Astrocyte elevated gene-1 (AEG-1) was cloned as an human immunodeficiency virus -1-inducible and tumor necrosis factor-α-inducible transcript in primary human fetal astrocytes by a rapid subtraction hybridization approach. AEG-1 down-regulates the expression of the glutamate transporter EAAT2, thus, it is implicated in glutamate-induced excitotoxic damage to neurons as evident in HIV-associated neurodegeneration. Meanwhile, AEG-1 expression is elevated in subsets of breast cancer, prostatic cancer, glioblastoma multiforme and melanoma cells, having a dual specificity phosphatase activity. Overexpression of AEG-1 increases and siRNA inhibition of AEG-1 decreases migration and invasion of human glioma cells, respectively. Recent observations indicate that AEG-1 exerts its effects by activating the nuclear factor kappa B (NF-κB) pathway and AEG-1 is a downstream target of Ha-ras and plays an important role in Ha-ras-mediated tumorigenesis. These findings are intensifying interest in AEG-1 as a crucial regulator of tumor progression and metastasis and as a potential mediator of neurodegeneration.

We constructed recombinant adenovirus vector expressing murine endostatin and evaluated the Inhibition of human umbilical vein endothelial cells (HUVEC). We proved that endostatin significantly suppressed the S phase fraction, inhibited proliferation and increased the apoptotic index of HUVEC.

4.The Study of Immune Pathogenesis and The Effective of Immune Therapy in Patients with Recurrent Vulvovaginal Candidiasis

Objective To study the role of immunoregulation in patients with Recurrent vulvovaginal candidiasis(RVVC) that evaluate the efficacy of application with combining clotrimazole and rhIFN-? therapy.Methods Patients were divided randomly into two groups :treatment group(60) and control group(60). The patients in treatment group were scheduled to receive clotrimazole therapy, 400mg,pv,qd,and add to rhIFN-? 1000000u im,biw; control group only using clotrimazole therapy.Results The curative rate betweet two groups had significant difference (68.33 (41/60) vs. 35.0(21/60). The efficiency rate in control group was significantly higher than treatment group( (56.67(34/60)vs 26.67(16/60)),In treatment group ,the serum levels of IL-18(pg/ml)(before treatment (111.71?18.45),after treatment (189.18?28.17))and TNF-? (pg/ml)(before treatment (52.66?18.75),after treatment (124.25?43.77))were significantly increased (P0.05). Conclusion The application with combining clotrimazole and rhIFN-? can significant ameliorate the curative rate of RVVC,by regulate immune fanction in the patients that were induced the produce of IL-18 and TNF-?. rhIFN-?can improve cell-mediated immunity in patients with RVVC.
5.A study on the clinicopathological features and prognosis of solid-pseudopapillary tumor of the pancreas

Objective To study the clinicopathological and immunohistochemical features, histogenesis and biological behavior of solid pseudopapillary tumor of the pancreas ( SPT ). Methods Routine HE and immunohistochemical ( SP) methods were used in 20 cases of SPT. Results There were 18 females and 2 males, age ranging from 13 to 48 years with mean age of 25. 3 years. Abdominal pain and palpable mass were among the main complains. Seventeen cases were followed-up from 9 to 120 monthes. Fourteen cases were alive. Tumors were encapsulated, mixed with solid and cystic tissues. Histological features were psudopapillary structure with a fibrovascular core. Immunohistogically, the tumors were positive for a-1-AT ( 17 cases) , vimentin ( 14 cases) , synaptophysin ( 10 cases) , CgA (5 cases) ,CK and insulin (2 cases) ,glucagon and S-100 (1 case) ,PR (14 cases) , ER (1 case) ,pS2 (6 cases) , but all were negative for CEA and gastrin. Conclusion SPT is of low-graded malignancy and a distinct clinicopathologic entity in young female patients with both exocrine as well as endocrine differentiation. The tumor is closely related with sex hormone receptors.
