Effects of berberine metabolites on differentiation and lipid metabolism of preadipocytes

Abstract:
Objective To investigate the effect of berberine(BBR)metabolites on 3T3-L1 preadipocytes differentiation and lipid metabolism.Methods 3T3-L1 preadipocytes were divided into control group and BBR metabolites treated-group.The cell viability was measured by cell proliferation counting method.The BBR,dihydroberberine(DHB),berberrubine(BRB),columbamine(COL),palmatine(PAL),jatrorrhizine(JAT)and demethyleneberberine(DEM)groups were treated with 5 μmol·L-1 BBR,2.5 μmol·L-1 DHB,10 μmol·L-1 BRB,20 μmol·L-1 COL,25 μmol·L-1 PAL,50 μmol·L-1 JAT and 50 μmol·L-1 DEM,respectively;the control group was cultured normally.After inducing differentiation,expression levels of lipoprotein lipase(LPL),and acetyl-CoA carboxylase A(ACACA)were detected by real-time fluorescence quantitative polymerase chain reaction.Results Compared with the control group,the DHB,COL,PAL,JAT and DEM did not affect the cell viability when incubated at the used concentration for 48 h.However,20 μmol·L-1 BRB significantly decreased the cell viability to(86.55±1.99)%.Twelve days after treatment,the relative expression levels of LPL mRNA in control,BBR,DHB,BRB,COL,PAL,JAT and DEM groups were 1.00±0.02,0.41±0.04,0.44±0.04,1.40±0.08,1.27±0.10,1.15±0.07,3.06±0.30 and 1.45±0.07;the relative expression levels of ACACA mRNA were 1.01±0.01,0.62±0.04,0.69±0.06,1.78±0.17,4.77±0.19,4.20±0.14,6.38±0.30 and 1.18±0.08,respectively.The relative expression levels of LPL in BBR,DHB and JAT groups and the relative expression levels of ACACA in BRB,COL,PAL and JAT groups were all compared with those of the control group,and the differences were statistically significant(all P<0.05).Conclusion BBR metabolites can inhibit differentiation and promote lipid metabolism in adipocytes,which therefore reduce lipid accumulation.
