1.Effects of medicated serum of Siwutang on autophagy of ovarian granulosa cells in polycystic ovarian syndrome

OBJECTIVE To investigate the effects of medicated serum of Siwutang on autophagy of ovarian granulosa cells (KGN cells) in polycystic ovarian syndrome (PCOS) and its underlying mechanism. METHODS Blank serum and different- concentration medicated serum of Siwutang were prepared by intragastric administration of normal saline and different doses of Siwutang [0.52, 1.04, 2.08 g/(kg·d)] in 3-month-old female SD rats. After screening the intervention concentration of Siwutang medicated serum, KGN cells were divided into control group (without any treatment), dehydroepiandrosterone (DHEA) group (treated with 50 μmol/L DHEA for 48 h), blank serum group (treated with 50 μmol/L DHEA for 48 h and with 10% blank serum for 72 h) and medium-concentration of Siwutang medicated serum group (treated with 50 μmol/L DHEA for 48 h and with 10% medium-concentration Siwutang medicated serum for 72 h). The number of autophagosomes was observed in each group, and protein expressions of pathway-related proteins [fructose-1, 6-bisphosphatase 1 (FBP1),mammalian target of rapamycin (mTOR), phosphorylated mTOR (p-mTOR)], autophagy-related proteins [p62, microtubule-associated protein 1 light chain 3 (LC3)] and mRNA expression of FBP1 were also detected. The (transfected) cells were further divided into Siwutang group (treated with 10% medium dose of Siwutang medicated serum for 72 h after 48 h intervention with 50 μmol/L DHEA), Siwutang+si-NC group [negative control small interfering RNA (siRNA) transfected cells treated with 50 μmol/L DHEA for 48 h, and then with 10% medium-concentration of Siwutang medicated serum for 72 h] and Siwutang+si-FBP1 group (FBP1 siRNA transfected cells treated with 50 μmol/L DHEA for 48 h, and then with 10% medium-concentration Siwutang medicated serum for 72 h). The effects of knocking down FBP1 on the above-mentioned effects of Siwutang were detected. RESULTS Compared with control group, DHEA group exhibited an increase in the number of autophagosomes, an elevated LC3-Ⅱ/LC3-Ⅰ and p-mTOR/mTOR, as well as increases in protein and mRNA expressions of FBP1, and decreased protein expression of p62 (P<0.05). Compared to both DHEA group and blank serum group, the medium-concentration of Siwutang medicated serum group showed a decrease in the number of autophagosomes, a decrease in LC3-Ⅱ/LC3-Ⅰ, and increases in p-mTOR/mTOR, protein expression of p62, protein and mRNA expressions of FBP1 (P<0.05). After knocking down FBP1, compared with Siwutang+si-NC group, Siwutang+si-FBP1 group showed a significant decrease in cell viability, protein expression of p62 , protein and mRNA expressions of FBP1 as well as p-mTOR/mTOR, and an increase in LC3-Ⅱ/LC3-Ⅰ (P<0.05). CONCLUSIONS Siwutang can promote the phosphorylation of mTOR protein by up- regulating the protein and mRNA expressions of FBP1 in KGN cells, thus inhibiting autophagy of KGN cells.

3.Luteolin inhibits the invasion and migration of endometrial stromal cells by regulating HPGD expression

Objective :
To investigate the effects of luteolin on invasion and migration of endometriosis stromal cells and whether its mechanism is related to the regulation of 15-hydroxyprostaglandin dehydrogenase (HPGD) expres- sion.
Methods :
The endometrial stromal cells HEM15A were divided into control group (cells were cultured in nor- mal medium) ,luteolin group ( cells were treated with different concentrations of luteolin) ,si-HPGD group ( cells were transfected with si-HPGD) ,si-NC group ( cells were transfected with si-NC ) ,luteolin + si-HPGD Group (cells were transfected with si-HPGD and treated with 20 μmol / L luteolin) ,Luteolin + si-NC Group ( cells were transfected with si-NC and treated with 20 μmol / L luteolin) . Real-time quantitative PCR was used to detect mRNA level of HPGD.Cell proliferation was detected by CCK-8 assay,Transwell and scratch assays were used to detect cell invasion and migration.The protein expression of proliferating cell nuclear antigen (PCNA) ,matrix metallopro- teinase (MMP-2) ,MMP-9 and HPGD were detected by Western blot,and the level of prostaglandin E2 ( PGE2) was detected by ELISA.
Results :
Compared with 0 μmol / L luteolin,luteolin at 20,50 and 100 μmol / L significantly inhibited the proliferation activity of hEM15A cells,and reduced PCNA expression ( all P<0. 05) .Compared with the control group,20 μmol / L of luteolin significantly inhibited cell invasion and migration (P <0. 05) ,decreased the expressions of MMP-2 and MMP-9 (P<0. 05) ,and up-regulated the mRNA and protein levels of HPGD (P < 0. 05) ,while inhibited cellular PGE2 level (P<0. 05) .Compared with the luteolin group,the luteolin + si-HPGD group increased cell invasion and migration (P <0. 05 ) ,increased the expressions of MMP-2 and MMP-9 (P < 0. 05) .
Conclusion
Luteolin regulates HPGD / PGE2 signaling pathway to inhibit the invasion and migration of en- dometrial stromal cells.
4.Influencing factors of medication adherence in patients with severe mental disorders in Zhengzhou

ObjectiveTo investigate the status and influencing factors of medication adherence in patients with severe mental disorders in Zhengzhou, so as to provide references for the formulation of prevention and treatment measures for severe mental disorders. MethodsFrom March to June 2021, a stratified multistage cluster sampling method was applied to select 342 patients from the National Information System for Severe Mental Disorders in Zhengzhou. The general demographic data of patients were collected via self-designed questionnaire, and the medication status was investigated, then the influential factors were summarized. The differences in influential factors of medication adherence were compared between the medication adherence group and the medication non-adherence group. Thereafter, Logistic regression analysis was applied to explore the factors influencing medication adherence. ResultsA total of 320 patients were included in the final analysis, altogether 76.56% of patients (n=245) complied with medication. The differences between patients in the medication adherence group and those in the medication non-adherence group were statistically significant in terms of residence, occupation, and outpatient chronic disease reimbursement (χ2=14.015, 7.502, 13.106, P<0.05 or 0.01). In the questionnaire of influential factors on medication adherence, there were statistically significant differences in the scores of lack of insight, stigma and drug-related factors between the two groups (Z=7.588, 2.379, 2.893, P<0.05 or 0.01). Outpatient chronic disease reimbursement was a protective factor for medication adherence (OR=2.727, 95% CI: 1.320~5.634, P<0.01), while rural residence (OR=0.465, 95% CI: 0.221~0.977, P<0.05) and lack of insight (OR=0.398, 95% CI: 0.286~0.553, P<0.01) were risk factors for medication adherence. ConclusionPatients with severe mental disorders in Zhengzhou have a high rate of medication adherence, moreover, the outpatient chronic disease reimbursement, lack of insight and residence may be influencing factors for medication adherence in patients with severe mental disorders.
5.MRI differential diagnosis of Brucella spondylitis and spinal tuberculosis

Objective To analyze the MRI features of Brucella spondylitis and spinal tuberculosis,to improve the ability of differential diagnosis. Methods MRI features of 22 cases with Brucella spondylitis and 26 cases with spinal tuberculosis confirmed by laboratory examination and operative pathology were analyzed retrospectively.Results Among 22 cases of Brucella spondylitis,1 7 cases occurred in the lumbar spine (1 3 cases in the lumbar 4 vertebrae),2 cases in the cervical spine,2 cases in the thoracic spine and 1 case in the sacral spine.1 9 cases had normal vertebral morphology,slight bone destruction and extensive edema,3 cases had severe vertebral wedge deformation, 16 cases had marginal bone hyperplasia.15 cases had slight changes in the intervertebral space,with narrow (or normal)or slight destruction of intervertebral disc,7 cases had severe narrowing or disappearance of intervertebral space and serious destruction of intervertebral disc. 13 cases had small paravertebral abscess,and 1 case had large paravertebral abscess.Among 26 cases of spinal tuberculosis,20 cases occurred in the lumbar spine (11 cases in the 4th and 5th vertebral bodies),5 cases occurred in the thoracic spine and 1 case occurred in the sacral spine,21 cases had severe vertebral wedge deformation due to bone destruction,9 cases had slight narrowing of intervertebral space, slight destruction of intervertebral discs,and 1 7 cases had obvious narrowing or narrowing of intervertebral space.7 cases had small paravertebral abscess and 1 6 cases had large paravertebral abscess with flowing phenomenon.Conclusion Vertebral morphology of Brucella spondylitis is basically normal,bone destruction is light and edema range is large,bone hyperplasia is common,slight narrowing (or normal)of intervertebral space occurs in intervertebral disc with less destruction,range of paravertebral abscess is limited.Bone destruction of spinal tuberculosis is heavy and edema range is small,vertebral body is easy to deform with no osteoporosis,significant stenosis (or disappearance)and severe disruption occur in intervertebral disc,paravertebral abscess is large often with flowing phenomenon.
6.MRI features of solitary fibrous tumors in the spinal canal

Objective To investigate the MRI features of solitary fibrous tumors(SFT)in the spinal canal.Methods MRI images of 5 cases with pathologically proved STF in the spinal canal were analyzed retrospectively.Results Of 5 lesions,there were 2 in the cervical spine,3 in the thoracic spine;1 in the epidural space and 4 in the subdural extramedullary space.On MRI plain scan,3 lesions showed homogeneous iso-intense signal on T1WI and hypo-intense signal on T2WI,2 lesions showed heterogeneous signal,1 showed patchy hypo-intense signal on T1WI and T2WI at the upper edge of lesion,which had been confirmed as hemorrhage and the other lesion showed internal cystic variation.All of the 5 lesions enhanced on enhancement scan,with moderate enhancement in 2 lesions and significant enhancement in 3 lesions.Cystic and hemorrhagic area were not enhanced.The"dural tail sign"was showed in 3 cases.Conclusion The diagnosis of SFT should be considered when a lesion shows a localized solitary mass in the spinal canal with hypo-intense on T2WI and moderate to significant enhancement.
7.Micro-invasive embedding combined with montelukast sodium for children cough variant asthma:a randomized controlled trial.

OBJECTIVETo observe the effects of micro-invasive embedding combined with montelukast sodium and simple montelukast sodium for children cough variant asthma (CVA).
METHODSA total of 240 patients were randomly assigned into an observation group and a control group, 120 cases in each one. Considering of cases dropping, 101 patients in the observation group and 105 cases in the control group were included. Montelukast sodium chewable tablets were applied before sleep for 3 months in the control group, 5 mg a time, once a day. Based on the treatment as the control group, micro-invasive embedding was used for 3 months in the observation group, twice in the first month and once in the other two months. The acupoints were Feishu (BL 13), Danzhong (CV 17), Dingchuan (EX-B 1), and Zusanli (ST 36). Follow-up was conducted 9 months after treatment in the two groups. The cough score, serum immunoglobulin (IgE, IgG, IgA), platelet activating factor (PAF) were observed before and after treatment. The indices were compared before and after treatment and at follow-up, including pulmonary function indices[peak expiratory flow rate (PEF), forced expiratory volume at the 1st second (FEV1)], and small airway function indices[forced expiratory flow rate with remaining 25% vital capacity (MEF25%), forced expiratory flow rate with remaining 50% vital capacity (MEF50%), forced expiratory flow rate with remaining 75% vital capacity (MEF75%) and mid expiratory flow rate (MEF25%-75%)]. Also, the total effects were evaluated.
RESULTS①The total effective rate in the observation group was 93.1% (94/101), which was better than 87.6% (92/105) in the control group (<0.05). The cough disappearance time of the cured children in the observation group was (10.38±2.64) d, and it was shorter than (10.72 ±2.60) d of those in the control group (<0.05). After treatment, the cough score apparently decreased compared with those before treatment in the two groups (both<0.05), with better result in the observation group (<0.05). At follow-up, the recurrence frequency of the observation group was (1.43±1.20), and it was less than (1.91±1.71) in the control group (<0.05). ②The levels of serum IgA and IgG after treatment in the two groups increased, and those of serum IgE and PAF decreased, compared with those before treatment. There was statistically significance except IgG in the control group before and after treatment (all<0.05), with better Results in the observation group after treatment (all<0.05). ③ Compared with those before treatment, all the pulmonary function indices were improved obviously after treatment and at follow-up in the two groups (all<0.05), without statistically significance between the two groups (both>0.05). ④ There was no statistically significance before and after treatment on small airway function indices in the two groups (all>0.05). The indices at follow-up increased compared with those before treatment in the two groups (all<0.05), with better Results in the observation group (all<0.05).
CONCLUSIONSMicro-invasive embedding combined with montelukast sodium achieved de-finite effect for children CVA, which can improve the body's immune and microcirculation. The effect is better than that of simple montelukast sodium on improving small airway function, etc.
8.Effects of rapamycin on promoting oral squamous cancer cell apoptosis by inhibiting TLR4, IL-6 and PGE2 expression

Objective To investigate whether the effects of rapamycin on promoting oral squamous cancer cell apoptosis is related with inhibiting TLR4 expression and inflammatory factor IL-6 and PGE2 expression.Methods Human oral squamous carcinoma SCC-15 cell line was cultured and interfered by rapamycin.Then the activity of SCC-15 cells was detected by CCK 8,the SCC-15 cells apoptosis was detected by the Hochest33342/PI double staining,the invasion ability was determined by the transwell method.The TLR4 protein expression level was detected by Western blot and IL-6 and PGE2 expressions were detected by ELISA.Results Rapamycin could promote cell apoptosis,the invasion ability of SCC-15 cells was significantly decreased.After rapamycin intervention,the TLR4 protein expression was decreased and expression levels of IL-6 and PGE2 were reduced,showing statistical difference as compared with the negative group (P<0.05).Conclusion Rapamycin promotes oral squamous cancer SCC-15 cell apoptosis,which may be related with inhibiting TLR4,IL-6 and PGE2 expression.
9.Expression of BAG3 protein in colon cancer tissue detected by tissue microarray method and its clinical significance

Objective:To explore the expression of Bcl related anti-apoptotic protein 3 (BAG3 protein)in colon cancer and its clinical significance,and to clarify the relationship between its expression and the occurrence and development of colon cancer.Methods:A total of 90 cases of colon cancer tissue and paracancerous tissue were selected, the expressions of BAG3 protein in different tissues were detected by tissue microarray and immunohistochemical SP method;Kaplan-Meier method,univariate analysis and multivariate analysis were used to investigate the relationship between the clinicopathological parameters and the prognosis of the patients with colon cancer.Results:The expression of BAG3 protein in the colon cancer tissue was high,and the positive expression rates of BAG3 protein in the colon cancer tissue and paracancerous tissue were 37.8% and 0.01%,respectively, and the difference was significant (P < 0.05).The expression of BAG3 protein was associated with gender and tumor size (P <0.05),but there was no correlation between the expression of BAG3 protein in the colon cancer tissue and age,pathologic grade,TNM stage,and lymph node metastasis (P >0.05).The Kaplan-Meier results showed that the expression of BAG3 protein in the colon cancer tissue was not related with the prognosis of the patients and the survival rates of the younger patients and the higher TNM staging patients were high. Conclusion:The expression of BAG3 protein in the colon cancer tissue is high,and its expression is associated with the tumor size;BAG3 protein may act as a tumor marker and therapeutic target for colon cancer.
10.A retrospective study on impact of healthcare-associated septicemia on hos-pitalization expense as well as length of hospital stay

Objective To explore the impact of healthcare-associated septicemia (HAS)on hospitalization expense as well as length of hospital stay,so as to optimize the allocation of healthcare resources,and provide scientific basis for reducing the economic burden caused by septicemia.Methods Hospitalized patients with confirmed HAS in a tertiary first-class teaching hospital between June 1 ,2012 and May 31 ,2015 were investigated retrospectively,con-trol group was set up in a 1 :1 ratio,hospitalization expense and length of hospital stay between two groups were compared.Results A total of 285 cases and 285 controls were enrolled in the study,the median of hospitalization expense in case group was higher than control group (¥19 718.39 vs ¥9 289.04,P <0.05);the median of length of hospital stay in case group was longer than control group (14.89 days vs 9.22 days,P <0.05).The disease bur-den caused by septicemia in different age groups and departments were different.The improvement rate of case group was lower than control group (76.49% [218/285 ]vs 83.51 % [238/285 ],χ2 = 2.562,P = 0.009 ). Conclusion As the common blood stream infection in hospitalized patients,septicemia not only increased the ex-pense of diagnosis and treatment,but also affected turnover rate of hospital bed.Rapid and effective diagnosis and treatment is significant o prevent and control septicemia.
