1.γ-Synuclein protects colon cancer cells through autophagy regulation

AIM:To investigate the effects of γ-synuclein on autophagy and apoptosis of colon cancer cells in-duced by endoplasmic reticulum stress,as well as the protective effect on the cells.METHODS:Gene expression profile chip analysis was performed to compare the cDNA expression profiles between human colon cancer HCT116 cells with γ-synu-clein knockdown and HCT116 cells with control siRNA,and to identify potential molecules related to autophagy and apop-tosis.In colon cancer cell lines,the functional effects of γ-synuclein on autophagy and apoptosis induced by thapsigargin(TG),an endoplasmic reticulum stress-inducing agent,were systematically explored by conducting immunofluorescence staining,Western blot,CCK-8 assay,flow cytometry,and transmission electron microscopy.Western blot was used to de-tect the expression of γ-synuclein protein,autophagy-related proteins[microtubule-associated protein 1 light chain 3(LC3),beclin-1,autophagy-related protein 5(ATG5)and ATG7],and apoptosis-related proteins[poly(ADP-ribose)polymerase(PARP),pro-caspase-3,and pro-caspase-9].To further analyze the mechanism of γ-synuclein in regulating autophagy and apoptosis,extracellular signal-regulated kinase(ERK)inhibitor PD98059,ERK inhibitor SP600125 and c-Jun N-terminal kinase(JNK)activator anisomycin were applied separately to test HCT116 cells transfected with γ-synu-clein siRNA.Subsequently,autophagy proteins,apoptosis proteins,and ERK and JNK pathway-related proteins were de-tected by Western blot.RESULTS:The TG-induced autophagy of colon cancer cells mainly occurred at the early stage(0~24 h),and apoptosis mainly occurred at the late stage(36~48 h).Endoplasmic reticulum stress up-regulated the ex-pression of γ-synuclein in colon cancer cells,which was associated with enhanced autophagy.γ-Synuclein promoted au-tophagy by activating ERK and JNK pathways at the early stage(0~24 h),and inhibited apoptosis by blocking JNK path-ways at the late stage(24~48 h)to protect HCT116 cells.In our model,γ-synuclein was observed to play a critical role in the transition from endoplasmic reticulum stress-induced autophagy to apoptosis.CONCLUSION:In the context of endo-plasmic reticulum stress,γ-synuclein promotes autophagy and inhibits apoptosis by regulating ERK and JNK signaling pathways,thus protecting colon cancer cells.This provides a potential idea for anti-tumor therapy.
2.Observation on the therapeutic efficacy of Tuina plus "three-bridge" exercise for non-specific low back pain

Objective: To investigate the efficacy of Tuina (Chinese therapeutic massage) plus "three-bridge" exercise for non-specific low back pain (NSLBP). Methods: A total of 80 patients with NSLBP were randomly divided into a control group and an observation group according to the random number table method, with 40 cases in each group. The two groups received Tuina treatment, and the observation group received an additional "three-bridge" exercise. The treatment lasted 4 weeks, and a follow-up evaluation was carried out 6 months after treatment. The visual analog scale (VAS), Oswestry disability index (ODI), and the Japanese Orthopaedic Association (JOA) scores for low back pain were used to evaluate patients' conditions before treatment, after 4-week treatment, and at the 6-month follow-up. Results: The total effective rate was 92.5% and 85.0% in the observation group, respectively, after 4-week treatment and at the 6-month follow-up, significantly different from the control group (P<0.05). After 4-week treatment and at the 6-month follow-up, the VAS, ODI, and JOA scores of the two groups were improved compared with those before treatment (P<0.05), and all the indicators in the observation group were significantly better than those in the control group (P<0.05). At the 6-month follow-up, the VAS, ODI, and JOA scores in the two groups were improved compared with those before treatment (P<0.05). Conclusion: Tuina plus "three-bridge" exercise can effectively alleviate pain, improve function, and have relatively sound long-term effects in treating patients with NSLBP.
3.Research Progress of Radiotherapy Combined with Immune Checkpoint Inhibitors in Treatment of Glioblastoma

Glioma is the most common primary intracranial tumor, and most patients present as glioblastoma, with high morbidity and mortality. The prognosis of patients with glioblastoma remains poor after surgical resection combined with standardized treatment of radiotherapy and chemotherapy. In recent years, although the breakthrough of immunotherapy have been achieved in the treatment of a variety of solid tumors, the existing data show that immunotherapy is not effective in improving the survival of patients with glioblastoma. However, studies have shown that immunotherapy can have a synergistic effect with radiotherapy which can increase antigen presentation and promote the formation of pro-inflammatory tumor microenvironment, providing more relevant targets for immunotherapy. The purpose of this paper is to discuss the effect of radiotherapy on tumor immune microenvironment and the role of radiotherapy combined with immune checkpoint inhibitors in the treatment of glioblastoma.
4.Effects of CYP2C9*3 Gene Polymorphism on Therapeutic Efficacy of Benzbromarone in Lowering Uric Acid and Its Hepatotoxicity

OBJECTIVE:To study the effects o f CYP2C9*3 gene polymorphism on therapeutic efficacy of benzbromarone in lowering uric acid and its hepatotoxicity. METHODS :A retrospective study was conducted to analyze the relevant clinical indicators and genotypes of 196 gout patients who received benzbromarone and CYP2C9*3 gene polymorphism test in Wuhan third hospital from Jan. 2018 to Sept. 2019. RESULTS :Among 196 patients,179,15 and 2 patients with CYP2C9*3 genotypes * 1/*1, *1/*3 and * 3/*3 genotypes were found ,respectively,and the distribution of each genotype was in line with Hardy-Weinberg balance(P>0.05). Before treatment ,there were no significant differences in the levels of UA ,Scr,ALT,AST and CRP between *1/*1 genotype and * 1/*3+*3/*3 genotype(P>0.05). After 4 weeks of treatment ,the UA ,Scr,CRP levels of patients with * 1/*1 genotype as well as the UA and CRP levels of patients with * 1/*3+*3/*3 genotype were significantly reduced ,the UA level of patients with * 1/*1 genotype was significantly lower than that of patients with * 1/*3+*3/*3 genotype(P<0.05 or P<0.01). The ALT and AST levels had no obvious changes in patients with different genotype before and after treatment ,and they were in the normal range. No serious abnormal liver function was observed during the treatment. CONCLUSIONS :Therapeutic efficacy of benzbromarone in lowering uric acid in gout patients with CYP2C9*3 genotypes * 1/*1 genotype is better than that of * 1/*3 and * 3/*3 genotypes. However ,the gene polymorphism may be not associated with its hepatotoxicity.
5.Two cases of intractable mycoplasma pneumoniae pneumonia complicated with intracranial venous sinus thrombosis

Mycoplasma pneumoniae pneumonia (MPP) complicated with cerebral venous sinus thrombosis (CVST) is rare.We retrospectively analyzed the clinical data of two patients with refractory mycoplasma pneumoniae pneumonia (RMPP) complicated with CVST who were hospitalized in Xi′an children′s Hospital from December 2018 to April 2019, inquired the relevant literature, analyzed the clinical diagnosis and treatment characteristics, and discussed the diagnosis and treatment measures of RMPP complicated with CVST.Two cases were 6-year-old girls with fever and cough as the main symptoms.After physical examination, the respiratory sounds of the affected lung decreased, and the sounds of phlegm and dampness could be heard in both lungs.Mycoplasma pneumoniae (MP) antibody and RNA were positive.Chest CT showed lobar pneumonia with a large number of pleural effusion.The effect of macrolide antibiotics anti infection treatment was not good.Headache symptoms occurred during the course of the disease, and serum D-dimer increased significantly.Brain MRI showed CVST, including 1 case with lower extremity pain, and B-ultrasound showed right lower extremity arterial embolism.After anti infection, thrombectomy, anticoagulation and symptomatic treatment, 2 cases were discharged.When children with MPP, especially those with RMPP, have extracranial thrombosis and/or neurological symptoms, accompanied by elevated serum D-dimer, the possibility of CVST should be considered, and brain MRI examination should be performed in time to confirm and actively treat, which can reduce or avoid the occurrence of sequelae.Thrombosis may be related to excessive inflammatory reaction and vascular endothelial injury caused by MP infection.
6.Detection and analysis of syphilis antibody in 80 478 cases of inpatients

Objective:To understand the infection and distribution of syphilis in hospitalized patients, thus to provide reference for syphilis prevention and control.Methods:TP-ELISA test was used to examine early syphilis antibody, and adopted the TPPA test to validation syphilis antibody, and TRUST was used to determine the titer of syphilis antibody in 80 478 hospitalized patients from January 2015 to November 2017, then the results were retrospectively analyzed.Results:Among 80 478 inpatients, 1 326 cases were positive by TP-ELISA test(1 223 cases positive, 101 cases weak positive and 2 cases negative by TPPA confirmed). The positive rates of TP-ELISA in different years were 1.62%(445/27 394), 1.72%(490/28 412) and 1.58%(389/24 672), respectively, and the difference was not statistically significant( P>0.05). The positive rates of male and female patients were 2.02%(689/33 985) and 1.37%(635/46 479), and the difference was statistically significant(χ 2=52.91, P=0.00). The positive rates of ≤18 years old, >18-59 years old, >59-79 years old and>79 years old were 0.32%(7/2 161), 1.44%(765/53 001), 2.31%(488/21 163) and 1.50%(62/4 153), respectively.The highest proportion of syphilis patients was in the group of >59-79 years old, and the differences were statistically significant compared with the other groups(χ 2=37.08, 67.05, 10.80, all P<0.01). Among the TP-ELISA positive patients, 54.90%(728/1 326) had TRUST titer negative, 36.50%(484/1 326) had titer less than 1∶8, and the others had 8.44%(112/1 326). Conclusion:The incidence of syphilis was higher in males than in females in 80 478 hospitalized patients.The highest positive rate was found in >59-79 years old group, and the number of elderly cases increased rapidly.Therefore, the effective interventions should be developed to control the transmission of syphilis according to the epidemiological features.
7. Dosimetric comparison between non-coplanar volumetric modulated arc therapy using flattening filter and flattening filter-free beams during stereotactic radiosurgery for brain tumors

Objective To compare the dosimetric and delivery efficiency between intensity-modulated radiation therapy (IMRT) plans based on Halcyon and Trilogy accelerator.Methods Clinical data of 12 patients with cervical cancer receiving Trilogy platform-based IMRT were retrospectively analyzed.The target area and organ at risk were introduced into the Eclipse version 15.1 TPS.The Halcyon-based IMRT was re-designed.The dose coverage of target area and organs at risk,monitor unit (MU) efficiency,delivery time and segment area and the differences between the proximal and distal segment area were statistically compared between two IMRT plans.Results The Halcyon treatment platform could meet the clinical objectives.Dose distribution of the target area was similar to that of the Trilogy plan (P>O.05).The volumes in the bladder and rectum receiving 30 Gy and the volumes in the left femoral head receiving 20 Gy of Halcyon plan and the mean dose (D) of bladder,rectum and left femoral head were significantly lower compared with those of the Trilogy plan (P=0.001-0.043).The MUs of the Halcyon plan was 2 316.7± 209.7,significantly higher than 1 692.5± 259.5 of the Trilogy plan (P=0.000).The delivery time significantly differed between the Halcyon [(3.01±0.28) min] and Trilogy [(12.38± 1.49) min] (P=0.000).The average segment area of Halcyon plan for proximal and distal MLC was (43.7±32.5) cm2 and (28.8±25.4) cm2,the average segment area of Trilogy plan was (25.7±16.8) cm2,the ratio of the proximal and distal segment area was 1.73±0.43,ranging from 0.71 to 6.28.Conclusions Halcyon treatment platform can meet the clinical requirements and reduce the dosage coverage of normal tissues with better delivery efficiency.The ratio of proximal and distal segment areas should be considered during the plan design.
9.Strategy of schistosomiasis elimination and its effects in Jinhu County, Jiangsu Province

Objective To understand the strategy of schistosomiasis elimination and its effects in Jinhu County, Jiangsu Province. Methods The data of schistosomiasis control in Jinhu County at different stages from 1970 to 2017 were collected and analyzed. Results From 1970 to 2017, there were three stages of schistosomiasis control, including transmission control, transmission interruption, and monitoring and elimination stages in Jinhu County. The main measures included Oncomelania hupensis snail control, infectious source control, and health education. A total of area of 290 691.78 hm2 was detected in Jinhu County, and the area with snails was 3 420.98 hm2. There were 8 729.37 hm2 area with snails was controlled. Since 2014, no O. hupensis snails were found. A total of 525 377 person-times were examined for schistosomiasis, with 2 815 schistosomiasis patients identified, and 2 844 person-times were treated by chemotherapy. In addition, 977 cases received the expand chemotherapy. Since 1990, no local schistosome-infected persons were found. In 2017, the awareness rate of schistosomiasis control knowledge and the correct rate of health behavior were increased by 54.59% and 14.23% respectively compared with those in 1992. Conclusions The comprehensive schistosomiasis control measures implemented in Jinhu County at different periods have achieved remarkable outputs and accelerated the schistosomiasis elimination process. However, the precise control measures should be implemented in the future to consolidate the prevention and control achievements.
10.The plan quality and delivery efficiency of intensity modulated radiotherapy on a new designed linac for left side breast cancer

Objective To analyze the dosimetric and delivery efficiency differences between IMRT plans based on Halcyon and Trilogy Accelerator for left side breast cancer.Methods A total of 10 patients with left side breast cancer based on the Trilogy platform were retrospectively analyzed.For each patient,plan based on Halcyon was redesigned using Eclipse Version 15.1 TPS.In order to evaluate plan quality,dose coverage of target and dose to organs at risk,monitor unit efficiency,segment size and delivery time were compared.Results Halcyon treatment platform could meet the clinical objective.Dose distribution of the target volume had no significant difference with the Trilogy plans (P>0.05).Volumes receiving 10 Gy,20 Gy and D of the left lung were significantly reduced(Z=-2.22--1.78,P<0.05).V5 of heart for Halcyon and Trilogy was(27.80±7.66)% and (23.18±8.19)%,respectively(Z=-0.71,P<0.05),while D was (7.03± 1.8)Gy and(7.11±2.40)Gy,respectively(P>0.05).Halcyon increased the monitor unit from 1 526.2±227.7 by Trilogy to 1 770.5±383.9(Z=-0.71,P<0.05),but decreased the delivery time from (12.38± 1.49) min by Trilogy to (3.01 ±0.28) min (Z =-3.42,P< 0.05).Conclusion Halcyon treatment platform can meet the clinical requirements,reduce dose to normal tissue with high delivery efficiency.
