		        	Abstract:
			       	
			       		
				        
				        	Objective To explore the mechanism of quercetin(Quer)inducing apoptosis of breast cancer cells.Methods Human breast cancer cells MCF-7 were divided into 4 groups:control group,experimental group,negative control(NC)group and miR-373-3p mimics group.The control group was cultured under normal conditions;the experimental group was treated with 80 μmol·L-1 quercetin;NC group was treated with 80 μmol·L-1 quercetin and transfected with 50 nmol·L-1 miR-373-3p negative control;miR-373-3p mimics group was treated with 80 μmol·L-1 quercetin and transfected with 50 nmol·L-1 miR-373-3p.Real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect microRNA 373-3p(miR-373-3p),thioredoxin-interacting protein(TXNIP),NOD-like receptor family pyrin domain containing protein 3(NLRP3),B-cell lymphoma-2(Bcl-2)and Bcl-2 associated X protein(Bax).Terminal dexynucleotidyl transferase(TdT)-mediated dUTP nick end labeling(Tunel)assay was used to detect the apoptosis rate of each treatment group.Results The relative expression levels of miR-373-3p in control group,experimental group,NC group and miR-373-3p mimics group were 1.00±0.15,0.61±0.08,0.67±0.05 and 1.19±0.16,respectively;the relative expression levels of TXNIP mRNA were 1.00±0.12,1.83±0.21,1.71±0.19 and 1.26±0.14,respectively;the relative expression levels of NLRP3 mRNA were 1.00±0.11,0.73±0.06,0.65±0.08 and 1.27±0.15,respectively;the relative expression levels of Bcl-2 mRNA were 1.00±0.15,0.43±0.06,0.37±0.04,0.83±0.11,respectively;the relative expression levels of Bax mRNA were 1.00±0.12,2.13±0.25,1.96±0.21,1.38±0.17,respectively;the apoptosis rates were(100.00±13.55)%,(217.24±25.13)%,(196.37±21.53)%and(134.71±16.42)%,respectively.The above indicators of the experimental group were compared with the control group,and the above indicators of miR-373-3p mimics group were compared with NC group,and the differences were statistically significant(P<0.05,P<0.01,P<0.001).Conclusion Quercetin can induce apoptosis of breast cancer cells by regulating miR-373-3p/TXNIP/NLRP3 pathway.
