1.Impact of Wenyang lishui formula on ventricular remodeling in rats with pulmonary hypertension-induced right heart failure

OBJECTIVE To discuss the impact of Wenyang lishui formula on ventricular remodeling in rats with pulmonary hypertension-induced right heart failure (RHF) based on the Hippo/Yes-associated protein (YAP) signaling pathway. METHODS Ten rats were randomly selected as the control group. The remaining 63 rats were given a single intraperitoneal injection of monocrotaline to establish the pulmonary hypertension-induced RHF model. The 50 rats that successfully underwent the model establishment were randomly divided into the RHF group, low-dose group of Wenyang lishui formula (4.25 g/kg), high-dose group of the Wenyang lishui formula (17.00 g/kg), furosemide group (20 mg/kg), and high-dose group of Wenyang lishui formula+ Hippo/YAP signaling pathway activator group (17.00 g/kg of Δ Wenyang lishui formula+16 mg/kg of PY-60), with 10 rats in each group. The rats in each group were given the corresponding drug solution or normal saline by gavage or/andtail vein injection, once a day, for 4 consecutive weeks. During the experiment, the general conditions of the rats in each group were observed; after the last administration, the right ventricular diameter, right atrial diameter, end-diastolic volume, pulmonary artery blood flow acceleration time (PAAT) and its ratio to ejection time (ET) (PAAT/ET), pulmonary artery pressure and its ratio to pulmonary arterial flow velocity (pulmonary artery pressure/velocity) were measured. The plasma levels of brain natriuretic peptide and angiotensin Ⅱ (Ang Ⅱ) were detected. The pathological changes of the right ventricular tissue were observed, and the collagen volume fraction, the phosphorylation levels of the large tumor suppressor 1/2 (LATS1/2) and YAP, and the protein expression of the transcriptional coactivator of PDZ-binding motif (TAZ) were also detected. RESULTS Compared with the RHF group, the rats in Wenyang lishui formula low-dose and high-dose groups showed improved hair color, movement, diet, and mental state. The atrophy of right ventricular myocardial cells, the increase of inflammatory cells, collagen deposition, and hypertrophy of myocardial fibers were significantly alleviated. The right ventricular internal diameter, right atrial internal diameter, end-diastolic volume, pulmonary artery pressure, pulmonary artery pressure/velocity, the plasma levels of brain natriuretic peptide and AngⅡ , collagen volume fraction, the phosphorylation level of YAP and protein expression of TAZ were significantly decreased, while the PAAT, PAAT/ET and the phosphorylation level of LATS1/2 were significantly increased (P<0.05). PY-60 could significantly reverse the improvement effects of high-dose Wenyang lishui formula on the above quantitative indicators (P< 0.05). CONCLUSIONS Wenyang lishui formula can restore the right heart function of pulmonary hypertension-induced RHF rats, reduce their pulmonary artery pressure, alleviate the pathological changes in their cardiac tissues, and the above effects may be related to the activation of Hippo expression and the inhibition of YAP phosphorylation.
2.Mediating effect of family function between first aid literacy and mental health of caregivers of children with asthma

Objective:To explore the mediating effect of family function between first aid literacy and mental health of caregivers of children with asthma.Methods:This study was a cross-sectional survey. From August 2020 to January 2021, convenience sampling was used to recruit 350 caregivers of children with asthma in the Pediatric Internal Medicine Clinic in a ClassⅢ Grade A hospital in Qingdao. The survey was carried out using the General Information Questionnaire, the Safety and First Aid of the China Residents Health Literacy Questionnaire, the General Function Subscale of the Family Assessment Device, and the Chinese version of the General Health Questionnaire. Structural equation model was used to analyze the mediating effect of family function between first aid literacy and mental health. A total of 350 questionnaires were distributed, of which 337 were valid, with a valid rate of 96.29%.Results:Among 337 caregivers of children with asthma, the total score of the Chinese version of the General Health Questionnaire, the scores of the first aid literacy and the General Function Subscale of the Family Assessment Device were (2.52±2.33) , (12.28±1.31) , and (28.61±3.71) , respectively. Structural equation model showed that first aid literacy could directly affect mental health ( β=0.542) , and it could also indirectly affect mental health ( β=0.239) through the partial mediating effect of family function, and the mediating effect of family function accounted for 30.06% of the total effect. Conclusions:First aid literacy is an important factor affecting the mental health of caregivers of children with asthma. Family function plays a partial mediating role between first aid literacy and mental health of caregivers of children with asthma. Medical and nursing staff should pay attention to first aid literacy and family function to improve the mental health of caregivers.
3.Effect of NACK knockdown on the proliferation and apoptosis of T-cell acute lymphoblastic leukemia Jurkat cells

Objective:To investigate the effect and mechanism of NACK knockdown on the proliferation and apoptosis of T-cell acute lymphoblastic leukemia (T-ALL) Jurkat cells. Methods:Lentivirus transfection technology was used to transfect Jurkat cells and knock down NACK gene.Real time fluorescent quantitative PCR and Western blot were used to detect the silencing efficiency of NACK gene.CCK-8 method and flow cytometry were used to detect the effects of NACK knockdown on the proliferation and apoptosis of Jurkat cells.The expressions of protein related with Notch1 pathway, such as Hes1 and c-Myc, were detected by Western blot. Results:After NACK-shRNA was successfully transfected into Jurkat cells by lentiviral vector, the expression of NACK mRNA and protein was reduced signi-ficantly ( P<0.05). Compared with the negative control group and the blank control group, the CCK-8 method showed that the cell proliferation in the experimental group was significantly inhibited [The inhibition rates of cell proliferation in the experimental group, negative control group and blank control group were (37.27±4.48)%, (4.25±2.10)% and (2.43±1.40)%, respectively]( F=132.640, P<0.05), and the flow cytometry test showed that the apoptosis in the experimental group increased significantly [The apoptosis rates of experimental group, negative control group and blank control group were (26.38±3.03)%, (6.07±2.61)% and (3.40±1.98)%, respectively]( F=90.534, P<0.05). Western blot results confirmed that the expression of Notch1 pathway-related proteins Hes1 and c-Myc was down-regulated compared with the negative control group and the blank control group, and the difference was statistically significant ( P<0.05). Conclusions:Targeting silent NACK can down-regulate the expression of Notch1 pathway-related proteins, which leads to the inhibition of Jurkat cell proliferation and increased apoptosis, thereby exerting its anti-T-ALL effect.
4.Quantitative Secretome Analysis Reveals Clinical Values of Carbonic Anhydrase Ⅱ in Hepatocellular Carcinoma

Early detection and intervention are key strategies to reduce mortality, increase long-term survival, and improve the therapeutic effects of hepatocellular carcinoma (HCC) patients. Herein, the isobaric tag for relative and absolute quantitation (iTRAQ)-based quantitative pro-teomic strategy was used to study the secretomes in conditioned media from HCC cancerous tissues, surrounding noncancerous tissues, and distal noncancerous tissues to identify diagnostic and prog-nostic biomarkers for HCC. In total, 22 and 49 dysregulated secretory proteins were identified in the cancerous and surrounding noncancerous tissues, respectively, compared with the distal non-cancerous tissues. Among these proteins, carbonic anhydrase Ⅱ (CA2) was identified to be signifi-cantly upregulated in the secretome of cancerous tissues; correspondingly, the serum concentrations of CA2 were remarkably increased in HCC patients compared with that in normal populations. Interestingly, a significant increase of serum CA2 in recurrent HCC patients after rad-ical resection was also confirmed compared with HCC patients without recurrence, and the serum level of CA2 could act as an independent prognostic factor for time to recurrence and overall sur-vival. Regarding the mechanism, the secreted CA2 enhances the migration and invasion of HCC cells by activating the epithelial mesenchymal transition pathway. Taken together, this study identi-fied a novel biomarker for HCC diagnosis and prognosis, and provided a valuable resource of HCC secretome for investigating serological biomarkers.
5.Congenital nemaline myopathy caused by KLHL40 gene complex heterozygous variations: a case report

This article reported a case of nemaline myopathy caused by KLHL40 gene complex heterozygous mutations. This baby girl presented with shortness of breath, low myodynamia, and low muscle tension immediately after birth. However, her symptoms became worse after conventional treatment. Physical examination found lower muscle strength and muscle tone in four limbs and no primitive reflexes. The biochemistry test showed increased serum creatine kinase (CK). A muscle biopsy was not performed. The second-generation gene test confirmed the KLHL40 gene complex heterozygous mutations, which was a known mutation c.932G>T (p.R311L) and a de novo mutation c.1487T>A (p.M496K), inherited from the father and mother, respectively. Nemaline myopathy is a rare congenital muscular disease characterized by nemaline bodies in muscle fibers. Pathological and genetic diagnoses are the gold standards for the diagnosis of this disease.

Objective To investigate the distribution and drug resistance of carbapenem-resistant Enterobacteriaceae ( CRE) isolated from children in China. Methods CRE strains were collected in 10 ter-tiary children's hospitals of China from January 1, 2016 to December 31, 2017. Antimicrobial susceptibility of the clinical strains was detected with disk diffusion method ( KB method) and automated method. The re-sults were analyzed according to the Clinical and Laboratory Standards Institute ( CLSI) Standards published in 2017. WHONET 5. 6 software was used to retrospectively analyze the distribution characteristics and drug resistance of these strains. Results A total of 3065 CRE clinical strains were isolated from children with an overall prevalence of 7. 7% and among them, 13. 5% were isolated in neonatal group and 5. 8% in non-neo-natal group. The detection rate of CRE in 2017 was higher than that in 2016 (9. 7% vs 5. 7%). Among the 3065 CRE strains, there were 1912 strains of Klebsiella pneumoniae (62. 0%), 667 strains of Escherichia coli (22. 0%), 206 strains of Enterobacter cloacae (7. 0%), 56 strains of Klebsiella aerogenes (1. 8%) and 47 strains of Serratia marcescens (1. 5%). Most of the strains were isolate in neonatology departments including neonatal intensive care units (NICU) and intensive care units (ICU), accounting for 44. 8% and 19. 7%, respectively. Respiratory tract (61. 8%), urine (19. 4%) and blood (5. 7%) specimens were the main sources of CRE isolates. Results of antimicrobial susceptibility test showed that the CRE strains were highly resistant to carbapenem antibiotics such as imipenem, meropenem and ertapenem, as well as penicillins and most cephalosporins (79. 6%-100%), especially those isolated in the neonatal group (P<0. 05). Children had relatively low resistance rates to aminoglycosides such as amikacin (19. 7%) and fos-fomycin (11. 9%), fluoroquinolones such as levofloxacin (37. 7%) and ciprofloxacin (43. 3%), and tige-cycline (3. 8%). Currently, no polymyxin B-resistant strains were isolated. Conclusions The prevalence of common CRE strains in children in 2017 was higher than that in 2016, especially in newborns. Drug re-sistance in CRE strains isolated from neonates to common antibiotics was more severe, suggesting that great attention should be paid to it and timely measures should also be taken.
7.Silencing LncRNA HOXD-AS1 enhances the radiosensitivity of H460 cells by regulating miR-454-3p expression

Objective To explore the effect of LncRNA HOXD-AS1 on the radiosensitivity of H460 cells by targeting the regulation of miR-454-3p expression. Methods H460 cells were irradiated with 0, 2, 4, 6 and 8 Gy, and the expression levels of HOXD-AS1 and miR-454-3p were detected by qRT-PCR. Cell apoptosis rate were measured by flow cytometry experiments. Cell cloning experiments were used to detect cell radiosensitivity. The protein expression levels of Caspase-3, Cyclin D1 and γ-H2AX were detected by Western blot. Results The expression of HOXD-AS1 in H460 cells was significantly increased after X-ray irradiation ( P<0.05) , while the expression of miR-454-3p was significantly decreased ( P<0.05) . Silencing HOXD-AS1 significantly promoted apoptosis, increased radiosensitivity of lung cancer cells, promoted Caspase-3, γ-H2AX protein expression, and inhibited Cyclin D1 expression. HOXD-AS1 could target the expression of miR-454-3p. Conclusion Silencing HOXD-AS1 can promote the apoptosis of lung cancer cells and inhibit cell survival by targeting miR-454-3p to increase the radiosensitivity of lung cancer cells.

Objective To investigate the effect of body mass index (BMI) on the clinical efficacy and prognosis of patients with acute exacerbation of chronic obstructive pulmonary disease(AECOPD), and to provide evidence for the use of BMI in predicting their disease situation and prognosis.Methods Eighty patients with AECOPD admitted to the Department of Respiration of the Fourth Central Hospital of Tianjin from October 2014 to October 2015 were enrolled, and they were divided into low BMI group (BMI < 18.5 kg/m2) and normal BMI group (BMI 18.5 - 24.9 kg/m2) by difference in BMI. The differences in body height, body weight, blood gas analysis indexes [arterial partial pressure of oxygen (PaO2), arterial partial pressure of carbon dioxide (PaCO2)], lung function indexes [the percentage of the first second forced vital capacity/predicted value (FEV1%), the ratio of the first second forced vital capacity/forced vital capacity (FEV1/FVC)], hospitalization time, recurrence time and smoking index were compared between the two groups, the correlations between BMI and the indexes of blood gas analysis, pulmonary function, hospitalization time, recurrence time and smoking index in low BMI group were analyzed.Results The BMI (kg/m2: 21.09±2.03 vs. 16.39±1.26), PaO2 [mmHg (1 mmHg = 0.133 kPa): 63.59±4.95 vs. 56.54±3.23], FEV1% [(64.18±3.82)% vs. (59.82±5.73)%] and FEV1/FVC (63.83±3.20 vs. 59.28±3.63) in normal BMI group were all significantly higher than those in the low BMI group, PaCO2 in normal BMI group was significantly lower than that in the low BMI group (mmHg: 57.05±5.25 vs. 63.70±7.29). The results indicated that the patients in low BMI group had severer hypoxia and carbon dioxide retention than those in normal BMI group, and the lung function was also worse than that in the normal BMI group; the hospitalization time of normal BMI group was shorter than that of low BMI group (days: 11.70±2.36 vs. 15.25±2.80), and the recurrence time in normal BMI group was obviously longer than that of low BMI group (days: 93.78±57.85 vs. 48.58±17.85), smoking index in normal BMI group was markedly lower than that of low BMI group (year·branch: 2550.0±917.6 vs. 3652.5±1015.8), the differences were statistically significant (allP < 0.05). The linear correlation analyses showed that: BMI in low BMI group was positively correlated with PaO2, FEV1% and recurrence time (r = 0.557, 0.507, 0.455,P = 0.000, 0.000, 0.003), and was negatively correlated with PaCO2, hospitalization time and smoking index (r = -0.670, -0.405, -0.440,P = 0.009, 0.000, 0.005), and had no significant correlation with FEV1/FVC (r = 0.061,P = 0.707).Conclusion BMI is an important index for evaluating the severity and prognosis of patients with AECOPD.
