Loganin(LOG),a bioactive compound derived from Cornus officinalis Siebold & Zucc,has been understudied in the context of osteoarthritis(OA)treatment.In this study,we induced an inflammatory response in chondrocytes using lipopolysaccharide(LPS)and subsequently treated these cells with LOG.We employed fluorescence analysis to quantify reactive oxygen species(ROS)levels and measured the expression of NLRP3 and nuclear factor erythropoietin-2-related factor 2(NRF2)using real-time quantitative polymerase chain reaction(qRT-PCR),Western blotting,and immunofluorescence(IF)techniques.Additionally,we developed an OA mouse model by performing medial meniscus destabilization(DMM)surgery and monitored disease progression through micro-com-puted tomography(micro-CT),hematoxylin and eosin(H&E)staining,safranin O and fast green(S&F)staining,and immunohisto-chemical(IHC)analysis.Our results indicate that LOG significantly reduced LPS-induced ROS levels in chondrocytes,inhibited the activation of the NLRP3 inflammasome,and enhanced NRF2/heme oxygenase 1(HO-1)signaling.In vivo,LOG treatment mitigated cartilage degradation and osteophyte formation triggered by DMM surgery,decreased NLRP3 expression,and increased NRF2 expres-sion.These findings suggest that LOG has a protective effect against OA,potentially delaying disease progression by inhibiting the ROS-NLRP3-IL-1β axis and activating the NRF2/HO-1 pathway.

Background and purpose:As one of the most fatal malignant tumors in China, the morbidity and mortality of lung cancer remain high. Early diagnosis and normative treatment is the key to improve the prognosis of lung cancer. The aim of this study was to explore the practice of early lung cancer screening with low-dose spiral computed tomography (CT) based on the current situation in community health service, with integration of superior resources of med-ical institutions at all levels in Shanghai.Methods:From Aug. 2013 to Aug. 2014, we screened high-risk population in selected communities of Minhang District, Shanghai, for early diagnosis of lung cancer with low-dose spiral CT combined with multidisciplinary comprehensive treatment models including minimally invasive surgery, exploring the medical service network covering prevention, diagnosis, treatment, rehabilitation and follow-up.Results:Screening population is 11 332 (male 7 144, female 4 188); Twenty-nine cases with pathological diagnosis of malignant tumor, including 27 cases of pri-mary lung cancer, 1 case of lung metastasis, 1 case of breast cancer. The morbidity of primary lung cancer is 238.26×10-5. There were 22 cases of Stage 0-Ⅰ lung cancer accounting for 81.48% of all diagnosed primary lung cancer.Conclusion:Based on community health service, screening with low-dose spiral CT could improve the early diagnosis rate of lung can-cer with feasibility and validity, which could be applicable in qualified eligible medical center and community in China.
4.Evaluation of therapeutic effects with inlra ( Need to be modified by authors ) -operative iodine-125 seed implantation for advanced pancreatic carcinoma

Objective To analyze the clinical effects of internal radiation of iodine-125 seed implantation in the treatment of advanced pancreatic carcinoma.Methods 22 patients with unresectable and advanced pancreatic carcinoma were treated by the internal and interstitial radiation using iodine-125 seed implantation.Results Survival of 20 months was observed in 5 patients,10 ~ 20 months in 9 patients,2 ~10 months in 8,patients with an average of (13.47 ± 8.12) months.Among the 22 patients,complete response was obtained in 5 patients,partial response in 8 patients,no response in 7 paticnts,and the other showing PD.The response rate( CR + PR) was 59%.Conclusions Intra - operative internal radiation by iodine - 125 seed implantation does show some therapeutic effects for advanced pancreatic carcinoma.
5.Expression of CD24 in esophageal squamous cell carcinoma and its relationship with prognosis

Objective To study the significance of CD24 protein expression in esophageal squamous cell carcinoma and its relationship with clinical pathological parameters and clinical prognosis.Methods The expression of CD24 protein was studied in 90 esophageal squamous cell carcinoma samples by EnVision immunohistochemical analysis and to study relationship of CD24 with clinical pathological parameters and prognosis.Results The expression of CD24 in esophageal squamous cell carcinoma was significantly higher than that in adjacent normal tissue (x 2 =0.832,P < 0.05).The expression of CD24 was not related with the age of patien,sexuality and the histologic grade (x2 =0.741,0.361,0.930,8.858,all P > 0.05),but with clinical stage of the cance (x2 =10.804,P =0.005),lymph node metastasis (x2 =12.349,P =0.000) and depth of tumor invasion (x2 =13.94,P =0.007).Significant difference in survival between the different CD24 expression group,the OR (95 % CI) values were 2.105 (61.022-69.273),2.313 (55.108-64.176),1.286 (32.388-37.429),the survival of patients with strong expression of CD24 was inferior to that of weak expression of CD24,depth of tumor invasion and lymph node metastasis were independent prognostic factors of esophageal squamous cell carcinoma.Conclusion CD24 may be related to the malignant process of esophageal squamous cell carcinoma.Overexpression of CD24 indicates poor prognosis in esophageal squamous cell carcinoma patients.CD24 may become a new independent prognostic factor for esophageal squamous cell carcinoma.
6.Study on the lethal effect of cytotoxic lymphocyte against A549 cells induced by dendritic cells in vitro

Objective The goal of the study was to assess the lethal effect of cytotoxic lymphocyte against A549 cells induced by dendritic cells (DC) pulsed with A549 lysate and transfected with GM-CSF recombinant adenovirus. Methods The cytotoxic lymphocyte against A549 cells were induced by culturing with DCs, which pulsed with A549 antigens and transfected with GM-CSF recombinant adenovirus. The of effector/target ratio, the killing rates of N-DC group, A-DC group and G-A-DC group were (1.9±0.7) %,effector/target ratio, the killing rates of N-DC group, A-DC group and G-A-DC group were (5.3±0.2) %, (40.5±7.7) % and (72.5±4.7) %, respectively. We found that the killing rate of G-A-DC group was the highest by statistics. Conclusion The cytotoxic lymphocyte against A549 cells can be induced by DCs pulsed with A549 lysate ,and the lethal effect of CTLs can be enhanced when DCs were infected with GM-CSF recombinant adenovirus.
7.Preparation and identification of the polyclonal antibody against the 3D polymerase of foot and mouth disease virus

To prepare the polyclonal antibody against the 3D polymerase of foot and mouth disease virus (FMDV), the 3D polymerase gene of this virus was amplified by PCR and doubly digested with BamH I and Nde I. Then, it were cloned into expression vector pET-30a(+) to obtain the recombinant plasmid pET-3D and this plasmid was transformed to E.coli BL21(DE3) with induction by IPTG.The target protein was identified and purified with SDS+PAGE, and the inclusion bodies were extracted. The purified target protein was used as antigen to immunize New Zealand rabbits to prepare the polyclonal antibody against 3D polymerase of FMDV, which was then characterized by indirect ELISA and Western blotting. As demonstrated by SDS-PAGE, the target protein with a molecular weight at 46 ku was expressed. The polyclonal antibody showed high affinity and obvious specificity and its titer was above 1:8 000. This polyclonal antibody may lay a foundation for the further studies on the biological functions and epitopes of the 3D polymerase of FMDV.
8.Isotretinoin erythromycin gel in the treatment of acne vulgaris: a multicenter randomized parallel-controlled clinical study

Objective To compare the clinical efficacy and safety of isotretinoin erythromycin gel, a gel containing isotretinoin (0.05%) and erythromycin (2%), versus adapalene gel in the treatment of mild to moderate acne vulgaris. Methods A multicenter, randomized, open, parallel-controlled clinical study was conducted. A total of 192 patients with mild to moderate (Grade Ⅰ -Ⅲ ) acne vulgaris were enrolled in this study according to the grading criteria for acne severity in guidelines for the treatment of acne in China. Efficacy analysis was carried out in 169 patients and safety analysis in 190 patients. The patients were classified into trial group (n = 86) and control group (n = 83 ) to be treated with isotretinoin erythromycin gel or adapalene gel once a night for 6 weeks. Patients were evaluated at the baseline, on week 2, 4 and 6 during the treatment for the count of comedones (both open and closed), inflammatory papules and pustules, severity of acne and local or general adverse effects. Results After the start of treatment, the response rate gradually increased and severity of acne decreased in both groups. On week 6, the total response rate was 51.16% in the trial group and 40.96% in the control group (P > 0.05), while a greater reduction in the count of pustules and inflammatory lesions was observed on week 4 and 6 in the trial group with a lower severity grade of acne compared with the control group (P < 0.05 or 0.01 ). Adverse reactions were similar in both groups and manifested as tolerable local irritation. Conclusions The efficacy of isotretinoin erythromycin gel is similar to that of adapalene gel in the treatment of mild to moderate acne vulgaris, however, isotretinoin erythromycin gel seems superior to adapalene gel in reducing inflammatory lesions and rapidly improving severity of acne vulgaris.
9.Expression of CD24 and FAK in breast carcinoma and their clinical significance

Objective To investigate the expression of CD24 and FAK in the invasive breast ductal carcinoma tissues and their clin-ical significance. Methods Tissues were obtained from 86 patients with breast carcinoma and 30 non-cancer specimens. The expression of CD24 and FAK was detected by immunohistochemical technique (ElivisionTMPlus) and their correlation with tumor clinicopathological char-acters was analyzed. Results The positive rate of CD24 and FAK expression in 86 cases of breast carcinoma was 87.2% (75/86) and 72.1% (62/86) respectively, which was significantly different from that in control group(P<0.01). Expression of CD24 was significantly correlated with axillary lymph node metastasis(P<0.01). Expression of FAK was correlated with histopathologic grade, axillary lymph node metastasis and clinical stage (P<0.05). There was a positive correlation between the expressions of CD24 and FAK in breast carci-noma tissues (rs = 0.222, P < 0.05). Conclusion Expression of CD24 and FAK are closely correlated with the development of breast carcinoma and there is a markedly positive correlation between CD24 and FAK expression. These indexes can be helpful in understanding the biological behaviors and predicting the prognosis of breast carcinoma.
10.Correlation between CD24 expression and its relation with Ki67,Bcl-2 in breast carcinoma

Objective To investigate the expression of CD24 and Ki67 in the invasive breast ductal car-cinoma tissues, so as to assess the role of CD24 protein in the carcinogenesis and progression of breast canc-er. Methods The expression of CD24 and Ki67, Bcl-2 was examined by immunohistochemically in 86 breast cancer specimens and 30 non-cancer specimens ( ElivisionTM Plus). The relationship between CD24 expression and Ki67, Bcl-2, patient age, tumor size, histopathologic grade, axillary lymphnode metastasis, and clinical stage of breast cancer was analyzed. Results In 86 eases of invasive breast ductal carcinoma, the positive incidences of CD24 was 87.2% (75/86), which distinguished apparently from those of the con-trol group( P <0.01 ). In breast carcinoma tissues, the expression of CD24 was positively related to axillary lymphnode metastasis(P <0.01 ), but was not related to patients age,tumor size, histopathologic grade and clinical stage(P>0.05). CD24 expression was positively related to Ki67 (P<0.01 ) and negatively relat-ed to Bcl-2(P>0.05). Conclusion CD24 expression was related to the proliferation, invasion and me-tastasis of breast cancer cells and it can serve as an important marker for predicting biological behavior and prognosis of tumors.
