Abstract:
Objective To study the effect of miR-21 on proliferation of hepatocellular carcinoma cell line HepG2 and its related signal pathway.Methods The liver cancer tissues and cancer adjacent tissues were separated from 98 cases of liver cancer patients that accepted in our hospital from June 2012 to June 2015.Immunohistochemistry was applied to detect the expression level of mitogen activated protein kinase kinase 3 (MAP2K3).HepG2 cells were divided into mimic-miR-21 groupand miR-21 inhibitor group,and transfected by mimic-miR-21 and miR-21 inhibitor.After 48 h of transfection,HepG2 proliferation status at 24,48,72 h were detected by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium,inner salt (MTS).The mRNA expressions of miR-21 and MAP2K3 were detected by quantitative real time polymerase chain reaction(qPCR).MAP2K3 protein expression was detected by Western blot.Results The expression levels of MAP2K3 in hepatocellular carcinoma and in its adjacent tissues were 1382 ± 105 and 5872 ± 289;mimic-miR-21 and miR-21 inhibitor HepG2 cells were transfected by mimic-miR-21 and miR-21 inhibitor for 24 h,the expression of miR-21 levels were 6.45 ± 1.21 and 0.43 ± 0.12.The mimic-miR-21 and miR-21 inhibitor proliferation activity in 24,48,72 h were (100 ± 12) %,(60 ± 18) %;(138 ±--12) %,(45 ± 8) %;(146 ± 9) %,(38 ±6)%,the differences of those items were all statistically significant(all P < 0.05).Mimic-miR-21 and miR-21 inhibitor HepG2 cells were transfected by mimic-miR-21 and miR-21 inhibitor for 24 h,MA2PK3 expression in mimic-miR-21 and miR-21 inhibitor were 0.17 ±0.03 and 0.97 ±0.09,and there was statistically difference between the two groups (P < 0.05).Conclusion miR-21 can induce the hepatocellular carcinoma cell proliferation through MAP2K3 inhibition.
