1.Exploration of Anti-depression Mechanism of Kai-Xin-San via Regulation of Neurogenesis of Hippocampus on Chronic Unpredictable Mild Stress Induced Mice

		                        			Objective To explore the anti-depression mechanism of Kai-Xin-San(KXS)via regulation of neurogenesis in hippocampus of depression-like mice.Methods The extracts of KXS were prepared and the anti-depression effects of KXS were evaluated by behavioral tests on chronic unpredictable mild stress(CUMS)induced depression-like mice.Evaluating depression-like behavior in CUMS mice through sucrose preference test,forced swimming test,tail suspension test,and other methods.Neurogenesis in hippocampus were determined by immunofluorescence assay.In addition,effects of KXS on regulating nestin expression and Wnt/b-catenin signaling pathway were explored by western blotting analysis.Amounts of cortisol,corticotropin-releasing factor(CRF),adrenocorticotropic hormone(ACTH),brain-derived neurotrophic factor(BDNF)and nerve growth factor(NGF)were determined by ELISA tests.Mouse primary neural stem cells(NSC)was used to evaluate the effect of KXS on promoting its proliferation by immunofluorescence assay.In addition,effects of KXS on regulating nestin and Wnt/β-catenin signaling pathway were also explored by Western blotting analysis.Results KXS significantly ameliorated the depression-like behaviors in presence of increased sucrose preference rate and decreased immobile time of tail suspension and forced swimming.KXS significantly promoted the neurogenesis in the hippocampus and expressions of nestin,reduced the expressions of cortisol,CRF,ACTH,increased the expressions of BDNF,NGF,and regulated Wnt/β-catenin signaling pathway.KXS also promoted the proliferation of NSCs and expressions of nestin,enhanced the translocation of b-catenin into nucleus,and regulated the expressions of proteins of Wnt/β-catenin signaling pathway.Conclusion KXS promoted neurogenesis in hippocampus and regulated Wnt/β-catenin pathway,which might contribute to its antidepressant effect.
		                        		
		                        		
		                        		
		                        	
2.Investigation and study of factors related to NCD health management of basic public health services in Chongqing and Guizhou Province

		                        			Objective To analyze the health management situation of non-communicable chronic disease(NCD)in Chongqing City and Guizhou Province and its influencing factors.Methods A stratified random sampling method was used to select 16 representative primary medical and health institutions from Chongqing City and Guizhou Province as the research sites,and the data were collected through the combination method of qualitative and quantitative research.Results A total of 760 patients with NCD were surveyed,and the awareness rate,utilization rate and satisfaction rate of the NCD health management program all exceeded 84.2%,while the awareness rate,utilization rate and satisfaction rate of follow-up and categorical intervention were the lowest.The awareness rate of hypertension physical examination in the patients with<60 years old was lower than that in the patients with 60-<70 years old(OR=4.28,95%CI:1.43-12.81)and the pa-tients with ≥70 year sold(OR=3.16,95%CI:1.12-8.91);the utilization rate of diabetes screening in the patients with<60 years-old was lower than that in the patients with ≥70 years old(OR=2.70,95%CI:1.08-6.76)and the awareness rate of hypertension physical examination was lower than that of the patients with 60-<70 years old(OR=4.24,95%CI:1.01-17.75);the awareness rate of hypertension physical ex-amination in the patients in Chongqing City was higher than that in Guizhou Province(OR=0.15,95%CI:0.04-0.54)and the utilization situation was better than that in Guizhou Province(OR=0.13,95%CI:0.05-0.34).Conclusion The overall situation of NCD management services in Chongqing City and Guizhou Province is good,but the service quality of follow-up and classified intervention projects needs to be further improved.
		                        		
		                        		
		                        		
		                        	
3.Expert consensus on postoperative care of patients with a left ventricular assistant device

		                        			Objective To standardize nursing management on postoperative patients with a left ventricular assist-ant device(LVAD).Methods The first draft of the Consensus was formed on the basis of literature review.2 rounds of expert consultations and a round of online meeting discussion were held for adjustments and modifications the draft of the Consensus.Results The recovery rate of the inquiry questionnaire was 93.75%.The authority coefficients of the 2 rounds of inquiry experts were 0.927 and 0.920.The concentration degree of expert opinions for each indicator was greater than 3.5 score,and the coefficient of variation was less than 0.25.The Kendall harmony coefficients for 2 rounds of correspondence were 0.402 and 0.407(P<0.01).The final Consensus formed through expert consultations and meetings includes 7 themes:hemodynamic monitoring,LVAD function monitoring,coagulation function monitoring,percutaneous cable and wound care,exercise rehabilitation care,health education and guidance,and pre-discharge assessment.Conclusion The Consensus is scientific,rigorous,and authoritative.The Consensus covers all aspects of postoperative care for patients with LVAD,and it will benefit to clinical practice.

		                        			Entinostat plus exemestane in hormone receptor-positive (HR+) advanced breast cancer (ABC) previously showed encouraging outcomes. This multicenter phase 3 trial evaluated the efficacy and safety of entinostat plus exemestane in Chinese patients with HR + ABC that relapsed/progressed after ≥1 endocrine therapy. Patients were randomized (2:1) to oral exemestane 25 mg/day plus entinostat (n = 235) or placebo (n = 119) 5 mg/week in 28-day cycles. The primary endpoint was the independent radiographic committee (IRC)-assessed progression-free survival (PFS). The median age was 52 (range, 28-75) years and 222 (62.7%) patients were postmenopausal. CDK4/6 inhibitors and fulvestrant were previously used in 23 (6.5%) and 92 (26.0%) patients, respectively. The baseline characteristics were comparable between the entinostat and placebo groups. The median PFS was 6.32 (95% CI, 5.30-9.11) and 3.72 (95% CI, 1.91-5.49) months in the entinostat and placebo groups (HR, 0.76; 95% CI, 0.58-0.98; P = 0.046), respectively. Grade ≥3 adverse events (AEs) occurred in 154 (65.5%) patients in the entinostat group versus 23 (19.3%) in the placebo group, and the most common grade ≥3 treatment-related AEs were neutropenia [103 (43.8%)], thrombocytopenia [20 (8.5%)], and leucopenia [15 (6.4%)]. Entinostat plus exemestane significantly improved PFS compared with exemestane, with generally manageable toxicities in HR + ABC (ClinicalTrials.gov #NCT03538171).
		                        		
		                        		
		                        		
		                        	
5.Relationship between idolatry and perceived social support in secondary school students: the mediating role of peer attachment and life satisfaction

		                        			BackgroundNowadays, idolatry is an important part of the spiritual life of secondary school students, making a significant impact on their physical and mental development. Previous research has examined the two-by-two relationship between idolatry, peer attachment, life satisfaction and perceived social support, but the potential mediating mechanisms between these variables remain to be explored. ObjectiveTo explore the mediating role of peer attachment and life satisfaction between idolatry and perceived social support, so as to provide references for mental health education for secondary school students. MethodsIn June 2022, 1 059 students currently enrolled in secondary school in Qiannan State, Guizhou Province were selected according to stratified random sampling method. In this study, assessment was performed using Celebrity Attitude Scale (CAS), Peer Attachment Subscale from Revised Inventory of Parent and Peer Attachment (IPPA-R), Satisfaction with Life Scale (SWLS) and Perceived Social Support Scale (PSSS). Pearson correlation analysis was used for correlation test. Model 6 in the SPSS Macro Process 3.3 was used to examine the mediating effect of peer attachment and life satisfaction between idolatry and perceived social support in secondary school students. Results①CAS score indicated positive correlation with scores of IPPA-R Peer Attachment Subscale, SWLS and PSSS (r=0.117, 0.097, 0.115, P<0.01). IPPA-R score indicated positive correlation with scores of SWLS and PSSS (r=0.279, 0.421, P<0.01). SWLS score was positively correlated with PSSS score (r=0.552, P<0.01). ②The direct effect of idolatry on perceived social support was significant (β=0.059, P<0.05). Both separate and chain mediating effects of peer attachment and life satisfaction between idolatry and perceived social support were significant (95% CI: 0.007~0.130, 0.004~0.054, 0.001~0.016, P<0.01). The three effects above respectively accounted for 33.56%, 13.54% and 3.93% of the total effect. ConclusionIdolatry can affect secondary school students' perceived social support both directly and indirectly through peer attachment and life satisfaction. 
		                        		
		                        		
		                        		
		                        	
6.Real emotional experience of subjective well-being of elderly people in nursing homes during the COVID-19 pandemic

		                        			Objective:To describe the real experience of the elderly who has felt happy during the time they stayed at nursing homes, and discuss the meaning which is behind their experience during the COVID-19 pandemic, to provide scientific basis for improving their subjective well-being.Methods:Qualitative explanatory phenomenological analysis methods, and semi-structured in-depth interviews were used to interview 21 elderly people living in Guangzhou Songhe Nursing Home. Data analysis and subject extraction were carried out with reference to the Colaizzi 7-step analysis.Results:The real experience of subjective well-being of the elderly in nursing homes has six themes: be satisfied with the nursing home management service, be satisfied with harmonious interpersonal relationship, be satisfied with good health, be satisfied with family support, grateful and expectations for the social support system, be satisfied with and grateful for the epidemic prevention work during the COVID-19 epidemic.Conclusions:The elderly who live in a nursing home with perfect management and service, care for each other, are healthy, receive family and social support, are grateful, and understand family and social support will be more likely to feel happy. 7.Determination of melamine and its conjugates in calcium oxalate stones by Fourier transform infrared spectroscopy

		                        			ObjectiveTo establish a method of two-times second derivative Fourier transform infrared spectroscopy (FTIR) for identifying melamine (MEL) and its conjugates with cyanuric acid (MEL-CYA) or with uric acid (MEL-UA) in human urinary calculi. MethodsMEL, MEL-CYA and MEL-UA were added to calcium oxalate stone samples, and then analyzed by two-times second derivative FTIR for identifying the characteristic response bands of MEL and its conjugates in stones as well as confirming the ratio of detection. ResultsThe second derivative FTIR could improve the detection of MEL in CaOx stones by two orders of magnitude. When CaOx being mixed with MEL (MEL∶ CaOx), the two-times second derivative FTIR showed the characteristic peak at 1 548 cm-1 and the minimum mass ratio of detection at 1∶100 (1%). When CaOx being mixed with MEL-CYA complex (MEL-CYA∶ CaOx), the two-times second derivative FTIR showed the characteristic peak on 1 740 cm-1 and 1 538 cm-1 and the minimum mass ratio of detection at 1∶500 (0.2%). When CaOx being mixed with MEL-UA complex (MEL-UA∶ CaOx), the two-times second derivative FTIR showed the characteristic peak at 1 117, 982 and 710 cm-1 and the minimum mass ratio of detection at 1∶250 (0.4%).ConclusionCompare to the original spectra, the two-times second derivative FTIR can improve the detection ratio MEL in the CaOx stones from 0.2% to 1.0%. 8.Shenbai Jiedu Prescription Inhibits Proliferation of Colorectal Cancer Cells by Regulating PTEN/PI3K/Akt Signaling Pathway

		                        			ObjectiveTo study the mechanism of Shenbai Jiedu prescription inhibiting the proliferation of HCT116 colorectal cancer (CRC) cells by regulating the phosphatase and tensin homolog deleted on chromosome ten (PTEN)/phosphatidylinositol 3-kinase (PI3K)/ protein kinase B (Akt) signaling pathway. MethodShenbai Jiedu prescription was extracted by water extraction and alcohol precipitation to prepare freeze-dried powder. HCT116 cells were cultured in vitro, and treated with different concentrations of Shenbai Jiedu prescription (2, 4, 8, 16 g·L-1). The inhibitory effect of Shenbai Jiedu prescription on the proliferation of HCT116 cells was tested by methyl thiazolyl tetrazolium (MTT). Real-time quantitative PCR was used to detect the mRNA expression levels of PTEN, PI3K, Akt, glycogen synthase kinase-3β (GSK-3β), c-Myc, survivin and Cyclin D1. Western blot was employed to measure the protein expression levels of PTEN, phosphorylated PTEN (p-PTEN), PI3K, Akt, phosphorylated Akt (p-Akt), GSK-3β, phosphorylated GSK-3β (p-GSK-3β), c-Myc, survivin and Cyclin D1, β-catenin nuclear import was explored by immunofluorescence assay. ResultCompared with the control group, Shenbai Jiedu prescription inhibited the proliferation of HCT116 cells in a dose-dependent manner (P<0.01). Compared with the control group, the mRNA expression levels of PTEN and GSK-3β were up-regulated whereas those of PI3K, Akt, c-Myc, survivin and CyclinD1 were down-regulated after treatment with Shenbai Jiedu prescription (P<0.01). The protein expression levels of PTEN, p-PTEN and GSK-3β were up-regulated whereas those of PI3K, Akt, p-Akt, GSK-3β, p-GSK-3β, c-Myc, survivin and CyclinD1 were down-regulated (P<0.05, P<0.01). Immunofluorescence assay showed that Shenbai Jiedu prescription suppressed β-catenin nuclear import in HCT116 cells. 9.An investigation of iodine nutritional status of children aged from 8 - 10 and pregnant women before and after adjustment of salt iodine concentration in Xiamen City

		                        			Objective:To investigate the iodine nutritional status among children aged from 8 - 10 and pregnant women in Xiamen City before and after adjustment of salt iodine concentration, and scientifically evaluate the suitability of the new standard iodized salt adjustment policy for Xiamen City.Methods:In the pre-adjustment period (2011) and the post-adjustment period (2018), cross-sectional research methods and stratified cluster sampling methods were adopted in six districts of Xiamen City. The salt samples of residents and pregnant women, urine samples of children aged from 8 - 10 and pregnant women were collected to determine the iodine level; thyroid of children aged from 8 - 10 was examined. Changes in salt iodine, urine iodine and goiter prevalence before and after adjusting iodine content of salt were compared.Results:The median of salt iodine in residents after the adjustment (23.5 mg/kg) was significantly lower than that of before (28.9 mg/kg, Z = - 10.512, P < 0.05), the median of salt iodine in pregnant women after the adjustment (23.7 mg/kg) was significantly lower than that of before (29.2 mg/kg, Z = - 12.622, P < 0.05); the consumption rate of qualified iodized salt in residents after the adjustment (95.7%, 1 196/1 250) was significantly lower than that of before (97.1%, 1 608/1 656; χ 2 = 4.250, P < 0.05); before and after adjustment of salt iodine concentration, the consumption rate of qualified iodized salt in pregnant women was 94.7% (124/131) and 95.9% (579/604), respectively. No significant difference was found when comparing the consumption rate of qualified iodized salt in pregnant women (χ 2 = 0.015, P > 0.05). There was significant difference in median urinary iodine (from 204.2 to 183.9 μg/L, detected in 652, 1 250 urine samples, respectively) of children aged 8 - 10 years ( Z = - 3.583, P < 0.05); the difference of iodine nutritional status in pregnant women before and after adjustment of salt iodine concentration(130.2, 130.8 μg/L, detected in 132, 604 urine samples, respectively) was not significant ( Z = - 1.715, P > 0.05). The thyroid goiter rate of children aged 8 - 10 years had reduced from 1.1% (14/1 239) to 0.2% (2/1 253), the difference was statistically significant (χ 2 = 9.195, P < 0.05). Conclusions:As a whole, the measure of control and prevention of iodine deficiency disorders through iodized salt is implemented well in Xiamen City. After adjustment of salt iodine concentration, the iodine status of children aged from 8 to 10 is adequate, but the pregnant women has showed iodine deficiency.
		                        		
		                        		
		                        		
		                        	
10.Study on Hypoglycemic Effects of Different Polar Parts from the Leaves of Dimocarpus longan on Type 2 Diabetes Mellitus Mice

		                        			OBJECTIVE:To optimize the hypoglycemic active parts from the leaves of Dimocarpus longan for type 2 diabetes mellitus mice,and to provide reference for material basis study of it. METHODS:Total extract was obtained from the leaves of D. longan with 95% ethanol percolation extraction. After confirming hypoglycemic effect of total extract,extract was suspended with water. Relevant extract was obtained by using petroleum ether,ethyl acetate and n-butanol in turn(respectively marked as ST,YT,ZT and WT). The model of type 2 diabetes mellitus mice was established by high glucose and high fat diet combined with STZ. Model mice were randomly divided into model group(distilled water),positive control group(metformin,0.1 g/kg), ST low-dose,medium-dose and high-dose groups(22.2,29.6,44 g/kg,calculated by crude drug,similarly hereinafter),YT low-dose,medium-dose and high-dose groups(10.7,16,32 g/kg),ZT low-dose,medium-dose and high-dose groups(10.5, 15.8,31.6 g/kg),WT low-dose,medium-dose and high-dose groups(18.5,24.6,37 g/kg),with 10 mice in each group. Other 10 normal mice were included in normal group(distilled water). Those groups were given relevant medicine intragastrically,once a day,for consecutive 28 d.The growth of mice and the change of body weight were observed and recorded. Fasting blood glucose(FBG)of mice was detected,and glucose tolerance test of mice was conducted. The contents of TC,TG,HDL-C and LDL-C in serum of mice were also determined. RESULTS:Compared with normal group,body weight and serum content of HDL-C in model group were decreased;FBG and the blood sugar after giving glucose for 0.5,1,2 h and area under glucose tolerance curve of mice were increased,and serum contents of TC,TG and LDL-C were alse increased,with statistical significance(P<0.01). compared with model group,body weight of mice in YT high-dose group after administration of 25 d was increased,FBG of mice in YT groups and ZT medium-dose,high-dose groups were decreased(P<0.05 or P<0.01);blood sugar of mice in YT medium-dose,high-dose groups and ZT high-dose group after giving glucose for 1 h as well as blood sugar of mice in YT high-dose group after giving glucose for 2 h were all decreased;area under glucose tolerance curve of mice and serum content of TC were decreased in YT groups and ZT high-dose group,while serum content of HDL-C was increased. Serum content of TG and LDL-C in YT high-dose group and ZT high-dose group as well as serum content of TG in YT medium-dose group were all decreased,serum content of HDL-C in ZT medium-dose group was increased,with statistical significance(P<0.05 or P<0.01).
