Abstract:
Objective To detect the gene expression profiles of miRNA in joint fluids of patients with rheumatoid arthritis(RA)and healthy people and clinical significance.Methods The expressions of miRNAs in joint fluids of 3 RA patients and 3 healthy adults were detected by miRNA chip technology,and miRNAs with significant differential expressions were selected to be validated by the qRT-PCR.Correlations between selected miRNA and clinical and laboratory indexes of RA were analyzed.Results In the gene expression profile of miRNA in joint fluids of patients with RA,the expressions of 23 miRNAs significantly changed when compared with those in the healthy people.The results of selected miRNAs by qRT-PCR were correspond with the Results of the miRNA chip technology.The difference of miR-146a expression in joint fluid between RA patients and healthy controls was the most significant.Pearson correlation analysis showed that serum miR-146a expression in joint fluid of RA patients was positively correlated with rheumatoid factor(RF)value and DAS28 score.ROC curve analysis showed that the area under the curve(AUC)of miR-146a was 0.71,and the sensitivity and the specificity of miR-146a in joint fluid in predicting RA were 85.29%and 61.54%respectively.Conclusion There is significant difference in expression of miRNAs in joint fluid between RA patients and healthy people.Expression of miR-146a increases significantly in joint fluid of RA patients,and it level is significantly correlated with the clinical and laboratory indexes of RA.
