1.Clinicopathological features of radioactive iodine-refractory differentiated thyroid cancer

Objective To investigate the clinicopathological characteristics of radioactive Iodine-refrac-tory differentiated thyroid cancer(RAIR-DTC)to provide a clinical evidence for early prediction of the thyroid cancer patients with radioactive Iodine-refractory(RAIR).Methods The data of 84 patients with undergoing thyroidectomy and 131I therapy in PLA 960 hospital from January 2010 to December 2019 were retrospectively analyzed.Thirty-nine patients with diagnosed RAIR-DTC served as the study group and 45 cases of radioactive iodine-avid differentiated thyroid cancer(RAIA-DTC)served as the control group.The clinicopathological characteristics were compared between the two groups.The logistic regression was used to analyze the inde-pendent risk factors of RAIR-DTC,and the RAIR-DTC prediction model was established.Results Compared with the RAIA-DTC group,the RAIR-DTC group had more iodine treatment times,the proportions of the pa-tients with age ≥55 years old,total iodine therapeutic dose,distant metastasis,TNM stage Ⅳ,high-risk sub-types and focal calcification were higher,the tumor maximum diameter was greater,the number of lymph node metastases was more and the probability of Ⅱ,Ⅰ+Ⅱ and non-central lymph node metastases was higher(P>0.05).The progression-free survival rate had statistical difference between the two groups(P<0.05).The total survival rate had no statistical difference between the two groups(P>0.05).The binary logistic re-gression analysis results showed that the distant metastasis,high-risk histological subtype and maximum tumor diameter ≥10.5 mm were the independent risk factors for RAIR-DTC.The obtained fitting equation logit(P)=-2.259+3.330X1+2.287X2+1.606X3,the ROC curve was used to calculate the truncation val-ue of the fitted equation as-0.312 5,when logit(P)>-0.312 5,it might develop into RAIR-DTC.Conclusion The clinicopathological characteristics of the patients with differentiated thyroid cancer could ef-fectively predict RAIR.
2.Construction of a prognostic model and drug prediction for clear cell renal cell carcinoma based on endoplasmic reticulum stress-related lncRNA

【Objective】 To construct a prognostic model of clear cell renal cell carcinoma (ccRCC) based on endoplasmic reticulum stress (ERS)-related long non-coding ribonucleotides (lncRNA),so as to explore the correlation between immune cell infiltration and prognosis of ccRCC patients,and to search for new drugs for the treatment of ccRCC. 【Methods】 The transcriptome and clinical data of cancerous and paracancerous tissues of ccRCC were obtained from the TCGA database.The ERS-associated gene set was obtained from the MSigDB database.ERS co-expressed lncRNAs were screened with Pearson correlation analysis.ERS-related lncRNA (ERSRL) with prognostic significance were screened with Lasso regression,univariate and multivariate Cox regression analyses,and a prognostic model was constructed.The risk value of each sample was calculated according to the prognostic model formula.The patients were divided into high- risk and low- risk groups for survival difference analysis.The predictive performance of the prognostic model was evaluated with survival curve,receiver operating characteristic (ROC) curve and calibration curve.The infiltration of immune cells in high-and low-risk groups was analyzed with CIBERSORT database.The relationship between ERSRL and drug sensitivity was analyzed with GDSC database to identify drugs with potential efficacy against ccRCC. 【Results】 A total of 9 lncRNAs with independent prognostic significance were screened to construct the prognostic model.Kaplan-Meier analysis showed significant survival differences between the high- and low-risk groups.Univariate and multivariate Cox regression analyses showed that age,grade,stage and risk score could be used as independent prognostic factors.The area under the ROC curve (AUC) of the 1-,3-,and 5-year survival rates of the training set were 0.754 (95%CI:0.659-0.848),0.744 (95%CI:0.667-0.815),and 0.759 (95%CI:0.662-0.820),respectively,and the C-index was 0.777 (95%CI:0.759-0.796).Immune infiltration results showed that plasma cells,activated memory CD4+T cells,regulatory T cells,M0 macrophages,and activated mast cell infiltration levels were higher in the high-risk group than those in the low-risk group.Drug susceptibility analysis identified 12 drugs with potential curative effects on ccRCC,including AZD8055. 【Conclusion】 Based on 9 ERSRLs,a prognostic model for ccRCC patients was constructed,and 12 drugs with potential therapeutic effects were screened,including AZD8055.
3.Maggot alleviates imiquimod-induced psoriasis-like skin lesions in mice by inhibiting immune stress and complement activation

Objective To explore the therapeutic mechanism of maggot for psoriasis-like lesions in mice from the perspective of immune stress and complement activation regulation.Methods Thirty-six male C57BL/6 mice were randomly divided into control group,model group,maggot(1.25%,2.5%,and 5%)groups,and Benvitimod(1%)group.Psoriasis-like lesions were induced by application of imiquimod cream,and the severity of skin lesions was assessed using the modified Psoriasis Area and Severity Index(MPASI)score.Auricular swelling of the mice was observed,and histopathological changes of the skin lesions were examined with HE staining.Scratching behavior of the mice was observed and the spleen index was calculated.Toluidine blue staining was used to detect mast cells in the skin lesions,and serum levels of IgG,IgM,the complements CH50,C1s,C3,C3a,C5 and C5a,and the inflammatory factors IL-23,IL-17A and TNF-α were determined with ELISA.Results In mice with imiquimod-induced psoriasis-like skin lesions,treatment with the maggot at the 3 doses significantly decreased MPASI score,alleviated auricular swelling and pathologies in the skin lesions,reduced scratching behaviors,spleen index,and the number of mast cells in the lesions.Treatment with high-dose maggot significantly lowered serum levels of IgG,C1s,C3a,C5a,IL-23,IL-17A and TNF-α and the levels of C1s,C3,C3a,C5 and C5a in the lesion tissue,and increased serum levels of CH50,C3,and C5.The therapeutic effect of maggot showed a dose-effect dependence.Conclusion Maggot can alleviate psoriasis-like skin lesions in mice by inhibiting immune stress and complement activation.
4.Effect of intravenous thrombolytic therapy mode on patients with acute ischemic stroke led by fast-track specialist nurses

Objective To investigate the effectiveness of intravenous thrombolytic therapy mode led by fast-track specialist nurses on patients with acute ischemic stroke(AIS).Methods This study involved 124 AIS patients who underwent intravenous thrombolytic therapy in the Department of Emergency of our hospital from March 2021 to February 2023.Among the patients,61 admitted between March 2021 and February 2022 received conventional AIS thrombolytic therapy were assigned to a control group.While the 63 patients who received AIS thrombolytic therapy under the specialist nurse-led intravenous thrombolytic therapy mode between March 2022 and February 2023 were assigned to an observation group.The two groups were compared in terms of the time from admission to completion of CT examination,time for signing the informed consent for thrombolytic therapy,door to needle time and percentage of DTN<60 minutes,as well as the post-thrombolysis scores according to the National Institute of Health Stroke Scale(NIHSS)and satisfaction to medical consultation.Results The observation group exhibited a significantly shorter time from admission to completion of CT examination,a shorter time for signing an informed consent for thrombolytic therapy,a shorter door to needle time and a higher percentage of DTN<60 minutes,all with significant difference in comparison with those in the control group.After thrombolysis,the NIHSS score of the observation group decreased more than that of the control group(P<0.05).The patients and their families in the observation group reported significantly higher satisfaction compared to those in the control group(both P<0.05).Conclusion The fast-track specialist nurse-led intravenous thrombolytic therapy mode demonstrates the superiority in reduction of the time from admission to completion of CT examination,time for signing an informed consent for thrombolytic therapy,door to needle time and the NIHSS scores,higher percentage of DTN<60 minutes as well as improvement of patient satisfaction.

6.Innovations and challenges in intraperitoneal chemotherapy for peritoneal metastatic carcinoma

Peritoneal metastatic carcinoma refers to the spread of malignant tumors from the primary site to the peritoneum.It is commonly observed in the advanced stages of various cancer types,including gastric cancer,colorectal cancer,ovarian cancer and pseudomyxoma peritonei(PMP).Peritoneal metastatic carcinoma seriously impair human health due to their high recurrence and mortality rates,and palliative treatment based on chemotherapy is usually provided after radical surgery or in the advanced stage of peritoneal metastases.The difficulties in the treatment of peritoneal metastatic carcinoma are the lack of targeted drugs and the difficulty of drugs to cross the blood-peritoneal barrier,resulting in poor systemic effects.Intraperitoneal chemotherapy,as an important therapeutic means,has a broad application prospect in the treatment of peritoneal metastatic carcinoma.In recent years,innovations in intraperitoneal chemotherapy techniques such as hyperthermic intraperitoneal chemotherapy(HIPEC),pressurized intraperitoneal aerosol chemotherapy(PIPAC)and the development of novel drugs have significantly improved patients'quality of life.The development of novel drugs has significantly improved patient survival.However,the diversity and complexity of these diseases have led to clinical variability and uncertainty in the efficacy of intraperitoneal chemotherapy in terms of treatment strategies such as mode of administration,drug type and dose.Although evidence-based guidelines and recommended treatment strategies have been developed,there is a need to further support these regimens through additional clinical trials and higher levels of evidence-based medicine.This review summarized the development history and recent advances in intraperitoneal chemotherapy,compared and analyzed the treatments such as surgery combined with traditional IPC,HIPEC and PIPAC,and summarized the research progress of recently conducted intraperitoneal chemotherapy techniques;moreover,in terms of the clinical application of intraperitoneal chemotherapy techniques,this paper elaborated on its use in diseases such as gastric cancer,colorectal cancer,gynecological oncology,PMP,cholangiocarcinoma and pancreatic cancer.For the limitations of intraperitoneal chemotherapy technology,it is proposed that the innovative development of nanomedicine is expected to provide a safer and more effective option for the treatment of peritoneal metastatic carcinoma.In conclusion,this review summarized the latest progress,shortcomings and future development of intraperitoneal chemotherapy,with the aim of providing a feasible direction for more effective clinical treatment of peritoneal metastatic carcinoma.
7.DNA nanoscale coordination polymers promote the activity of bone marrow stem cells

Objective:A DNA coordination polymer(Ca-pHis-Apt1 9S)was synthesized and its effect on the activity of bone marrow stem cells(BMSCs)was investigated. Methods:BMSCs were extracted from the epiphysis of the femoral shaft of mice by adherence screening method and identified by the expression of surface antigens through flow cytometry.Cell penetrating peptide pHis was covalently linked with the BMSCs aptamer Aptl9S,and a DNA coordination polymer(Ca-pHis-Aptl 9S),which was expected to improve the viability of BMSCs,was prepared through coordination assembly with calcium ions.BMSCs were treated with different concentrations of Ca-pHis-Apt1 9S,and the effects of Ca-pHis-Apt1 9S on the biological activities and the phenotype of BMSCs were evaluated by CCK-8 assay,live/dead cell staining assay and flow cytometry assay.The targeting ability of Ca-pHis-Apt1 9S toward BMSCs was observed using a fluorescence confocal microscope.Meanwhile,CCK-8 assay was used to study the main components of Ca-pHis-Aptl 9S that could promote the viability of BMSCs. Results:Spherical Ca-pHis-Apt1 9S with the particle size of 50-120 nm was successfully synthesized.The synthesized Ca-pHis-Apt1 9S could significantly promote the viability of BMSCs(P<0.05)without affecting the apoptosis or the expression of surface antigens(CD29,CD44 and CD34).Confocal microscopy analysis showed that the BMSCs aptamer Apt19S could further enhance the cellular uptake of Ca-pHis-Apt1 9S by BMSCs.The results of CCK-8 assay revealed that pHis in Ca-pHis-Apt1 9S was the main component that could promote the viability of BMSCs,and this promotional effect was dose-dependent(P<0.05). Conclusion:Ca-pHis-Aptl 9S can target BMSCs in vitro and significantly promote their biological activity without affecting their phenotype,providing a new method for the in vitro culture of BMSCs.
8.Protein-based Bioinformatics Analysis of Cervical Cancer Related Genes

Purpose/Significance To explore the characteristics and clinical significance of differentially expressed genes closely re-lated to HPV E6/E7 by using bioinformatics.Method/Process The cervical tissue and clinical information of cervical cancer in TCGA and GTEx of UCSC are used as the training set.The expression profile chip GSE63514 related to cervical cancer in GEO is used as the validation set.Firstly,the limma package of R software is used to screen DEGs of tumor and normal samples,and Venn map of genes re-lated to E6/E7 protein in MigDB is made.Survival analysis is performed by survival kit and verified by ROC and protein expression lev-els.Secondly,key genes are obtained by copy number variation and methylation correlation.Finally,the specific co-expression network is constructed and enrichment analysis and immune infiltration analysis are performed.Result/Conclusion There are 101 differentially expressed genes related to HPV E6/E7 protein,and three genes are found to have significance after screening,namely E2F1,MCM4 and PCNA.At the same time,it is found that the genes in the specific coexpression network are significantly enriched in the DNA replication and chromosome organization pathways.Immune correlation analysis shows that key genes are significantly associated with CD4 T cells,B cells and neutrophils.DNA replication,chromosome organization,etc.,are the molecular mechanisms and key genes significantly related to the development of cervical squamous cell carcinoma and HPV E6/E7 encoded proteins.
9.Analysis of SCI paper output and research hotspots of Chinese dental research institutions

Objective:To statistically analyze SCI paper output and research hotspots of Chinese stomatology research institutes from 2011 to 2020.Methods:The papers of Chinese stomatology research institutions in the Web of Science database were searched and analyzed by bibliometric methods in various dimensions including publication time, institutional and journal distribution, international (regional) cooperation, and research hotspots.Results:The number and quality of publications in Chinese dental research institutions have increased significantly in the past decade, and the top four institutions in terms of publication number were Sichuan University, Peking University, Shanghai Jiaotong University, and Air Force Military Medical University. The distribution of the journals to which the articles belong was relatively scattered, and the research hotspots mainly focused on squamous cell carcinoma, periodontitis, bone regeneration, and materials science.Conclusions:The output of SCI papers and the number of high-quality papers in Chinese stomatology research institutions increased year by year during 2011-2020, and the scientific research strength continued to increase.
