Abstract:
Objective To establish the high performance liquid chroma-tography ( HPLC ) method for determination of the concentration of imi-penem in sputum.Methods HPLC was performed on a C18 column, ( Venusil MP, 4.6 mm ×250 mm, 5 μm).The mobile phase was com-posed of phosphate buffer 10 mmol? L-1 ( pH 7.2 ) and methanol ( with 0.3 mmol? L-1 tetraethyl ammonium bromide ) ( 96∶4 ) .The flow rate was 1 mL? min-1 .Stabilizer was a mixture of 0.5 mmol? L-1 3-mor-pholine propanesulfonic acid buffer ( pH 6.8 ) , glycol and pure water (2∶1∶1).The column temperature was set at 30 ℃.The detection wavelength was set at 300 nm.The flow rate was 1.0 mL? min-1 .And then the specificity , and quantitative lower limit standard curve , preci-sion and recovery rate and stability of the method was investigated . Results A good linearity was shown in the concentration range 0.25-50 μg? mL-1 ( r=0.999 0-0.999 6 ).The limit of quantitation concen-tration of imipenem was 0.25 μg? mL-1 .The intra-day relative stan-dard deviation ( RSD) were 3.29%, 4.94%, 1.35%, and inter -day RSD were 2.57%, 6.15%, 6.98% at 3 concentrations ( 0.3 , 15 , 45μg? mL-1 ) , the extraction recovery of each quality control concentra-tion of the diluted sputum was ( 86.77 ±0.88 )%, ( 98.31 ±0.47 )%and (95.51 ±1.54 )%, respectively. Conclusion The method is simple and accurate .It can be used to determine the concentration of imipenem in sputum .
