Abstract:
Objective:To prepare monoclonal antibodies against Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) for developing antigen diagnostics.Methods:The supernatants of hybridoma cell lines secreting monoclonal antibodies against SARS-CoV were collected and were subjected to examinate the reaction between the monoclonal antibodies and SARS-CoV-2 N protein by using western blot and indirect enzyme-linked immunosorbent assay (ELISA). The reactive monoclonal antibodies were used to detect SARS-CoV-2 in Vero cell cultures by using immunofluorescence assay.Results:Two monoclonal antibodies, termed A1 and A2, were obtained from the hybridoma cell lines. They can both react with SARS-CoV-2 N protein, while A2 can also recognize SARS-CoV N protein. These monoclonal antibodies can be used in ELISA and immunofluorescence experiments.Conclusions:Two monoclonal antibodies against SARS-CoV-2 N protein were obtained, providing materials for future development of SARS-CoV-2 antigen detection.
