Objective To analyze the correlation of perfusion parameters obtained by functional multi-slice computed tomography (fMSCT) with vascular endothelial growth factor (VEGF) detected with immunohistochemistry on rabbit model with VX2 breast cancer and noninvasively evaluate the vascularization of untreated VX2 breast cancer in vivo. Methods Sixteen New Zealand femal rabbits were selected and suspension (1 ml) of mass was injected around the breast areola. CT perfusion was performed after two weeks and perfusion parameters including blood flow (BF), blood volume (BV), mean transit time (MTT)and permeability surface (PS) were assessed. Expression of VEGF in neoplasm was detected with immunohistochemistry. Paried t test was used for the comparison of perfusion parameters between the tumor and muscle and Pearson correlation analysis was used to assess the correlation of VEGF with perfusion parameters. Results The mean value of BF, BV, MTT and PS were (228.21 ± 13. 13 ) ml · min-1100 mg-1 ,(13.45 ± 1.01) ml · 100 mg-1 ,(3. 50 ±0. 20) sand (7.85 ±1.18) ml · min-1 · 100 mg-1 in tumor, respectively. They were (66. 10 ±22. 11 ) ml · min-1 · 100 mg-1 , ( 1.88 ± 1.80) ml · 100 mg-1,(23. 87 ±0. 63)s,(1.55 ±0. 38)ml · min-1 · 100 mg-1 in muscular tissue, respectively. The mean value of BF, BV and PS in tumor were obviously higher than those in muscle, and the mean value of MTT in tumor was lower than that in muscle. There were significant differences in CT perfusion parameters between tumor and muscle (t = 61.83,13.63,27.72,20. 54, P < 0. 01 ). The mean value of VEGF in tumor was 7. 33 ±0. 27 and there were positive correlation with BF ( r = 0. 712, P < 0. 01 ), BV ( r = 0. 647, P < 0. 01 ), PS ( r =0. 627 ,P < 0. 01 ), and negative correlation with MTT ( r = - 0. 564, P < 0. 05 ). Conclusion MSCT perfusion imaging can be used to noninvasively evaluate the vascularization in rabbits with untreated VX2 breast cancer in vivo.

BACKGROUND: The mitochondrial DNA with multiple copy number, maternal inheritance, high mutation rate, restructuring and other rare features, has important significance at levels of molecular ecology, molecular genetics, forensic and anthropological research.OBJECTIVE: To comprehensively analyze mitochondrial DNA in the development of research methods, and to explore the application of anthropological research.METHODS: With the key words of mitochondrial DNA, polymorphism, and haplogroup in English, a computer-based online search was conducted in Pubmed database from January 1999 to April 2009. Anthropology relevant articles were included, but animal experiments were excluded.RESULTS AND CONCLUSION: Among the first inspection of 235 documents, 33 met the inclusion criteria. Mitochondrial DNA characterized by multiple copy number, maternal inheritance, high mutation rate, and very few re-occurring features, thus by analyzing the scope and frequency of information, it could be used to infer the relationship between the different populations, as well as the reconstruction of mass incidents. Mitochondrial DNA variation information extraction method had experienced low RFLP, high-resolution RFLP, sequencing and RFLP and sequencing of hypervariable region. Mitochondrial DNA polymorphisms played an important role in interfering phylogenetic relationships, different groups of national origin and migration routes.Mitochondrial DNA database of various groups around the world did not restrict the full application in anthropology. Mathematical statistical model which was used to improve existing, build and enrich various groups of mitochondrial DNA database all over the world is the main direction for future research. Mitochondrial DNA is significance in molecular ecology, molecular genetics, forensic science and anthropology, as well as the study of tissue engineering, especially in the study of human origins and comparative phylogenetic relationships of different populations.

Objective To explore the influence on isthmic spondylolisthesis (IS) from lumbar vertebras facet joint orientation (FJO) variation. Methods The lumbar vertebras of 60 patients scanned by 16-slices spiral CT were collected, 30 cases with IS was defined as isthmic group;30 cases with normal lumbar vertebras was defined as control group, the FJO at L3-4, L4-5 and L5-S1 were measured in two groups. The difference between two groups were compared at three levels, the difference between both sides of the facet joint orientation also was compared;the measured data and the data of 60 patients from foreign literature were compared at three levels. Results The FJO in isthmic group were (47.9±6.3)° ,(37.5±7.3)° , (37.9±7.7)° at the right of L3-4, L4-5, L5-S1 levels respectively, the FJO in control group were (53.1 ± 7.3)° , (40.5±6.3)° , (38.5±7.3)° respectively, the FJO in isthmic group were (48.1±6.0)° , (37.9 ± 7.4)° , (37.6 ± 7.6)° at the left of L3-4, L4-5, L5-S1 levels respectively, the FJO in control group were (52.3 ± 7.6)° , (41.6 ± 6.0)° , (38.2 ± 7.2)° respectively. The significant difference was found at L3-4 and L4-5 levels (P < 0.05), the orientation was similar at L5-S1 level (P > 0.05). The difference between the both sides FJO of lumbar vertebras was not found at L3-4,L4-5 and L5-S1 levels (P >0.05). The same as the measured data and the corresponding data from foreign literature(P > 0.05 ). Conclusions It seems possible that the coronal FJO may be the phenotypic representation of the familial etiology of IS. It is helpful for the measurement of lumbar vertebras FJO to find IS early,it is important to reduce and release the IS.

BACKGROUND: Polymorphism in promoter region can change the expression of genes, which may be associated with susceptivity of diseases. Gene polymorphism of interleukin-6 (IL-6) promoter is associated with nationality and many diseases. Different nationalities often display different characteristics of gene polymorphism.OBJECTIVE: To observe the distribution of polymorphism at sites -597 and -572 in IL-6 promoter region in Tibet Tibetan population and to provide the theoretical data for Tibetan population genetics and background of immunity.DESIGN: Randomized investigation.SETTING: Institute of Anthropology, Jinzhou Medical College.PARTICIPANTS: Totally 108 healthy Tibetan teenagers were selected from Lasa and Naqu region in Tibet autonomous region from October 2003 to July 2004, including 60 males and 48 females, aged from 14-21 years. Inclusive criteria:The parents of the volunteers were healthy Tibetans after body examination. The volunteers knew the fact, agreed to participate into the trail and signed the informed consent.METHODS: 5 mL peripheral vein blood was collected from 108 Tibetan teenagers. DNA from human leucocytes was extracted by salt fractionation. IL-6 promoter including -597 and -572 fragments was amplified by polymerase chain reaction (PCR). Representative fragments were cloned then sequenced after restriction fragment length polymorphism (RFLP).MAIN OUTCOME MEASURES: Distribution of polymorphism in Tibet Tibetan population; Results after comparison with those of other nationalities including Han population.RESULTS: Data of 108 Tibetan teenagers were involved in the result analysis. ①Distribution of polymorphism on -572C/G site of IL-6 promoters in population of either sex: There were no GA and AA genotypes at site -597, but only GG genotype appeared. There were CC, CG and GG genotypes at site -572, and the frequencies were 0.63, 0.35 and 0.02 in order. Distribution of genotype with representativeness met the Hardy-Weinberg equilibrium. Allele frequencies were 0.81 and 0.19, respectively. There was no significant difference of either sex in genotype and allele frequency (P > 0.05). ②Distribution of polymorphism on -597 G/A and -572 C/G in different nationalities: GG, GA, AA genotypes appeared on -597 site in England and France, and G and A allele frequencies were 0.60 and 0.40,respectively. It was significantly different from that of Tibetan in Tibet. Furthermore, Japanese had no polymorphism, which was similar to that of Hans in China (P > 0.05). ③Genotype of different straps and results of DNA sequencing:Only GG genotype was found on -597 site (without the restriction site, one fragment after restriction, PCR amplification products), no GA and AA genotypes. CC, CG and GG genotypes appeared at site -572, and frequencies were 0.64,0.35 and 0.01, respectively. Distribution of genotype with representativeness met the Hardy-Weinberg equilibrium (P >0.05). Allele frequencies were 0.81 and 0.19, respectively. There was no significant difference of either sex in genotype and allele frequency. Distribution of gene frequency and allele frequency in IL-6 were similar between Tibetan and that of Hans, but it was significantly different from that of population in England, France and America.CONCLUSION: There are nationality differences of IL-6 gene polymorphism at sites -597 and -572. No polymorphism is found at site -597 in Tibetan. Race differences are seen at site -572, having CC, CG and GG genotypes and G allele is rate. Compared with white population, there is significant difference in genotype and allele frequency at site -572. Their characteristics are close to Hah population and Japanese, which may be associated with genetic gene of persons living in plateau.
