1.Effect of RORγ gene on proliferation and migration of colon cancer cells

Objective:
To investigate the effects of retinoid-related orphan receptor γ (RORγ) gene on proliferation and metastasis of human colon cancer cells.
Methods:
RORγ knockdown cell lines were constructed and the knockdown efficiency was detected by RT-qPCR and Western blot assays ; MTT,colony formation,Transwell and wound healing assays were used to detect cell proliferation and metastasis ; the expression of epithelial-mesenchymal transition (EMT) related proteins was detected by Western blot.The relationship between RORγ gene expression and immune cell infiltration in tumor microenvironment was analyzed using TIMER 2. 0 database.
Results :
The knockdown of RORγ enhanced the viability (F = 157. 40,P<0. 01) ,clonogenesis (F = 61. 35,P<0. 01) ,migration (F = 13. 00,P<0. 01) ,invasion (F = 21. 26,P<0. 01) and wound healing ability (F = 877. 2,P<0. 01) of colon cancer cells,inhibited the expression of E-Cadherin,and promoted the expression of vimentin and N-Cadherin.TIMER 2. 0 database analysis showed that RORγ expression in colon adenocarcinoma ( COAD) tissues was associated with multiple immune cell infiltrates.
Conclusion
Downregulation of RORγ expression promoted the proliferation and metastasis of colon cancer cells.

Objective To evaluate the development, hot spots and trends of the fields of neurogenic bladder.Methods The relevant articles of neurogenic bladder from January, 2000 to June, 2021 in CNKI and Web of Science were retrieved.The countries, authors, institutions, cited reference and keywords were extracted with CiteSpace to draw knowledge mapping. Results and Conclusion A total of 5 064 articles were enrolled. At present, the research on the field of neurogenic bladder is in a stable period of development, and this field has been widely concerned by scholars at home and abroad. The cooperation between domestic authors and institutions is not close enough compared with foreign countries, and domestic cooperation is more between medical schools and their respective affiliated hospitals. In the future, China can further strengthen cross-regional and cross-agency cooperation. Low-frequency electrical stimulation and sacral nerve regulation are seem to be research hotspots, and children39;s neurogenic bladder and robot-assisted technologies are also needed more attention.
3.E3 ubiquitin ligase alleviates DSS-induced colitis via regulating NLRP3 inflammasome activation

Objective To investigate the role of TRIM65 on DSS induced colitis and the underlying molecular mechanisms. Methods Trim65+/+ and Trim65-/- mice were administered with 3% (w/v) DSS in their drinking water for 5 consecutive days and then were switched to sterile water for 2 days. DSS treated mice were monitored daily for the clinical symptoms (bodyweight, stool consistency and rectal bleeding score). Mice were sacrificed on day 7 to measure colon length. Colon homogenates were collected to measure MPO activity and detect cleaved caspase-1 and mature IL-1β by Enzyme linked immunosorbent assay (ELISA) and Western blot. Trim65-/- mice were intraperitoneally injected with NLRP3 inflammasome inhibitor MCC950, and were given the above treatment to determine the effect of MCC950 on colitis in Trim65-/- mice. Results The results showed that deletion of Trim65 significantly enhanced weight loss and colon shortening in DSS mice, increased disease activity index and histopathological score, induced the activity of MPO, and promoted the F4/80+ immune cell infiltration, the activation of caspase-1 and the secretion of mature IL-1 in the colon of DSS mice. The NLRP3 inflammasome inhibitor MCC950 alleviated DSS induced colitis symptoms and inflammation levels in trim65 deficient mice. Conclusion TRIM65 plays an anti-inflammatory role in DSS induced colitis mice by inhibiting the activation of NLRP3 inflammasome.
4.Serological epidemiological characteristics of EV71 and pathogen spectrum of hand , foot and mouth disease in Deyang

Objective To explore the pathogen spectrum of hand, foot, and mouth disease (HFMD) in Deyang, and the serological epidemiological characteristics of enterovirus 71 (EV71). Methods The general clinical data and etiological data of 287 patients with HFMD in Deyang from June 2019 to June 2021 were statistically analyzed. Blood samples were collected to detect serum EV71 neutralizing antibodies by neutralization test. Results Among the 287 patients with HFMD, most of the critically ill patients were between 1 and 6 years old. There was a statistically significant difference in clinical classification between different age groups (P < 0.05). A total of 254 (88.50%) EV-positive cases were detected, including 131 (45.64%) positive for EV71, 98 (34.15%) positive for CVA16, and 25 (8.71%) positive for other EVs. The EV71 positive rate was higher in critically ill patients (P < 0.05). The positive rate of EV71 antibody increased with age within 15 years old, with statistically significant differences among different age groups (P < 0.05). Among the 287 serum samples, there were 156 (54.36%) samples with neutralizing antibody titer <1:8, 86 (29.97%) with titer between 1:8 and 1:32, and 45 (15.68%) with titer >1:32. Patients with EV71 neutralizing antibody titer>1:32 were over 15 years old. There were statistically significant differences in EV71 neutralizing antibody titer between different age groups (P<0.05). Conclusion The positive rate of EV is different in patients with different clinical types. EV71 and CVA16 are main pathogens of HFMD in Deyang. HFMD patients under 6 years old have lower EV71 antibody level. It is recommended to strengthen EV71 vaccination for children under 6 years old.
5.Preliminary exploration of the domestic balloon-expandable valve in the treatment of degenerated tricuspid bioprosthetic valve via transcatheter "valve-in-valve" technology

Objective To discuss the operation skill and clinical effects of using domestic balloon-expandable Prizvalve® transcatheter "valve-in-valve" to treat the degenerated bioprosthesis in the tricuspid position. Methods All the admitted surgical tricuspid valve bioprosthetic valve replacement patients were evaluated by computerized tomography angiography (CTA), ultrasound, and 3D printing technology, and 2 patients with a degenerated bioprosthesis were selected for tricuspid valve "valve-in-valve" operation. Under general anesthesia, the retro-preset Prizvalve® system was implanted into degenerated tricuspid bioprosthesis via the femoral vein approach under the guidance of transesophageal echocardiographic and fluoroscopic guidance. Results Transcatheter tricuspid valve implantation was successfully performed in both high-risk patients, and tricuspid regurgitation disappeared immediately. The operation time was 1.25 h and 2.43 h, respectively. There was no serious complication in both patients, and they were discharged from the hospital 7 days after the operation. Conclusion The clinical effect of the degenerated tricuspid bioprosthetic valve implantation with domestic balloon-expandable valve via femoral vein approach "valve-in-valve" is good. Multimodality imaging and 3D printing technology can safely and effectively guide the implementation of this innovative technique.
6.Effect of orthokeratology combined with 0.01% Atropine Sulfate Ophthalmic Gel on curative effect and analysis influencing factors on children39;s myopia

AIM: To investigate the influence of orthokeratology combined with 0.01% Atropine Sulfate Eye Gel on the recovery of myopia in children and the factors influencing the curative effect of children.
METHODS: Totally 200 children with myopia admitted to our hospital in 2017-03/2019-03 were selected. According to the requirements of children and guardians, the observation group(orthokeratology combined with 0.01% Atropine Sulfate Eye Gel)and the control group(only wearing orthokeratology)were selected. Totally 108 cases in the observation group were included in the study, 8 cases were abandoned after 6mo, and 100 cases were finally included to complete the follow-up. 92 cases in the control group were included in the study, 5 cases abandoned after 6mo, and 87 cases completed the follow-up. The patients39; best corrected far vision, best corrected near vision, naked eye vision, intraocular pressure, pupil diameter and adjustment amplitude and discomfort symptoms after treatment were collected before and after 6mo.
RESULTS: The uncorrected visual acuity of the observation group after treatment was 0.12±0.05, and that of the control group was 0.19±0.07, which were significantly higher than those before treatment(P<0.05), and the observation group was significantly higher than that of the control group(P<0.05). After treatment, the intraocular pressure of the two groups had no significant change(P>0.05); the pupil diameter of the observation group after treatment was(7.01±0.66)mm, which was significantly higher than(6.09±0.69)mm before treatment(P<0.05); the adjustment amplitude was(14.06±4.03)D, which was significantly lower than(15.31±4.40)D before treatment(P<0.05); there were no significant changes in the pupil diameter and adjustment amplitude of the control group after treatment(P>0.05). Pearson correlation analysis showed that good myopia control was negatively correlated with age, uncorrected visual acuity and astigmatism before treatment(P<0.05). Multivariate Logistic regression analysis showed that myopia control was negatively correlated with age and uncorrected visual acuity before treatment(P<0.05).
CONCLUSION: Orthokeratology combined with 0.01% Atropine Sulfate Eye Gel is helpful for the control of myopia in children. The effect of treatment is influenced by the age of children and the visual acuity before treatment.
7.Analysis of mineral contents in the colostrum and peripheral blood and their correlation in pregnant women in Beijing

Objective To investigate the mineral contents of colostrum and peripheral blood and their correlation in pregnant women in Beijing. Methods A total of 80 pregnant women who gave birth in the Affiliated Fuxing Hospital of Beijing Capital Medical University in May 2019 were selected as research subjects, and their colostrum and peripheral blood samples were collected. The contents of 46 minerals in the colostrum and peripheral blood samples were determined by Inductively Coupled Plasma Mass Spectrometry (ICP-MS) and Atomic fluorescence spectrum (AFS). The correlation between the contents of minerals in the colostrum and peripheral blood was analyzed by SPSS 21.0. Results Among the 46 minerals detected, the minerals with higher contents in the peripheral blood were sodium, potassium, iron, magnesium, calcium, zinc, rubidium, copper, aluminum and selenium. The minerals with higher colostrum contents included potassium, sodium, calcium, magnesium, zinc, iron, rubidium, copper, strontium and aluminum. The contents of calcium, strontium, cesium, molybdenum, cobalt, uranium and thorium in the colostrum were significantly higher than those in the peripheral blood (P<0.05). There were significant correlations in the contents of sodium, rubidium, selenium, strontium, cesium, arsenic, mercury and cadmium between the colostrum and peripheral blood (P<0.05). In addition, there were significant correlations between some minerals in the colostrum, for example potassium vs manganese (r=-0.236, P=0.043), rubidium (r=0.816, P<0.001) and magnesium (r=0.400, P<0.001); sodium vs selenium (r =0.509, P<0.001), cerium (r=0.353, P=0.002), praseodymium (r =0.337, P=0.003) and thulium (r=0.331, P=0.004); calcium vs iron (r=0.251, P=0.031) and strontium (r =0.365, P=0.001); magnesium vs cesium (r=0.314, P=0.006) and copper (r =0.395, P=0.001); zinc vs selenium (r=0.310, P=0.007), cerium (r=-0.229, P=0.050), gadolinium (r=-0.372, P=0.001) and yttrium (r=-0.380, P=0.001); rubidium vs titanium (r=-0.413, P<0.001); copper vs cesium (r=0.275, P=0.018); strontium vs uranium (r=0.439, P<0.001); cadmium vs molybdenum (r=0.379, P =0.001). Conclusion The correlations of the mineral contents between the colostrum and peripheral blood suggested that some mineral elements could be accumulated from blood to breast milk through various transport mechanisms. The correlations between the mineral elements in the colostrum showed that there were some interactions between these elements, indicating that maintaining the dynamic balance of mineral elements in human milk is very important for the health of their offspring.
8.Hypertension and diabetes synergistically strengthen the association with cataracts

AIM: To examine the relationship between age-related cataracts(ARC)and comorbid hypertension and diabetes.
METHODS: We analyzed the administrative records of 6 467 patients aged 50 years and older admitted to the ophthalmological department of a tertiary hospital from January 1st, 2011 to May 20th, 2017. With either eye considered, an ARC(n=3 343)was defined as the presence of lens opacity or previous cataract surgery without evidence of trauma, congenital anomalies or using certain medications. Patients admitted to the same department during the same period due to ocular traumas without clinical evidence of cataracts(n=379)were recruited as the cataract-free controls. Unconditional Logistic regressions were ran to obtain the odds ratio(OR)of hypertension and diabetes among ARC patients adjusted for age, sex and health care accessibility.
RESULTS: Hypertension was diagnosed in 29.54% of men with any type of cataracts, in 30.12% of men with an ARC, and 10.82% of men of cataract-free controls. Diabetes was diagnosed in 16.64% of men with any type of cataracts, in 16.48% of men with ARC and 4.22% of men of cataract-free controls. Similar patterns were observed among women. After adjusting for age, sex, and health care accessibility, hypertension was weakly \〖OR=1.83(95% CI: 1.23, 2.74)\〗 and diabetes was strongly \〖3.38(1.86, 6.15)\〗 associated with ARCs. The adjusted OR of comorbid hypertension and diabetes among adults with ARC was 18.20(4.38, 75.59).
CONCLUSION: Hypertension and diabetes were independently associated with ARC. Hypertension and diabetes, if co-existing, multiplicatively strengthened the association with ARC.
9.EYA1 inhibits malignant biological behavior of gastric cancer SGC-7901 cells by regulating PTEN/PI3K/AKT signaling pathway

Objective:To explore the mechanism of EYA1 (eyes absent 1) inhibiting the malignant progression of gastric cancer SGC7901 cells through regulating PTEN/PI3K/AKT signaling pathway. Methods: Twenty-nine pairs of gastric cancer tissues and para-cancerous tissues collected at the General Surgery center, Southwest Hospital Affiliated to Military Medical University during June 2016 and June 2018 were used in this study. Wb and RT-PCR assays were used to test the mRNA and protein expressions of EYA1 in gastric cancer tissues and the paired para-cancerous tissues; Transfection with plasmid or siRNAs were used to up-regulate or down-regulate EYA1 or PTEN expression in gastric cancer SGC-7901 cells; MTT, Flow Cytometry, Wound Healing and Transwell assays were carried out to detect cell proliferation, apoptosis, metastasis and invasion abilities, respectively. Results: EYA1 expression was decreased in gastric cancer tissues as compared with the para-cancerous tissues at both mRNA and protein levels (P<0.01); EYA1 over-expression significantly enhanced the proliferation, metastasis and invasion of SGC-7901 cells (all P<0.05), and inhibited cell apoptosis (P<0.05); moreover, its over-expressionsignificantly increased the expression of PTEN, and inhibited the activation of PI3K/AKT pathway (all P< 0.05 or P<0.01). However, the above effects mediated by EYA1 up-regulation were significantly impaired after the knockout of PTEN (all P<0.05 or P<0.01). Conclusion: EYA1 can inhibit the malignant progression of gastric cancer SGC-7901 cells through promoting the expression of PTEN and activating PI3K/AKT pathway.
10.Clinical research of intravitreal injection of Conbercept combined with different methods in the treatment of neovascular glaucoma

AIM:To observe the clinical effects of intravitreal injection of conbercept combined with complex trabeculectomy and panretinal photocoagulation(PRP)in the treatment of neovascular glaucoma.
METHODS: Eleven patients(11 eyes)with neovascular glaucoma were injected with conbercept 0.50mg/0.05mL in vitreous chamber. Complex trabeculectomy was performed after the neovascularization dissipated, and PRP was performed in all cases at the right time. The follow-up time was 6mo. The vision, intraocular pressure(IOP)and complications were observed after complex trabeculectomy.
RESULTS: The IOP were all decreased at 1wk, 1, 3 and 6mo after operation, and the difference was statistically significant(P<0.01). After 6mo, the best-corrected visual acuity of 2 eyes(18%)was improved, 8 eyes(73%)were stable, and 1 eye(9%)was declined. The postoperative complications included hyphema, shallow anterior chamber, and vitreous hemorrhage.
CONCLUSION: Intravitreous injection of conbercept combined with complex trabeculectomy and PRP can control the IOP and preserve the visual function effectively in patients with neovascular glaucoma.
