Aim To observe the effect of lipopolysac-charide(LPS)induced supernatant of BV-2 cells on the inflammatory response and apoptosis of HT22 neu-rons.Methods After the concentration and time of LPS were determined by CCK-8 method,BV-2 cells were cultured with medium without LPS and medium containing LPS,the morphological changes of BV-2 microglia were observed by inverted microscope, and the CD86/CD206 ratio of BV-2 microglia was detected by immunofluorescence.Subsequently,BV-2 cell cul-ture supernatants were isolated and added to HT22 neuronal culture to observe the effect on the inflamma-tory response of HT22 neurons. The proliferation of HT22 neurons was detected by CCK-8 method and EdU method. The structural changes of HT22 neurons were observed under the microscope and examined by urani-um-lead staining. The levels of cytokines interleukin-1β(IL-1β),interleukin-10(IL-10),nuclear factor kappa-B(NF-κB)and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent as-say(Elisa).Neuronal apoptosis was detected by the TUNEL method. The protein expressions of Bax,Bcl-2 and inflammatory factors were detected by Western blot. Results After induction with 1 mg·L-1 LPS,BV-2 cells exhibited increased cell body size, thicker protrusions on both side, and some cells showed de-formed protrusions, the CD86/CD206 ratio in BV-2 cells decreased, promoting the transformation of BV-2 cells from M2 type to M1 type. After treating with the culture supernatant of BV-2 cells,HT22 neuronal cell activity and proliferation were reduced, axons short-ened,and the number of cells decreased.Neuronal cell bodies were enlarged and some cells were de-formed,with damaged cell membranes, round cell nu-clei but displaced nucleoli from the normal position, swollen mitochondria with vacuoles, reduced internal ridge structures, and increased levels of inflammatory factors NF-κB,IL-1 β,and TNF-α(P<0.05 or P<0.01),while the anti-inflammatory factor IL-10 de-creased(P<0.05),protein expression of the pro-apoptotic indicator Bax increased(P<0.01),and the protein expression of the anti-apoptotic indicator Bcl-2 decreased(P<0.05).Conclusion After induction of BV-2 cell polarization by LPS,the supernatant could inhibit HT22 neuronal cell viability, upregulate inflam-matory factor expression and promote apoptosis.

OBJECTIVE To investigate the clinical distribution and dynamic change of drug resistance of K.pneu-moniae and carbapenem-resistant Klebsiella pneumoniae(CRKP)isolated from a three-A hospital of Suzhou so as to provide scientific bases for prevention and control of hospital-associated infections and reasonable application of antibiotics.METHODS The K.pneumoniae and CRKP strains that were isolated from the submitted specimens were collected from the patients who treated in the Affiliated Suzhou Hospital of Nanjing Medical University from 2019 to 2023.The clinical characteristics of the patients with infection and the trend of drug resistance were statis-tically analyzed.RESULTS Totally 5631 strains of K.pneumoniae were isolated,1205(21.40%)of which were CRKP,and the isolation rate of CRKP showed an upward trend in the five years(x2=236.352,P<0.001).Among the K.pneumoniae isolates,51.59%were isolated from sputum,13.51%from urine;19.43%were isolated from intensive care unit(ICU),7.64%from emergency department, and 7.19%from respiratory department. There were significant differences in gender, age and season between the patients detected with CRKP and the patients detected with non-CRKP(P<0.05).The drug resistance rates of the K.pneumoniae strains to cephalosporins, quinolones and carbapenems con-tinuously increased from 2019 to 2023(P<0.001),the drug resistance rate to imipenem increased from 11.69%to 34.24%,meropenem from 10.92%to 34.24%.CONCLUSIONS The K.pneumoniae isolates show severe drug re-sistance from 2019 to 2023,and the isolation rate of CRKP strains rises increasingly. It is necessary for the hospi-tal to focus on the continuous monitoring of key populations and departments and optimize the management of an-tibiotics and infection control strategies so as to provide guidance for reasonable clinical use of antibiotics, effective control of transmission of drug-resistant strains and cope with the increasingly severe drug resistance.

Alzheimer's disease(AD),a major cause of dementia,is a common neurodegenerative disease, beginning with memory loss and difficulties with thinking, language and problem-solving skills.Intensification of population aging and the increasing incidence of AD have imposed a heavy burden on healthcare systems.Currently, the main pathological explanation remains the excessive accumulation of β-amyloid plaques, formation of neurofibrillary tangles and neuronal loss, but the true etiology and pathogenesis of AD remain unknown. The advances of research in AD,with respect to genetics, pathophysiological mechanism, imaging diagnosis and laboratory diagnosis, are reviewed in this article, aiming to provide references for relevant research and potential clinical applications.

Objective Ticks serve as vectors for transmitting Babesia microti. However, the specific mechanism remains unclear. This study aimed to investigate the effect of tick autophagy molecules on the proliferation of Babesia microti. Methods An experimental model of infected and uninfected mice was used to collect tick materials for proteomic analysis to identify differentially expressed autophagy-related molecules in Haemaphysalis longicornis. The cloning of the HlATG8 gene, protein expression, and production of polyclonal antibodies were completed. The HlATG8 gene was then knocked down using RNAi interference technology. Results The tick autophagy molecule,HlATG8,was identified and found to be significantly upregulated in ticks infected with Babesia microti. The load of Babesia microti in ticks increased significantly following the knockdown of the HlATG8 gene. Conclusions The tick autophagy molecule in Hae.longicornis,HlATG8,inhibits the proliferation of Babesia.
