1.Investigation on vehicle occupant dummy applicability for under-foot impact loading conditions

		                        			Purpose::Under-foot impact loadings can cause serious lower limb injuries in many activities, such as automobile collisions and underbody explosions to military vehicles. The present study aims to compare the biomechanical responses of the mainstream vehicle occupant dummies with the human body lower limb model and analyze their robustness and applicability for assessing lower limb injury risk in underfoot impact loading environments.Methods::The Hybrid III model, the test device for human occupant restraint (THOR) model, and a hybrid human body model with the human active lower limb model were adopted for under-foot impact analysis regarding different impact velocities and initial lower limb postures.Results::The results show that the 2 dummy models have larger peak tibial axial force and higher sensitivity to the impact velocities and initial postures than the human lower limb model. In particular, the Hybrid III dummy model presented extremely larger peak tibial axial forces than the human lower limb model. In the case of minimal difference in tibial axial force, Hybrid III's tibial axial force (7.5 KN) is still 312.5% that of human active lower limb's (2.4 KN). Even with closer peak tibial axial force values, the biomechanical response curve shapes of the THOR model show significant differences from the human lower limb model.Conclusion::Based on the present results, the Hybrid III dummy cannot be used to evaluate the lower limb injury risk in under-foot loading environments. In contrast, potential improvement in ankle biofidelity and related soft tissues of the THOR dummy can be implemented in the future for better applicability.
		                        		
		                        		
		                        		
		                        	
2.Comparison of three different modes of hysteroscopic surgery:outpatient,daily and inpatient

		                        			Objective To explore the safety and management mode of hysteroscopy in three different modes:outpatient,daily and inpatient.Methods The quality control data of patients who underwent hysteroscopic surgery in Hysterscopy Centre,Obstetrics and Gynecology Hospital,Fudan University from Jan 2019 to Dec 2021 were collected through the electronic information system of the hospital and the monthly quality control report of hysteroscopy center.The amount of surgery,the proportion of grade Ⅳsurgery,the analysis of operation types,the indicator including complications,and unanticipated secondary surgery were retrospectively analyzed.Results From 2019 to 2021,5 162 outpatient hysteroscopic patients,15 331 daily hysteroscopic patients and 5 942 inpatient hysteroscopic patients were admitted in our hospital.The age of inpatient hysteroscopic patients was significantly older than those of outpatient and daily patients(P<0.001).In the past three years,the proportion of daily hysteroscopy gradually increased,and the proportion of inpatient hysteroscopy gradually decreased(P<0.001).The total percentage of grade Ⅳ hysteroscopic surgery was 12.9%,in which inpatient was higher than daily,and daily was higher than outpatient(P<0.001).The incidence of complications and accidents during hysteroscopy was 0.117%(31/26 435),including 17 cases of uterine perforation,7 cases of hysteroscopy failure,3 cases of excessive intraoperative bleeding,2 cases of fluid overload,1 case of intestinal injury,and 1 case of anesthesia accident.The incidence of hysteroscopy in outpatient,daily and inpatient were 0.020%(1/5 162),0.137%(21/15 331)and 0.151%(9/5 942)respectively.Conclusion Hysteroscopy in outpatient,daily and inpatient are all safe and reliable.Outpatient and daily hysteroscopy can improve the efficiency of medical services,which has gradually become a trend.
		                        		
		                        		
		                        		
		                        	
3.Analysis of HA and NA gene variation characteristics of A(H1N1)pdm09 influenza virus in Shandong Province from 2022 to 2023

		                        			This study was aimed at characterizing the variations in hemagglutinin(HA)and neuraminidase(NA)genes of influenza virus subtype A(H1N1)pdm09 isolated during the 2022-2023 influenza monitoring year in Shandong Province,to provide a scientific basis for influenza prevention and control.A total of 14 A(H1N1)pdm09 subtype influenza strains were se-lected randomly by city by the influenza monitoring network laboratory.The vaccine strains recommended by the WHO served as references for whole gene sequencing analysis.A fluorescence method was used to conduct neuraminidase inhibition experi-ments to evaluate drug sensitivity.The A(H1N1)pdm09 influenza virus in Shandong Province,2022-2023 belonged to the 5a.2a evolutionary cluster in the 6B.1A branch.Nucleotide sequence analysis indicated that the HA and NA genes were closely re-lated to the Northern Hemisphere vaccine strain A/Victoria/2570/2019 in the years 2021-2023,and showed homology of 98.5%to 98.7%and 98.8%to 99.1%,respectively.Amino acid sequence analysis revealed 20 amino acid sequence mutations in the HA protein,but only one virus strain was found to have antigen drift,and three virus strains showed loss of HA protein glycosylation sites.No mutations were found at important sites affecting NA enzymes.The neuraminidase inhibition experiment indicated viral sensitivity to anti-influenza drugs.In conclusion,the monitored virus strains had high overall homology with vac-cine strains but showed some amino acid variation.In the future,continued monitoring of the genetic variation characteristics of influenza viruses will be necessary to understand the risk of influenza epidemics,and the effectiveness of influenza vaccines and therapeutic drugs.
		                        		
		                        		
		                        		
		                        	
4.Monitoring and analysis of avian influenza virus in poultry related environments in Shandong Province from 2020 to 2023

		                        			This study was aimed at understanding the pollution distribution pattern of avian influenza virus in the environ-ment in poultry related places in Shandong Province,to provide a scientific basis for the prevention,control,prediction,and early warning regarding human infection with avian influenza.From 2020 to 2023,6 523 environmental samples were collected in 16 cities in Shandong Province from four types of poultry-related places.Fluorescence quantitative PCR was used for nucleic acid testing of influenza A virus.Positive samples were further identified for the H5,H7,and H9 subtypes of avian influenza virus.The epidemiological characteristics of avian influenza viruses in the poultry related environment of Shandong Province were described,and inter-rate comparisons were performed with the x2 test.During 2020-2023,6 523 environmental samples were collected,and 1 007 cases positive for avian influenza virus were detected,with a positivity rate of 15.44%.H5,H7,and H9 subtypesand mixed infections were detected.H9 was the main subtype(88.48%)in positive specimens.A significant difference in positivity rates was observed among regions(x2=431.956,P<0.001),and the highest positivity rate was 28.93%.Significant differences in positivity rates were observed among monitoring sites(x2=304.604,P<0.001),sample types(x2=109.678,P<0.001),and quarters(x2=64.963,P<0.001).The positive detection rate was highest at monitoring sites in urban and rural live poultry markets(20.12%),and the positive detection rate of samples collected by wiping meat cut-ting board surfaces was higher than that of other samples(22.56%).The peak positive detection rate occurred in spring(20.31%).Widespread contamination with avian influenza virus was observed in poultry environments in Shandong Prov-ince.The H9 subtype,the main pathogen,coexisted with H5 and H7 subtypes,thus posing a risk of human infection with avian influenza.Therefore,prevention and control of avian influenza must be strengthened in key seasons,areas,places,and links.
		                        		
		                        		
		                        		
		                        	
5.Effect of astragaloside Ⅳ on depression-like behavior in rats with post-stroke depression and its related mechanisms

		                        			Objective To explore the effects and related mechanisms of astragaloside Ⅳ on depression-like behavior in rats with post-stroke depression(PSD)induced by chronic unpredictable mild stress(CUMS).Methods Fifty-four healthy male Sprague Dawley rats were randomly divided into blank group,sham surgery group,physiological saline group,astragalosideⅣ group,fluoxetine group,and combination therapy group,with 9 rats in each group.Rats in the blank group were raised normally without any treatment.After anesthesia,rats in the sham surgery group were exposed to the common carotid artery through a longitudinal incision about 2 cm along the right neck.The incision was not processed and sutured directly,and the rest were the same as those in the blank group.Rats in the remaining four groups were first used to prepare rat stroke models according to the suture method,and then CUMS induction was performed to prepare PSD models.After successful modeling,the rats in the physiological saline group were given physiological saline(5 mL·kg-1)daily by gavage,the rats in the astragaloside Ⅳ group were given astragaloside Ⅳ suspension(25.0 mg·kg-1)daily by gavage,the rats in the fluoxetine group were given fluoxetine suspension(12.5 mg·kg-1)daily by gavage,and the rats in the combination therapy group were given astragaloside Ⅳ(25.0 mg·kg-1)and fluoxetine(12.5 mg·kg-1)suspensiondaily by gavage.All rats in the four groups were administered once a day for a total of 4 weeks.The rats in the blank group and sham surgery group did not undergo gastric lavage treatment.The body mass was measured once a week,and the sucrose preference test,open field test,and Morris water maze test were also conducted once a week until the end of the experiment.After the administration,the rats were euthanized.The expression levels of adenosine monophosphate-activated protein kinase(AMPK)and brain-derived neurotrophic factor(BDNF)mRNA in the hippocampus of rats in each group were detected by reverse transcription polymerase chain reaction.The relative content of AMPK and BDNF positive cells in the hippocampus of rats in each group was detected by immunohisto-chemical staining.The expression levels of AMPK and BDNF protein in the hippocampus of rats in each group were detected by Western blot.Results After 4 weeks of treatment,there was no statistically significant differences in body mass,sucrose preference rate,horizontal movement distance in open field test,and Morris water maze crossing frequency of rats between the blank group and the sham surgery group(P>0.05);compared with the blank group and sham surgery group,the body mass,sucrose preference rate,horizontal movement distance in open field test,and Morris water maze crossing frequency of rats in the physiological saline group,astragaloside Ⅳ group,fluoxetine group,and combination therapy group were significantly reduced(P<0.05);compared with the physiological saline group,the body mass,sucrose preference rate,horizontal movement distance in open field test,and Morris water maze crossing frequency of rats in the astragaloside Ⅳ group,fluoxetine group,and combination therapy group significantly increased(P<0.05);there was no statistically significant differences in body mass,sucrose preference rate,horizontal movement distance in open field test,and Morris water maze crossing frequencyof rats among the astragaloside Ⅳ group,fluoxetine group,and combination therapy group(P>0.05).After 4 weeks of treatment,there was no statistically significant differences in the relative content of positive cell,and the relative expressions of AMPK,BDNF mRNA and protein in the hippocampus of rats between the blank group and the sham surgery group(P>0.05);compared with the blank group and sham surgery group,the relative content of positive cell,and the relative expressions of AMPK,BDNF mRNA and protein in the hippocampus of rats in the physiological saline group,astragaloside Ⅳ group,fluoxetine group,and combination therapy group significantly decreased(P<0.05);compared with the physiological saline group,the relative content of positive cell,and the relative expressions of AMPK,BDNF mRNA and protein in the hippocampus of rats in the astragaloside Ⅳ group,fluoxetine group,and combination therapy group increased(P<0.05);there was no statistically significant differences in the relative content of positive cell,and the relative expressions of AMPK,BDNF mRNA and protein in the hippocampus of rats among the astragaloside Ⅳ group,fluoxetine group,and combination therapy group(P>0.05).Conclusion Astragaloside Ⅳ can improve depression-like behavior in CUMS-induced PSD rats,and its mechanism may be related to increasing the expression of AMPK and BDNF in the hippocampus.

9.Effects of Chaihu Shugan Powder on behaviors and expression of 5-HT2A and BDNF in brain region of a rat model of post-stroke depression

		                        			AIM To investigate the effects of Chaihu Shugan Powder on the behaviors rat models of post-stroke depression(PSD)and the expression of 5-HT2A and BDNF in prefrontal cortex and striatum.METHODS The rats were randomly divided into the blank group,the sham operation group,the model group,Chaihu Shugan Powder group,the fluoxetine group and the combination drug group,with 9 rats in each group.The PSD rat models were established by the right middle cerebral artery occlusion(MCAO)by thread embolism method followed by further exposure to 4 weeks chronic and unpredictable mild stimulation.Subsequently,the rats started their corresponding regime of normal saline 5 mL/kg,Chaihu Shugan Powder 2 mg/kg,fluoxetine 12.5 mg/kg,and Chaihu Shugan Powder 2 mg/kg and fluoxetine 12.5 mg/kg(the combination group),respectively,and had their body mass measured and their behaviors(sugar water preference test,open field test and Morris water maze test)assessed simultaneously during the trial.After the experiment,the rats had their 5-HT2A and BDNF in prefrontal cortex and striatum detected of the mRNA expressions by RT-qPCR;of their expressions of the positive cells by immunohistochemical staining;and of their protein expressions by Westren blot.RESULTS Compared with the blank group,the model group displayed no improvement of depression-like behaviors,and decreased in mRNA expressions,positive cell expressions and protein expressions of 5-HT2A and BDNF(P<0.01).Compared with the model group,the groups intervened with Chaihu Shugan Powder,fluoxetine and combination drug showed improved depression-like behaviors,and increased mRNA expressions,positive cell expressions and protein expressions of 5-HT2A and BDNF as well(P<0.01).CONCLUSION Chaihu Shugan Powder can improve the depression-like behaviors in the PSD rat model,and its mechanism may be associated with the improvement in the expressions of 5-HT2A and BDNF in prefrontal cortex and striatum.
		                        		
		                        		
		                        		
		                        	
10.Effect of Tongdu Tiaoshen electroacupuncture pretreatment on PPARγ-mediated pyroptosis of cerebral cortex in rats with cerebral ischemia reperfusion injury.

		                        			OBJECTIVE:
		                        			To observe the effect of Tongdu Tiaoshen (promoting the circulation of the governor vessel and regulating the spirit) electroacupuncture (EA) pretreatment on pyroptosis mediated by peroxisome proliferators-activated receptor γ (PPARγ) of the cerebral cortex in rats with cerebral ischemia reperfusion injury (CIRI) and explore the potential mechanism of EA for the prevention and treatment of CIRI.
		                        		
		                        			METHODS:
		                        			A total of 110 clean-grade male SD rats were randomly divided into a sham-operation group, a model group, an EA group, an EA + inhibitor group and an agonist group, 22 rats in each group. In the EA group, before modeling, EA was applied to "Baihui" (GV 20), "Fengfu" (GV 16) and "Dazhui" (GV 14), with disperse-dense wave, 2 Hz/5 Hz in frequency, 1 to 2 mA in intensity, lasting 20 min; once a day, consecutively for 7 days. On the base of the intervention as the EA group, on the day 7, the intraperitoneal injection with the PPARγ inhibitor, GW9662 (10 mg/kg) was delivered in the EA + inhibitor group. In the agonist group, on the day 7, the PPARγ agonist, pioglitazone hydrochloride (10 mg/kg) was injected intraperitoneally. At the end of intervention, except the sham-operation group, the modified thread embolization method was adopted to establish the right CIRI model in the rats of the other groups. Using the score of the modified neurological severity score (mNSS), the neurological defect condition of rats was evaluated. TTC staining was adopted to detect the relative cerebral infarction volume of rat, TUNEL staining was used to detect apoptosis of cerebral cortical nerve cells and the transmission electron microscope was used to observe pyroptosis of cerebral cortical neural cells. The positive expression of PPARγ and nucleotide-binding to oligomerization domain-like receptor protein 3 (NLRP3) in the cerebral cortex was detected with the immunofluorescence staining. The protein expression of PPARγ, NLRP3, cysteinyl aspartate specific protease-1 (caspase-1), gasdermin D (GSDMD) and GSDMD-N terminal (GSDMD-N) in the cerebral cortex was detected with Western blot. Using the quantitative real-time fluorescence-PCR, the mRNA expression of PPARγ, NLRP3, caspase-1 and GSDMD of the cerebral cortex was detected. The contents of interleukin (IL)-1β and IL-18 in the cerebral cortex of rats were determined by ELISA.
		                        		
		                        			RESULTS:
		                        			Compared with the sham-operation group, the mNSS, the relative cerebral infarction volume and the TUNEL positive cells rate were increased (P<0.01), pyroptosis was severe, the protein and mRNA expression levels of PPARγ, NLRP3, caspase-1 and GSDMD were elevated (P<0.01); and the protein expression of GSDMD-N and contents of IL-1β and IL-18 were increased (P<0.01) in the model group. When compared with the model group, the mNSS, the relative cerebral infarction volume and the TUNEL positive cells rate were decreased (P<0.01), pyroptosis was alleviated, the protein and mRNA expression levels of PPARγ were increased (P<0.01), the protein and mRNA expression levels of NLRP3, caspase-1 and GSDMD were decreased (P<0.01), the protein expression of GSDMD-N was reduced (P<0.01); and the contents of IL-1β and IL-18 were lower (P<0.01) in the EA group and the agonist group; while, in the EA + inhibitor group, the protein expression of PPARγ was increased (P<0.01), the protein and mRNA expression levels of NLRP3 and GSDMD were decreased (P<0.01, P<0.05), the mRNA expression of caspase-1 was reduced (P<0.01); and the contents of IL-1β and IL-18 were lower (P<0.01). When compared with the EA + inhibitor group, the mNSS, the relative cerebral infarction volume and the TUNEL positive cells rate were decreased (P<0.05, P<0.01), pyroptosis was alleviated, the protein and mRNA expression levels of PPARγ were increased (P<0.01), the protein and mRNA expression levels of NLRP3, caspase-1 and GSDMD were decreased (P<0.01), the protein expression of GSDMD-N was reduced (P<0.01); and the contents of IL-1β and IL-18 were declined (P<0.01) in the EA group. Compared with the agonist group, in the EA group, the relative cerebral infarction volume and the TUNEL positive cells rate were increased (P<0.05, P<0.01), the mRNA expression of PPARγ was decreased (P<0.01) and the protein expression of GSDMD-N was elevated (P<0.05); and the contents of IL-1β and IL-18 were higher (P<0.01).
		                        		
		                        			CONCLUSION
		                        			Tongdu Tiaoshen EA pretreatment can attenuate the neurological impairment in the rats with CIRI, and the underlying mechanism is related to the up-regulation of PPARγ inducing the inhibition of NLRP3 in the cerebral cortex of rats so that pyroptosis is affected.
