1.The effective components of Chinese medicine combined with scaffold materials promote bone tissue regeneration

BACKGROUND:With the proven ability of traditional Chinese medicine such as icariin and berberine to promote bone regeneration by regulating various mechanisms and targets,researchers have combined active ingredients of traditional Chinese medicine with bone tissue engineering and found that they have unique advantages in treating bone defects. OBJECTIVE:Starting from the active ingredients of traditional Chinese medicines that promote bone formation,to screen cases of their effective combination with different drug-carrying scaffold materials,and summarize the active ingredients of traditional Chinese medicines that have the potential to be applied to bone tissue engineering. METHODS:CNKI,WanFang,PubMed,and Web of Science were searched for relevant literature published from 2000 to 2023,using the keywords of"bone tissue engineering,bone tissue-engineered scaffold materials,bone defect,bone repair,bone regeneration,traditional Chinese medicine"in Chinese and English.According to the inclusion and exclusion criteria,87 papers were finally included for review. RESULTS AND CONCLUSION:There are various kinds of active ingredients of traditional Chinese medicine to promote bone regeneration,mainly including flavonoids,non-flavonoid polyphenols,alkaloids,glycosides.These active ingredients have anti-inflammatory and analgesic effects,promote osteoblasts,inhibit osteoclasts and promote early angiogenesis.The combination of active ingredients of traditional Chinese medicine with bone tissue engineering is effective in anti-inflammation,accelerating collagen and bone formation,and promoting the expression of osteogenic genes,which provides a theoretical basis for the application of traditional Chinese medicine in bone tissue regeneration,and at the same time provides a new idea for the repair of bone defects.
2.Improvement effects of glycyrrhizin on Helicobacter pylori-associated gastritis in rats and its mechanism

OBJECTIVE To investigate the improvement effects of glycyrrhizin (GL) on Helicobacter pylori (HP)-associated gastritis in rats and its mechanism. METHODS HP-associated gastritis rat model was induced by inoculating with 1×109 cfu/mL HP. The model rats were randomly divided into model group, positive control group (HP standard quadruple group), GL low-dose, medium-dose and high-dose groups (5, 20, 50 mg/kg), with 12 rats in each group. Another 12 healthy rats were selected as normal control group. Except the normal control group and model group were given constant volume of normal saline intragastrically, the other groups were given corresponding drugs intragastrically, once a day, for 30 consecutive days. After administration, rats received 13C urea breath test, and delta-over-baseline (DOB) was recorded; the pathological and cellular morphological changes of gastric mucosa in rats were observed, and pathological scoring was performed; the levels of interleukin-8 (IL-8), IL-1β, tumor necrosis factor-α (TNF-α), reactive oxygen species (ROS) and malondialdehyde (MDA) were detected in gastric mucosa of rats; mRNA expressions of high mobility group box-1 protein (HMGB1) and nuclear factor-κ-B (NF-κB), relative expressions of nitric oxide synthases (iNOS) and HMGB1, the phosphorylation level of NF- κBp65 were also detected in rats. RESULTS Compared with normal control group, the DOB value, histopathological score of gastric mucosa, the levels of IL-8, IL-1β, TNF-α, ROS and MDA, relative expressions of HMGB1 and NF- κB mRNA, relative expressions of iNOS and HMGB1 protein and the phosphorylation level of NF-κB p65 were all increased significantly in model group (P<0.05); the epithelial cells of gastric mucosa in rats were incomplete in structure and decreased in the number, with an increase in cell fragments and vacuoles, and significant cell pyknosis. Compared with model group, the changes of the above indexes in GL groups and positive control group were significantly reversed (P<0.05); the changes in the above indicators in the GL high-dose group were more significant than GL low-dose and medium-dose groups (P<0.05); the pathological changes of gastric mucosal cells in rats had all improved. CONCLUSIONS GL may inhibit inflammation and oxidative stress by inhibiting the activation of HMGB1/NF-κB pathway, thus relieving HP-induced gastric mucosal injury.
3.Effects of community TCM health management on patients with chronic obstructive pulmonary disease at a stable stage

Objective:To observe the effects of community traditional Chinese medicine (TCM) health management of patients with chronic obstructive pulmonary disease (COPD).Methods:From June 2017 to June 2018, 79 patients were selected as study subjects. These patients were randomly divided into two groups. The TCM management group had 39 patients (of which 26 were male), with an average age of (70.77±5.91) years and the course of disease of (7.97±3.12) years. The control group had 40 patients (of which 25 were males), with an average age of (70.60±5.93) years and the course of disease of (7.70±3.01) years. Both groups were given conventional western medicine treatment. The TCM management group was administered TCM health management for 1 year on the basis of the conventional western medicine treatment. This management included establishing electronic archives for COPD patients, Hu Xi Tu-Na, Baduanjin, acupoint massage, acupoint plaster, and dietary guidance according to the TCM physique identification. Management and follow-up of patients was continuous. The TCM syndrome score, CAT score, number of acute exacerbations, and lung function of COPD patients were recorded before and after 1 year of treatment.Results:In the TCM management group, there were 6 cases of clinical control, with 8 and 19 cases displaying obvious and effective effects, respectively. The total effective rate was 84.61%. In the control group, there were 2 cases of clinical control, with 3 cases and 25 cases displaying obvious and effective effects, respectively. The total effective rate was 75.00%. The curative effect of the TCM syndrome in the TCM management group was better than that in the control group. In the TCM management group,the CAT score was (7.46±3.28) points and the number of acute exacerbations was (4.21±2.61) times, the forced expiratory volume (FEV 1) in the first second was (1.99±0.79) L, and the forced vital capacity (FVC) was (2.63±0.92) L. In the control group, the CAT score was (10.38±4.68), the number of acute exacerbations was (6.03±3.00), FEV 1 was (1.66±0.65) L, and FVC was (2.41±0.81) L. The CAT score and the number of acute exacerbations in the TCM management group decreased, while the number of FEV 1 increased, which was statistically significant compared to the control group. Conclusion:Community TCM health management can improve the symptoms of COPD patients at a stable stage, reduce the number of acute exacerbations, and improve lung function.
4.Establishment of liver transplantation model with improved two-cuff in rat

Objective To build rat orthotopic liver transplantation model and use this model as an experimental animal model for further research about liver transplantation.Methods Based on the classic two-cuff technique,a variety of skills were integrated,such as abdominal aorta perfusion first before ligation,suprahepatic inferior vena cava single continuous suture,preoperative and postoperative rehydration,hepatic portal vein and infrahepatic vena cava blocked by mosquito's haemostatic forceps.At last,the postoperative survival rates of 2 days and 2 weeks were recorded.Results Donors' operation time was (35±2) min,cuffing time was (7.0±1.5) min,receptors' operation time was (80±15) min,anhepatic phase was (21±3) min.The 2-day survival rate was 92 % (46/50) and 2-week survival rate was 86 % (43/50).Conclusion To some extent,the improved methods can not only reduce the difficulty of orthotopic liver transplantation operation,but also to create a uniform,stable and reliable orthotopic liver transplantation model.
5.Effect of arterial and venous subarachnoid hemorrhage on voltage-dependent calcium channel currents of cerebral artery smooth muscle cells in rats

Objective To investigate the effect of arterial and venous subarachnoid hemorrhage ( SAH)on voltage-dependent calcium channel( VDCC)currents of cerebral artery smooth muscle cells and the relationship between the concentration of oxyhemoglobin( OxyHb)in arterial and venous blood and cerebral blood flow. Methods Thirty-six clean grade rats were colleted. A rat SAH model was induced by injection of autologous arterial or venous blood in suprasellar cistern using assisted stereotaxic apparatus. The rats were divided into three groups:an arterial SAH( n=14 ),a venous SAH( n=13 ),and a sham operation( n=9 )group. The arterial and venous OxyHb concentrations were measured. Three days after SAH modeling,a patch clamp was used to detect the relative surface area of the cerebral artery smooth muscle cells,resting potential,and VDCC currents in rats. A fluorescent microsphere method was used to quantitatively analyze cerebral blood flow(CBF). Results (1)Arterial SAH OxyHb concentration (127 ± 4 g/L)was significantly higher than venous SAH OxyHb concentration(54 ± 6 g/L),and that of the sham operation group was 50 ± 5 g/L. The differences were statistically significant among the 3 groups( P<0. 01).(2)The maximum current of VDCC of the arterial SAH group(3. 22 ± 0. 31 pA)was significantly higher than that of the venous SAH group(2. 19 ± 0. 27 pA)and the sham operation group(2. 18 ± 0. 29 pA). The differences were statistically significant among the 3 groups( P<0. 01 ). The VDCC currents of the arterial SAH group were consisted of L- and R-currents,while the currents of the venous SAH group were only consisted of L-VDCC.(3)The cerebral blood flow of the arterial SAH group(0. 83 ± 0. 14 mL/[g·min])was significantly higher than that of the venous SAH group(1. 28 ± 0. 28 mL/[g·min])and the sham operation group(1. 35 ± 0. 19 mL/[g·min]). The differences were statistically significant(P<0. 01). Conclusions The changing effect of arterial SAH on the expression and function of the cerebral artery smooth muscle cells are greater than that of the venous SAH. This difference may be associated with the concentration and composition of vasospasm factors of OxyHb in arterial and venous blood.
6.The different suppression on voltage-dependent potassium channels currents of smooth muscle cells from cerebral pial arteries and penetrating arteries by subarachnoid hemorrhage

Objective To study the effect of subarachnoid hemorrhage (SAH) on voltagedependent potassium channels (Kv) currents of smooth muscle cells,which is hypothesized to be different between cerebral pial arteries and penetrating arteries.Methods Smooth muscle cells from cerebral pial arteries and penetrating arteries in rats were enzymatically isolated 72 h after SAH,and patch clamp was used to test the relative cell size,resting potential and Kv currents.Results Resting potential of either pial ((45.63 ±1.18) mV) or penetrating artery ((41.55-± 1.19) mV) was shifted positively by SAH,even more significantly in latter (F =8.24,P < 0.05 ; F =9.36,P < 0.01) ; Resting potential of pial artery of control ((38.76 ± 1.03) mV),penetrating artery of control ((38.53 ± 0.67) mV),pial artery of SAH ((36.87 ± 1.49) mV) and penetrating artery of SAH((37.89 ± 1.24) mV) were shifted positively to the same level by 1 μmol/L 4-aminopyridine (4AP; F =3.08,P >0.05).Maximum Current Density (Imax) of either pial ((20.82 ±0.59) pA/pF) or penetrating artery ((15.15 ±0.37) pA/pF) was compromised by SAH,also more significantly in latter (F =6.22,P < 0.05) ; Imax of pial artery of control (9.15 ± 0.16),penetrating artery of control (9.04 ± 0.36),pial artery of SAH (8.77 ± 0.26) and penetrating artery of SAH (9.12 ± 0.17) were decreased to the same level by 1 μmol/L 4AP (F =2.96,P > 0.05).Conclusions SAH probably shares the similar pathway with Kv blocker (4AP) in Kv currents inhibition.Further,SAH differently inhibits smooth muscle cells Kv currents and resting potential of cerebral pial arteries and penetrating arteries,which may be related with their different sensitivity towards cerebral vasospasm following SAH.
7.The protective effects of hydrogen inhalation on renal injury in severe septic mice

Objective To investigate the protective effects of hydrogen gas (H2) inhalation on acute renal injury in severe septic mice,and the associated mechanism.Methods Sepsis model was made by cecal ligation and puncture (CLP) operation in mice.A total of 24 mice were randomly (random number)divided into four groups:sham operation group (Sham),sham operation with H2 inhalation group (Sham +H2),sepsis group (Sepsis),and sepsis with H2 inhalation group (Sepsis + H2).The procedure for inhalation of 2% H2 for 1 h was initiated at 1 h after CLP and 6 h after sham operation,respectively.The pathological changes and apoptosis of kidney,the blood creatinine (Cr) and blood urea nitrogen (BUN)concentration,the activities of superoxide dimutase (SOD) and catalase (CAT) in serum and kidney,as well as the levels of 8-iso-prostaglandin F2α (8-iso-PGF2α) and high mobility group box 1 (HMGB1) in serum and kidney were observed at 24 h after operation.Results H2 inhalation could significantly alleviate the renal injury of septic mice,reduce the renal cell apoptosis,increase the activities of SOD and CAT,as well as decrease the levels of 8-iso-PGF2α and HMGB1 in serum and kidney (P < 0.05).Conclusions H2 inhalation can alleviate acute renal injury in septic mice,resulting from the increased activities of antioxidant enzymes and the reduced levels of oxidative products and inflammatory cytokines.
8.Effect of hydrogen-rich saline on lipopolysaccharide-induced acute lung injury in mice

Objective To investigate the effects of hydrogen-rich saline on lipopolysaccharide (LPS)-induced acute lung injury (ALI) in mice.Methods Thirty-two male C57BL/6 mice,weighing 20-25 g,were randomly divided into 4 groups (n =8 each):control group (group C),hydrogen-rich saline group (group H2),ALI group and ALI + hydrogen-rich saline group (group ALI+ H2).ALI was induced by inhaled aerosolized LPS 25μg (dissolved in PBS) in groups ALI and ALI+ H2.Aerosolized PBS 50μl was inhaled in groups C and H2.0.6mmol/L hydrogen-rich saline 5 ml/kg was injected intraperitoneally at 1 and 12 h after aerosolized PBS or LPS was inhaled in groups H2 and ALI+ H2,respectively.At 24 h after PBS or LPS treatment,the animals were mechanically ventilation for 15 min,and arterial blood gas analysis was performed.The oxygenation index (PaO2/FiO2) was calculated.Bronchoalveolar lavage fluid (BALF) was collected for determination of total protein concentration and polymorphonuclear neutrophils (PMN) count.The concentrations of TNF-α,IL-1β,IL-6 and high mobility group box-1 (HMGB-1) protein in BALF were determined by ELISA.Then the animals were sacrificed and the lungs were obtained for determination of W/D lung weight ratio (W/D ratio),and myeloperoxidase (MPO) and caspase-3 activities.The pathological changes of the lung were scored.Apoptosis index (AI) was calculated.Results Compared with group C,no significant difference was found in PaO2/FiO2,the concentrations of protein,TNF-α,IL-1β,IL-6 and HMGB1,and PMN count in BALF,lung injury score,W/D ratio,MPO and caspase-3 activities and AI in group H2 (P > 0.05),and PaO2/FiO2 was significantly decreased,the concentrations of total protein,TNF-α,IL-1β,IL-6 and HMGB1,and PMN count in BALF,lung injury score,W/D ratio,MPO and caspase-3 activities,and Al were increased in groups ALI and ALI + H2 (P < 0.05).Compared with group ALI,PaO2/FiO2 was signifieantly increased,the concentrations of total protein,TNF-α,IL-1β,IL-6 and HMGB1,and PMN count in BALF,lung injury score,W/D ratio,MPO and caspase-3 activities,and AI were decreased in group ALI+ H2 (P < 0.05).Conclusion Hydrogen-rich saline can alleviate LPS-induced ALI via inhibition of inflammatory response and apoptosis in mice.
9.Effects of hydrogen gas on lipopolysaccharide-induced damage to human umbilical vein endothelial cells

Objective To evaluate the effects of hydrogen gas (H2) on lipopolysaccharide (LPS)-induced damage to human umbilical vein endothelial cells (HUVECs) in vitro.Methods HUVECs were cultured in DMEM culture medium containing 10% fetal bovine serum.The cells were seeded in 6-well plates (2 ml/hole) at a density of 2 × 106 cells/ml and randomly divided into 4 groups (n =35 each) using a random number table:control group (group C),group H2,LPS group and LPS + H2 group.The cells were cultured in the plain culture medium in C and LPS groups or in hydrogen-saturated culture medium in H2 and LPS + H2 groups.In addition,LPS 1 μg/ml was added simultaneously in LPS and LPS + H2 groups,and the equal volume of normal saline was added instead in C and H2 groups.The cells were incubated for 24 h.At 6,12 and 24 h of incubation,the cells of 5 wells were chosen and stained with Wright-Giemsa to observe the destruction of HUVEC barrier.At 6,12 and 24 h of incubation,the cells of 5 wells were chosen to detect the expression of VE-cadherin and β-catenin using Western blot.At 24 h of incubation,the cells of 5 wells were chosen to detect the expression of VE-cadherin and βcatenin using immunofluorescence.Results Pathological changes of endothelial cells were observed in LPS group.The pathological changes of the cells were lighter in LPS + H2 group than in LPS group.Compared with group C,VE-cadherin expression was significantly down-regulated (P < 0.05),while no significant change was found in β-catenin expression in LPS and LPS + H2 groups (P > 0.05).Compared with group LPS,VE-cadherin expression was significantly up-regulated (P < 0.05),while no significant change was found in ~catenin expression in group LPS + H2 (P > 0.05).Conclusion H2 can effectively reduce LPS-induced damage to HUVECs through inhibiting down-regulation of VE-cadherin expression.
10.Effect of hydrogen on acute lung injury in septic mice

ObjectiveTo investigate the effect of hydrogen (H2 ) on acute lung injury (ALI) in septic mice.MethodsOne hundred and twelve male C57BL/6 mice,aged 5 weeks,weighing 20-25 g,were randomly divided into 4 groups ( n =28 each):sham operation group (group A),sham operation + H2 group (group B),sepsis group (group C) and sepsis + H2 group (group D).Sepsis was produced by cecum ligation and puncture (CLP).Groups B and D received 1 h inhalation of 2% H2 at 1 and 6 h after CLP operation or sham operation.Twenty animals in each group were selected and observed for the 7 d survival rate.The left 8 animals in each group were sacrificed at 24 h after CLP operation.Venous blood samples and lung tissues were obtained to determine the levels of superoxide dismutase (SOD),catalase (CAT) and 8-iso-prostaglandin F2alpha (8-iso-PGF2α) in the serum and lungs,the concentration of protein in bronchoalveolar lavage fluid (BALF),and the activity of myeloperoxidase (MPO) in the lungs.The lung injury score (LIS) was assessed and W/D lung weight ratio was calculated.ResultsCompared with group A,the 7 d survival rate and activities of SOD and CAT in the serum and lungs were significantly decreased,and LIS,W/D ratio,the concentration of protein in BALF,MPO activity and 8-iso-PGF2α level in the serum and lungs were significantly increased in group C ( P < 0.05 ).Compared with group C,the 7 d survival rate and activities of SOD and CAT in the serum and lungs were significantly increased,and LIS,W/D ratio,the concentration of protein in BALF,MPO activity and 8-iso-PGF2α level in the serum and lungs were significantly decreased in group C ( P < 0.05).ConclusionH2 can alleviate ALI in septic mice via inhibiting oxidative stress response.
