1.Effects of three sterilization methods on the magnetic flux of magnetic surgical devices and analysis of sterilization cost

		                        			Objective To analyze the effects of three sterilization methods,namely,pressure steam,low-temperature plasma and ethylene oxide,on the magnetic flux of magnetic surgical devices and their sterilization costs.Methods A total of 234 magnetic surgical devices of different specifications and models(magnetic rings)were randomly divided into Group A,Group B and Group C after the paired number was labelled,and each group consisted of 78 pieces(39 pairs).After packaging each pair of devices according to sterilization specifications,Group A was sterilized by pressure steam,Group B was sterilized by low-temperature plasma,and Group C was sterilized by ethylene oxide.We measured the magnetic flux of three sets of magnetic rings before and after sterilization,and comparatively analyzed the sterilization cost and sterilization time of the single package.Results There was no statistically significant difference in the impact of the three sterilization methods on the magnetic flux of the magnetic surgical devices(P>0.05),but there was a significant difference in the magnetic flux before and after sterilization for each sterilization method(P<0.001);the sterilization cost was(1.96±0.16)yuan for Group A,(23.17±0.32)yuan for Group B,and(8.16±0.18)yuan for Group C,showing statistically significant differences among the three groups(P<0.01).The sterilization time was(65.21±3.36)min for Group A,(45.46±1.39)min for Group B,and(1020.38±12.21)min for Group C,with statistically significant differences among the three groups(P<0.01).Conclusion None of the three sterilization methods affects the magnetic flux of the magnetic surgical devices.Pressure steam method shows the lowest cost of single package,low-temperature plasma method shows the highest cost of single package,while ethylene oxide method shows the highest sterilization time.Pressure steam should be the preferred sterilization method for magnetic surgical devices.
		                        		
		                        		
		                        		
		                        	
2.Clinical characteristics and genetic analysis of two children with Autosomal dominant mental retardation type 21 due to variants of CTCF gene.

		                        			OBJECTIVE:
		                        			To explore the clinical and genetic characteristics of two children with developmental delay.
		                        		
		                        			METHODS:
		                        			Two children who had presented at the Children's Hospital Affiliated to Shandong University on August 18, 2021 were enrolled as the study subjects. Clinical and laboratory examination, chromosomal karyotyping and high-throughput sequencing were carried out for both children.
		                        		
		                        			RESULTS:
		                        			Both children had a 46,XX karyotype. High-throughput sequencing showed that they have respectively carried a c.489delG (p.Q165Rfs*14) and a c.1157_1158delAT (p.Y386Cfs*22) frameshifting variant of the CTCF gene, both had a de novo origin and were unreported previously.
		                        		
		                        			CONCLUSION
		                        			The CTCF gene variants probably underlay the development delay in the two children. Above discovery has enriched the mutational spectrum of the CTCF gene and has important implications for revealing the genotype-phenotype correlation for similar patients.

3.Eukaryotic expression,purification and activity identification of human His-GRK2 recombinant protein

		                        			Objective:
		                        			To construct a human  G protein-coupled receptor kinase 2   ( GRK2) eukaryotic expression system.
		                        		
		                        			Methods:
		                        			The  primers  were  designed ,and  the  His-GRK2  target  gene  was  amplified  by  PCR  using  the  Pans-EGFP-GrK2  (full-length) gene as the template.The His-GRK2 target gene was connected to the pcDNA3.1EGFP eukaryotic  expression  vector. The  pcDNA3. 1-EGFP-His-GRK2  plasmid was transfected into  HEK  293T  cells.48 h later,the expression of GRK2 protein was detected by Western blot,and the GRK2 protein was purified  by nickel chelated magnetic bead method.The purification of GRK2 protein was detected by Coomassie bright blue  staining and Western blot,and the activity of GRK2 protein was detected by His pull down.  
		                        		
		                        			Results :
		                        			The results of double enzyme digestion and sequencing  showed that pcDNA3. 1-EGFP-His-GRK2  eukaryotic  expression plasmid  was successfully constructed.Western blot analysis showed that the molecular weight of GRK2 protein was about 80  ku,indicating that GRK2 protein was successfully expressed in HEK 293T cells  (t = 6. 433,P = 0. 003) .GRK2  protein was purified by nickel chelated magnetic beads.His pull down experiment results showed that GRK2 was  bound to prostaglandin E2 receptor 4 (EP4) ,suggesting that GRK2 protein had biological activity  (t = 13. 5,P =  0. 000 2) .
		                        		
		                        			Conclusion
		                        			The pcDNA3.1-EGFP-His-GRK2  eukaryotic expression plasmid was correctly sequenced  and the GRK2 recombinant plasmid was successfully constructed.The GRK2 recombinant plasmid was successfully  expressed in eukaryotic cells HEK 293T and the protein expressed was biologically active.
		                        		
		                        		
		                        		
		                        	
4.Analysis of clinical and genetic characteristics of a child with ring chromosome 4 syndrome.

		                        			To explore the genetic basis for a child featuring short stature.
		                        		
		                        			METHODS:
		                        			G-banded karyotyping, chromosomal microarray analysis (CMA) and high-throughput sequencing were carried out on peripheral blood sample from the child.
		                        		
		                        			RESULTS:
		                        			The karyotype of the child was ascertained as 45,XY,-4[3]/46,XY,r(4)(p16q35)[84]/47,XY,-4,r(4)(p16q25)*2[7]/48,XY,-4,r(4)(p16q35)*3[1]/46,XY,dic r(4;4)(p16q35;p16q35)[2]/46,XY,add(4)(p16)[3]. A 647 kb deletion at 4p16.3 was identified by CMA, which encompassed 6 OMIM genes including ZNF141, PIGG, PDE6B, ATP5I, PCGF3 and MYL5. High-throughput sequencing has identified no pathogenic/likely pathogenic variants consistent with the clinical symptoms.
		                        		
		                        			CONCLUSION
		                        			A rare ring chromosome 4 syndrome was identified by combined chromosomal karyotyping, CMA and high-throughput sequencing. Conventional cytogenetic analysis and genetic testing in combine have enabled the diagnosis in this case.
		                        		
		                        		
		                        		
		                        	
6.Correlation between aortic elasticity and coronary artery calcification evaluated by CT

		                        			Objective To discuss the correlation between aortic elasticity and coronary artery calcification by CT.Methods Totally 111 patients who were diagnosed of coronary artery disease underwent coronary artery CTA.The images were qualified for aortic elasticity measurement.All patients were divided into calcification negative group (n=43) and calcification positive group (n =68).The calcification positive group was further divided into light,medium,and serious groups according to their calcification scores.The ascending aortic images were reconstructed every 5 % R-R intervals.The cross-sectional areas and diameters of aortic in each R-R interval were measured automatically,then diameter variation rate (% A0),aortic distensibility (A0D),aortic compliance (A0C) and aortic stiffness (A0SI) were calculated to evaluate aortic elasticity.Correlation between aortic elasticity and coronary artery calcification were analyzed.Results % A0,A0 D,A0C were lower and A0SI was higher in calcification positive group than those in calcification negative group (all P<0.05).There was no significant differences in the four reference indexes of aortic elasticity among light,medium,and serious groups in calcification positive group (all P>0.05).Correlation analysis demonstrated negative correlations between % A0,A0 D,A0 C and calcification scores,and a positive correlation between A0SI and calcification scores.Conclusion Aortic elasticity is correlated with coronary artery calcification,and the combination of them will be beneficial for the early diagnosis of coronary heart disease.
		                        		
		                        		
		                        		
		                        	
7.Compatible Stability of Levetiracetam Injection with Three Injections

		                        			OBJECTIVE:To investigate the compatible stability of Levetiracetam(Lev)injection with 3 injections. METHODS:Each Lev injection 1000 mg mixed with 0.9% Sodium chloride injection 100 mL,5% Glucose injection 100 mL or Sodium lactate Ringer's injection 100 mL respectively. Under the light condition,at 25 ℃,the color and clarification degree of mixtures were ob-served at different time points within 24 h after mixing;pH value and the number of insoluble particles were determined. The contents of related impurities(impurity A,B,C,D,2-hydroxypyridine)and Lev in mixtures were determined by HPLC. RESULTS:Under above condition,all mixtures were colorless clear liquid within 24 h;pH value had no significant change (RSD<1%,n=7);the number of insoluble particles was no more than the range stated in Chinese Pharmacopeia(2015 edition). Impurity B and C were not detected;the contents of other impurities were in line with the requirements of foreign pharmacopeia. No marked change was noted for relative content of Lev(RSD<1%,n=7). CONCLUSIONS:After mixing with 0.9% Sodium chloride injection,5% Glucose injec-tion or Sodium lactate Ringer's injection,Lev injection keep stable at 25℃within 24 h under the light condition.

9.The appliacation of retrospective ECG-gated 256 slices spiral CT technique in inferior phrenic artery angiography

		                        			Objective To evaluate the value of retrospective ECG-gated 256 slices spiral CT technique in the scanning of inferior phrenic artery (IPA).Methods 80 patients with underdone abdominal CTA were divided randomly into two groups (40 patients each)as regular abdominal CTA scanning group and retrospective ECG-gated technique group.And further analysis was done to evaluate the display of IPA branches and the image quality.Results 80 patients were all scanned successfully.40 cases LIPA and 40 cases RIPA were showed in the regular group.80 cases showed 1st level branches of IPA,73 cases with 2nd level,59 cases with 3rd and 43 ca-ses showing 4th level.40 cases LIPA and 40 cases RIPA were showed in ECG-gated group.And the numbers of IPA branches levels were 80,79,71,65,respectively.There is no significant difference in the ability of showing the1st level IPA or their image quality between those two groups (P >0.05);and there is a statistical significance in the ability of showing other levels of IPA and their im-age quality.Conclusion It is feasible that we use retrospective ECG-gated 256 slices spiral CT scanning technique to show the IPA. 10.Colonoscopic and endoscopic ultrasonography diagnosis of appendix mucocele

		                        			Objective To investigate the features and clinical value of endoscopic ultrasonography and colonoscopy for appendix mucocele. Methods The patients with mucocele of appendix who were diagnosed by endoscopic ultrasonography and colonoscopy in three hospitals all underwent surgery from January 2008 to March 2015. Appendix mucocele in these patients was confirmed by postoperative pathology and clinical data were retrospectively analyzed. Results A total of 22 patients with appendix mucocele were analyzed retrospectively. The average size of intralumen mucocele was 1. 84±1. 42 cm (0. 6-4. 5 cm) . The colonscopic finding of appendiceal mucocele showed submucosal protuberance at the appendiceal orifice with smooth surface. The appendiceal orifice was found at the edge of appendiceal mucocele. Endoscopic ultrasonogrphy showed low echo with smooth cyst wall in 8 patients, mixed equal echo and low echo in 14 cases. Appendicectomy was performed in 11 patients and resection of ileocecum in 11 others. Conclusion Endoscopic ultrasonography and colonoscopy are valuable for diagnosis and treatment in appendiceal mucocele.
