Effect of Qingnao Yizhi on expression of NogoA and Rho-associated kinase in hippocampus of vascular dementia rats

Abstract:
Objective To investigate the effect of Qingnao Yizhi formula (QNYZF) on NogoA and Rho-associated kinase(ROCK-2)expression in hippocampus of vascular dementia (VaD) of rats.Methods The rat model of VaD was established by bilateral ligation of the common carotid artery (2-VO).The male Sprague-Dawley rats were randomly divided into 6 groups,each group had 6 rats:sham operation group,model group,control group,experimental-L,experimental-M and experimental-H groups.The sham operation group and model group were fed with equal volume of 0.9% NaCl;the control group were fed with donepezil hydrochloride 0.52 mg · kg-1 · d-1;the experimental-L,experimental-M and experimental-H groups were fed with QNYZF 3,6,12 g · kg-1 · d-1,respectively for 4 weeks.After 4 weeks of intragastric administration,the heart was perfused and the hippocampus of the brain was rapidly taken,and then the protein expression of NogoA and ROCK-2 in hippocampus were detected by immunohistochemistry and reverse transcription-PCR respectively.Results The relative gray values of NogoA + cells in the hippocampus:the sham operation group,model group,control group,experimental-L,experimental-M and experimental-H groups were 435.30 ± 127.98,5365.94 ± 1351.96,1585.92 ±360.64,1953.77 ±653.58,2300.94 ±742.25,1681.35 ±437.87.Comparison between the sham group and the model group,the difference was significantly increased (P < 0.05);comparison between the model group and control group,the difference was significantly increased (P < 0.05);but experimental-L,experimental-M and experimental-H groups was not statistically significant (P > 0.05).The relative gray values of ROCK -2+ cells in hippocampus of rats:the 6 groups were 68.11 ± 12.58,556.79 ± 141.85,130.20 ± 31.05,357.24 ± 100.07,193.39 ±40.79 and 503.64 ±219.87,respectively.Comparison between the sham group and the model group,the difference was significantly increased (P < 0.05);comparison between the control group,experimental-M group and model group,the difference was significantly increased (P < 0.05,P < 0.01).Conclusion QNYZF can promote synaptic remodeling of neurons by inhibiting NogoA-NgR/Rho-ROCK signaling pathway.
