1.Research Progress of Radiation-induced Brain Injury for Nasopharyngeal Carcinoma

Radiation-induced brain injury (RBI) is a common long-term complication of radiotherapy for nasopharyngeal carcinoma (NPC) and seriously affects the quality of life and overall survival of patients. In the era of intensity-modulated radiation therapy (IMRT), the long-term complications after radiotherapy, especially RBI, are becoming increasingly concerning because a number of treated patients with NPC obtain long-term survival. At present, the understanding of RBI is still being explored, and its pathogenesis and treatment methods are continuously updated. This article reviews the research progress of RBI in patients with NPC.
2.Effects of auranofin on the activity of ovarian cancer cells and its molecular mechanisms

Objective:
To explore the activity of auranofin against ovarian cancer cells and its possible molecular mechanism.
Methods :
The dose-response survival curve and IC50 of auranofin on ovarian cancer cell lines ,SKOV3,Caov3 and SW626 cells and immortalized normal human embryonic kidney HEK-293T cells were determined by CCK-8 method.Cell cycle was determined by flow cytometry.The levels of total glutathione ( GSH) ,reduced GSH and glutathione disulfide ( GSSG) ,thioredoxin reductase (TrxR) and reactive oxygen species (ROS) in cells were determined by microplate reader,and the reduced GSH / GSSG ratio was calculated.Western blot was used to determine the expression of cyclin dependent kinases( CDK) 4,CDK6,Cyclin D1,P53,p-P53 and MDM2 in SKOV3 and Caov3 ovarian cancer cells.
Results :
Compared with HEK-293T cells,the dose-response survival curves and IC50 values of SKOV3,Caov3 and SW626 cells showed that ovarian cancer cells were more sensitive to auranofin (P<0. 05) .After SKOV3 and Caov3 cells were treated with the dose of respective IC50 concentrations of auranofin,compared with the untreated cells group,the Auranofin IC50 group cells' intracellular levels of GSH,the ratio of reduced GSH / GSSG and the activity of TrxR decreased (t = 25. 11 /31. 18,14. 72 /19. 92,43. 30 /10. 74, all P<0. 05) ,and the levels of ROS increased (t = 23. 82 /27. 71,P<0. 05) ; cells number at G0 / G1 phases increased,with cells number at S and G2 phases decreased (P<0. 05) ; and the expression levels of cell cycle-related proteins CDK4,CDK6,Cyclin D1 and the P53-specific E3 ubiquitin ligase MDM2 were down-regulated (t = 7. 51 /15. 59,17. 32 /11. 26,20. 78 /20. 78,24. 25 /17. 32,all P<0. 05) ,while the expression levels of P53 and p-P53 were up-regulated (t = 17. 32 /24. 25,12. 12 /10. 39,all P <0. 05) .
Conclusion
Auranofin causes oxidative stress in ovarian cancer cells by inhibiting TrxR activity,and by partially degrading MDM2 to stabilize and acti- vate P53,so as to block the cancer cells in G0 / G1 phase,and exert anti-ovarian cancer activities.
3.Intravenous route to choroidal neovascularization by macrophage-disguised nanocarriers for mTOR modulation.

Retinal pigment epithelial (RPE) is primarily impaired in age-related macular degeneration (AMD), leading to progressive loss of photoreceptors and sometimes choroidal neovascularization (CNV). mTOR has been proposed as a promising therapeutic target, while the usage of its specific inhibitor, rapamycin, was greatly limited. To mediate the mTOR pathway in the retina by a noninvasive approach, we developed novel biomimetic nanocomplexes where rapamycin-loaded nanoparticles were coated with cell membrane derived from macrophages (termed as MRaNPs). Taking advantage of the macrophage-inherited property, intravenous injection of MRaNPs exhibited significantly enhanced accumulation in the CNV lesions, thereby increasing the local concentration of rapamycin. Consequently, MRaNPs effectively downregulated the mTOR pathway and attenuate angiogenesis in the eye. Particularly, MRaNPs also efficiently activated autophagy in the RPE, which was acknowledged to rescue RPE in response to deleterious stimuli. Overall, we design and prepare macrophage-disguised rapamycin nanocarriers and demonstrate the therapeutic advantages of employing biomimetic cell membrane materials for treatment of AMD.
4.Feasibility of a single catheter intervention using the transradial approach for ST-segment elevation myocardial infarction

Objective:To evaluate the safety and effectiveness of primary percutaneous coronary intervention using the transradial approach in patients with acute ST-segment elevation myocardial infarction (STEMI) based on electrocardiography results.Methods:The clinical data of 298 patients with STEMI who had indications of emergency coronary angiography and percutaneous coronary intervention who received treatment in The Second People's Hospital of Shantou between January 2015 and June 2019 were retrospectively included in this study. These patients were assigned into traditional transfemoral intervention (TTFI, n = 56), traditional transradial intervention (TTRI, n = 167), and single transradial intervention (STRI, n = 75) groups. Door-to-balloon (D2B) time, needle-to-balloon (N2B) time, hospital days, and the incidence of major adverse cardiac events within 1 year after treatment were compared among the three groups. Results:The D2B time in the STRI, TTFI and TTRI groups was (67.6 ± 2.1) minutes, (73.3 ± 15.3) minutes, and (77.4 ± 16.7) minutes, respectively. There was a significant difference in D2B time among the three groups ( F = -2.24, P = 0.013). The D2B time was significantly shorter in the STRI group than in the TTFI and TTRI groups ( t = -1.84, -1.84, both P = 0.033). The N2B time in the STRI, TTFI and TTRI groups was (7.6 ± 2.1) minutes, (15.3 ± 6.5) minutes, and (14.1 ± 5.7) minutes, respectively. There was a significant difference in N2B time among the three groups ( F = -4.34, all P < 0.001). The N2B time was significantly shorter in the STRI group than in the TTFI and TTRI groups ( t = -2.06, P = 0.020; t = -3.12, P < 0.001). The proportion of patients with D2B time less than 90 minutes in the STRI group was 74.7% (56/75), which was significantly higher than that in the TTRI [46.1% (77/167)] and TTFI [51.8% (29/56)] groups ( χ2 = 4.07, P < 0.001). The incidence of major adverse cardiac events within 1 year after treatment in the TTFI, TTRI, and STRI groups was 16.1% (9/56), 13.2% (22/167), and 9.3% (7/75), respectively. The incidence of major adverse cardiac events within 1 year after treatment was significantly lower in the STRI group than in the TTFI and TTRI groups ( χ2 = 5.67, P < 0.05). Conclusion:STRI is safe and effective for STEMI and is expected to improve long-term prognosis.
5.Synchronous double primary malignant tumours of the tongue and lung: a case report and literature review

Objective:
To investigate the diagnosis and treatment of synchronous double primary malignant tumours of the tongue and lung.
Methods:
A case of adenoid cystic carcinoma(ACC) and lung adenocarcinoma with double primary malignancy was retrospectively analyzed.
Results:
The tumor of patient′s tongue base gradually grew. MRI showed multiple enlarged lymph nodes on both sides of neck. CT of the chest showed obvious lesions in the anterior basal segment of the right lower lobe. The pathological biopsy of the tongue mass identified ACC, and pathological biopsy of the lung mass identified lung adenocarcinoma. The tongue and lung tumors were both surgically resected, and the tongue defect was repaired at the same time. No residue was found after surgery, and no recurrence was found during the follow-up period. The aesthetic and functional restoration of the lingual region was good.
Conclusion
There are few cases of adenoid cystic carcinoma and lung adenocarcinoma with double primary malignancies, and the related diagnosis and treatment are very difficult; the simultaneous removal of double primary malignant tumors may achieve good prognosis.
6.Effects of sleeve gastrectomy in mice by using titanium clip

Objective To introduce details of a new procedure of sleeve gastrectomy in mice by using titanium clip,and to observe whether this model has the same post-surgical metabolic effects as traditional procedure of sleeve gastrectomy in mice by using running stitch.Methods Thirt-one male FVB-Tg (NPY-hrGFP) mice,6 weeks (29-31 g) were randomly divided into SG (running stitch) group (n =8),SG (titanium clip) group (n =15) and Sham Operation (SO) group (n =8) by using random number table.Titanium clip was applied to close the side incision of stomachin SG (titanium clip) group,while running stitch was used in SG (running stitch) group which was detailed described in current references.Ligaments around stomach were detached without any stomach incision in SO group.Operation duration,mortality,and changes of post-surgical body weight and food intake were observed.Measured data were expressed as mean ± standard deviation ((x) ± s).Independent sample t test was used for comparison between the two groups.The variance of the data between the three groups was equal and analysis of variance (least significant LSD method) was used to compare each group.Preoperative and postoperative data were compared using a paired t test.Results Operation duration of SG (running stitch) group and SG (titanium clip) group were (44.60 ± 3.13) min and (28.25 ± 2.77) min,respectively.Compared with SG (running stitch) group,operation duration was significantly reduced in SG (titanium clip) group (P ≤ 0.01).Mortality of SG (running stitch) group and SG (titanium clip) group were 37.5% and 20%,respectively,which has no significant difference (P =0.621).Compared with SO group,postoperative food intake was significantly reduced in both two SG groups (P ≤ 0.01),but there's no difference between two SG groups (P =0.149).At the end of the 6-week-postoperative,body weights of SG (running stitch) group and SG (titanium clip) group were reduced (17.67 ± 5.06) % and (18.76 ± 7.87) %,respectively,while body weight of SO group increased (7.17 ± 2.31)%.Compared with SO group,postoperative change of body weight was significantly reduced in both two SG groups (P≤ 0.0 1),but there's no difference between two SG groups (P =0.746).Conclusions SG by using titanium clip in mice can reduce food intake and body weight as traditional SG by using running stitch does.But with shorter operation duration,this new model of SG in mice can provide a better and stable surgical model in research of mechanism of SG.
7.A 22-year-follow-up cohort study on primary liver cancer in Haimen city of Jiangsu province

Objective A prospective cohort study was carried out to assess the mortality and potential risk factors for primary liver cancer (PLC) in Haimen city of Jiangsu province.Methods The cohort involved 89 789 adult residents aged 25-69 years.Upon the entry of this project,each subject was asked to complete a questionnaire and to provide a blood sample of 10 ml.Surface antigen of hepatitis B virus (HBsAg) was tested by radioimmunoassay.All the subjects were followed-up every year for vital statistics and death certificate information until 2014.Cox proportional hazard model was used to estimate the hazard ratio (HR) and 95% confidence interval (95% CI) for PLC mortality associated with HBsAg status and other risk factors.Results During the 1 299 611 person-years of follow-up,a total of 2 583 PLC cases were identified,including 2 149 men and 434 women.Mortality of the PLC for men and women were 247.80/100 000 person-years and 100.38/100000 person-years,respectively.Among those who died of PLC,73.87% had been tested HBsAg positive.HBV infection seemed the predominant risk factor for PLC and the HRs were 15.97 for men (95%CI:14.29-17.85) and 21.63 for women (95%CI:16.16-28.96) respectively.Ageing,cigarette smoking,previous history of hepatitis,and family history of HCC were factors associated with the increased risk for PLC.Conclusion People living in Haimen city had a high risk on PLC.HBV infection appeared the most important risk factor for HCC mortality in this area.

METHODSA clinical isolate of EV71 was inoculated to Vero cells and the EV71 antigen was detected by immunofluorescence assay. The morphological changes of Vero cells were observed using optical microscopy and transmission electron microscopy. The diameter and area density of the viral particles and the ratio and area density of vacuolated mitochondria in the cells were measured on the ultrastructural images.
RESULTSEV71-infected Vero cells underwent obvious changes and to a spherical morphology followed by cell death EV71 particles were detected in the cytoplasm by immunofluorescence. Ultrastructurally, the infected cells contained a large number of viral particles in the cytoplasm, with a clustered distribution and lattice-like arrangement. The diameter of the particles were 16.3 nm and the mean area density was 38.3%. Most of the mitochondria presented with swelling, vacuoles and degeneration. The ratio of the vacuolated mitochondria was 90.9% with a mean area density of 89.2%. Viral particles were also found in some mitochondria.
CONCLUSIONEV71 proliferates in the cytoplasm and invades the mitochondria of infected Vero cells leading to mitochondrial injury and cell death, suggesting that mitochondria are the targets for EV71 infection.
