(+)-Syringaresinol regulates the growth and metastasis of triple-negative breast cancer cells through NF-κB signal

Abstract:
Objective:To reveal the effect of(+)-Syringaresinol on the growth and metasta-sis of triple negative breast cancer(TNBC)cells,and to analyze its effect on nuclear factor-κB(NF-κB)signal.Methods:According to the concentration of(+)-Syringaresinol,MDA-MB-231 cells were divided into 0 μM group,2 μM group,4 μM group,8 μM group,16 μM group,32 μM group,64 μM group and 128 μm group.MDA-MB-231 cells were incubated with corresponding concen-tration of(+)-Syringaresinol for 48 h.Cell proliferation and(+)-Syringaresinol semi-inhibitory con-centration(IC50)were detected by MTT method.Apoptosis was detected by Annexin V-FITC/PI kit.Cell migration and invasion were detected by Transwell.The relative expressions of Bax,Bcl-2,matrix metalloproteinase(MMP)-2,MMP-9,E-cadherin,N-cadherin and Vimentin mRNA were detected by qRT-PCR.The phosphorylation level of NF-κB p65 was detected by Western blot.Re-sults:Compared with 0 μM group,the growth inhibition rate of MDA-MB-231 cells in 2~128μm group increased(P<0.05).The IC50 value of(+)-Syringaresinol on MDA-MB-231 cells was 25.24 μ.m.Compared with 0 μM group,the apoptosis rate of MDA-MB-231 cells in 2~128 μm group increased,the relative expression of Bax mRNA increased,the relative expression of Bcl-2 mRNA decreased,the number of migratory and invasive cells decreased,the relative expression of MMP-2 and MMP-9 mRNA decreased,the relative expression of E-cadherin mRNA in-creased,the relative expression of N-cadherin and Vimentin mRNA decreased,relative expression of p-NF-κB p65/NF-κB p65 protein decreased(P<0.05).Conclusion:(+)-Syringaresinol inhibits the growth and metastasis of TNBC cells in a concentration-dependent manner.The anti-TNBC ef-fect of(+)-Syringaresinol may be related to the inhibition of the activation of NF-κB signal pathway.
