1.Clinical efficacy of repeated transcranial magnetic stimulation combined with acupuncture for chronic insomnia

		                        			Objective To explore the clinical efficacy of repeated transcranial magnetic stimulation(rTMS)and acupuncture therapy in the treatment of chronic insomnia disorder(CID)patients.Methods A total of 80 patients with CID,who were treated at Nanchang First Hospital from January 2022 to December 2023,were selected for the study.The patients were randomly divided into control group and treatment group,with 40 cases in each group.The control group patients were treated with dexmedetomidine,while the treatment group patients received rTMS and acupuncture therapy in addition to control group.The treatment course was 4 weeks,and the sleep quality,sleep related indicators,and psychological condition improvement of both groups of patients were observed before and after treatment.Results After treatment,the Pittsburgh sleep quality index scores of both groups of patients decreased(P<0.05);The sleep latency and number of awakenings were lower than before treatment(P<0.05),and the total sleep time,sleep efficiency,and proportion of rapid eye movement sleep were higher than before treatment,treatment group showed more significant improvement than control group(P<0.05).After treatment,the Hamilton anxiety and depression scale scores of both groups of patients decreased compared to before treatment,but there was no statistically significant difference in control group before and after treatment(P>0.05).However,there was a statistically significant difference in treatment group before and after treatment(P<0.05).Conclusion The combination of rTMS and acupuncture treatment can significantly improve the sleep quality of CID patients,while also reducing the accompanying symptoms of anxiety and depression.
		                        		
		                        		
		                        		
		                        	
2.Effects of Electroacupuncture on Motor Function and the Expressions of Irisin,Decorin and Myostatin in SAMP8 Mice

		                        			Objective To observe the effects of electroacupuncture on the motor function and expressions of Irisin,Decorin and Myostatin in skeletal muscle of SAMP8 mice;To explore the mechanism of electroacupuncture in the treatment of the motor dysfunction of Alzheimer disease(AD).Methods Totally 247-month-old male SAMP8 mice were randomly divided into model group and electroacupuncture group,with 12 mice in each group,and the 12 male SAMR1 mice with the same age were set as the control group."Baihui","Dazhui"and"Shenshu"were selected in the electroacupuncture group,once a day,8 days as one course of treatment,with an interval of 2 days,for a total of 3 courses.The control group and the model group were not intervened.The motor function of mice was tested by grip strength test,pole climbing test and open field test,the mRNA expressions of Irisin,Decorin and Myostatin in quadriceps muscle were detected by RT-qPCR,and the protein expressions of Irisin,Decorin and Myostatin in quadriceps muscle were detected by immunohistochemistry and Western blot.Results Compared with the control group,the grip peak and duration of the mice in the model group decreased,head turning time and pole climbing time were prolonged(P<0.01),the mRNA and protein expressions of Irisin and Decorin decreased(P<0.05,P<0.01),and the expressions of Myostatin mRNA and protein increased(P<0.05).Compared with the model group,the grip peak and duration of the mice in the electroacupuncture group increased,head turning time and pole climbing time were decreased(P<0.05),the mRNA and protein expressions of Irisin and Decorin increased(P<0.05),and the expressions of Myostatin mRNA and protein decreased(P<0.05).Conclusion Electroacupuncture can improve the motor dysfunction of SAMP8 mice,and its mechanism may be related to regulating the expressions of Irisin,Decorin and Myostatin in skeletal muscle.
		                        		
		                        		
		                        		
		                        	
3.Establishment and characterization of bone marrow mesenchymal stem cell lines stably synthesizing high-level dopamine.

		                        			A triple-transgenic (tyrosine hydroxylase/dopamine decarboxylase/GTP cyclohydrolase 1, TH/DDC/GCH1) bone marrow mesenchymal stem cell line (BMSCs) capable of stably synthesizing dopamine (DA) transmitters were established to provide experimental evidence for the clinical treatment of Parkinson's disease (PD) by using this cell line. The DA-BMSCs cell line that could stably synthesize and secrete DA transmitters was established by using the triple transgenic recombinant lentivirus. The triple transgenes (TH/DDC/GCH1) expression in DA-BMSCs was detected using reverse transcription-polymerase chain reaction (RT-PCR), Western blotting, and immunofluorescence. Moreover, the secretion of DA was tested by enzyme-linked immunosorbent assay (ELISA) and high-performance liquid chromatography (HPLC). Chromosome G-banding analysis was used to detect the genetic stability of DA-BMSCs. Subsequently, the DA-BMSCs were stereotactically transplanted into the right medial forebrain bundle (MFB) of Parkinson's rat models to detect their survival and differentiation in the intracerebral microenvironment of PD rats. Apomorphine (APO)-induced rotation test was used to detect the improvement of motor dysfunction in PD rat models with cell transplantation. The TH, DDC and GCH1 were expressed stably and efficiently in the DA-BMSCs cell line, but not expressed in the normal rat BMSCs. The concentration of DA in the cell culture supernatant of the triple transgenic group (DA-BMSCs) and the LV-TH group was extremely significantly higher than that of the standard BMSCs control group (P < 0.000 1). After passage, DA-BMSCs stably produced DA. Karyotype G-banding analysis showed that the vast majority of DA-BMSCs maintained normal diploid karyotypes (94.5%). Moreover, after 4 weeks of transplantation into the brain of PD rats, DA-BMSCs significantly improved the movement disorder of PD rat models, survived in a large amount in the brain microenvironment, differentiated into TH-positive and GFAP-positive cells, and upregulated the DA level in the injured area of the brain. The triple-transgenic DA-BMSCs cell line that stably produced DA, survived in large numbers, and differentiated in the rat brain was successfully established, laying a foundation for the treatment of PD using engineered culture and transplantation of DA-BMSCs.

4.Effect of home care based on personalized multimedia technology guidance in ophthalmology nursing management

		                        			Objective:To explore the application effect of home care based on personalized multimedia technical guidance in ophthalmology nursing management.Methods:A total of 120 patients with eye diseases in the First Affiliated Hospital of Zhengzhou University from January 2020 to December 2021 were selected as research objects by the convenient sampling method. Patients were numbered according to the order of inclusion and were divided into the observation group and the control group with 60 cases each for home nursing by random number table method. The control group was guided by conventional health education mode, and the observation group was guided by personalized multimedia technology. The Exercise of Self-Care Agency Scale (ESCA) score, Positive and Negative Affect Scale (PANAS) score, Low Vision Quality of Life Questionnaire (LVQOL) score were compared between the two groups before and after the intervention.Results:During intervention, 2 cases (3.33%) were dropped out in the observation group and 4 cases (6.67%) in the control group. After intervention, the total score of ESCA of the two groups were higher than before intervention, and the total score of the observation group was higher than that of the control group, and the differences were statistically significant ( P<0.05) . Positive emotion score of PANAS in both groups was higher than before intervention, negative emotion score was lower than before intervention, and positive emotion score of observation group was higher than control group, negative emotion score was lower than control group, and the differences were statistically significant ( P<0.05) . The total score of LVQOL and scores of each dimension of the two groups were higher than before intervention, and the observation group were higher than the control group, and the differences were statistically significant ( P<0.05) . Conclusions:The application of personalized multimedia technology in home nursing management of ophthalmic disease patients is conducive to improving their self-care ability, improving their psychological state, and improving the quality of life.
		                        		
		                        		
		                        		
		                        	
5.Analysis of brachial-ankle pulse wave velocity in 1 607 adolescents

		                        			Objective:To explore the distribution characteristics of brachial-ankle pulse wave velocity (BaPWV) in adolescents.Methods:Cross sectional survey research.A total of 1 607 adolescents aged 12-17 years from 5 middle schools in the Haidian District, Beijing from June 2020 to May 2021 were enrolled for measuring their blood pressure, weight, height and the body mass index (BMI), including 782 males and 825 females.BaPWV was measured using an arteriosclerosis detector.They were divided by the age, sex and obesity.Differences of measurement data among multiple groups were compared by one-way ANOVA, followed by the LSD test, and those between groups were compared by the two-sample t-test.The correlation between BaPWV and other indexes was assessed by the Pearson correlation analysis. Results:The median level of BaPWV in adolescents aged 12-17 years increased with age, which was 982.50 cm/s in adolescents aged 12 years, and maximum 1 113.50 cm/s those aged 17 years.BaPWV in adolescents aged 12-17 years was positively correlated with age ( r=0.936, P=0.006). Subgrouped by the sex, the median BaPWV of male and female adolescents aged 12 years was 962.25 cm/s and 997.50 cm/s, respectively, which reached 1 122.50 cm/s and 1 096.00 cm/s in those aged 17 years, respectively.BaPWV of male and female adolescents was positively correlated with the age ( r=0.903, P=0.014; r=0.945, P=0.004). In male adolescents, BaPWV was positively correlated the systolic and diastolic blood pressure, age, weight, height, BMI( r=0.308, P<0.001; r=0.289, P<0.001; r=0.478, P<0.001; r=0.190, P<0.001; r=0.315, P<0.001; r=0.109, P=0.002). In female adolescents, BaPWV also was positively correlated the systolic and diastolic blood pressure, age, weight, height, BMI( r=0.340, P<0.001; r=0.285, P<0.001; r=0.379, P<0.001; r=0.115, P=0.001; r=0.170, P=0.001; r=0.097, P=0.014). In the overall population, BaPWV was statistically significant between the obese and normal groups ( t=-3.428, P=0.001). No significant difference in BaPWV between male and female adolescents aged 12-13 years was identified ( t=0.123, P=0.902 ), but it was significantly lower in female adolescents aged 14-15 years and 16-17 years than those of age-matched males( t=2.315, P=0.021; t=2.152, P=0.032). Conclusions:The median BaPWV level in adolescents aged 12-17 years, increases with the age.Systolic blood pressure, diastolic blood pressure, body weight, BMI, age and height are positively correlated with BaPWV in adolescents.Obesity can increase the stiffness of blood vessels, which is influenced by the sex.
		                        		
		                        		
		                        		
		                        	
6.Effect of calmodulin and its mutants on binding to Na1.2 IQ.

		                        			OBJECTIVE:
		                        			To investigate the effect of calmodulin (CaM) and its mutants on binding to voltage-gated Na channel isoleucine-glutamine domain (Na1.2 IQ).
		                        		
		                        			METHODS:
		                        			The cDNA of Na1.2 IQ was constructed by PCR technique, CaM mutants CaM, CaM and CaM were constructed with Quickchange site-directed mutagenesis kit (QIAGEN). The binding of Na1.2 IQ to CaM and CaM mutants under calcium and calcium free conditions were detected by pull-down assay.
		                        		
		                        			RESULTS:
		                        			Na1.2 IQ and CaM were bound to each other at different calcium concentrations, while GST alone did not bind to CaM. The binding affinity of CaM and Na1.2 IQ at [Ca]-free was greater than that at 100 nmol/L [Ca] ( < 0.05). In the absence of calcium, the binding amount of CaM wild-type to Na1.2 IQ was greater than that of its mutant, and the binding affinity of CaM to Na1.2 IQ was the weakest among the three mutants ( < 0.05).
		                        		
		                        			CONCLUSIONS
		                        			The binding ability of CaM and CaM mutants to Na1.2 IQ is Ca-dependent. This study has revealed a new mechanism of Na1.2 regulated by CaM, which would be useful for the study of ion channel related diseases.

7.Establishment of predictive model for blood pressure elevation in outpatients with hypertension

		                        			Objective To establish the regression equation of blood pressure in population based on the pulse wave transit time (PWTT) and verify its accuracy. Methods A convenient sampling method was used to collect gender, age, arm circumference, arm length, PWTT, history of hypertension, and body height, etc. of 4 121 outpatients' information from the Hainan Branch of Chinese PLA General Hospital from June 2016 to May 2018 to establish a binary variable logistic regression equation for blood pressure elevation or not and screen out the influencing factors of time point blood pressure. The accuracy of the equation was then verified in 252 outpatients. Results Logistic regression analysis showed that PWTT, gender, age, history of hypertension present or not, body height might be the influencing factors of blood pressure elevation [odds ratio (OR) values were 0.995, 0.530, 0.980, 107.128, 0.979, 95% confidence interval (95% CI) were 0.991-0.999, 0.405-0.695, 0.971-0.989, 73.935-155.223, and 0.962-0.996, respectively, all P < 0.05]. The classification prediction equation: Ln [P / (1-P)] = 2.087-0.005×PWTT-0.635×gender (man = 1, woman = 2)-0.02×age + 4.674×hypertension history present or not-0.021×body height [P indicates the probability of a positive result: systolic blood pressure≥140 mmHg (1 mmHg = 0.133 kPa) and/or diastolic blood pressure≥90 mmHg]. The overall test results showed that χ2 = 1 835.305, P < 0.05, statistically significant; The goodness of fit test results:χ2 = 5.881, P > 0.05, the data was conform to the equal proportion distribution. Model-confirmed results showed that in patients with a history of hypertension, the probability of predicting accuracy was 95%-99%. In patients without a history of hypertension, the probability of predicting accuracy was 45%-89%. Conclusion The predictive value of blood pressure elevation with this model in patients with a history of hypertension is superior to that of patients without the history of hypertension.
		                        		
		                        		
		                        		
		                        	
8.Effects of intervention based on WeChat on blood pressure control in patients with hypertension:Meta-analysis

		                        			To evaluate the effects of the intervention based on WeChat on blood pressure control in patients with essential hypertension. 
		                        		
		                        			Methods:
		                        			The databases of CNKI,Wanfang,VIP,PubMed and the Cochrane Library were researched for the literatures about effects of the intervention based on WeChat on blood pressure control in patients with essential hypertension published from 2011 to 2018. These papers were retrieved and evaluated according to inclusion and exclusion criteria. Stata 12.0 software was used for meta analysis.
		                        		
		                        			Results:
		                        			Eleven out of 880 papers were included into meta analysis. There were 1 174 cases,with 587 cases in the experimental group and 587 cases in the control group. The results of the meta analysis showed that compared with traditional health intervention method,the intervention based on WeChat significantly reduced the level of systolic blood pressure(WMD=14.77,95%CI:11.76-17.77)and diastolic blood pressure(WMD=8.17,95%CI:5.67-10.67)of hypertensive patients. Subgroup analysis showed that the longer the intervention,the better the effects(P<0.05). 
		                        		
		                        			Conclusion

10.Correlation between autophagy and polarization of macrophages in atherosclerosis plaque in arteriosclerosis obliterans amputees.

		                        			This study was designed to investigate the correlation between autophagy and polarization of macrophages in atherosclerosis (AS) plaque in arteriosclerosis obliterans amputees. Femoral artery specimens from arteriosclerosis obliterans amputees were performed hematoxylin and eosin (HE) staining, oil red O and immunofluorescence staining to observe the morphology of atherosclerotic plaque, phenotype of macrophages and autophagy in plaque; using real-time quantitative RT-PCR technology to detect the mRNA level of M1 and M2 type markers in arterial tissue; to analyze polarized signal pathway and autophagy protein levels in macrophages by Western blotting. Arterial specimens staining showed obvious lipid deposition and obvious infiltration of amount of foam cells and inflammatory cells. Macrophages were mainly expression M1 type in percentage in fibrous plaque. Although both M1 and M2 macrophages were upregulated in atheromatous plaque, the increase was dominant in M2 type in percentage. The level of autophagy was significantly higher in the atheromatous plaque than that of fibrous plaque. The expression of tumor necrosis factor- α (TNF-α), monocyte chemotactic protein-1 (MCP-1), inducible nitric oxide synthase (iNOS), interleukin-6 (IL-6) and interleukin-12 (IL-12) mRNA was significantly higher in fibrous plaque than that of atheromatous plaque (P < 0.01 or 0.05), and arginase-1 (Arg-1), transforming growth factor-β (TGF-β), CD163 and interleukin-10 (IL-10) mRNA was significantly lower than that in atheromatous plaque (P < 0.01). The levels of p-STAT1 and NF-κB were significantly increased in fibrous plaque (P < 0.01), while p-STAT6 expression was significantly increased in atheromatous plaque (P < 0.01). The level of LC3-II was significantly higher in atheromatous plaque than that in fibrous plaque (P < 0.01). Macrophages in early atherosclerotic plaque were induced to M1 type through p-STAT1/NF-κB pathway and expressed moderate levels of autophagy; while macrophages in advanced plaques were induced to polarization of M2 type through p-STAT6 pathway.
