1.Improvement of quality control methods and “quality evaluation via color discrimination”of Hypericum perforatum

OBJECTIVE To provide a reference for the quality control of Hypericum perforatum. METHODS High- performance liquid chromatography (HPLC) was used to establish fingerprints for 20 batches of H. perforatum and determine the contents of its main components: chlorogenic acid, rutin, hyperin, isoquercitrin, avicularin, quercitrin and quercetin. Cluster analysis was conducted using SPSS 26.0 software. The chromaticity values (luminance value L*, red-green value a*, and yellow- blue value b*) of H. perforatum powder were measured using electronic eye. A prediction model for the contents of seven components in H. perforatum based on its appearance chromaticity values was established using machine learning algorithms. The predictive performance of the models was evaluated using root-mean-square-error (RMSE). RESULTS A total of 16 common peaks were calibrated in the fingerprints of 20 batches of H. perforatum, and 9 peaks were identified, which were chlorogenic acid, rutin, hyperin, isoquercitrin, avicularin, quercitrin, quercetin, hypericin and hyperforin; the similarities of the 20 batches of samples and reference fingerprint ranged from 0.889-0.987. The contents of chlorogenic acid, rutin, hyperin, isoquercitrin, avicularin, quercitrin and quercetin were 0.025%-0.166%, 0.048%-0.339%, 0.082%-0.419%, 0.017%-0.209%, 0.011%-0.134%, 0.020%-0.135%, 0.041%-0.235%, respectively. Cluster analysis results showed that 18 batches of qualified H. perforatum were grouped into three categories, when the Euclidean distance was set to 1.4. L* of the 20 batches of H. perforatum ranged from 62.814 to 75.668, a* ranged from 1.409 to 3.490, and b* ranged from 25.249 to 30.759. RMSE of three prediction models, namely XGBoost, LightGBM, and AdaBoost, ranged from 0.008 to 0.070, indicating good fitting performance. XGBoost model predicted the contents of the other six components with high accuracy, except for rutin. CONCLUSIONS The established fingerprints and content determination methods are accurate, reproducible, and reliable. The content prediction model based on appearance chromaticity values, combined with machine learning algorithms, can be used for the quality control of H. perforatum.
2.Comparison on Quality of Honey Processed Lilii Bulbus Prepared by Traditional Empirical and Pharmacopoeial Methods Based on Chemical Analysis and Sensory Techniques

ObjectiveTo compare the quality of two kinds of honey-processed Lilii Bulbus(HPLB) prepared by empirical and pharmacopoeial methods based on chemical analysis and intelligent sensory technology, and to improve and upgrade the quality standard of HPLB on the basis of excavating and inheriting the traditional experience of famous experts in traditional Chinese medicine(TCM). MethodSamples of HPLB were prepared according to the method in the 2020 edition of Chinese Pharmacopoeia(honey was added before frying) and the empirical method of the old pharmacists(honey was added after frying), and the appearance characteristics, thin layer chromatography(TLC) identification, inspection, leachables and other items of the two kinds of samples were detected according to the methods under the Lilii Bulbus in the 2020 edition of Chinese Pharmacopoeia. The fingerprints of HPLB with different honey addition methods were established by high performance liquid chromatography(HPLC), and the contents of regaloside A, regaloside B and 5-hydroxymethylfurfural(5-HMF) were determined, the quality difference between the two HPLB samples was evaluated by multivariate statistical analysis. Meanwhile, electronic nose and electronic tongue were used to measure the sense of smell and taste, and combined with multivariate statistical analysis, the difference sensors were screened with the variable importance in the projection(VIP) value>1 in order to compare the differences between the two kinds of HPLB. ResultCompared with HPLB prepared by the pharmacopoeial method, the sample prepared by the empirical method had lower water content, slightly higher soluble extract content, better appearance and easy storage. HPLC fingerprints of two kinds of HPLB samples were established, and the similarity between them was≥0.999, indicating that the composition of two species was similar. And 29 common peaks were calibrated, among which peaks 8 and 18 were regaloside A and regaloside B. The quantitative analysis of the index components in the two kinds of HPLB samples showed that the difference in the contents of regaloside A and regaloside B was not statistically significant, whereas the content of 5-HMF in empirical HPLB was significantly higher than that of the pharmacopoeial HPLB(P<0.01). The electronic tongue and electronic nose test results showed that the umami, umami richness and saltiness of the empirical HPLB were higher than those of the pharmacopoeial HPLB. The results of electronic nose detection showed that the four sensors W1W, W2S, W5S and W1S were the differential odor sensors of the two samples, among which the response values of W1S and W2S sensors were higher than those of the pharmacopoeial HPLB, which might be related to the content of 5-HMF. ConclusionCompared with the pharmacopoeial HPLB, the empirical sample is of better quality, but the two can be clearly distinguished by electronic nose and electronic tongue, which can provide a reference for excavating and inheriting the traditional experience of famous experts in TCM and further improving the processing methods and quality standards of HPLB.
3.Effect of oridonin on cell proliferation,migration,and apoptosis of human nasopharynx carcinoma HONE-1 cells

Objective:To discuss the effect of oridonin on the proliferation,migration,epithelial-mesenchymal transition(EMT),and apoptosis of the human nasopharyngeal carcinoma HONE-1 cells,and to clarify its related antitumor mechanism.Methods:The HONE-1 cells were treated with different concentrations(0,5,10,20,40,80,and 160 mg·L-1)of oridonin for 48 h.CCK-8 method was used to detect the inhibitory rates of proliferation of the cells in various groups and the drug concentration for subsequent experiment was confirmed.The HONE-1 cells were divided into control group,3 mg·L-1 oridonin group,and 6 mg·L-1 oridonin group.After 24 and 48 h of culture,CCK-8 method was used to detect the proliferation activities of the cells in various groups;5-ethynyl-2'-deoxyuridine(EdU)method was used to detect the rates of EdU-positive cells in various groups;colony formation assay was used to detect the numbers of clone formation in the cells in various groups;Transwell chamber experiment and cell wound assay were used to detect the numbers of migration cells and the scratch healing rates of the cells in various groups;real-time fluorescence quantitative PCR(RT-qPCR)method was used to detect the expression levels of cyclin-dependent kinase 1(CDK1)and cyclin-dependent kinase 4(CDK4)mRNA in the cells in various groups;Western blotting method was used to detect the expression levels of E-cadherin,Vimentin,Caspase-3,and poly ADP-ribose polymerase 1(PARP1)proteins in the cells in various groups.Results:The CCK-8 method results showed that the half-maximal inhibitory concentration(IC50)of oridonin at 48 h was 12.18 mg·L-1,and 1/4 IC50 and 1/2 IC50 values were used as the concentrations for subsequent experiments.Compared with control group,after treated for 24 and 48 h,the proliferation activities of the cells in 3 and 6 mg·L-1 oridonin groups were decreased(P<0.05 or P<0.01),the rate of EdU-positive cells were decreased(P<0.05 or P<0.01),the numbers of clone formation and migraton cells were decreased(P<0.05 or P<0.01),the scratch healing rates were decreased(P<0.05 or P<0.01),the expression levels of CDK1 and CDK4 mRNA in the cells were decreased(P<0.05 or P<0.01),the expression levels of E-cadherin,Caspase-3,and PARP1 proteins were increased(P<0.05 or P<0.01),and the expression levels of Vimentin protein were decreased(P<0.05).Conclusion:Oridonin can inhibit the proliferation,clone formation,and migration of the human nasopharyngeal carcinoma HONE-1 cells by downregulating the expression of cell cycle-related proteins and EMT,and promote the apoptosis to exert an antitumor effect.
4.The correlation between hemoglobin,monocyte to high density lipoprotein cholestero ratio and post-stroke cognitive impairment

Objective The correlation between Hb content,monocyte to high density lipoprotein cholesterol ratio(MHR)and post-stroke cognitive impairment(PSCI)was analyzed,and the application value of Hb and MHR in assisting diagnosis and evaluation of PSCI was discussed.Methods The clinical data of first-time stroke patients admitted to the Rehabilitation Department of the Second Affiliated Hospital of Nanjing Medical University from January 2021 to December 2022 were collected.The patients were divided into PSCI group and post-stroke no cognitive impairment(PSNCI)group by using MMSE scores.The differences in serum related indicators between the two groups were analyzed to find the risk factors of PSCI.Results There were a total of 81 stroke patients,including 43 in the PSCI group and 38 in the PSNCI group.There were no statistically significant differences in gender,age,history of hypertension,diabetes,smoking,drinking history,white blood cell count,neutrophil count,lymphocyte count,platelet count,total cholesterol,triglyceride,high density lipoprotein cholesterol and low density lipoprotein cholesterol between the two groups.The Hb content in PSCI group was significantly lower than that in PSNCI group,and the MHR was significantly higher than that in PSNCI group(P<0.05).Spearman correlation analysis showed that Hb was positively correlated with MMSE score of stroke patients(rs = 0.353,P<0.05),and MHR was negatively correlated with MMSE score of stroke patients(rs =-0.325,P<0.05).According to whether PSCI occurred as the dependent variable,the analysis showed that lower Hb content and higher MHR were independent risk factors for PSCI(OR =1.056,95%CI:1.025-1.088,P<0.05;OR =0.059,95%CI:0.006-0.577,P<0.05).The area under ROC curve of Hb in predicting PSCI in stroke patients was 0.76(95%CI:0.656-0.865).The area under ROC curve of MHR for predicting PSCI in stroke patients was 0.62(95%CI:0.497-0.742).Conclusions Hb content and MHR are significantly correlated with PSCI.The decrease of Hb content and the increase of MHR are independent risk factors for PSCI.Hb content and MHR have certain reference value for the prediction and diagnosis of PSCI.
5.Analysis of the influencing factors on the perceptions of e cigarette among adolescents in Shanghai

Objective:
To assess the status of current e cigarette perception and its influencing factors among adolescents in Shanghai, so as to provide reference for the refinement of the prevention and control measures of teenagers e cigarette use.
Methods:
From May to June 2021, a stratified random cluster sampling was used to investigate 7 456 junior high and high school students in Shanghai. Harm and benefit perception of e cigarette as well as its social environment benefits were collected.
Results:
The rate of adolescents ever and current e cigarette use was 3.19% and 1.09%, respectively. The top four risk factors for low harm perception of e cigarette were adolescent e cigarette use( OR=2.74, 95%CI =2.10-3.59), high school students ( OR=1.47, 95%CI = 1.32 - 1.64 ), family members ( OR=1.45, 95%CI =1.24-1.70) and friends ( OR=1.36, 95%CI =1.20-1.54) using e cigarette. Adolescent ecigarette use ( OR=2.77, 95%CI =1.97-3.89), high school students( OR=2.11, 95%CI =1.89-2.36), friends ( OR= 1.63, 95%CI =1.42-1.87) and family members using e cigarette( OR=1.39, 95%CI =1.18-1.65) were the top four associated factors for high benefit perception of e cigarette. And, adolescent e cigarette use ( OR=1.95, 95%CI =1.47-2.59), high school students ( OR= 1.73, 95%CI =1.55-1.93), friends ( OR=1.60, 95%CI =1.40-1.82) and pocket money≥200 yuan using e cigarette( OR= 1.29 , 95%CI =1.17-1.43) were the top four risk factors for high social environmental benefit perception of e cigarette. Moreover, perception of e cigarette harm, benefit and social environmental benefit were associated with the risk of future use of e cigarette( OR = 0.78,1.44,1.21, P <0.01).
Conclusion
Being high school students and using e cigarette by oneself, friends, and family members are the important influencing factors for adolescents e cigarette perception. Both low harm and high benefit perception of e cigarette elevate the risk of future e cigarette use among adolescents, so effective measures should be taken to promote control education about e cigarette and smoke free environment construction.

7.Cognition of e-cigarettes and its influencing factors among secondary school personnel in Shanghai

ObjectiveTo investigate the cognition of e-cigarette compositions and hazards and its influencing factors among secondary school personnel (junior, senior and vocational high school) in Shanghai, so as to provide ideas for improving the cognition level of e-cigarettes among the school personnel and tobacco control efforts on campus in the future. MethodsA multi-stage stratified cluster random sampling method was used to conduct an anonymous online questionnaire survey among 1 767 secondary school personnel in Shanghai. ResultsThe proportion of school staff who had used e-cigarettes was 3.79%. The participants who were not sure whether the e-cigarette smoke was “mainly water vapor (incorrect)” and whether the e-cigarette smoke “contained nicotine (correct)” and “contained formaldehyde (correct)” accounted for 62.71%, 54.44% and 61.91%, respectively. Among them, 68.48%, 55.46% and 50.37%, respectively, agreed that e-cigarettes would be “addictive”, “harmful” and “that second-hand smoke was harmful”. Binary logistic regression analysis showed the following risk factors for the low level of school personnel’s awareness of e-cigarette compositions: age ≥40 years (OR=1.65, 95%CI: 1.32‒2.06), working in junior high school (OR=1.37, 95%CI: 1.09‒1.73), educational attainment level as junior college or below (OR=1.67, 95%CI: 1.02‒2.74), and not having participated in tobacco control education activities (OR=1.37, 95%CI: 1.08‒1.73). Meanwhile, working in junior (OR=1.43, 95%CI: 1.12‒1.83) or senior (OR=1.44, 95%CI: 1.08‒1.92) high school, educational attainment level as junior college or below(OR=2.10, 95%CI: 1.24‒3.56), having used e-cigarettes (OR=2.98, 95%CI: 1.63‒5.42), not having participated in tobacco control education activities (OR=1.49, 95%CI: 1.16‒1.92) and low level of awareness of e-cigarette compositions (OR=4.24, 95%CI: 3.44‒5.23) were risk factors for the low level of school personnel’s awareness of e-cigarette hazards. ConclusionsThe level of e-cigarette awareness among school personnel in Shanghai is low, especially those who are older, had low educational attainment level and had used e-cigarettes. In the future, tobacco control education for school personnel should be strengthened to improve their cognition and ability of tobacco control on campus.
8.Environmental exposure to e-cigarettes among adolescents and its preventive countermeasures

In recent years, with the vigorous marketing of e-cigarette manufacturers, e-cigarettes have become a popular tobacco product for adolescents. The problem of e-cigarette environmental exposure among adolescents is also getting worse and its associated adverse impacts cannot be ignored. However, domestic research on the environmental exposure to e-cigarettes among youth is insufficient, and experience on e-cigarette regulation is also limited. This review first briefly introduced the definition and sources of e-cigarette environment exposure, then focused on the differences of e-cigarette environmental exposure among adolescents with different characteristics to identify possible influencing factors, as well as the impacts of e-cigarette environmental exposure on adolescents, and finally summarized international countermeasures to prevent e-cigarette environmental exposure in adolescents, aiming to provide directional guidance for China to conduct e-cigarette control activities among adolescents.
9.Comparison of Zhuyao and Douchi Ligustri Lucidi Fructus Based on Quality Investigation of Commercial Samples

ObjectiveTo investigate the quality of Ligustri Lucidi Fructus in the market, and the moisture, extract, determination of Zhuyao and Douchi Ligustri Lucidi Fructus were compared to increase the utilization rate of Ligustri Lucidi Fructus. MethodThe properties, moisture, total ash, alcohol-soluble extract content and thin layer chromatography (TLC) identification were determined by the methods of Ligustri Lucidi Fructus included in the 2020 edition of Chinese Pharmacopoeia, and high performance liquid chromatography (HPLC) fingerprint and determination of specnuezhenide and salidroside were established with the mobile phase of 0.2% phosphoric acid aqueous solution (A)-acetonitrile (B) (0-70 min, 92%-65%A) for gradient elution, and the detection wavelength of 220 nm at 0-14 min and 225 nm at 14-70 min. The two different characters of Ligustri Lucidi Fructus were comprehensively compared by the above indicators. ResultExcept for one batch which did not meet the requirements due to the quality of harvesting, the other 12 batches of samples all met the requirements of the 2020 edition of Chinese Pharmacopoeia, but there were two different characters. Comparing the two different characters of Ligustri Lucidi Fructus, it is found that the moisture, total ash, extract, salidroside and specnuezhenide contents of Zhuyao samples were 2.22%-5.19%, 3.91%-4.49%, 32.56%-40.95%, 0.073%-0.170% and 1.45%-4.14%, and these values of Douchi samples were 3.57%-5.61%, 3.65%-4.44%, 41.31%-46.70%, 0.041%-0.067% and 3.01%-4.20%, respectively. ConclusionThe contents of extract and specnuezhenide of Douchi Ligustri Lucidi Fructus are mostly higher than those of Zhuyao Ligustri Lucidi Fructus, while the content of salidroside is lower than that of Zhuyao samples, and there are no significant differences in moisture, TLC identification and total ash content. Based on the above research, if the main purpose is to extract salidroside, it is recommended to choose Zhuyao Ligustri Lucidi Fructus. If the main purpose is to use Ligustri Lucidi Fructus as medicine, it is recommended to choose Douchi Ligustri Lucidi Fructus.
10.Effect of urolithin B on the biological behavior of glioblastoma U251 cell

Objective :
To investigate the effect and mechanism of Urolithin B ( UB) on proliferation , migration , invasion , apoptosis and cell cycle of glioblastoma cells.
Methods :
The effects of UB on the proliferation , migration and invasion of glioblastoma U251 cells were detected by CCK⁃8 , clone formation assay , scratch healing assay , Transwell invasion assay respectively. The regulation effect of UB on cell cycle and apoptosis was detected by flow cytometry. Western blot was used to detect the effects of UB on the phosphorylation levels of downstream signaling pathway proteins ERK , AKT , p38 and JNK.
Results :
Compared with the control group , at 24 h and 48 h , the absorbance value of U251 cells was decreased by UB (P < 0. 01) in a dose dependent manner. UB reduced the percentage of cell clone formation (P < 0. 01) . The percentage of the scratch healing area was reduced by UB (P < 0. 05 or P < 0. 01) . The percentage of the number of invaded cells was reduced by UB (P < 0. 01) . UB could induce apoptosis (P < 0. 01) and cause the cells to stagnate in G2/M phase (P < 0. 01) . UB could significantly inhibit the phosphorylation of ERK (P < 0. 05 or P < 0. 01) and AKT (P < 0. 05 or P < 0. 01) .
Conclusion
UB can inhibit proliferation , migration , invasion , apoptosis and cell cycle of glioblastoma cells , and regulate the phosphorylation levels of ERK and AKT.
