1.Cordycepin Inhibits Fat Infiltration after Rotator Cuff Tear Injury by Regulating Wnt/β-catenin Signaling Pathway

ObjectiveTo investigate the effect and mechanism of cordycepin in inhibiting fat infiltration after rotator cuff injuries in rats by regulating the Wnt/β-catenin signaling pathway, providing a theoretical basis for clinical treatment of rotator cuff injuries. MethodsFifty SPF-grade female SD rats were used in this study, with 10 randomly selected as the blank group. A rotator cuff injury repair model was established by supraspinatus tendon and suprascapular nerve compression. The successfully modeled rats were randomized into model and low-dose (20 mg·kg-1), medium-dose (40 mg·kg-1), and high-dose (80 mg·kg-1) cordycepin groups. After 6 weeks of treatment, the gait analysis was performed to assess the limb function in rats. Oil red O staining and Masson staining were employed to observe pathological changes in the muscle tissue. Enzyme-linked immunosorbent assay (ELISA) was used to measure the levels of interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α) in the serum. Immunohistochemistry (IHC) was employed to detect the expression of peroxisome proliferator-activated receptor γ (PPARγ) and CCAAT/enhancer-binding protein α (C/EBPα), which are markers of adipogenesis. Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and Western blot were employed to determine the mRNA and protein levels, respectively, of Wnt3a, Wnt10b, and β-catenin. ResultsCompared with the blank group, the model group showed decreases in stride length and paw print area (P<0.01), an increase in ratio of wet muscle mass reduction and a decrease in muscle fiber cross-sectional area (P<0.05), and decreased ratios of fat infiltration area and collagen fiber area (P<0.01). Additionally, the model group showed elevated levels of IL-1β, IL-6, and TNF-α (P<0.05), up-regulated protein levels of PPARγ and C/EBPα (P<0.01), and down-regulated mRNA and protein levels of Wnt3a, Wnt10b, and β-catenin (P<0.05, P<0.01). Compared with the model group, the low-, medium-, and high-dose cordycepin groups showed increases in stride length and paw print area (P<0.01), a decrease in ratio of wet muscle mass reduction and an increase in muscle fiber cross-sectional area (P<0.05), and increases in ratios of fat infiltration area and collagen fiber area (P<0.05, P<0.01). In addition, cordycepin lowered the serum levels of IL-1β, IL-6, and TNF-α (P<0.05, P<0.01), down-regulated the protein levels of PPARγ and C/EBPα (P<0.01), and up-regulated the mRNA and protein levels of Wnt3a, Wnt10b, and β-catenin (P<0.05, P<0.01). ConclusionCordycepin can improve the limb function, alleviate rotator cuff muscle atrophy, fat infiltration, and fibrosis, and inhibit inflammation in rats by regulating the Wnt/β-catenin signaling pathway.
2.Effects of subclinical hypothyroidism during pregnancy treated with LT 4 on the growth and neuropsychology of offspring aged 0-36 months: A cohort study

Objective:To evaluate the effects of subclinical hypothyroidism during pregnancy treated with LT 4 on the growth and neuropsychology of offspring aged 0-36 months. Methods:A maternal-infant cohort was established in healthy singleton pregnant women aged 20-45 years without history of thyroid disease. Women developing subclinical hypothyroidism during pregnancy were treated with LT4. The weight, length, and head circumference of the offspring were recorded between 0 to 36 months after birth. Meanwhile, infant nutrition and family support were investigated. The Neuropsychological Development Questionnaire of 0-6 year old children was used to evaluate the neurodevelopment of offspring.Results:A total of 186 mother-infant pairs were included. All subjects were divided into the euthyroidism(ETH) group( n=136) and subclinical hypothyroidism(SHT) group( n=50) according to maternal thyroid function during pregnancy. The Z-scores(adjusted by months of age and gender) of weight, length, weight/length at birth, weight/length at 1 month, head circumference at 6 months, length at 8 months, weight/length at 24 months of SHT group were lower than those of the ETH group( P<0.05). Furthermore, the language competence of the SHT offspring at 12 months of age was also lower than that of the ETH group( P<0.05). Maternal subclinical hypothyroidism treated with LT 4 did not significantly affect preterm delivery, low birth weight, and developmental quotient <85, but reduced the risk of macrosomia(AOR 0.206, 95% CI 0.046-0.929, P=0.040). Conclusion:Although women with subclinical hypothyroidism received LT 4 treatment during pregnancy, the offspring still may suffer adverse effects on their growth and neural development.
3.Dynamics of parenting styles of adolescent students from the perspective of intergenerational conflict

Objective:
To explore dynamics of parenting styles of adolescents from 1999 to 2019 from the perspective of intergenerational conflict, to provide support for family education and adolescent healthy development.
Methods:
Using a multistage stratified cluster random sampling method, the unified questionnaire was administered to 2 590 students in the same sampling junior and senior high schools in 1999, 2009, and 2019 using the Egna Minnen av Barndoms Uppfostran own memories of parental rearing practices in childhood(EMBU).
Results:
Overall there were differences in the nine factors of parenting styles across generations ( F = 12.07-72.52, P <0.01), with decreasing ratings of warmth and understanding of father and mother (F1, M1), over interference of father (F3) over generations(F1:46.72±9.41, 45.87±11.33, 43.61±11.27; M1:51.56±9.38, 51.03±11.59, 46.23± 12.27 ; F3:19.03±4.00, 18.29±4.32, 17.95±4.51), and all other parenting styles rated higher in 2019 than in 2009 and 1999(except for the over protection and over interference of mother, and punishment, firm control of mother). Parenting styles across generations (except for the rejection and denial of father among girls) showed gender difference.The overall gender trend coincided with the total population trend. Parenting styles across generations varied significantly among middle and high school students( F =3.92-47.27, P <0.05 ), changes in F1 and F3 factors coincided with the overall decreasing trend. Factor analysis showed that parenting styles could be classified into two dimensions, with varied factor loading across generation.
Conclusion
Intergenerational decreases in parental emotional warmth and paternal interfering are observed in a sex and grade specific manner. Based on the diversity of needs and population differentiation, optimal intervention for comprehensive health development of adolescents are in great need to keep pace with the times and promoting the high quality development of adolescents.
4.Research progress of non-alcoholic fatty liver disease in postmenopausal women

Non-alcoholic fatty liver disease (NAFLD) is a common chronic liver disease with a risk of developing non-alcoholic hepatitis, cirrhosis and liver cancer. Postmenopausal women have an increased risk of developing metabolic diseases including NAFLD under the influence of various factors such as aging, endocrine and metabolic changes. This article summarizes the latest research of NAFLD in postmenopausal women to elaborate and analyze the epidemiological characteristics, correlation between NAFLD and postmenopause, possible pathogenesis as well as feasible prevention and treatment methods.
5.Comparison of quality of life in patients with advanced Siewert type Ⅱ adenocarcinoma of the esophagogastric junction treated by three different operations

Objective To compare the quality of life (QOL) of advanced Siewert type Ⅱ adenocarcinoma esophagogastric junction (AEG) patients treated by the total gastrectomy, traditional proximal gastrectomy or proximal gastrectomy with gastric tube reconstruction opertations, and to provide some clinical basis for the choice of surgical methods for AEG. Methods A total of 90 patients with Siewert type Ⅱ AEG were retrospectively collected from the Affiliated Hospital of North Sichuan Medical College. Patients were divided into 3 groups according to different surgical methods (n=30 in each group), a total gastrectomy group (23 males, 7 females, aged 47-79 years), a traditional proximal gastrectomy group (treated with the traditional proximal gastrectomy procedure, 19 males, 11 females, aged 44-80 years), and a narrow gastric tube group (treated with the proximal gastrectomy with gastric tube reconstruction procedure, 25 males, 5 females, aged 47-83 years). The Chinese version of Quality of Life Questionnaire Core-30 (QLQ-C30) and Quality of Life Questionnaire Oesophagogastric-25 (QLQ-OG25) designed by European Organization for Research and Treatment of Cancer (EORTC) were used to collect the patients’ information in the three groups about their QOL during the first six months and one year after the three procedures. Results There was no statistical difference in the clinical data among the three groups (all P>0.05). QOL during the first six months after the operations assessed by the QLQ-C30 questionnaire table showed that the narrow gastric tube group was significantly best in total QOL, physical function, fatigue, and emotional function among the three groups (all P<0.05). The total gastrectomy group was the worst in role function, dyspnea, fatigue and diarrhea among the three groups (all P<0.05). The traditional proximal gastrectomy group had a worse evaluation in lose of appetite than the other two groups (P<0.05). QOL during the first six months after the operations assessed by the QLQ-OG25 questionnaire table showed that the traditional proximal gastrectomy group had a significantly worse evaluation in palirrhea than the other two groups (both P<0.05). The total gastrectomy group had a significantly worse evaluation in anxiety than the other two groups (both P<0.05). QOL during the first year after the operations assessed by the QLQ-C30 questionnaire table showed that the narrow gastric tube group had a significantly highest evaluation in total QOL physical function and emotional function among the three groups (all P<0.05). The total gastrectomy group had a significantly worst evaluation in diarrhea among the three groups (P<0.05). QOL during the first year after operations assessed by QLQ-OG25 questionnaire table showed that the traditional proximal gastrectomy group had a significantly worse evaluation in palirrhea than the other two groups (all P<0.05). Conclusion The narrow tubular esophagogastric anastomosis is better than the total gastrectomy and the traditional proximal gastrectomy for the treatment of the advanced Siewert type Ⅱ adenocarcinoma of esophagogastric junction, so this operation is worth being recommended.
6.Research progress of FBLN-5 in the role of tumorigensis

Fibulin protein family has widely existed in the extracellular matrix, and plays an important role in maintain the stable of basement membrane, elastic fibers and loose connective tissue. Fibulin-5 (FBLN-5) is one of the members of the Fibulin protein family, which is essential to the structure function and cell mediated action. The lack of FBLN-5 will cause many diseases such as internal organ deformity. Current research suggests that changes of expression level of FBLN-5 are associated with the occurrence of tumors such as lung cancer, and also can influence the tumor proliferation, invasion and prognosis, thus may become a new molecular marker for cancer diagnosis. Therefore, in this review, we summarize and update biological characteristics and functions of FBLN-5, and its role in the development of tumor formation mechanism.
7.Systemic inflammatory response syndrome after percutaneous nephrolithotomy: an assessment of risk factors

Objective To identify risk factors of systemic inflammatory response syndrome (SIRS) after percutaneous nephrolithotomy (PCNL).Methods We retrospectively reviewed 438 renal calculi patients after PCNL from August 2015 to July 2016.Among them,there were 251 men and 187 women,the mean age was (49.4 ± 11.1) years.The positive preoperative urine WBC,culture and nitrite rates were 29.7% (130),12.1% (53) and 15.1% (66),respectively.The stone size was (851.2 ± 663.6) mm2,the stone CT value was (960.4 ± 303.4) HU,the operative time was (63.5 ± 33.4) min,124 (28.3 %) were infection stones and multiple-tracts PCNL was performed in 69 (15.8%) patients.Univariate and multivariate logistic regression analysis were used to analyze perioperative predictors after PCNL.Results Thirty-nine patients developed SIRS (8.9%) after PCNL.The univariate analysis showed that positive preoperative urine WBC,nitrite,culture,operation time,stone size and transfusion had significantly impacts on the outcome of postoperative SIRS after PCNL (P < 0.05).Multivariable logistic analysis showed that positive preoperative urine nitrite (OR =5.990,P < 0.001),stone size (OR =2.251,P =0.027) and transfusion (OR =7.501,P =0.007) were independently related to the postoperative SIRS.Conclusion The positive preoperative urine nitrite,stone size and transfusion are independent risk factors for postoperative SIRS after PCNL.
8.Recognition of scientific literature novelty assessment in medical and health staff of a military area com-mand and their information need

Objective To provide reference for effective scientific literature novelty assessment by investigating its recognition in medical and health staff of a military area command and their use of information resources and need of information .Methods Knowledge of scientific literature novelty assessment , satisfaction of scientific literature novelty assessment report, need and use of information resources were investigated in 149 staff of 9 hospitals with self-de-signed questionnaire .Results The general recognition and satisfaction levels of scientific literature novelty assess-ment were good in medical and health staff of a military area command .However , the information resources were insufficiently used , which was related with the scientific research level and scientific literature novelty assessment popularization .Conclusion Scientific literature novelty assessment should be popularized , staff engaged in scientific literature novelty assessment should be trained, sharing of information resources should be strengthened , and infor-mation service should be expanded and deepened in military area commands.
9.Effect of rice oil on secretory immunoglobulin A content in feces and disease outcome in children with acute diarrhea

Objective To study the effect of rice oil on secretory immunoglobulin A (sIgA) content in feces and disease outcome in children with acute diarrhea.Methods We selected 180 children with acute diarrhea hospitalized in Tianjin Children's Hospital during the period from January 2013 to October 2014.They were randomly divided with a random number table into rice oil group,montmorillonite powder group and control group (all n =60).The effect of treatment,duration before subsiding of diarrhea and vomit,length of hospital stay and sIgA content in feces before and after treatment were analyzed.Results The total effective rates in the rice oil group and the montmorillonite powder group (93.3%,96.7%) were significantly higher than that in the control group (76.7%,P =0.011,P =0.001).Length of hospital stay,duration before subsiding of diarrhea and vomit in the rice oil group and the montmorillonite powder group were significantly shorter than those in the control group[(5.6±1.4) d,(5.7±1.6) d vs.(6.9±1.8) d;(3.7±0.9) d,(3.5±0.9) d vs.(5.4±1.2) d;(2.8 ± 0.6) d,(3.2 ± 0.8) d vs.(5.1 ± 0.8) d;all P =0.000].The total effective rates,length of hospital stay,duration before subsiding of diarrhea showed no statistically significant differences between the rice oil group and the montmorillonite powder group (P =0.402,0.716,0.226),but the duration before subsiding of vomit in the rice oil group was significantly shorter than that in the montmorillonite powder group (P =0.016).Before treatment,the sIgA contents in feces in the control group,the montmorillonite powder group,the rice oil group were 0.527 ± 0.133,0.487 ± 0.109,0.534 ± 0.150,with no significant difference (P > 0.05).Two days after treatment,sIgA contents in the rice oil group and the montmorillonite powder group (0.669 ±0.176,0.612 ± 0.161) were significantly higher than in the control group (0.541 ±0.149,P =0.000,0.014).There was no significant difference between the montmorillonite powder group and the rice oil group (P =0.067).Four days after treatment,sIgA content in the montmorillonite powder group (1.981 ±0.462) was significantly higher than in the control group (1.762 ±0.378,P =0.005),while sIgA content in the rice oil group (2.331 ± 0.494) was significantly higher than in the other 2 groups (P =0.000).Compared with the levels before treatment,sIgA content 2 days after treatment was not significantly changed in the control group (P =0.295),but was significantly elevated in the montmorillonite powder group and the rice oil group (both P =0.000).sIgA contents of all the three groups 4 days after treatment were significantly higher than the contents 2 days after treatment (all P =0.000).Conclusion The effect of rice oil on acute diarrhea in children is as same as that of montmorillonite powder,and may be better in mitigating vomit and promoting the secretion of intestinal sIgA.
10.RNAi-silenced Survivin and hTERT gene inhibits proliferation and induces apoptosis of human colorectal carcinoma cell line SW 480

Objective To investigate the Influence of Survivin and hTERT gene on cell proliferation and apoptosis in human colorectal carcinoma cell line SW 480 and to find experiment evidence for gene therapy of colorectal carci-noma .Methods Plasmids carrying shRNAs targeting survivin and hTERT were designed , constructed and trans-fected into SW480 cells.SW480 cells were then divided into blank group , blank Plasmid control group , survivin RNAi group , hTERT RNAi group and Survivin-hTERT RNAi group .The telomerase activity was examined by TRAP-PCR-ELISA analysis 48h after hTERT-shRNA transfection.Survivin and hTERT mRNA and protein expres-sion was analyzed by RT-PCR and Western blot .Cell apoptosis , proliferation were measured by flow cytometry , CCK-8 assay.Results Telomerase activity of SW480 cells in Survivin-hTERT RNAi groups were significantly decreased compared with the blank group ( P<0.01 ) .The expression of survivin and hTERT mRNA, proteins in the Survivin-hTERT RNAi group was reduced by 82.8%and 73.6%( P<0.01 ) ,79.2%and 66.7%( P<0.01 ) respectively .The inhibitory rate of cell proliferation of Survivin-hTERT RNAi group was 43.6% ±0.1%( P <0.01 ) .The apoptosis rate was 39.2%±2.3%( P<0.01 ) in the Survivin-hTERT RNAi group .Conclusions The Survivin-hTERT RNAi group could significantly reduces the protein expression of survivin and hTERT mRNA, in-hibit cell proliferation and induces cell apoptosis in human colorectal carcinoma cell line SW 480 .
