The effect of L-gossypol on proliferation of CNE2 cells was estimated by MTT assay.Flow cytometry was used to analyz cell apoptosis induced by(-)-gossypol and the expression of Bcl-2 、Bax proteins.Caspase-3 activity was determined by caspase-3 colorimetric assay kit.Results Lgossypol was able to inhibit the proliferation of CNE2 cells in a dose-dependent and time-dependent fashion at concentrations more than 10 μmoL/L.L-gossypol regulated cell cycle and GO/G1 arrest could be observed in CNE2 cells.In the process of apoptosis,the expression of Bcl-2 was down-regulated and Bax was up-regulated,caspase-3 activity was in peak at 24h.Conclusion L-gossypol could induce apoptosis in nasopharyngeal carcinoma cell line CNE2 in vitro,which may be associated with down-regulation of Bcl-2,up-regulation of Bax genes expression and caspase-3 activation.

Objective:To investigate radiological features in early diagnosis for multiple primary keratocystic odontogenic tumor (MPKCOT). Methods: The radiologic data of 9 cases of MPKCOT were analyzed retrospectively, and used to give comprehensive diagnosis in 2 subsequent cases. Results: The primary lesions of MPKCOT showed some special imaging features, such as symmetry, constant position and asynchronism which appeared in the most cases. Conclusion: Basing on the features mentioned above, the minor primary lesions of MPKCOT in other regions can be diagnosed earlier.

Objective To prepare a new type of chitosans nanoparticles(PEG/CS-EPI NPs),which contains epirubicin and modified by PEG.Furthermore,to investigate the anticancer activity of the NPs in vitro and in vivo.Methods The ionic gelation technique was employed to prepare the PEG/CS-EPI NPs and CS-EPI NPs.The particle size and shape were illustrated respectively by laser scattering and transmission electron microscopy.The proliferation of nasopharyngeal carcinoma cells was detected by MTT assay;The mouse model of implantation murine sarcoma cells(S-180) was applied to evaluate the anticancer effectiveness of PEG/CS-EPI NPs and CS-EPI NPs in vivo.Results The PEG modified CS NPs were discrete and uniform spheres with average diameter of 322.1 nm.The rate of drug loading and encapsulation is 13% and 74% respectively.The results of the anticancer tests showed a sustained cytotoxicity of loading drug NPs on nasopharyngeal carcinoma cells in vitro and the stealth nanoparticles was more powerful than ordinary nanoparticles on the inhibitory potency in vivo.Conclusion Stealth chitosan nanoparticles as compared with ordinary chitosan nanoparticles seems to be a potential candidate of chemotherapy drug carriers.

Objective To observe the therapeutic efficacy of continuous arterial infusion chemotherapy combined with radiofrequency hyperthermia in the treatment of patients with advanced colorectal cancer. Methods A total of 21 advanced colorectal cancer patients with various distant metastasis were treated by the combined continuous intra arterial infusion chemotherapy and radiofrequency hyperthermia. For the chemotherapy, a dose of 200mg/m 2 surface body area of calcium folinate (CF) was given for 1 to 3 days. 80mg/m 2 of cisplation was infused intravenously for the first day and replaced by etoposide Vp 16, 60mg/m 2 for 1～3days in patients with renal dysfunction. For intra arterial infusion, a dose of 500mg/m 2 5 FU 2 was given for 72 hours. For patients with liver metastases, chemoembolization(ADM 30mg/m 2+MMC 6mg/m 2 mixed with ultra liquefied lipiodol) was carried out. Radiofrequency hyperthermia with a frequency of 41MHz was performed on the second day after chemotherapy. Results Response rates were assessed by CT scan and ultrasonography. The overall response rate of the cases was 66.67%. No serious side effect or complication was found in the course of chemotherapy. Local pain and lipid nodule were occasionally observed in some patients after hyperthermia. Conclusion Continuous intra arterial infusion chemotherapy in combination with radiofrequency hyperthermia is an useful and safe method for the treatment of patients with advanced colorectal cancer.

Objective: To investigate the role and mechanism of histamine in regulation of C57BL/6 mice spleen derived LAK activity in vivo. Methods: The C57BL/6 mice carrying B16 pulmonary metastatic melanoma were treated with LAK/IL 2/histamine therapy or LAK/IL 2 therapy with the aim of evaluating both anti tumor responses in vivo. Results: It was found that the addition of histamine effectively potentiated the anti metastatic effect produced by LAK/IL 2 therapy and induced regression of NK resistant B16 pulmonary metastatic melanoma. Survival period was significantly prolonged in mice receiving LAK/IL 2/histamine as compared with LAK/IL 2 therapy alone. The effect of histamine was completely blocked by H 2 R antagonist ranitidine, and mimicked by dimaprit, a H 2 R agonist. Conclusion:Histamine, via specific activation of H 2 R, may be an important regulator of LAK cells activity; histamine and LAK/IL 2 synergistically induce more potent antitumor efficacy in vivo .

PC84045 human lung carcinoma cells (PC84045 HLCCs) were mixed with the BM cells activated by anti-CD3 McAb and IL-2 and then CD3-ALTO4 bi-specific McAbs was put into the cell suspension. The mixed cells were cultured for 3 days in presence of IL-2 and IL-3. Tumor cell colony formation method in half-solid culture system was used to assess the purging efficiency (PE) of the activated T cells to PC84045 HLCCs. The ratio of T cells to PC84045 HLCCs was 8 to 1, the PE was 4 Logs. The rario of T cells to PC84045 HLCCs was 16 tol, the PE was 5 Logs. When the purged BM cells were injected into the NC nude mice, none of subcutaneous node was found. The number of the CFU-GM, BFU-E of the purged BM cells was more than 85 % compared with that of the fresh BM cells (P
