1.Research Progress on Glutamate Receptor-Mediated Changes in Synaptic Plasticity of Central Nervous System and Acupuncture Regulation Mechanism

Synapse is the key part of information transmission between neurons,which has certain plasticity.Glutamate receptor plays an important role in the synaptic plasticity of the central nervous system,and acupuncture can regulate the glutamate receptor to normal under pathological conditions,and its research is also deepening.This paper summarizes the recent research literature on the effects of two types of glutamate receptors,ionic glutamate receptor and metabotropic glutamate receptor,on synaptic plasticity,as well as acupuncture regulating the expression of ionic glutamate receptor and metabotropic glutamate receptor,in order to provide a theoretical basis for acupuncture regulating synaptic plasticity based on glutamate receptor expression,and provide reference for subsequent research.
2.Textual research on Bambusae Concretio Silicea.

By studying various ancient texts such as herbal classics and medical literature from different eras, it was found that there were discrepancies in the records about Bambusae Concretio Silicea(Tian Zhu Huang). In order to establish an accurate foundation, this research was based on ancient herbal literature and combined with plant morphology and investigative studies to examine its earliest mentions in ancient texts, nomenclature, medicinal properties, indications, and quality assessment standards. In the early records, Bambusae Concretio Silicea was referred to by several different names, such as "Zhu Huang" "Tian Zhu Huang" "Zhu Gao" "Zhu Tang", and "Zhu Huang". The earliest known formal usage of the name "Tian Zhu Huang" was found in the book Ri Hua-zi's Materia Medica(Ri Hua Zi Ben Cao). Throughout various ancient texts, the earliest recorded information about Bambusae Concretio Silicea also appeared in Ri Hua-zi's Materia Medica, not in Materia Medica of Sichuan(Shu Ben Cao) or other ancient texts. Ri Hua-zi's Materia Medica provided relevant descriptions of its origin, medicinal properties, and indications, albeit with some errors due to limited knowledge. However, this has been a valuable starting point for future research on Bambusae Concretio Silicea and holds pioneering significance in forming a mature system. As the research delved deeper, the medicinal properties of Bambusae Concretio Silicea have been consistent since Ri Hua-zi's Materia Medica, and the understanding has gradually improved through years of clinical verification. During the investigation process, the authors found limited records on the quality evaluation of Bambusae Concretio Silicea in ancient texts. Although the information is scarce, it serves as a foundational basis for establishing corresponding quality grading standards for Bambusae Concretio Silicea in the future.

3.Study on discovery of efficacy markers for Dachaihu Decoction and its action mechanism.

Dachaihu Decoction is a classical Chinese herbal prescription that is effective in harmonizing lesser yang and purging internal accumulated heat. At present, it has been widely used in clinical practice, and the resulting outcomes are satisfactory. However, its quality indicators and action mechanism are still not clear. Therefore, this paper explored the efficacy markers of Dachaihu Decoction and its action mechanism based on literature mining, molecular biology, and network pharmacology, so as to better control its quality and ensure its clinical efficacy. The efficacy markers of Dachaihu Decoction were predicted and analyzed according to the "five principles" for Q-markers of Chinese herbs. Then the anti-inflammatory activity of the efficacy markers of Dachaihu Decoction was evaluated with Griess reagent after the establishment of RAW264.7 cell inflammation model in vitro with lipopolysaccharide(LPS). The potential targets of efficacy markers were predicted by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP), ChEMBL, and SwissTargetPrediction, followed by the construction of the protein-protein interaction(PPI) network of the efficacy markers of Dachaihu Decoction. Topological, GO, and KEGG enrichment analysis was carried out to construct the "key target-signaling pathway-biological process" network, thus elucidating the action mechanism of the efficacy markers of Dachaihu Decoction. Saikosaponin B_2, baicalin, baicalein, wogonoside, neohesperidin, naringin, hesperidin, and paeoniflorin were considered as the potential efficacy markers of Dachaihu Decoction. The anti-inflammatory activity evaluation showed that the potential efficacy markers effectively inhibited the release of NO, exhibiting good anti-inflammatory activities. As demonstrated by network pharmacology, the efficacy markers of Dachaihu Decoction regulated the inflammatory response by acting on MAPK and NF-κB signaling pathways, the carbohydrate metabolism by HIF-1 and PI3 K-AKT signaling pathways, and the lipid metabolism by AMPK and PI3 K-AKT signaling pathways. This study discovered the efficacy markers of Dachaihu Decoction based on literature mining combined with molecular biological experiments and explored its action mechanism at the molecular level based on network pharmacology, which would provide reference for the quality control of Dachaihu Decoction and scientific basis for its clinical application.

4.Screening, identification and functional analysis of differential expression genes in rat visual cortex after monocular deprivation by RNA-Seq technique

Objective:To screen differentially expressed genes (DEGs) in rat visual cortex after monocular deprivation by RNA sequencing technology, and to analyze the function of the DEGs.Methods:Eighteen 14-day-old SD rats were randomly divided into blank control group and monocular deprivation model group according to random number table method, with 9 rats in each group.The monocular deprivation model was established through lid suture of the right eye for 14 days.Patten visual evoked potential (PVEP) in the right eyes of the rats was recorded before and 14 days after modeling, respectively.Bilateral visual cortex tissues of the rats were dissected from the two groups, and specific genes associated with the pathogenesis of amblyopia were screened out for RNA-seq analysis.The biological functions of differentially expressed genes were evaluated by Gene Ontology (GO) enrichment analysis, and metabolic pathways involved were analyzed by Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis.The use and care of the animals complied with ARVO statement.This study protocol was approved by an Ethics Committee of Gansu University of Chinese Medicine (No.2016-58).Results:Compared with blank control group, the latency of P 100 wave was significantly prolonged, and the amplitude was reduced in the eyes of monocular deprivation model group (both at P<0.05). Forty DEGs in the left visual cortex and 63 DEGs in the right visual cortex were determined, among which 9 genes were overlapped.GO analysis indicated that the DEGs were mainly involved in biological processes, such as DNA-templated transcription, glutamate secretion, transcriptional regulation of RNA polymerase Ⅱ promoter, protein phosphorylation etc., as well as molecular functions, such as DNA binding, ATP binding, protein serine/threonine kinase activity, calcium ion binding, zinc ion binding, phospholipase A 2 activity, nucleic acid binding and cell components involved in the formation of intracellular and membrane of endoplasmic reticulum.The abnormal expressions of Grm2 and Pla2g2a genes might be closely associated with visual function impairment. Grm2 gene was mainly involved in visual signaling pathway processes including glutamate synapse, long-term potentiation (LTP), long-term depression (LTD) etc. Pla2g2a gene was mainly involved in α-linolenic acid metabolism and arachidonic acid pathway. Conclusions:There are abnormal expressions of genes in the bilateral visual cortices of monocular deprivation rats in the sensitive period of visual development, mainly leading to the disorder of visual signal transduction pathway.Metabolic pathway changes based on specific response gene regulation may be one of the important molecular biological mechanisms in the pathogenesis of amblyopia.
5.Study on Q-markers of Da Chaihu decoction based on specific chromatogram and network pharmacology

Da Chaihu decoction is a classic prescription for the treatment of cholecystitis that is widely used in clinical practice, and has a definite curative effect. However, due to its diverse components and complex functions, the traditional indexes fail to capture its overall efficacy. Therefore, this study analyzed and predicted the quality markers (Q-markers) of Da Chaihu decoction based on specific chromatogram and network pharmacology to provide a reference for the comprehensive control of the quality. The study obtained 35 potential practical components of Da Chaihu decoction through virtual screening. The specific chromatogram of 15 batches of Da Chaihu decoction was established by HPLC-DAD with neohesperidin as a reference. Compared with the chromatographic peaks and the reference substance, the chemical components were assigned to predict the nine components of albiflorin, paeoniflorin, naringin, hesperidin, neohesperidin, baicalin, wogonoside, saikosaponin b2, saikosaponin b1 as Q-markers of Da Chaihu decoction. Finally, the network of the "components-key targets-signal pathways-biological processes" was constructed by network pharmacology to explore the mechanism of Da Chaihu decoction in treating cholecystitis to clarify the accuracy of Q-markers. The results indicated that potential Q-markers could act on multiple targets to regulate inflammatory and metabolism, and then combine to treat cholecystitis. Q-markers could combine with the pharmacologic action of Da Chaihu decoction, which could elucidate the overall efficacy of Da Chaihu decoction. This study explored the Q-markers of Da Chaihu decoction combined with the specific chromatogram and network pharmacology, which provided a basis for the quality control and evaluation of Da Chaihu decoction.
6.Regulatory effect of acupuncture on electrical activity level of optic cortex in amblyopia model rats

Objective: To explore the abnormal changes in neuroelectric activity in the primary visual cortex of rats deprived of vision in one eye and to investigate the regulatory effect of acupuncture in the sensitive period on the abnormal coding and conduction of electrical signals of rats' optic neurons.Methods: Sixty 14-day-old Sprague-Dawley rats were randomly divided into a blank group, a model group, an early-stage acupuncture group, a middle-stage acupuncture group, and a late-stage acupuncture group, with 12 rats in each group. Rats in every group except the blank group received right eyelid suturing to create a monocular deprivation model in the sensitive period of visual development (from the day rats open their eyes to the 45th day after their birth). Rats in the three acupuncture groups started to undergo acupuncture respectively on the 3rd, 12th, and 21st days after the model replication was done, with each group receiving nine-day treatment. The activity level of the neuroelectrical signal of the primary visual cortex in each group, including the latency and amplitude of P100 wave, average discharge frequency and amplitude of neurons, the power spectral density (PSD), and interspike interval (ISI), were measured by neuroelectric evaluation technology after the acupuncture treatment was finished. Results: Compared with the blank group, the latency of P100 wave in the visual center of vision-deprived eyes was significantly prolonged, and the amplitude was significantly reduced (P<0.05); the average discharge frequency and amplitude of the neurons in the visual cortex also decreased significantly (P<0.05); PSD decreased and ISI was prolonged significantly (P<0.05). Compared with the model group, the abnormal electrical activity of optic neurons in the three acupuncture groups ameliorated, the latency of P100 shortened, and the amplitude of P100 increased (P<0.05), the discharge frequency and amplitude increased significantly (P<0.05), the PSD reduced, and the ISI shortened (P<0.05). In addition, among the three acupuncture groups, the early-stage acupuncture group had the best effect on various indicators. Conclusion: Abnormal electrophysiological activity is significant in the visual center of vision-deprived rats, and acupuncture treatment in the sensitive period of visual development can enhance the bioelectrical activity of visual nerve cells, improve the efficiency of optic nerve conduction, and regulate the inhibition and retardation of visual response caused by visual deprivation.

Through analyzing the research literature about the mechanism of acupuncture intervention on vision in recent years, this article reviewed the mechanism of acupuncture intervention on vision from the aspects of central processing of visual information, the influnce of information transmission in the retina, the changes in visual microcirculation, optic nerve and neurotransmitter transmission, visual electrophysiology, visual plasticity. This article also proposed the future research idea for this field.
8.Meta-analysis of clinical effectiveness on Chinese herbs combined with moderate comprehensive therapy for childhood amblyopia

Background The conventional therapies for childhood amblyopia mainly include optometry,occluding therapy,atropine application,fine eyesight training,instrument or regular comprehensive therapy,but the clinical effectiveness of each method had its limitation.Chinese herbs have been used to the treatment of childhood amblyopia,however,their effects are still in controversy.Objective This study was to assess the evidence of efficacy of Chinese herbs combined with moderate methods for childhood amblyopia.Methods The published papers of randomized controlled trials (RCT) and controlled clinical trials (CCT) of Chinese herbs combined with moderate comprehensive therapy for childhood amblyopia from 2000 to 2015 were searched in China National Knowledge Infrastructure (CNKI),Wanfang database and PubMed database with computer.The methodology quality of included papers was evaluated according to the criteria of Cochrane and advanced Jadad scale.Data synthesis was facilitated using Reviewer Manager 5.1 software.The overall effect,or odds ratio (OR) of the treatment was analyzed by fixed effective model.Results Thirty-three RCT or CCT papers meeting inclusion criteria were included with the total sample size 6 666,and matched demography was showed in 25 papers;referential diagnosis criteria and effective criteria were described in 29 papers and 30 papers,respectively.Advanced Jadad scale showed 6,3 and 1-2 scores in 1,2 and 30 papers,respectively.The evidence showed that the total effectiveness of treatment within the group receiving Chinese herbs combined with conventional methods was higher than that in conventional group,with a statistically significant difference between groups (polled fixed effects model [OR] =3.92,95% confidence interval [CI]:3.30-4.64,Z=15.74,P<0.01).Conclusions The total effectiveness rate of Chinese herbs combined with conventional methods for childhood amblyopia is superior to only conventional treatment,indicating that Chinese herbs is effective for childhood amblyopia.More well-designed RCTs are needed to confirm the effect.
9.Point application in literature ofstone chamber and its application.

In the paper we summarize its medical prescriptions and the existing acupoint application contents. It is considered that its acupoint application is widely clinically used in dermatological department, cosmetology department, ophthalmology and otorhinolaryngology departments, paediatrics department, gynaecology department, anorectal department, orthopedics and traumatology department. It achieves remarkable effects in dermatological department, orthopedics and traumatology department. Medical literature ofis a treasure of TCM with distinctive feature and it deserves to be further explored so as to serve the clinic and promote the inheritance and development ofmedicine, such as finding out high effect prescription.
10.Effect on Immunologic Function of Chronic Abacterial Prostatitis Rats Treated by Electroacupuncture at Sanyin Points

Objective To study the immunologic mechanism of electroacupuncture at Sanyin points on chronic abacterial prostatitis rats. Methods Forty-eight male rats were divided randomly into normal group, model group, drug group and electroacupuncture group, 12 rats in each group. The animal model was made by SC purified prostate protein twice with immunologic adjuvant, and the rats in electroacupuncture group and drug group were treated by electroacupuncture at Sanyin points and feeding Cernilton for 15 days. The changes of prostate weight, index, pathology and contents of IgG, expressions of IL-2 and IL-8 in prostate homogenate were observed. Results Compared with normal group, the prostate weight and index were significantly increased in model group (P<0.05), the content of IgG was significantly increased (P<0.05), cytokine IL-2 and IL-8 were highly expressed (P<0.05, P<0.01), prostate tissue’s morphology and structure were significantly impaired in model group. Compared with model group, the prostate weight and index were significantly decreased in electroacupuncture group after treatment (P<0.05 or P<0.01), pathological changes reduced significantly, the content of IgG decreased significantly (P<0.05), the expressions of cytokine IL-2 and IL-8 were significantly decreased (P<0.05, P<0.01), and the pathological manifestation of chronic inflammation was significantly improved. Conclusion Electroacupuncture at Sanyin points can reduce the secretion of local prostate antibody IgG, regulate the expression of cytokine IL-2 and IL-8, protect the morphology and structure of prostate tissue from damage. Electroacupuncture at Sanyin points can adjust local immunologic function and promote the recovery of prostate.
