1.Discussion on the effects of glycyrrhetinic acid on cough variant asthma mice by adjusting Th2 deviation based on molecular docking technique

Objective:To evaluate the therapeutic effect of glycyrrhetinic acid on cough variant asthma (CVA) mice based on molecular docking technique; To explore the possibility of its treatment for cough variant asthma.Methods:The software of Autodock Vina was used for molecular docking. The mice were divided into control group, model group, prednisone acetate group, glycyrrhetinic acid high-, medium-, and low-dosage groups according to the random number table method, with 8 mice in each group. Except for the blank control group, all other groups were induced by egg protein to establish cough variant asthma models. Glycyrrhetinic acid high-, medium-, and low-dosage groups were orally administered glycyrrhetinic acid suspension at 20, 10, and 5 mg/kg, while the prednisone acetate group was orally administered prednisone acetate at 5 mg/kg. The blank control group and model group were orally administered equal volumes of physiological saline, once per day for 14 consecutive days. The animal asthma behavior was observed after drug administration. The secretion of bronchial mucus in lung tissue were observed by AB-PAS staining and the index of spleen were recorded. The protein expressions of Gata3, IL-4 and IL-13 in the spleen tissue were determined by Western blot.Results:Molecular docking results showed that glycyrrhetinic acid had good binding ability to Th2-related factors Gata3, IL-4 and IL-13. Results of animal experiment showed that compared with the model group, the mucus secretion decreased in glycyrrhetinic acid groups, the index of the spleen of mice obviously decreased, protein expression levels of IL-4 and IL-13 in the spleen tissue of mice in glycyrrhetinic acid high-, medium-, and low-dosage groups decreased ( P<0.05), and Gata3 in glycyrrhetinic acid medium- and low-dosage groups decreased ( P<0.05). Conclusion:Glycyrrhetinic acid can correct the shift of Th2 in the immune system of cough variant asthma mice and has a certain therapeutic effect.
2.Genetic analysis of a child with 46, XY Disorder of sex development due to a novel variant of NR5A1 gene

Objective:To analyze the clinical features and genetic basis of a child with Disorder of sex development (DSD).Methods:A child who was admitted to the Linyi People′s Hospital for primary amenorrhoea on July 29, 2019 was selected as the study subject. Clinical data of the child was collected. Chromosomal karyotyping and quantitative real-time PCR were used to detect Y chromosome microdeletions and other chromosomal aberrations. Next-generation sequencing was carried out for the child and her parents. Candidate variant was verified by Sanger sequencing and bioinformatic analysis.Results:The child, a 13-year-old girl, has featured primary amenorrhoea and onset of secondary sex characteristics of males. Ultrasound exam had detected no uterus and definite ovarian structure, but narrow band vaginal hypoecho and curved cavernoid structure. The child was found to have a 46, XY karyotype without an AZF deletion. DNA sequencing revealed that she has harbored a maternally derived c. 323delA (p.Q108Rfs*188) variant in the nuclear receptor subfamily 5 group A member 1 ( NR5A1) gene, which may result in a truncated protein. The variant was classified as pathogenic (PVS1+ PM2_Supporting+ PP4) based on the guidelines from the American College of Medical Genetics and Genomics. Conclusion:The NR5A1: c. 323delA variant probably underlay the pathogenesis of 46, XY DSD in this child. The discovery of the novel variant has enriched the mutational spectrum of NR5A1 gene and provided a basis for clinical diagnosis, treatment and prenatal diagnosis.

Notoginseng Radix et Rhizoma as a traditional Chinese herbal medicine has now been recognized and paid attention to by the pharmaceutical community.Modern phytochemical studies have shown that Panax notoginseng saponin is the main chemical compo-nent of Notoginseng Radix et Rhizoma.Modern pharmacological studies and clinical applications have revealed that it has anti-cancer,antioxidant and cardiovascular disease effects.In this study,we reviewed the research progress of the main chemical components and pharmacological effects of Notoginseng Radix et Rhizoma,with the aim of providing assistance for the clinical application and later stud-ies of Notoginseng Radix et Rhizoma.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection has spread worldwide and threatened human's health. With the passing of time, the epidemiology of coronavirus disease 2019 evolves and the knowledge of SARS-CoV-2 infection accumu-lates. To further improve the scientific and standardized diagnosis and treatment of maternal SARS-CoV-2 infection in China, the Chinese Society of Perinatal Medicine of Chinese Medical Association commissioned leading experts to develop the Recommendations for the Diagnosis and Treatment of Maternal SARS-CoV-2 Infection under the guidance of the Maternal and Child Health Department of the National Health Commission. This recommendations includes the epidemiology, diagnosis, management, maternal care, medication treatment, care of birth and newborns, and psychological support associated with maternal SARS-CoV-2 infection. It is hoped that the recommendations will effectively help the clinical management of maternal SARS-CoV-2 infection.
5.Anti-tumor Effect of Chinese Herbal Polysaccharides: A Review

Tumor has become one of the major diseases threatening human health at present. Conventional therapies for tumor have serious side effects and are prone to drug resistance, while new therapies are expensive and impose a heavy burden on patients. Chinese herbs effective in the treatment of tumor contain a variety of natural active ingredients, and many anti-tumor drugs used in clinical practice are derived from Chinese herbs. Polysaccharides, ubiquitous in Chinese herbs, are easy to extract and have antitumor, antiviral, and antioxidant activities. Studies have demonstrated that the polysaccharides from Chinese herbs have significant anti-tumor effect and are characterized by multi-target and multi-mechanism functioning, which can avoid the development of drug resistance, exhibiting a great research value and the potential to be developed into novel anti-tumor drugs. Their anti-tumor mechanisms mainly include the inhibition of tumor proliferation, promotion of apoptosis, induction of autophagy, influence on cell cycle, inhibition of tumor angiogenesis, modulation of epithelial-mesenchymal transition (EMT) and immunity, promotion of tumor oxidative stress, and regulation of phosphatidylinositol 3-kinase/ protein kinase B (PI3K/Akt), Toll-like receptor 4 (TLR4), mitogen-activated protein kinase/nuclear factor-kappa B (MAPK/NF-κB), adenosine 5'-monophosphate (AMP)-activated protein kinase/mechanistic target of rapamycin (AMPK/mTOR), epidermal growth factor receptor/extracellular signal-regulated kinase (EGFR/ERK), p53, and Wnt/β-catenin signaling pathways.
6.Effect of family integrated care on the psychological status of parents of premature infants: a systematic review

Objective:To systematically evaluate the effect of family integrated care on the psychological status of parents of premature infants.Methods:Randomized and semi randomized controlled trials related to the effect of family integrated care on the psychological status of parents of premature infants were systematically searched in PubMed, the Cochrane Library, Web of Science, Embase, Scopus, Wiley Online Library, CINAHL, China Biomedical Literature Database, China National Knowledge Infrastructure (CNKI), WanFang Data and VIP, and their references were tracedback. The retrieval time limit was from the establishment of each database to March 3, 2022. Two researchers screened and evaluated the article according to the inclusion and exclusion criteria, and extracted the data. RevMan 5.3 was used for meta-analysis.Results:A total of 14 articles were included, including 3 296 subjects. Meta-analysis showed that family integrated care could alleviate anxiety [ SMD=-1.50, 95% CI (-2.11, -0.89), P<0.000 01], depression [ SMD=-2.08, 95% CI (-3.32, -0.85), P=0.000 9] and stress [ SMD=-1.09, 95% CI (-1.61, -0.57), P<0.000 1] of parents of premature infants. Conclusions:Family integrated care can alleviate anxiety, depression and stress of parents of premature infants. Due to the limitation of the number and quality of included studies, it still needs to be confirmed by large samples and well-designed randomized controlled studies.
7.Effects of computer-assisted cognitive behavioral therapy in orthopedic traumatic patients with anxiety

Objective:To explore the effects of computer-assisted cognitive behavioral therapy (CCBT) on anxiety, sleep and pain in orthopedic traumatic patients.Methods:Totally 80 traumatic patients receiving selective surgery in the Department of Orthopedics at First Hospital of Shanxi Medical University between February and September 2019 were selected by convenient sampling and randomized into an observation group and a control group, with 40 patients in each group. Patients in the control group received routine health education and psychological counseling, while patients in the observation group received CCBT 5 times on this basis. The Self-Anxiety Scale (SAS) , Asens Insomnia scale (AIS) and Visual Analogue Scale (VAS) were used to evaluate the anxiety, sleep and pain status of the two groups before and after intervention.Results:The anxiety score of the observation group after receiving CCBT was (45.85±4.04) , lower than (49.60±2.27) of the control group, and the difference was statistically significant ( t=5.242, P< 0.05) . The sleep score of the observation group after intervention was (5.02±1.44) , lower than (6.42±1.44) of the control group, and the difference was statistically significant ( t=4.334, P< 0.05) . The pain score of the observation group after intervention was (3.75±1.17) , lower than (4.40±1.03) of the control group, and the difference was statistically significant ( t=2.816, P< 0.05) . Conclusions:CCBT can effectively ameliorate the anxiety, sleep and pain response of orthopedic traumatic patients, make psychological treatment more standardized, and benefit a wider population.
8. Repair of rabbit articular cartilage defect by a novel injectable bioactive glass scaffold

Objective:
By comparing different repairing effects of different methods on articular cartilage defects in rabbit model, the ability of a new bioactive glass scaffold to repair cartilage defect was studied.
Methods:
We prepared the PSC/CS bone cement by mixing the bioactive glass (BG) powder composed of 10.8%P2O5-54.2%SiO2-35%CaO (PSC), chitosan solution (CS) and calcium sulfate hemihydrate (CSH), then the bone cement was sized by a mold to form cylindrical scaffold. In vivo experiment, 18 male rabbits were divided into three groups randomly, including blank group, BG group and TGF-β1 group, which was added TGF-β1 into BG scaffold. Both knees of each rabbit were made cartilage defect for the same group, and no intervention was applied in B group, then implanted scaffolds into defects in both experimental groups. At 6 and 12 weeks after surgery, observed the macroscopic growth, histologic staining and collagen II immunohistochemistry (IHC), and the International Cartilage Repair Society (ICRS) and Wakitani score were used to analyze the experimental results quantitatively.
Results:
At 6 weeks after surgery, there was no obvious difference between blank group and BG groups, but the macroscopic result of TGF-β1 group was better than the other two groups and its ICRS score 4.67±0.52 points was statistically higher than BG group 2.83±0.75 points (
9.Clinical features of cerbrotendinous xanthomatosis

Objective To analyze the clinical manifestations, imaging features, histopathology and genes of cerebrotendinous xanthomatosis (CTX) to improve the understanding of clinical workers on the disease. Methods The imaging examination, histopathological and gene detection methods were synthetically applied, and the disease characteristics of two patients with CTX, admitted to our hospital in March 2018 , were analyzed. Results (1) Patient one was a 52-year-old male, with typical neurological symptoms: weakness of both lower limbs, damage of pyramidal tract and extrapyramidal system, cognitive impairment, and ataxia; non-neurological symptoms included cataract, arch foot, and Achilles tendon mass; cranial MR imaging indicated symmetrical abnormal signal of cerebellar dentate nucleus, low signal on T1WI, and slightly high and low signal intensity on T2WI/FLAIR; achilles tendon biopsy showed fibrous connective tissues with multiple xanthoma cells and multinucleated giant cells aggregation, accompanied by cholesterol crystallization. (2) Another 16-year-old male presented with cerebellar ataxia, recurrent seizures, mental and motor retardation, and congenital cataract; two heterozygous mutations of CYP27A1 gene were detected by gene detection; and the mutations were c.373-379 delCCAGTAC and c.1420C>T. Conclusion The clinical manifestations of CTX are varied; early imaging examination lacks specificity; it can be clearly diagnosed by histopathology and CYP27A1 gene detection.
10.Study on the Lipid-lowering Effect and Potential Mechanism of the Couplet Medicines of Flastem Milkvetch Seed and Tribulus Terrestris on Hyperlipidemia Rats

Objective: To explore the lipid-lowering effect and potential mechanism of the couplet medicines of Flastem Milkvetch Seed and Tribulus terrestris on hyperlipidemia rats. Methods: Forty-eight Sprague-Dawley rats were randomly divided into 6 groups, normal control group, high-fat model group, simvastatin group, Tribulus terrestris group, Flastem Milkvetch Seed group, and couplet medicines of Flastem Milkvetch Seed and Tribulus terrestris group, with 8 rats in each group. The normal control group was fed with basal diet, and the other groups were fed with high-fat diet to establish a hyperlipidemia rat model. At the same time, different group rats were treated with different drug respectively. 6 weeks later, the body weight and visceral index of the rats were measured. Levels of serum cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), and low density lipoprotein cholesterol (LDL-C) were detected by automatic biochemical analyzer. The mRNA and protein expression of HMG coenzyme A reductase (HMGCR), cholesterol 7-alpha hydroxy-lase (CYP7A1) and low density lipoprotein receptor (LDL-R) were detected by RT-PCR and western blot. Results: Compared with model group, the serum levels of TC, TG, and LDL-C were significantly decreased in all Chinese medicine groups, and the serum level of HDL-C was significantly increased in Flastem Milkvetch Seed group and couplet medicines of Flastem Milkvetch Seed and Tribulus terrestris group. Moreover, the mRNA and protein expression of HMGCR, CYP7A1, and LDL-R in rat liver also significantly increased in all Chinese medicine treatment groups. And couplet medicines of Flastem Milkvetch Seed and Tribulus terrestris group shows more significantly effect. Conclusion: Couplet medicines of Flastem Milkvetch Seed and Tribulus terrestris significantly regulate the serum lipid levels on hyperlipidemia rat model, and its mechanism may be related to the regulating expressions of HMGCR, CYP7A1, LDL-R gene.
