1.GLUT1-targeted Nano-delivery System for Active Ingredients of Traditional Chinese Medicine:A Review

Tumor cells use glycolysis to provide material and energy under hypoxic conditions to meet the energy requirements for rapid growth and proliferation, namely the Warburg effect. Even under aerobic conditions, tumor cells mainly rely on glycolysis to provide energy. Therefore, glucose transporter protein 1(GLUT1), which is involved in the process of glucose metabolism, plays an important role in tumorigenesis, development and drug resistance, and is considered to be one of the important targets in the treatment of malignant tumors. In recent years, research on tumor glucose metabolism has gradually become a hot spot. It has been shown that various factors are involved in the regulation of tumor energy metabolism, among which the role of GLUT1 is the most critical. In this paper, the authors reviewed the latest research progress of GLUT1-targeted traditional Chinese medicine(TCM) active ingredient nano-delivery system in tumor therapy, aiming to reveal the feasibility and effectiveness of this system in the delivery of chemotherapeutic drugs. The GLUT1-targeted TCM active ingredient nano-delivery system can overcome the bottleneck of the traditional targeting strategy as well as the high-permeability long retention(EPR) effect. In summary, the authors believe that the GLUT1-targeted TCM active ingredient nano-delivery system provides a new strategy for targeted treatment of tumors and has a broad application prospect in tumor prevention and treatment.
2.Compound ANBP promotes diabetic wound healing by regulating TCA circulation-related enzymes

AIM:To investigate the effects of the compound ANBP on wound healing in diabetic rats and ex-plore its mechanism of action.METHODS:Ninety male SD rats were randomly divided into blank,model,compound ANBP,Beifuxin,and nicotinamide mononucleotide(NMN)groups,with 16 rats in each group.Wound healing in each group was observed and samples were taken on days 3,7 and 14 to analyze the wound healing rate.Local histopathological changes were observed using HE and Masson staining.The expressions of pyruvate dehydrogenase E1 subunit alpha 1(PDHA1),citrate synthase(CS),isocitrate dehydrogenase(IDH1)and oxoglutarate dehydrogenase(OGDH)were de-tected through immunofluorescence and Western blot.The number and morphology of mitochondria in the wound tissue were observed using transmission electron microscopy.RESULTS:Histomorphological changes revealed significant im-provement in diabetic wound healing in the blank and compound ANBP groups compared to that of the model group.The wound healing rates of the blank,compound ANBP,Beifuxin,and NMN groups were significantly increased on days 3,7,and 14(P<0.01).Compared to the model group,granulation tissue generation was higher in the other groups,cover-ing the wound defect and producing abundant collagen fibers.At 3,7,and 14 days after intervention,the blank,com-pound ANBP,Beifuxin,and NMN groups showed significantly enhanced fluorescence intensities of TCA cycling-related enzymes PDHA1,CS,IDH1,and OGDH indicating increased expression of these enzymes.The levels of the TCA cy-cling-related enzymes were significantly increased(P<0.01)in the compound ANBP,Beifuxin and NMN groups but were significantly decreased(P<0.01)in the model group.An increase in the number and density of mitochondria and a de-crease in the cavitation rate of mitochondria with improved morphology(P<0.05)was observed in the group treated with compound ANBP.CONCLUSION:Compound ANBP may increase the number of mitochondria,improve mitochondrial morphology and function,upregulate the expression levels of PDHA1,CS,IDH1,and OGDH proteins,and accelerate the regeneration of wound granulation tissue,thus promoting the healing of diabetic wounds in rats.
3.Clinical analysis on platinum-based combined chemotherapeutical regimens for treating relapsed or refractory non-Hodgkin lymphoma

Objective To evaluate the efficacy and adverse reactions of platinum-based combined chemotherapeutical regimens in treating relapsed or refractory non-Hodgkin lymphoma(NHL).Methods The clinical data of 68 patients with relapsed or refractory NHL treated with platinum-based combined chemotherapeutical regimens in the Affiliated Tumor Hospital of Guangxi Medical University from January 2008 to December 2014 were retrospectively analyzed.The curative effect of related regimens,adverse reactions and related influence factors were analyzed.Results Sixty-eight cases received 283 cycles of chemotherapy.In all cases,11 cases(16.18 %) achieved the complete response(CR),31 cases(45.59 %) achieved the partial response(PR),the overall response rate(ORR) was 61.76%;the median progression-free survival(PFS) was 6.51 months(95%CI:4.97-8.04 months).ORR and PFS in the cases of stage Ⅱ-Ⅲ,IPI score 0-2 and receiving only one chemotherapeutical regimen were superior to those in the cases of corresponding subgroup(P<0.05);ORR and PFS had no statistical difference between the B cells lymphoma and Tcells lymphoma(P>0.05).The medion PFS in the combined R group was 11.16 months,which was longer than 5.84 months in the non-combined R group(P =0.004).The major adverse events (stage Ⅱ-Ⅲ) included leukopenia (41.18 %),thrombocytopenia (27.94%),hemoglobin decrease(11.76%),vomiting(8.82%) and diarrhea(1.47%).Conclusion The platinum-based combined chemotherapeutical regimens are effective with good safety in the treatment of relapsed or refractory NHL.
4.Establishment and evaluation of isoprenaline-induced myocardial fibrosis mouse model

Aim To establish and evaluate a mouse model of my-ocardial fibrosis by hypodermic injection of isoprenaline. Meth-ods Kunming mice were randomly divided into 2 groups, namely, the myocardial fibrosis model group and the control group, 10 mice in each group. The mice in the model group were given isoprenaline 5 mg · kg-1 by hypodermic injection. From the following day, the dose of isoprenaline was reduced to 2. 5 mg·kg-1 , and lasted for 30 days. The mice in the control group were treated with the physiological saline in the same way. Final-ly, heart weight was then weighed and the cardiac weight index (CWI) was calculated. Hydroxyproline (Hydro) level in myo-cardium was determined by a colorimetric method. HE and Mas-son’s trichrome staining were used to estimate the extent of myo-cardial fibrosis and calculate the collagen volume fraction ( CVF) . RT-PCR was used to measure the myocardial Collagen I mRNA expression. Results Compared with the control group, the CWI and Hydro content in the myocardial tissues in the model group were increased(P<0.01). The content of col-lagen in the myocardial tissues and the CVF were increased obvi-ously(P<0.01). The RT-PCR results showed that the left ven-tricle Collagen I mRNA expression in the model group increased obviously( P<0.01 ) . Conclusion Isoprenaline-induced myo-cardial fibrosis model has been established and the method is very simple, economic and reliable.
5.Correlated Study of CRP、IL-12 and Insulin resistance in Gestational Diabetes Mellitus

Objective to investigate the relationship between CRP,IL-12 and gestational diabetes mellitus.Methods Fifty normal pregnant women (NGT group) and forty with GDM (GDM group) were enrolled in the study.The serum IL-12、fasting blood sugar、CRP were mesured respectively.Results GDM fasting blood glucose,fasting insulin,insulin resistance index (IRI),CRP,IL-12 were higher than those in the control group [(4.95 ± 0.76 vs 4.57 ± 0.58) mmol/L,(13.65 ± 1.74 vs 8.78 ± 1.65) uIU/ml,(2.91 ± 1.05vs 1.72 ± 0.87),(5.56 ± 1.24 vs 3.25 ± 1.13) mg/L,(71.47 ± 4.35 vs 56.03 ± 4.06) PG,/ml,P < 0.05),the results of single factor analysis of CRP showed a positive correlation with IRI (r =0.865,P < 0.05),IL-12 was positively correlated with IRI (r =0.913,P < 0.05).Conclusions Our study show inflammatory cytokins take part in the genesis of gestational diabetes.Serum levels of IL-12 and is one factor in pathogensis of GDM.
6.Study of aspirin on inhibiting the atherosclerotic plaque rupture and MMP-2 expression of abdominal aorta in atherosclerotic rabbits

Aim To study the effects of aspirin on increasing the atherosclerotic plaque stability and its possible mechanisms.Methods The hyperlipidemic atherosclerotic model was generated in male New Zealand rabbits given high fat diet and endothelial abrasion of abdominal aorta.These rabbits were then treated with aspirin 5~20 mg?kg-1 for 4 weeks.At experimental end,the plaques were evoked into rupture by injection of Russell's viper venom and histamine.Areas of thrombosis on atherosclerotic aorta were determined by image analysis,morphologic character of plaque rupture was examined by light microscope,the protein expression of macrophages was detected by immunohistochemistry,and the mRNA expression of COX-2 and MMP-2 was determined by hybridization in situ,respectively.Results Aspirin at doses of 5~10 mg?kg-1 was able to inhibit thrombosis on atherosclerotic plaque(P

9.Regulation of SREBPs and its research progress in the development of nonalcoholic fatty liver

Nonalcoholic fatty liver(NAFL)is characterized by the hepatocyte steatosis and lipid accumulation,and is a clinical syndrome of unrelated to excessive alcohol consumption. At present,patients with the syndrome obviously increase in the whole world. Sterol regulatory element-binding proteins (SREBPs) are the key transcription factors of controlling the biosynthesis of cholesterol,fatty acids and triglycerides,and adipocyte differentiation. In this paper,the regulation of SREBPs and its research progress in the development of NAFL were reviewed.
