1.Association of physical activity and sugar sweetened beverage consumption with psychological sub health among middle school students in Bao an District, Shenzhen

Objective:
To explore the association of physical activity and sugar sweetened beverage consumption with psychological sub health among middle school students in Bao an District, Shenzhen, so as to provide a reference for adolescent mental health promotion.
Methods:
A questionnaire survey was conducted in November 2024 by a stratified cluster random sampling method to select 6 926 junior and senior middle school students from 5 middle schools in Shenzhen. The questionnaire from Youth Risk Behavior Surveillance System was used to assess the consumption of sugar sweetened beverages, and physical activity Rating Scale was used to assess the level of physical activity, and Brief Instrument on Psychological Health of Youths was used to evaluate the psychological sub health status. The Chi -square test was used to analyze the differences in the detection rates of psychological sub health among different groups of middle school students, and a multivariate Logistic regression model was established to analyze the effects of physical activity and sugar sweetened beverage consumption and their combined effects on the psychological sub health of middle school students.
Results:
The detection rate of psychological sub health among middle school students in Bao an District, Shenzhen was 18.93%. Multivariate Logistic regression analysis showed that, after controlling for confounding factors such as gender, school stage, family residence, family economic status, parental literacy, academic stress and number of friends, lack of physical activity or excessive sugar sweetened beverage consumption were associated with increased risks of psychological sub health among middle school students ( OR =1.36, 1.45); and the highest risk of psychological sub health was found in middle school students who were lack of physical activity and excessive sugar sweetened beverage consumption ( OR =2.59) ( P <0.01). Further analysis by school stages showed that junior high school students with sufficient physical activity and excessive intake of sugary drinks ( ROR =2.10), lack of physical activity and excessive intake of sugary drinks ( ROR =2.31) were at higher risks of psychological sub health than senior high school students( P <0.05).
Conclusions
Insufficient physical activity and excessive sugar sweetened beverage consumption are closely associated with an increased risk of psychological sub health among middle school students. Effective interventions should be targeted to reduce the risk of psychological sub health problems among middle school students.
2.Evaluation on the effectiveness of comprehensive control of a bedbug infestation incident in Jiading District, Shanghai

ObjectiveTo investigate the infestation of bedbugs in a staff dormitory in Jiading District, Shanghai, to explore the measures to dispose Cimex lectularius linnaeus, so as to provide a scientific basis for the prevention and control of bedbugs. MethodsThe infestation of bedbugs in the dormitory of the company was determined through field investigation, accompanied by scientific guidance under the comprehensive control measures and an effect evaluation of the control results. ResultsA total of 114 rooms distributed in 3 dormitory buildings were investigated, with an average infestation rate of 42.11%, of which building B has the highest infestation rate of 51.52%. Six bedbug specimens were collected by visual inspection in the room, and all of them were identified as Cimex lectularius linnaeus. After a series of comprehensive control measures including environmental cleanup, aerosol elimination, replacement of wooden beds with iron frame beds, and purchase of all-inclusive mattress, the bedbug infestation rate dropped to 5.26%. ConclusionComprehensive control can effectively prevent the breeding and spread of bedbugs. Dissemination and education effort should be strengthened in case of the occurrence of bedbug infestation, together with an implementation of long-term and continuous surveillance and monitoring.
3.Establishment and optimization of a hyperuricemic nephropathy mouse model

The aim of this study was to establish an efficient and stable mouse model of hyperuricemic nephropathy (HN) by testing different modes of administration of potassium oxonate (PO) combined with hypoxanthine (Hx). Animal welfare and experimental procedures were in accordance with the regulations of the Animal Ethics Committee of Guangdong Pharmaceutical University. Male C57BL/6 mice were randomly divided into a control group, a PO+Hx group (i.g.; 100 mg·kg-1·d-1 and 500 mg·kg-1·d-1, respectively), and a PO+Hx group (i.p.; 100 mg·kg-1·d-1, and 500 mg·kg-1·d-1). This HN model was induced by combination of PO and Hx administration once daily for 21 days. Objective::To study whether long-term administration of Gastrodiae Rhizoma powder can improve the learning and memory ability of APPswe/PSldE9 double transgenic (APP/PS1) Alzheimer' s disease(AD) model mice and delay the progress of AD whether these effects are related to the regulation of antioxidant stress pathway in Kelch-like epoxylopropylamine-related protein 1(Keap1)-nuclear factor E2 related factor 2 (Nrf2)/heme oxygenase(HO)-1, and further explore the neuroprotective mechanism of Gastrodiae Rhizoma powder and its role in the prevention and treatment of AD. Method::APP/PS1 double transgenic mice model, the mice consisted of five groups: normal, normal administration group, model group, Gastrodiae Rhizoma powder prevention group, Gastrodiae Rhizoma powder treatment group.The mice in the normal administration group and the Gastrodiae Rhizoma powder prevention group were given the same dose of Gastrodiae Rhizoma powder (1.5 g·kg-1) daily at the age of 8 weeks.The normal group and model group were given the same amount of normal saline at the same time, until 24 weeks old, Morris water maze was used to test the learning and memory ability of mice, and the treatment group was treated with Gastrodiae Rhizoma powder at 22 weeks old.The mice were given the same dose of Gastrodiae Rhizoma powder (1.5 g·kg-1) every day for 2 weeks.The number of crossing platform, escape latency and platform residence time of mice were detected by Morris water maze from 24 weeks old to 24 weeks old.RNA, Real-time PCR was extracted from mouse hippocampus to detect the mRNA level of Nrf2, HO-1, Keap1, and Western blot was used to detect the expression of Nrf2, HO-1, Keap1 protein in mouse hippocampus. 5.Effects of external fixation as a terminal versus staged treatment on prognosis in patients with open tibiofibular fracture

Objective:To compare the effects of external fixation as a terminal versus staged treat-ment on the health-related quality of life and function in patients with open tibiofibular fracture.Methods:From March 2017 to October 2018, 52 patients with open tibiofibular fracture were admitted to Department of Orthopaedic Surgery, Honghui Hospital.They were 37 males and 14 females, aged from 19 to 62 years(mean, 39.9 years).They were assigned into 2 groups subjected to 2 different treatment plans.In group A of 35 cases primary external fixation was replaced by terminal internal fixation while in group B of 17 cases primary ex-ternal fixation continued to the end.The 2 groups were compared in terms of the physiological total score (PCS) and mental component summary (MCS) in the 12-item short-form health survey (SF-12) at 1, 3, 6, 9 and 12 months postoperation, fracture healing time, time for external fixation, incidence of complications, and the lower extremity functional scale (LEFS).Results:The 2 groups were compatible due to insignificant differences in the general clinical data ( P>0.05).The follow-ups for the 2 groups were all beyond 12 months.PCS and MCS peaked both at 12 months postoperation in group A, but peaked respectively at 6 and 3 months postoperation in group B.Compared with group B, group A showed significantly higher PCS and MCS at 6, 9 and 12 months postoperation, significantly shorter fracture healing time (19.2 weeks ± 4.6 weeks versus 23.3 weeks ± 5.6 weeks), significantly shorter time for external fixation (15.6 days ± 4.2 days versus 270.0 days ± 15.4 days), significantly lower incidence of complications[5.7%(2/35) versus 35.3%(6/17)], significantly higher LEFS scores (88.3±7.2 versus 78.5 ± 5.2), and significantly higher the excellent and good rate by the Johner-Wruh scoring [94.3%(33/35) versus 70.6%(12/17)] (all P< 0.05). Conclusion:Compared with external fixation as a terminal treatment, external fixation followed by internal fixation can significantly improve the health-related quality of life and function in patients with open tibiofibular fracture and effectively reduce the incidence of postoperative complications.
6.Establishment and Application of HPA1-6, 15 Platelet Donor Bank in Beijing Area.

OBJECTIVE:
To establise the bank of platelet donors with the human platelet antigen (HPA) 1-6, 15 genes so as to provide the HPA-matched platelets for the patients.
METHODS:
The HPA genotyping of platelets donors and patients with platelet antibody positive confirmed by sercening was performed by using the SSP-PCR; the efficacy of transfusing the HPA-matched platelets for 37 cases platelet antibody positive was analyzed.
RESULTS:
The most common genotype in platelet donors were HPA-1a/1a-2a/2a-3a/3b-4a/4a-5a/5a-6a/6a-15a/15b, followed by HPA-1a/1a-2a/2a-3a/3a-4a/4a-5a/5a-6a/6a-15a/15b; the most common genotype in 53 cases of platelet antibody positive confirened by screening were HPA-1a/1a-2a/2a-3a/3b-4a/4a-5a/5a-6a/6a-15a/15b. Among 37 patients with platelet antibody positive confirened by screeming, 28 showed that the transfusion of HPA-matched platelets was effective with statistically significant difference in comparison with random transfusion group. The HPA-3, HPA-15 were the main factors leading to polymorphisms.
CONCLUSION
HPA-3 and HPA-15 are polymorphic, which should be focused on. HPA-matched platelets can improve the efficiency of platelet transfusion, and avoid the waste of blood resources. The genotypes of platelet donors can basically meet the requirements for common genotype transfusion.
7.Effects of laser stimulation with different power on oxygen consumption rate of CC myoblasts in mice.

OBJECTIVE:
To investigate the effects of 650 nm laser irradiation on cell oxygen consumption rate in CC myoblasts following different doses.
METHODS:
CC cells were irradiated with 650 nm laser(λ=650 nm, p=5 mW) with energy densities of 0, 0.4, and 0.8 J/cm. Cell oxidative function was measured by oxygen consumption rate kit. Protein expression of myogenic determination factor (MyoD), peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α), mammalian target of rapamycin (mTOR) and its phosphorylation were detected by Western blot.
RESULTS:
Compared to the control group, the expression levels of MyoD, PGC-1α protein were increased and cell oxygen consumption rate was promoted in the low dose group(<0.05). MyoD and PGC-1α protein expressions were also increased(<0.05), the ratio of mTOR and its phosphorylationwere decreased significantly in the high dose group(<0.05).
CONCLUSIONS
650 nm laser irradiation that dose is 0.4 J/cm enhances cell oxidative function, it related to that proper dose laser irradiation promoted the expression of PGC-1a protein.

8.Clinical efficacy of tripterygium wilfordii in the treatment of early diabetic nephropathy and its mechanism

Objective To investigate the clinical efficacy of tripterygium wilfordii in the treatment of early diabetic nephropathy and its mechanism .Methods 86 patients with early diabetic nephropathy admitted to Jiaxing Hospital of Zhejiang Provincial Armed Police Corps from December 2015 to December 2017 were selected as research subjects.They were randomly divided into two groups according to the digital table ,with 43 cases in each group .The control group was treated by conventional internal medicine , while the observation group was added tripterygium glycosides on the basis of the control group .The clinical efficacy of the two groups and the changes of serum matrix metalloproteinase-9 (MMP-9) and plasminogen activator -1 (PAI-1) before and after treatment were compared . Results The total effective rate of the observation group was significantly higher than that of the control group (86.04%vs.58.14%)(χ2 =8.323,P=0.004).The MMP-9 and PAI-1 levels and urinary protein of the two groups after treatment were significantly improved ,but the changes of the observation group [ ( 57 .36 ±10 .24 ) ng/L, (24.39 ±7.66)ng/mL,(70.35 ±12.58)μg/min] were more significant than those of the control group [(85.62 ± 15.42)ng/L,(29.64 ±8.61)ng/mL,(102.57 ±20.36)μg/min](t=10.011,P<0.001;t=2.987,P=0.004;t=3.828,P<0.001).Conclusion The therapeutic effect of tripterygium wilfordii in the treatment of early diabetic nephropathy is significant .The mechanism is related to improving the expression of MMP -9 in kidney tissue and reducing the level of PAI-1.

10.Effects of cyclic mechanical stretch stimulation on proliferation and aerobic capacity of C2C12 myoblasts

Objective To investigate the influence of mechanical stretch at different frequencies on proliferation and aerobic capacity of mice myoblast cells C2C12. Methods C2C12 cells cultured in vivo were exposed to mechanical strain with the magnitude of 15% at the frequency of 1 and 2 Hz, respectively, for 2 hours per day over a period of 4 days by using Flexercell Cell Tension System, while in control group C2C12 cells were cultured statically. The C2C12 cells were observed by inverted phase contrast microscope. CCK-8 Cell Counting Kit was used to estimate the proliferation of cells. After the experiment, the cells were obtained by trypsin digestion. MitoXpress-Xtra system with oxygen sensitive probe was induced to detect the extracellular oxygen consumption level. Results The morphology of C2C12 cells presented a typical long spindle under the microscope following the mechanical stretch stimulation. The cells were arranged in a certain direction, parallel to the direction of tension stimulation and growing in good condition. Compare with the control group, the cell numbers in 1 Hz group and 2 Hz group were significantly increased (P<0.05). At the 3rd and 4th day, number of cells in 1Hz group was significantly increased than that in 2 Hz group. However, the oxygen consumption capacity increased in both 1 Hz group and 2 Hz group, but no significant difference was found between experiment groups (P>0.05). Conclusions Cyclic mechanical stretch stimulation can effectively induce proliferation of C2C12 cells, which is related to the frequency of mechanical stretch, with the frequency of 1 Hz being optimum. But stretch stimulation has no significant impact on the aerobic ability of C2C12 cells.
