1.Expression of miR-33a-5p and SIRT6 in the serum of esophageal cancer patients and their relationship with clinical pathological characteristics

Objective To investigate the expression levels of serum microRNA-33a-5p(miR-33a-5p)and Sirtuin6(SIRT6)in patients with esophageal cancer,and their relationship with clinical pathological characteristics and prognosis.Methods From March 2018 to March 2020,123 newly diagnosed patients with esophageal cancer(esophageal cancer group)and 94 patients with esophagitis(benign lesion group)admitted to our hospital were collected as the research subjects,another 72 healthy individuals who underwent physical examination were regarded as the health control group;the clinical and pathological characteristics of esophageal cancer patients were collected.Real-time fluorescence quantitative PCR method was applied to detect the expression of miR-33a-5p and SIRT6 mRNA in three groups of serum,the relationship between the expression levels of serum miR-33a-5p and SIRT6 mRNA in esophageal cancer patients and clinical pathological characteristics was analyzed;TargetScanHuman website was applied to predict the targeting relationship between miR-33a-5p and SIRT6;Pearson method was applied to analyze the correlation between serum miR-33a-5p and SIRT6 mRNA expression levels in esophageal cancer patients;Kaplan-Meier method was applied to analyze the relationship between the expression levels of serum miR-33a-5p and SIRT6 mRNA in esophageal cancer patients and their prognosis;Cox regression was applied to analyze the prognostic factors of esophageal cancer patients.Results The expression level of serum miR-33a-5p in the esophageal cancer group was greatly higher than that in the benign lesion group and healthy control group(P<0.05),the expression level of serum SIRT6 mRNA was greatly lower than that of the benign lesion group and healthy control group(P<0.05);TargetScanHuman website predicts that there was a targeted binding site between miR-33a-5p and SIRT6,and there was a negative correlation between serum miR-33a-5p and SIRT6 mRNA expression levels in esophageal cancer patients(r=-0.468,P<0.05);the expression levels of serum miR-33a-5p and SIRT6 mRNA in esophageal cancer patients were related to TNM staging,lymph node metastasis,and degree of differentiation;the 3-year survival rate of esophageal cancer patients with low expression of serum miR-33a-5p and high expression of SIRT6 mRNA was greatly higher than that of esophageal cancer patients with high expression of miR-33a-5p and low expression of SIRT6 mRNA(P<0.05);Cox regression analysis showed that high expression of miR-33a-5p and low expression of SIRT6 mRNA were independent risk factors for death in esophageal cancer patients(P<0.05).Conclusion The expression level of serum miR-33a-5p in esophageal cancer patients is greatly increased,while the expression level of SIRT6 mRNA is greatly reduced;the expression levels of both are closely related to the clinical pathological characteristics and prognosis of esophageal cancer patients.
2.Correlation and significance of the C-reactive protein to albumin ratio,angiopoietin-2 and receptor for advanced glycation end product with the outcomes of patients infected with carbapenem-resistant Enterobacteriaceae

Objective To explore the correlation and significance of the C-reactive protein/albumin ratio(CRP/ALB),angiopoietin-2(Ang-2),and receptor for advanced glycation end product(RAGE)with the outcomes of patients infected with carbapenem-resistant Enterobacteriaceae(CRE).Methods A total of 103 patients with severe and long-term repeated CRE infections who were admitted to the Sengong Hospital of Shaanxi Province from January 2020 to May 2023 were selected as the research subjects.The patients were divided into poor outcome group(n=23)and good outcome group(n=80)according to the different outcomes within 28 days.The clinical data of patients were collected by reviewing the electronic medical record system,including age,gender,body mass index,medical history,bacterial species,body temperature,whether invasive mechanical ventilation was performed,and whether complicated infectious shock.The serum CRP level was detected with a fully automated electrochemiluminescence immunoassay analyzer,the ALB level was detected with a fully automated biochemical analyzer,and the CRP/ALB was calculated;the levels of serum Ang-2 and RAGE were detected using a multi-function microplate reader.Clinical data and levels of CRP/ALB,Ang-2,and RAGE before treatment,3 days and 7 days after treatment were compared between the two groups.The influencing factors associated with the outcomes of patients with CRE infection were analyzed using Cox regression.The predictive value of CRP/ALB,Ang-2 and RAGE levels before treatment for outcomes of patients with CRE infection was analyzed using the receiver operating characteristic curve.The prognosis at 28 days of patients with different levels of CRP/ALB,Ang-2,and RAGE was analyzed using the Kaplan-Meier curve.Results The proportions of patients with invasive mechanical ventilation and infectious shock in the poor outcome group were significantly higher than those in the good outcome group(P<0.05).The CRP/ALB,Ang-2,and RAGE levels were significantly lower after 3 and 7 days of treatment than those before treatment and were significantly lower after 7 days of treatment than those after 3 days of treatment in the good outcome group(P<0.05).There was no significant difference in CRP/ALB,Ang-2,and RAGE levels at different time points in the poor outcome group(P>0.05).The C RP/ALB,Ang-2,and RAGE levels in the poor outcome group were significantly higher than those in the good outcome group after 3 and 7 days of treatment(P<0.05).Invasive mechanical ventilation,complicated with infectious shock,and increased CRP/ALB,Ang-2 and RAGE levels were independent risk factors for the outcome of CRE infection(P<0.05).After 7 days of treatment,CRP/ALB+Ang-2+RAGE had the highest area under the curve(0.931)for predicting the outcome of CRE infection,with a sensitivity of 86.96%and a specificity of 88.75%.The survival rates of patients in the high-expression subgroups of C RP/ALB,Ang-2,and RAGE were significantly lower than those in the low-expression subgroups(P<0.05).Conclusion CRP/ALB,Ang-2 and RAGE are related to the outcome of CRE infection.Combined detection can be used as an early prediction scheme for the outcome of CRE infection,providing accurate and quantified guidance information for clinical stratified management.

Patients with severe trauma require an extremely timely treatment and transfusion plays an irreplaceable role in the emergency treatment of such patients. An increasing number of evidence-based medicinal evidences and clinical practices suggest that patients with severe traumatic bleeding benefit from early transfusion of low-titer group O whole blood or hemostatic resuscitation with red blood cells, plasma and platelet of a balanced ratio. However, the current domestic mode of blood supply cannot fully meet the requirements of timely and effective blood transfusion for emergency treatment of patients with severe trauma in clinical practice. In order to solve the key problems in blood supply and blood transfusion strategies for emergency treatment of severe trauma, Branch of Clinical Transfusion Medicine of Chinese Medical Association, Group for Trauma Emergency Care and Multiple Injuries of Trauma Branch of Chinese Medical Association, Young Scholar Group of Disaster Medicine Branch of Chinese Medical Association organized domestic experts of blood transfusion medicine and trauma treatment to jointly formulate Chinese expert consensus on blood support mode and blood transfusion strategies for emergency treatment of severe trauma patients ( version 2024). Based on the evidence-based medical evidence and Delphi method of expert consultation and voting, 10 recommendations were put forward from two aspects of blood support mode and transfusion strategies, aiming to provide a reference for transfusion resuscitation in the emergency treatment of severe trauma and further improve the success rate of treatment of patients with severe trauma.
4.Analysis and reflection on the effect difference between the two teaching modes caused by the difference of discipline nature

In order to further cultivate bioscience talents, we compared and analyzed the teaching effects of different teaching modes of Bioscience courses in our university. Under the condition that the difficulty of the examination subjects and papers remains unchanged, we collected and compared the examination results of the online teaching mode covering three grades of students with the corresponding examination results of the traditional teaching mode of the previous academic year. After comparing the final examination results of 21 courses, we found that the online teaching effect of examination courses with long courses and experimental courses is better (such as physiology, pathology, etc.). The phenomenon that online teaching cannot be carried out normally has affected most students (66.28%, 57/86), indicating that the experimental course cannot be replaced by online teaching. At the same time, students who expected systematic teaching reform of biological science major courses accounted for the majority in the survey (80.23%, 69/86). Based on this, we conclude that the mixed teaching mode with traditional teaching as the main body of teaching and online platform technology as the auxiliary can become a new exploration direction in undergraduate teaching of Bioscience majors in medical colleges and universities.
5.Effect of KIF22 on promoting proliferation and migration of gastric cancer cells via MAPK-ERK pathways.

BACKGROUND:
Gastric cancer (GC) is one of the most globally prevalent cancers in the world. The pathogenesis of GC has not been fully elucidated, and there still lacks effective targeted therapeutics. The influence of altered kinesin superfamily protein 22 (KIF22) expression in GC progression is still unclearly. The aim of this study was to investigate the KIF22 effects on GC and related mechanisms.
METHODS:
Gastric carcinoma tissues and matching non-cancerous tissues were collected from patients with GC who have accepted a radical gastrectomy in Lanzhou University Second Hospital from May 2013 to December 2014. The expression of KIF22 was examined in GC of 67 patients and 20 para-carcinoma tissues by immunochemical staining. The relationship between the expression of KIF22 and clinicopathologic characteristics was next investigated in the remaining 52 patients except for 15 patients who did not complete follow-up for 5 years. Cell viability was performed via 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) test and colony formation assay in the MGC-803 and BGC-823 GC cells. Cell scratch and trans-well invasion assay was performed to assess migration ability in the MGC-803 and BGC-823 GC cells. Gene set enrichment analysis (GSEA) pathway enrichment analysis was performed to explore the potential functions. Cell cycle was detected by flow cytometry. In addition, the two GC cell lines were used to elucidate the underlying mechanism of KIF22 in GC in vitro via assessing the effects on mitogen-activated protein kinase and extracellular regulated protein kinases (MAPK/ERK) signal transduction pathway-related expressions by Western blotting assays. The differences were compared by t tests, one-way analysis of variance, and Chi-squared tests.
RESULTS:
The study showed that KIF22 was up-regulated in GC, and KIF22 high expression was significantly related to differentiation degree (χ = 12.842, P = 0.002) and poorly overall survivals. GSEA pathway enrichment analysis showed that KIF22 was correlated with the cell cycle. Silence of KIF22 decreased the ability of the proliferation and migration in gastric cells, induced G1/S phase cell cycle arrest via regulating the MAPK-ERK pathways.
CONCLUSIONS
KIF22 protein level was negatively correlated with prognosis. KIF22 knockdown might inhibit proliferation and metastasis of GC cells via the MAPK-ERK signaling pathway.
6.Therapeutic Efficacy of Ezetimibe Combined with Tirofiban in the Treatment of Acute Coronary Syndrome Patients and Its Influence on the Content of Ischemia Modified Albumin

OBJECTIVE:To investigate therapeutic efficacy of ezetimibe combined with tirofiban in the treatment of acute coronary syndrome patients and its influence on the content of ischemia modified albumin (IMA) in peripheral blood.METHODS:A total of 82 patients with acute coronary syndrome in our hospital from Jan.2016 to Jan.2017 were divided into observe group and control group according to random number table,with 41 cases in each groups.All patients were given general treatment containing statins,glyceryl trinitrate and so on.Control group was additionally given Ezetimibe tablet 10 mg orally once a day.Observation group was additionally given Tirofiban hydrochloride sodium chloride injection 5 mg intravenously once a day,on the basis of control group,for consecutive 15 d.Clinical efficacies of 2 groups were evaluated.The dose of glyceryl trinitrate,duration and frequency of angina pectoris attack,left ventricular function indexes (SV,LVDD,CO,LVEF) were compared before treatment and after last medication.The contents of IMA in peripheral blood were detected before treatment and 3,6,9,12,15 h after the first day medication.ADR of the 2 groups were observed closely during the treatment.RESULTS:The total response rate of observation group was 92.7%,which was significantly higher than 72.3% of control group (P<0.05).Compared with before treatment,dose of glyceryl trinitrate and the content of IMA in 2 groups were decreased significantly after last medication,duration and frequency of angina pectoris attack were shortened significantly;the levels of SV,CO and LVEF were increased significantly while the level of LVDD was decrease significantly;the effect of observation group was significantly better than that of control group,with statistical significance (P<0.05).No severe ADR was found in all patients during the treatment.CONCLUSIONS:Based on general treatment of glyceryl trinitrate therapeutic efficacy of ezetimibe combined with tirofiban is better than that of ezetimibe alone in the treatment acute coronary syndrome patients,and can decrease the content of IMA in peripheral blood more quickly.
7.Application of wireless wearable intelligent body temperature monitoring system in temperature monitor-ing of patients with abdominal surgery

Objective To evaluate the effect of wireless wearable intelligent body temperature monitoring system on temperature monitoring of perioperative patients. Method The body temprature of 572 patients with abdoiminal surgery who hospitalized in our hospital from June 2017 to Juanuary 2018 was measured by mercury thermometers and wireless intelligent body temprature monitoring system four times a day for three days with toally 6864 times after surgery.The measured time and data by both ways were compared and its relativity were analyzed. Results The measured data by mercury thermometers and wireless intelligent body temprature monitoring system was compared with no statistical significance (P>0.05), but the time measured by wireless intelligent body temprature monitoring system was shorter (P<0.05). The measured data of two ways was postively related (r=0.962,P<0.05). Conclusions The measurement results by the wireless wearable intelligent body temperature monitoring system are the same as by mercury thermometers. The accuracy of the system is well validated, and it can improve the working efficiency of nurses.
8.Mercury accumulation of Tibetan medicine Zuotai in mice

AIM To explore the mercury accumulation in KM mice after being given Zuotai at different doses and time.METHODS KM mice were randomly divided into blank group,Zuotai low-,middle-and high-dose (6.07,60.70 and 606.97 mg/kg,42 d;606.97 mg/kg,14 d) groups.The mercury contents in brain (olfactory bulb,cortex,hippocampus,hypothalamus,brain stem,cerebellum),heart,lung,kidney,liver,spleen,serum,muscle of mice were measured after administration.RESULTS Compared with the blank group,Zuotai at low-dose significantly increased the mercury contents in hippocampus,cerebellum,lung,kidney,liver and serum of mice after 42-day treatment;Zuotai at middle-dose markedly increased the mercury contents in olfactory bulb,cortex,hippocampus,brain stem,cerebellum,heart,lung,kidney,liver,spleen and serum of mice after 42-day treatment;the mice treated with high-dose of Zuotai for 42,14 days significantly increased the mercury contents in olfactory bulb,cortex,hippocampus,hypothalamus,brain stem,cerebellum,heart,lung,kidney,liver,spleen,muscle and serum.CONCLUSION Mercury can be accumulated in different tissues of mice after intragastric administration of Zuotai in a dose-and time-dependent manner,which suggests that Zuotai and its compound preparations should not be used in high-dose and long-term.
9.Application of atherogenic index of plasma and intima-media thickness of carotid artery in renal arteriolar sclerosis patients with chronic renal failure

Objective To evaluate the application of atherogenic index of plasma (AIP) and intima-media thickness of carotid artery (CA-IMT) in renal arteriolar sclerosis patients with chronic renal failure.Methods One hundred and twenty nine patients with chronic renal failure patients underwent ultrasound-guided percutaneous renal biopsy from October 2013 to June 2014,the biopsy results showed that renal arteriolar sclerosis was identified in 72 patients (atherosclerosis group) and no renal arterioles sclerosis was detected in 57 patients (non-atherosclerosis group);71 healthy adults were enrolled in the study as controls.The age,height,body weight,systolic and diastolic blood pressure,the indexes of blood lipid and renal function were documented and compared among three groups.The correlation of AIP and CBMmax of common carotid artery and carotid bifurcation with blood lipid level and renal function was analyzed.Results There was significant difference in body weight among patients with atherosclerosis [(70.77 ± 14.27) kg],without atherosclerosis [(60.63 ± 12.12) kg] and the controls [(64.20 ± 8.13) kg] (t =3.071,3.391,all P < 0.05).The TG [(2.43 ± 1.61) mmol/L vs.(1.02 ± 0.37) mmol/L],TC [(7.40 ± 8.80) mmol/L vs.(4.53 ±0.67)mmol/L],LDL-C[(4.40 ±2.13) mmol/L vs.(2.85 ±0.70) mmol/L],AlP[(0.15 ± 0.351) vs.(-0.127 ± 0.184)] of the atherosclerosis group were higher than those of control group (t =5.975,2.252,2.614,-5.467,all P < 0.05).The HDL-C of atherosclerosis group was lower than that of control group [(0.78 ±0.16) mmol/L vs.(1.29 ±0.21) mmol/L,t =4.750,P <0.05].The Scr[(117.24 ± 94.27) mmol/L vs.(64.16 ± 13.42) mmol/L],BUN [(6.73 ± 3.58) mmol/L vs.(4.66 ± 1.08) mmol/L] of the atherosclerosis group were higher,and the GFR was lower [(65.60 ±23.00)ml · min-1 · 1.73 m-2 vs.(124.78 ± 24.35)ml · min-1l.73 m-2,t =5.118] than those of control group (t =4.730,4.702).The Scr of the atherosclerosis group was higher,and the GFR was lower [(65.60 ± 23.00) ml · min-1 · 1.73 m-2 vs.(95.60±53.00)ml · min-1 · 1.73 m-2,t =3.514] than those of the non-atherosclerosis group [(117.24 ± 94.27) mmol/L vs.(71.35 ± 42.18) mmol/L,t =3.690].There were positive correlation between TG and LDL-C (r =0.828,0.323,P < 0.05) and negative correlation between AIP and HDL-C (r =-0.489,P <0.05).There was positive correlation of CBMmax with Scr,BUN and AIP (r =0.394,0.289,0.528,all P < 0.05),and negative correlation between CBMmax and GFR (r =-0.277,P < 0.05).Conclusion Body weight,GFR,AIP and CBMmax are useful indicators in evaluation of renal arteriolar sclerosis in patients with chronic renal failure.AIP is a sensitive index for abnormal blood lipid level.AIP and CBMmax are important risk factors in chronic renal failure patients with renal arteriolar sclerosis.
10.Serum vitamin D status in 668 elderly men in Beijing and variations in related factors

Objective To evaluate the levels of serum 25 hydroxyvitamin D (25(OH)D), parathyroid hormone(PTH)and osteocalcin,as well as bone mineral density(BMD),in aged men in Beijing, and the relationship among these factors.Methods A total of 668 elderly male patients who were treated at the Beijing Friendship Hospital were evaluated, and their medical data were collected. The serum concentrations of 25(OH)D,PTH,and osteocalcin were measured.Additionally,bone mineral density at the lumbar spine1- 4(L1- 4) and left femoral neck were measured using dual-energy X-ray absorptiometry. Participants were divided into 4 groups by age,3 groups according to BMD,and 4 groups according to serum 25(OH)D levels. The relationship among these factors was investigated. Results The mean serum concentrations of 25(OH)D and PTH were (40.2 ± 17.3) nmol/L and (45.19 ± 15.54) ng/L, respectively. The serum 25(OH)D levels in the group aged≥90 years[(27.1±16.8)nmol/L]was significantly lower compared to that in the other groups. Among different BMD groups, serum 25(OH)D levels reduced gradually, but differences were not significant(P>0.05).Age and PTH levels were not significantly associated with 25(OH) D groups (P>0.05). However, BMD of the femoral neck in the groups with severe [(0.782±0.121)g/m2] and mild[(0.775±0.247)g/m2]vitamin D deficiency were significantly lower than that in the group with normal vitamin D levels [(0.837 ± 0.192) g/m2] (P<0.05). With the increase in vitamin D levels, osteocalcin levels decreased significantly(P<0.05).Conclusion Vitamin D deficiency is highly prevalent among aged men in Beijing and the 25(OH)D levels have some correlation with BMD and osteocalcin.
