Establishment and Evaluation of the IBS-D Visceral Hypersensitivity Mouse Model

Abstract:
Objective To establish and evaluate a mouse model of visceral hypersensitivity in diarrhea-predominant irritable bowel syndrome(IBS-D).Methods A mouse model of IBS-D visceral hypersensitivity was established by gavage of different doses of Citrobacter rodentium(CR)combined with chronic water avoidance stress(WAS).Sixty C57BL/6J mice were randomly divided into the normal control group(NC group)and the model group.The model group was established by different doses of CR(0.12ml,0.15ml,0.18ml,0.21ml,0.30ml)combined with WAS.After modeling,the fecal trait score,fecal water content,total intestinal transit time,colonic transit time,abdominal withdrawal reflex(AWR)score,and pathological changes of colon tissue were observed.Results Com-pared with the NC group,the mice in the model group had soft stool,increased fecal water content,reduced total intestinal transit time(P<0.05),significantly reduced colonic transit time(P<0.005),and reduced threshold water injection of AWR score 3.In addition,in the 0.21ml and 0.30ml CR groups,the edge of feces was blurred and mush,the threshold of AWR score 3 was significantly reduced(P<0.01),the colon tissue cells were arranged disorderly,inflammatory cell infiltration,crypt hyperplasia,and the plica was short-ened.Conclusion The visceral hypersensitivity mouse model of IBS-D can be successfully established by gavage of CR combined with WAS.According to the evaluation results of the model,it is recommended to use 0.21ml CR dose by gavage to establish the model.
