1.Research progress of miRNA in ulcerative colitis

Ulcerative colitis(UC)is a chronic im-mune-mediated disease characterized by diffuse mucosal inflammation and clinical symptoms of ab-dominal pain,diarrhea,mucopurulent bloody stool and tenesmus.Due to its complex etiology,difficult treatment and poor prognosis,UC has been listed as one of the modern refractory diseases by the World Health Organization.MicroRNAs(microR-NAs)regulate various biological processes such as cell growth,differentiation,and development by regulating gene expression.Studies have shown that miRNA is involved in the pathogenesis of UC through various pathways such as intestinal muco-sal barrier homeostasis disorder,intestinal flora im-balance and excessive differentiation of immune cells,and is expected to become a new target for clinical treatment of UC.In order to better under-stand the role of miRNA in the pathogenesis of UC and provide new research directions for the treat-ment of UC,this article reviews the correlation be-tween miRNA and the pathogenesis of UC,the po-tential of miRNA as a biomarker of UC,and the treatment strategies of miRNA in UC in recent years.
2.Application research of mixed teaching based on DingTalk platform in clinical practice teaching of respiratory department

Objective:To explore the application effect of online and offline mixed teaching based on DingTalk platform in clinical practice teaching of respiratory department.Methods:The Batch 2016 ( n= 40) and Batch 2017 ( n=47) undergraduates of clinical medicine in Qiqihar Medical College were selected as the research objects, and they were divided into a control group and a research group. The control group adopted traditional offline theoretical teaching, while the research group adopted online-offline mixed teaching, focusing on teaching design and teaching objectives, teaching subjects, teaching resources, network platforms and teaching activities (classroom learning and evaluation). The theoretical and practical test scores of the two groups of interns were compared, and the evaluations of the students in the research group on the effect of online and offline mixed teaching were collected. Excel 2013 was used to sort out the data, and R software was used for data processing and statistical analysis. T-test was used for measurement data and Chi-square test was used for counting data. Results:The operational test scores [(28.16±1.70) points] and theoretical scores [(82.07±6.40) points] of the students in the study group were higher than those in the control group [(23.35±2.88) points and (76.20±5.67) points], with significant differences ( P<0.001). Compared with the students in the control group, the students in the research group were more satisfied with the mixed teaching method. Conclusion:The online and offline mixed teaching is conducive to improving students' comprehensive performance, learning effect, and promoting students' clinical diagnosis and treatment skills, self-study, innovation abilities and clinical competence.
3. Determination of methyl ethyl ketone in urine by headspace gas chromatography

To establish a method for determination of methyl ethyl ketone in urine by headspace gas chromatography.
Methods:
In the urine sample(hereinafter referred to as urine sample), methyl ethyl ketone is pretreated by headspace technology, and a certain amount of head air is injected into the gas chromatograph, separated by capillary column, detected by hydrogen flame ionization detector, and the retention time is qualitative and the peak height is high. Peak area.
Results:
Good linearity was in the range of 0.01 to 6.0 μg/ml with a regression equation of
4.New progress in the anti-tumor mechanism of curcumin

Curcumin is derived from traditional Chinese medicine turmeric and can be used for chemoprevention and treatment of various cancers. Recent studies have shown that curcumin can participate in the regulation of various molecular signal transduction pathways, regulate the levels of reactive oxygen species, and regulate the inflammatory microenvironment to inhibit the occurrence of tumors. In addition, curcumin can inhibite the proliferation of tumor cells and promote apoptosis of tumor cells. Here, the paper reviews the mechanism of action of curcumin in cancer prevention and treatment and the new progress in the clinical trials, aiming to provide a basis for research in related fields.
5.Effects of electroacupuncture on synaptic plasticity in hippocampal CA3 area of rats with chronic stress depression.

OBJECTIVETo explore the effects of electroacupuncture (EA) on behavioral function and synaptic plasticity in hippocampal CA3 area in rats with chronic stress depression.
METHODSAccording to the random number table method, 144 SD male rats were assigned into a blank group, a model group, an EA group and a fluoxetine group, then each group was divided into a 7 d subgroup, a 14 d subgroup and a 21 d subgroup, 12 rats in each subgroup. The chronic mild unpredictable stress stimulus combined with lonely breeding were applied to establish the depression model of rats, which was performed simultaneously with intervention treatment. The rats in the EA group were treated with EA (dilatational wave) at "Shenting" (GV 24) and "Baihui" (GV 20), while the rats in fluoxetine group were treated with intragastric administration of fluoxetine, once daily. With open-field test, sugar consumption experiment and transmission electron microscope, the changes of behavior and neuronal synapse inhippocampal CA3 area were observed.
RESULTSOn 7 d, 14 d and 21 d, compared with the blank group, the open-field test score, sugar consumption and body mass were significantly lower in the model group (all<0.01); compared with the model group, the open-field test score, sugar consumption and body mass were significantly higher in the EA group and the fluoxetine group on 14 d and 21 d (<0.01,<0.05). On 14 d and 21 d, compared with the blank group, the synapse in hippocampal CA3 area was significantly lower in the model group (both<0.01); compared with the model group, the synapse in hippocampal CA3 area was significantly higher in the EA group and the fluoxetine group (<0.01,<0.05). The neurons cells in hippocampal CA3 area in the model group showed pyknosis and deformation from 7 d with fusion structure and unclear boundary of synapse, which were significantly improved on 21 d; the neurons cells in hippocampal CA3 area in the EA group and the fluoxetine group were significantly improved from 14 d and restored to normal level on 21 d, in addition, the structure of synapse restored to normal level.
CONCLUSIONSEA is involved in the regulation of synaptic plasticity in hippocampal CA3 area, and promotes the recovery of depression symptoms.
6.Study on correlation of waist circumference with intra-abdominal fat area

To explore the best measurement of waist circumference related with intra-abdominal fat area evaluated by magnetic resonance imaging ( MRI). Totally 207 participants aged 20-60 years were enrolled. Waist circumference were measured at the levels of navel ( WC1) and the midpoint between costal brim and iliac cest (WC2). Intra-abdominal fat area was evaluated by MRI scan. Intra-abdominal fat area was significantly higher in men than in women [(132. 17 ± 59. 49 vs 70. 56 ± 35. 33)cm2 , P<0. 01]. Pearson correlation analysis showed that WC1 and WC2 were positively correlated with intra-abdominal fat area (r = 0. 779, r = 0. 809, both P<0. 01). Multiple linear stepwise regression analysis demonstrated that WC1 and WC2 were independently associated with intra-abdominal fat area(β=0. 553, R2 =0. 714, P<0. 01; β = 0. 603, R2 = 0. 735, P<0. 01). All of the two different measurements of waist circumference parameters may reflect intra-abdominal fat area, while WC1 seems to be the simpler one.
7.Research on Saliva Biochemical Changes about “Normal Constitution - Kidney Deficiency Constitution - Kidney Deficiency Syndrome”

Objective To investigate the changes of saliva biochemical parameters in the dynamic evolution of “normal constitution - kidney deficiency constitution - kidney deficiency syndrome”.Methods Totally 24 male and female SD rats of 3 months old (12 male rats and 12 female rats) were put in packet pair cage. Pregnant rats were randomly divided intoⅠ andⅡ groups, 6 rats in each group.Ⅰ group was without intimidation during pregnancy, with normal feeding, and then after the birth, 20 rats were randomly selected as normal control group and kidney deficiency syndrome model group, 10 rats in each group, half male and half female.Ⅱ group, from gestation day 2, was given intimidation until postnatal. 10 half male and half female rats were randomly selected as model group of kidney deficiency constitution. Rats in each group received normal feeding conditions for 6 weeks. The kidney deficiency syndrome model group was given intramuscular injection of hydrocortisone 25 mg/kg body weight every day, while normal control group and kidney deficiency constitution model group were injected with the same amount of normal saline for 10 consecutive days. The changes of salivary biochemical parameters were observed in each group of rats.Results Compared with the normal control group, salivary pH in the kidney deficiency constitution group and kidney deficiency syndrome group significantly increased (P<0.01). CK, ALT, AST, ALP levels in the kidney deficiency constitution group were reduced, but the contents of K, P, TP, AMY increased. The levels of CK, ALT, AST, ALP and LDH in the kidney deficiency syndrome decreased, while the contents of K, P, TP, and AMY increased (P<0.05,P<0.01). Compared with the kidney deficiency constitution group, the content of LDH in the kidney deficiency syndrome group decreased (P<0.05).Conclusion Rat saliva composition changes did occur in the kidney deficiency constitution group and kidney deficiency syndrome group.
8.Relationship between hypertriglyceridemic-waist phenotype and clustering of cardiovascular risk factors in adolescents

Objective The aim of the study was to explore the relationship between hypertriglyceridemic-waist(HTWC) phenotype and clustering of cardiovascular risk factors in adolescents. Methods A cross-sectional study was conducted in 3 136 Han adolescents aged 13-17 years in Qinhuangdao. The HTWC phenotype was defined as serum triglyceride concentrations ≥1. 47 mmol/ L and waist circumference ≥90 th percentile for age and sex. Results The prevalence of HTWC phenotype was 3. 3% . The prevalence of clustering of cardiovascular risk factors was 0. 4%in adolescents with normal waist circumference and serum triglyceride and 14. 7% in adolescents with HTWC phenotype. After adjustment for sex, age, body mass index, and family history of diabetes and hypertension, the prevalence of clustering of cardiovascular risk factors in adolescents with HTWC phenotype was 9. 286 fold(95% CI 3. 161-27. 282, P<0. 01) of that in adolescents with normal waist circumference and triglyceride. Conclusion There was a significant correlation between HTWC phenotype and clustering of cardiovascular risk factors, suggesting that this phenotype may be applied as a simple marker for identifying adolescents with clustering of cardiovascular risk factors.
9.Determination of five components in Pueraria labta decoction with reference extraction method.

OBJECTIVETo establish the reference extraction method for determining five components contained in Pueraria labta decoction and detecting the applicability and feasibility of the application of reference extracts according to the quality standard of traditional Chinese medicine.
METHODZorbax C18 chromatographic column (4.6 mm x 250 mm, 5 microm) was used, with methanol-0.02% phosphoric acid (25:75) as the mobile phase and the detection wavelength was set at 250 nm. Self-prepared reference extracts of P. labta was used for determining the content of 3'-hydroxy puerarin, puerarin, 3'-methoxy puerarin, isoflavoues aglycone-8-C-apiose-(1--> 6) glucoside and daidzin and compare with the experimental results obtained with the single reference substance determination method.
RESULTThe reference extraction determination method showed high precision, stability and reproducibility, with coincident determination results with the results obtained with the single reference substance determination method.
CONCLUSIONThe reference extraction determination method of P. labta can be used for quality control of P. labta herbs. The study preliminarily proves the feasibility of the application of P. labta in traditional Chinese medicine.
