Amyotrophic lateral sclerosis (ALS) is the most common motor neuron disease. At present, no definite ALS biomarkers are available. In this study, exosomes from the plasma of patients with ALS and healthy controls were extracted, and differentially expressed exosomal proteins were compared. Among them, the expression of exosomal coronin-1a (CORO1A) was 5.3-fold higher than that in the controls. CORO1A increased with disease progression at a certain proportion in the plasma of patients with ALS and in the spinal cord of ALS mice. CORO1A was also overexpressed in NSC-34 motor neuron-like cells, and apoptosis, oxidative stress, and autophagic protein expression were evaluated. CORO1A overexpression resulted in increased apoptosis and oxidative stress, overactivated autophagy, and hindered the formation of autolysosomes. Moreover, CORO1A activated Ca²⁺-dependent phosphatase calcineurin, thereby blocking the fusion of autophagosomes and lysosomes. The inhibition of calcineurin activation by cyclosporin A reversed the damaged autolysosomes. In conclusion, the role of CORO1A in ALS pathogenesis was discovered, potentially affecting the disease onset and progression by blocking autophagic flux. Therefore, CORO1A might be a potential biomarker and therapeutic target for ALS.

Objective:To establish an analysis method for determining the contents of 7 components including ganoderic acid C2, ganoderic acid B,ganoderic acid G,ganoderic acid A,lucidenic acid A,ganoderma D and ganoderic acid F in ganoderma by QAMS. Methods:An HPLC method was used. The column was WatersX-bridge C18(250 mm × 4.6 mm, 5 μm), the mobile phase was acetonitrile(A)-1% acetic acid(B) (28:72) with gradient elution(0-30 min:A-28% to 38%;30-45 min:A-38% to 55%) at a flow rate of 1.0 ml·min-1,the column temperature was 30℃, and the detection wavelength was 254 nm. Ganoderic acid A was used as the internal reference, the correction factors of ganoderic acid C2, ganoderic acid B, ganoderic acid G, lucidenic acid A,ganoderma D and ganoderic acid F were calculated,and the contents of the 7 components were calculated by an external standard method to verify the feasibility and applicability of QAMS. Results:The relative correction factors of multiple assessments were reproducible,and the relative deviation of the contents of 6 components in 16 batches of Ganoderma lucidum was less than 2% when compared with that of the external standard method. Conclusion:QAMS is accurate and reliable in the quality evaluation of Ganoderma lucidum.

Biomechanical test in vitro was performed to study the biomechanical characteristics of the femoral component in the non-stemmed anatomical total hip prosthesis (A-Fix) arthroplasty. It mimics two stages, the early stage with cementless and the later stage with cement fixation that imitates bone ingrowth. The results were compared with that of normal proximal femur by normalized coefficient analysis. Translation of femoral head after hip replacement is slightly more than normal. The strain of femoral neck after replacement is lower than that of normal. It changes flatly. The security coefficient in early stage is raised by 13% on the tensile side and by 27.6% on the compressive side of femoral neck; that in later stage is raised by 29.6% and by 22.1% separately. The maximal torsional angle of femoral head is minimal, it is 0.02 times that of normal in early stage and 0.01 times in later stage. It demonstrated that A-Fix arthroplasty has the characteristics of low stress and deformation, high intensity and rigidity, and anti-loosening, thus affording mechanical evidences for its clinical use.

Objective To evaluate the effect of operative treatment of hypertensive brain hemorrhage.Methods 46 cases wer operated on ultra-early or early with perfect operating skills.It is also important to select cases properly and use a series of comprehensive treatment after surgical operation,such as controlling blood pressure,preventing alimentary canal hemorrhge,pulmonary infection,reducing blood sugar and hypothermia mild.Results 7 out of 46 patients died.The evaluation of recovery of survival function depends on ADL.Grade Ⅰ 14 cases,Grade Ⅱ 12 cases,Grade Ⅲ 7 cases,Grade Ⅳ 4 cases,Grade Ⅴ 2 cases.Conclusion The operating curative effect for hypertensive brain hemorrhage can be improved if we grasp its indication,time,methods and comprehensive treatment correctly.

Objective To evaluate the effects of the synthetic osteogenic growth peptide (sOGP) on the healing of tibial fractures in rabbits. Methods Fifty two tibial fracture models were produced in the tibiae of New Zealand white rabbits and immobilized with a 2 mm diameter Kirschner wire as intramedullary nail and grouped by random. sOGP was administered to experimental animals by intravenous injection (0.5 ?g?kg-1 ?d-1) from the first day after surgery until the day before sacrifice, while the control animals were injected with saline solution. 3-5 rabbits from each group were killed at 1, 2, 3, 4, 6 weeks after fracture. The serum was obtained to determine the level of ALP and BGP by biochemistry and RIA respectively. The bone mineral density at fracture healing site was detected by DXA, the anteroposterior and lateral radiographs were taken, the histological examination was done and quantitative analysis of the callus area and the relative amounts of bone, cartilage, and fibrous tissue in the callus of each section were calculated by computer program. The 6 weeks tibial sample were tested biomechanically by universal material test machine. Results The serum ALP, BGP level were higher in sOGP injected groups than those in the controls. All tibia osteotomies healed uneventfully on radiograph. The area of the external callus was larger in the experimental groups than that in controls at 3,4 weeks after fracture; the average area was 265.44 mm2 at 3 weeks, 233.10 mm2 at 4 weeks in the experimental groups compared with that of 209.95 mm2 at 3 weeks and 209.21 mm2 at 4 weeks in the controls. Objective To observed the alteration of the glomerular charge-barrier (GCB) in children with idiopathic nephrotic syndrome(INS) and relationship between GCB and severity of proteinuria. Mothods The clearance ratios of pancreatic to salivary isoamylase(CPAM/CSAM) were detected in 21 cases with INS and 29 healthy controls by use of a selective inbibitor, then use the clearance ration to show the CCB. Results The ratio of CPAM/CSAM in the cases with massive proteinuria was mean 1. 18 (range 0.82 - l .54), in the cases with no-massive proteinuria was mean 2.06(1 .22 - 3. 14), in healthy controls was 3 .33 (l. 18 - 6.54). There was a significant difference in ratios CPAM/CSAM between three groups. As all cases with INS, the ratio was very significantly negatively correlated with 24-hour urine protein quantity, but not correlated when 24 hour proteinuria was in excess of 50 mg/kg. Conclusion GCB of the cases with INS is reduced, and of the cases with massive proteinuria almost dissappeared.

Aqueous extract of Dracocephalum tanguticum Maxim. (DtM ),when given ig or ip to experimental hypoxia mice,markedly increased their tolerance to normobaric and hypobaric hypoxia. Erythrocyte superoxide dismutasc (SOD ) activity of mice or rats determined at high altitude (animaquing mountain 4700 m above sea level ) showed an elevated dismutation activity of peroxide anions. At an experimentally simulatcd altitude of 6000 m above sea level,determunation of MDA, Hb and Hct showed an obvious reduction of all the abnormally elevated criteria. It is suggested that the antihypoxia activity of DTM may be partly due to its free radical scavenging activity
