1.A comparative study of regulation of the homing ability of mesenchymal stem cells by hypoxia and hydrogen peroxide pretreatment

Objective To determine the optimal conditions for CXCR4 upregulation by comparing the expression levels of chemokine(C-X-C motif)receptor 4(CXCR4)in MSCs cultured with varying concentrations of oxygen and hydrogen peroxide(H2O2).Methods MSCs were cultured with 0.1%,1%,or 3%O2 and 50 μmol/L H2O2 for different lengths of time(3,6,12,and 24 h).The mRNA and protein expressions of CXCR4 in MSCs were measured by real-time quantitative PCR(qPCR),Western blotting,and immunofluorescence staining.The viability and chemotactic ability of MSCs were measured using CCK-8,wound-healing and Transwell migration assays.Results Both hypoxia and H2O2 treatment were found to upregulate MSC expressions of CXCR4 to some extent.The mRNA and protein levels of CXCR4 were higher after 6-12 h of culture of MSCs with 3%O2,and significantly higher when treated with H2O2 for 6 h.Cell viability was significantly increased after culture with 3%O2 compared with the control group and both 3%O2 and H2O2 pretreatment could enhance chemotactic migration in MSCs.Conclusion Culture with 3%O2 and H2O2 pretreatment can upregulate CXCR4 expressions in MSCs and enhance migration in cells,with superior effects observed with 3%O2.Therefore,treatment with 3%O2 represents the best choice for upregulating the chemotactic ability of MSCs.
2.Prenatal diagnosis and genetic analysis of two fetuses with Wolf-Hirschhorn syndrome

Objective:To explore the prenatal ultrasound phenotype and genetic basis of two fetuses with Wolf-Hirschhorn syndrome (WHS).Methods:A retrospective analysis was conducted on the ultrasound imaging data of two fetuses suspected for WHS at the Prenatal Diagnostic Center of Qingyuan People′s Hospital in July 2017 and August 2019, respectively. Amniotic fluid samples of the two fetuses were subjected to chromosomal karyotyping and chromosomal microarray analysis (CMA). This study was approved by Medical Ethics Committee of the Qingyuan People′s Hospital (Ethics No. IRB-2022-064).Results:Prenatal ultrasound examination of the two fetuses had consistently revealed WHS-associated traits including intrauterine growth restriction (IUGR), craniofacial abnormalities and cardiovascular anomalies. Karyotyping analysis suggested that both fetuses had harbored cryptic chromosomal translocations involving partial deletion of 4p. And parental verification revealed that it was de novo for fetus 1 and paternal for fetus 2. CMA has confirmed that fetus 1 had an approximately 8.7 Mb deletion at 4p16.3p16.1 and a 6.8 Mb duplication at 8p23.1p23.1, whilst fetus 2 had a 20.05 Mb deletion at 4p16.3p15.31 and a 7.66 Mb duplication at 9p24.3p24.1. The karyotype of fetus 1 was determined as 46, XN, der(4)t(4; 8)(p16.1; p23.1)dn.arr[hg19]4p16.3p16.1(68345_8721580)×1, 8p23.3p23.1(158048_6933745)×3, and that of fetus 2 was determined as 46, XN, der(4)t(4; 9)(p15.3; p24)dpat.arr[hg19]4p16.3p15.31(68345_20116061)×1, 9p24.3p24.1(208454_7868292)×3. Conclusion:The 4p deletion is probably the main cause for the WHS phenotype in both fetuses. WHS should be suspected when IUGR, renal anomalies, craniofacial and cardiovascular abnormalities are detected upon prenatal ultrasound screening.
3.Consistency analysis between pathological and imaging evaluations of gastrointestinal stromal tumor after preoperative targeted therapy

Objective:To assess the efficacy and prognosis of gastrointestinal stromal tumors(GIST)after preoperative targeted therapy us-ing the Choi criteria compared to pathological effects,and to observe the consistency between them.Methods:The clinicopathological data of 37 patients,who underwent preoperative treatment with targeted imatinib therapy for GIST,were retrospectively analyzed.Survival ana-lysis of the Choi criteria and pathological effects was conducted using the Kaplan-Meier method and Log-rank test.The consistency between the Choi criteria and the pathological effects was assessed using Spearman's correlation and Kappa tests.Results:The median preoperative treatment duration for the 37 patients was 10 months(range,2-36 months).According to the Choi criteria,there were no cases of complete response(CR),26 cases of partial response(PR),five cases of stable disease(SD),and six progressive disease(PD)cases.The difference in overall survival(OS)between the effective group(CR+PR)and the ineffective group(SD+PD)was statistically significant(P<0.01).Pathologic-al effects were evaluated as one complete effect,11 high effects,18 partial effects,and seven zero effect cases.The OS significantly differed between the effective(full effect+high effect+partial effect)and ineffective(zero effect)groups was statistically significant(P<0.01).The Choi showed moderate consistency with the pathological effects(r=0.592,P<0.01)with a(κappa=0.566).Conclusions:The Choi criteria were moderately correlated and consistent with the pathological effects.Both can be used to evaluate the efficacy and prognosis of preoperative targeted therapy for GIST.The combined use of these two criteria has better clinical application value than that of either alone.
4.Drug resistance characteristics and spatial clustering of Mycobacterium tuberculosis in Qinghai Province from 2016 to 2019

Abstract: Objective To analyze the resistance and spatial distribution of Mycobacterium tuberculosis (MTB) to six commonly used anti-tuberculosis drugs in Qinghai Province from 2016 to 2019, so as to provide a reference for tuberculosis treatment and drug-resistant tuberculosis control. Methods A total of 1 182 identified strains of Mycobacterium tuberculosis in Qinghai Province from 2016 to 2019 were collected, and 6 anti-tuberculosis drugs were subjected to drug susceptibility tests and strain confirmed by the proportional method. By means of ArcMap10.7 and SaTScan10.1 software, map visualization, spatial autocorrelation analysis and spatial scanning of MTB drug resistance were performed to identify MTB drug resistance clusters in Qinghai Province. Results From 2016 to 2019, the total drug resistance (TDR) rate of 1 182 Mycobacterium tuberculosis strains in Qinghai Province was 23.77% (281/1 182), with a mono-resistance (MR) rate of 11.08% (131/1 182), a poly-resistance (PDR) rate of 3.89% (46/1 182), a multi-drug resistance (MDR) rate of 8.80% (104/1 182), and an extensive drug resistance (XDR) rate of 0.85% (10/1 182). The rates of MDR, XDR and TDR all showed a decreasing trend year by year (P<0.01). The drug resistance spectrum displayed 21 combinations. The TDR rate and MDR rate in the retreatment patients were higher than those of the initial treated patients, and the difference was statistically significant (χ2
TDR=22.784, χ2MDR=45.082, P<0.01). In terms of demographic characteristics, the TDR rate in males was higher than that in females, and the middle-aged group was higher than other age groups, and the differences were statistically significant (χ2=7.541, 10.825, P<0.05). The results of global spatial autocorrelation analysis showed that there was no statistical significance in the autocorrelation and obvious spatial clustering of MTB drug resistance in Qinghai Province from 2016 to 2019 (P>0.05), which indicated a random distribution. The results of spatiotemporal scanning showed that there was a kind of clustering area, but the clustering effect was not significant (P>0.05), indicating a random distribution. Conclusions The TDR of MTB in Qinghai Province from 2016 to 2019 showed a downward trend year by year. In comparison with the national average, the rate of multi-drug resistance and extensive drug resistance was still high, and most of the multi-drug resistance resulted from rifampicin and isoniazid. In response to the Opinions of the CPC Central Committee and the State Council on Promoting the Inheritance, Innovation, and Development of traditional Chinese medicine(TCM) and the spirit of the National Conference on TCM, Chinese Association of Chinese Medicine organized experts in Otorhinolaryngology Head and Neck Surgery of traditional Chinese and western medicine to discuss the clinical advantages of TCM and integrated traditional Chinese and western medicine in the treatment of allergic rhinitis (AR) and they reached a basic consensus. In recent years, the prevalence of AR has been on the rise, threatening the quality of life of patients and giving rise to a heavy burden to both the patients and the society. AR is resulted from immune imbalance rather than reduced immunity or hyperimmunity, and the imbalance is similar to the Yin-yang disharmony in TCM. In the treatment of this disease, western medicine features rapid onset. However, it is cost-intensive and causes severe surgical trauma, and the recurrence is common. TCM boasts diverse methods for AR, which can be used in all stages of this disease. It has advantages in controlling symptoms such as nasal congestion, runny nose, or dysosmia in the attack stage, preventing recurrence in the remission stage, and treating refractory AR or steroid-resistant AR. In particular, acupuncture enjoys a reputation in treatment of AR, which has been supported by evidence-based medicine and recommended by guidelines. While treating local symptoms of AR, TCM regulates the psychosomatic conditions, which facilitates chronic disease management and long-term follow-up. We should integrate the advantages of TCM and western medicine, give full play to the unique nonnegligible and irreplaceable advantages of TCM, formulate a comprehensive diagnosis and treatment scheme for learning and promotion, and summarize the research outcomes to promote the theoretical innovation of TCM on AR from the perspective of integrated traditional Chinese and western medicine.
6.Diagnostic value of real-time shear wave elastography for liver fibrosis in patients with autoimmune hepatitis

Objective To explore the diagnostic value of Young's modulus obtained by real-time shear wave elastography (SWE) for liver fibrosis in autoimmune hepatitis (AIH) patients. Methods A total of 75 AIH patients in the First Affiliated Hospital of Zhengzhou University from January 2013 to April 2022 were retrospectively enrolled. Scheuer scoring system was used to evaluate degrees of liver fibrosis (S0-S4). By using pathological examination of liver tissues as the golden standard, the receiver operating characteristic curve (ROC) was plotted and the area under the curve (AUC) was used to evaluate the diagnostic value of SWE for the significant fibrosis (≥S2), advanced liver fibrosis (≥S3), and liver cirrhosis (S4), respectively. Independent sample t test was used for comparison of continuous data with normal distribution between the two groups. The Kruskal-Wallis H test was used for comparison of non-normally distributed continuous data between multiple groups and Bonferroni method was used for further comparison between two groups. The Spearman correlation coefficient was used for correlation analysis. The logistic regression analysis was used to predict the impact factors in diagnosis accuracy. Results The Young's modulus measured by SWE was statistically significant different among various fibrosis groups ( H =35.186, P < 0.001) although there was no statistical significance in patients' age and platelet, alanine aminotransferase, aspartate aminotransferase, total bilirubin, alkaline phosphatase, and glutamyl transpeptidase levels (all P > 0.05). The Young's modulus measurement was positively correlated with liver fibrosis ( r =0.675, P < 0.05). The AUCs of SWE in the diagnosis of≥S2, ≥S3, and S4 were 0.839, 0.820 and 0.898, respectively and the corresponding optimum cut-off values were 9.2, 10.9, and 14.4 kPa, respectively. The overall concordance rate of the liver Young' s modulus measurements vs . fibrosis stages was 57.33%. Moreover, the alkaline phosphatase level was an independent predictor for diagnostic accuracy of SWE for stage 0-1 fibrosis ( OR =1.009, 95% CI : 1.001-1.018, P =0.029). Conclusion The SWE possessed a diagnosis value for the significant fibrosis (≥S2), advanced liver fibrosis (≥S3) and liver cirrhosis (S4), although there was a low overall concordance rate in the liver Young's modulus measurements obtained using SWE vs. fibrosis stages.
7.Correlation analysis of school bullying and depressive symptoms among multi ethnic adolescents in western China

Objective:
To understand the prevalence of school bullying and depressive symptoms among the Han, Yi, and Tibetan adolescents in western China, and to explore the relationships between bullying roles, bullying frequency and depression symptoms.
Methods:
A combination method of typical sampling and cluster sampling was used to recruit 3 115 participants from six primary and middle schools in Cangxi County and Ganluo County in Sichuan Province, and Lhasa City in Tibet Autonomous Region from April to November 2020. All participants completed self filled questionnaires. Multiple Logistic regression was used to analyze the association between school bullying and depression symptoms.
Results:
A total of 881 participants (28.3%) had experienced school bullying in the past 6 months, and 892 participants(28.6%) were detected with depressive symptoms. The rates of school bullying in the Han, Yi, and Tibetan adolescents were 20.5%, 46.4% and 23.8%, respectively. Among them, the rates of victims were 15.3%, 25.0% and 11.8%, respectively; the rates of bully were 2.3%, 5.7% and 6.7%, respectively; the rates of bully victims were 2.9%, 5.7% and 5.4%, respectively. The prevalence of depressive symptoms in the Han, Yi, and Tibetan adolescents were 20.4%, 37.9% and 36.5%, respectively. After adjusting for age, grade, ethnicity, smoking, and drinking, the odds for depressive symptoms among victims ( AOR=1.98, 95%CI =1.61- 2.45 ), bully ( AOR=2.63, 95%CI =1.81-3.82), and bully victims ( AOR=3.33, 95%CI =2.44-4.54) were significantly higher than those without school bullying experience( P <0.01). And the risks increased with the increasing bullying frequency.
Conclusion
School bullying and depressive symptoms of the Yi and the Tibetan adolescents in western China were higher than those of the Han adolescents. Meanwhile, participation in school bullying and the high frequency of bullying were closely related to depressive symptoms. Future interventions should pay attention to various bullying roles and minority adolescents.

Ferroptosis is defined as an iron-dependent, non-apoptotic cell death pathway, with specific morphological phenotypes and biochemical changes. There is a growing realization that ferroptosis has significant implications for several neurodegenerative diseases. Even though ferroptosis is different from other forms of programmed death such as apoptosis and autophagic death, they involve a number of common protein molecules. This review focuses on current research on ferroptosis and summarizes the cross-talk among ferroptosis, apoptosis, and autophagy that are implicated in neurodegenerative diseases. We hope that this information provides new ideas for understanding the mechanisms and searching for potential therapeutic approaches and prevention of neurodegenerative diseases.

9.Rehabilitation study of somatosensory game combined with high frequency repetitive transcranial magnetic stimulation on mild cognitive impairment after stroke

Objective:To explore effects of somatosensory games combined with high frequency repetitive transcranial magnetic stimulation on cognitive function rehabilitation in patients with mild cognitive impairment after stroke.Methods:Using the convenience sampling method, a total of 64 patients with post stroke cognitive impairment (PSCI) admitted to Rehabilitation Department of the First Affiliated Hospital of Wenzhou Medical University from January to June 2021 were selected as the research objects. According to the order of admission and the 1∶1 matching principle, they were randomly divided into the control group and the observation group, with 32 cases in each group. The patients in the control group received high-frequency repetitive transcranial magnetic stimulation and conventional cognitive rehabilitation training, while the observation group received somatosensory game training on the basis of the control group. Scores of National Institute of Health Stroke Scale (NIHSS) , Montreal Cognitive Assessment (MoCA) , Mini-mental State Examination (MMSE) and Modified Barthel Index (MBI) were compared between the two groups before and after the intervention.Results:After 20 days of intervention, the NIHSS score of the observation group was lower than that of the control group, and the difference was statistically significant ( P<0.05) . The scores of MoCA, MMSE and MBI in the observation group were higher than those in the control group, and the differences were statistically significant ( P<0.05) . The compliance rate of rehabilitation therapy in the observation group was higher than that in the control group, and the difference between the two groups was statistically significant (χ 2=4.267, P=0.039) . Conclusions:Somatosensory games combined with high-frequency repetitive transcranial magnetic stimulation can improve the cognitive ability of patients with PSCI, improve the compliance rate of patients with rehabilitation exercises and help patients recover.
10.Progress of JMJD5 in tumors

JMJD5 belongs to the protein family of JMJD and is closely related to tumorigenesis. JMJD5 plays a carcinogenic role in some tumors, such as oral cancer, prostate cancer, breast cancer, colon cancer and atypical meningiomas, JMJD5 promotes the proliferation, migration and invasion of cancer cells. However, JMJD5 not only plays a carcinogenic role in some tumors, but also plays an anticancer role. For example, in bladder cancer, JMJD5 inhibits the metastasis of bladder cancer; in liver cancer, on the one hand JMJD5 promotes the replication of hepatitis B virus (HBV), and on the other hand it inhibits the growth of cancer cells; in lung cancer, on the one hand JNJD5 promotes the proliferation of cancer cells, and on the other hand it inhibits the metastasis of cancer cells by promoting the stabilization of microtubules. This article will focus on the mechanisms of JMJD5 in tumors, in order to provide a new target for tumor prevention and treatment.
