				        	Objective:To study the role and molecular mechanism of long non-coding RNA(lncRNA)LINC00472 in the prolifera-tion,migration and invasion of oral squamous cell carcinoma(OSCC)cells by targeting miR-155-5p.Methods:Bioinformatics pre-diction and dual-luciferase reporter test were carried out to examine the relationship between LINC00472 and miR-155-5p.The ex-pression levels of LINC00472 and miR-155-5p in 40 OSCC samples and SCC-15 cells were detected by qRT-PCR.SCC-15 cells was cultured and allocated into:control group(blank),LINC00472 overexpression group(OE-LINC00472),miR-155 overexpression group(miR-155-5p mimics)and overexpression of LINC00472 and miR-155-5p group(OE-LINC00472+miR-155-5p mimics).MTT,scratch and Transwell assays were utilized to determine the proliferation,migration and invasion of SCC-15 cells.The expres-sion levels of E-cadherin,Vimentin and matrix metallopeptidase9(MMP9)were detected by qRT-PCR and western blotting.Results:miR-155-5p was a direct target gene of LINC00472.LINC00472 was down expressed(P<0.01),while miR-155-5p was highly ex-pressed in OSCC tissues and SCC-15 cells(P<0.01).LINC00472 was negatively correlated with miR-155-5p in OSCC tissues(r=-0.766,P<0.01).Compared with control group,OE-LINC00472 group showed lower proliferation,migration invasion,and lower expression of Vimentin and MMP9,and higher expression of E-cadherin(P<0.01).miR-155-5p mimics group showed higher prolif-eration,migration,invasion,and higher expression of Vimentin and MMP9 expression,and lower expression of E-cadherin(P<0.01).miR-155-5p mimics reverted the effects of overexpressed LINC00472 on the proliferation,migration and invasion of SCC-15 cells.Conclusion:LINC00472 plays a protective role in the progression of OSCC,LINC00472/miR-155-5p axis may be a potential therapeutic target for OSCC.
