1.Impacts of cordycepin on isoproterenol-induced cardiomyocyte apoptosis by regulating cAMP/PKA signaling pathway

Objective To investigate the impacts of cordycepin(Cor)on isoproterenol(ISO-induced cardiomyocyte apoptosis and cyclic AMP/protein kinase A(cAMP/PKA)signaling path-way.Methods After H9c2 cardiomyocytes were treated with ISO(10 μmol/L),the cells were further treated with Cor(1,5,10,15 μg/ml),respectively to detect cell proliferation.Then the H9c2 cells were grouped into Control group,ISO group,5 μg/ml Cor group(Cor-L group),10μg/ml Cor group(Cor-H group),and 10 μg/ml Cor+20 μmol/L H-89 group(combined group).Flow cytometry was applied to detect cell apoptotic rate in above groups.Phalloidin staining was used to detect the surface area of the cells.RT-PCR was used to detect the expression of hypertro-phy related genes,such as ANP,BNP,and β-myosin heavy chain(β-MHC)in the cardiomyocytes.ELISA was conducted on test the levels of inflammatory factors,TNF-α,IL-6 and IL-1β and cAMP.Western blot assay was employed to measure the expression levels of B cell lymphoma/leukemia-2(Bcl-2),Bcl-2 associated X protein(Bax),Caspase-3,and PKA.Results Compared with the Control group,the apoptotic rate[(33.89±3.68)%vs(2.57±0.26)%],cell surface area,and the expression levels of TNF-α,IL-6,IL-1β,Bax,Caspase-3,ANP,BNP,and β-MHC were in-creased,and the expression levels of Bcl-2,cAMP,and PKA were decreased in the ISO group(P<0.05).The Cor-L and Cor-H groups had significantly lower apoptotic rate[(20.58±1.93)%and(7.58±0.76)%vs(33.89±3.68)%],smaller cell surface area,and decreased expression levels of TNF-α,IL-6,IL-1β,Bax,Caspase-3,ANP,BNP,and β-MHC,and increased levels of Bcl-2,cAMP,and PKA when compared with the ISO group(P<0.05).The apoptotic rate[(28.69±2.82)%vs(7.58±0.76)%],cell surface area,and expression levels of TNF-α,IL-6,IL-1β,Bax,Caspase-3,ANP,BNP,and β-MHC were elevated,and the expression levels of Bcl-2,cAMP and PKA protein were decreased in the combined group than the Cor-H group(P<0.05).Conclusion Cor inhibits ISO-induced cardiomyocyte apoptosis by activating the cAMP/PKA signaling pathway.

3.Whole blood collection at street fixed blood donation sites in Chengdu

【Objective】 To explore the whole blood collectionstatusat street fixed blood donation sites in different time periods and regions, among different populations with different recruitment methods in Chengdu. 【Methods】 The whole blood donation data at street fixed blood donation sites in Chengdu from 2014 to 2018 were collected, and then statistically analyzed in terms of weekdays/ weekends/holidays, the urban/suburban counties, the first/ repeated blood donation and local/ non-local household registration.On weekdays, blood donors were recruited at fixed blood donation sites while on weekends/holidays they were recruited via on-site recruitment combined with "computer-network-mobile phone" mode. 【Results】 There are 36 fixed blood donation sites in Chengdu, among which 22 were in suburban counties and 14 in urban area.There were more blood drives on weekdays than weekends / holidays every year, with fewer average daily collection on weekdays in comparison with weekends/holidays(P<0.05). The collection units on weekends/holidays were high in January, July, August and December, medium in March, April, June, September and October, and low in February, May and November (P<0.05). There were more blood drives in urban area than suburban counties on weekends/holidays, with fewer average daily collection in urban areain comparison with suburban counties. From 2014 to 2018, the total collection units at street fixed blood donation sites in suburban counties increased year by year (P<0.05). The proportion of first-time blood donors in urban area (59.62%) was significantly higher than that in suburban counties (43.45%) (P<0.05). The proportion of non-local registered blood donors in urban area (53.39%) was significantly higher than that in suburban counties (20.39%)(P<0.05). The number of text messages we sent for recruitment on weekends and holidays and the number of blood donors who responded increased year by year (P<0.05). 【Conclusion】 Whole blood collection at street fixed blood donation sites in Chengdu suburban counties on weekends and holidays was enhanced by using information interactive mode as "computer-network-mobile phone", which allows blood station managers to schedule blood collection in different periods and formulate reasonable blood collection plan.
4.Effects of type D personality on anxiety, depression and quality of life of hospitalized patients with coronary heart disease

Objective To explore the relationship between type D personality,anxiety and depression of coronary heart disease(CHD) and the effects on quality of life (QOL).Methods 296 CHD patients were randomly chosen to answer the questionnaires of general situation,type D personality scale (DS14),Self-rating Anxiety Scale(SAS),Self-rating Depression Scale (SDS) and Short-Form Health Survey (SF-36).Results Of all subjects 37.2% were identified as type D personality,detection rates of anxiety and depression was were higher than the patients with non-type D personality (33.6% vs 14.0%,49.1% vs 23.1%,separately) (P< 0.01).The scores of role-physical (RP),bodily pain (BP),general health (GH),vitality (VT),social functioning (SF),role-emotional(RE),mental health (MH) in patients with type D personality ((32.73 ± 42.52),(57.68 ± 26.93),(42.14 ±18.23),(47.15 ±22.60),(59.43 ±26.67),(45.91 ±46.22),(49.25 ±22.21),separately) were lower than the patients with non-type D personality ((56.00 ± 46.61),(64.62 ± 22.27),(48.27 ± 19.19),(61.56 ±20.17),(69.70 ± 24.35),(70.79 ± 44.53),(70.52 ± 18.91),separately) (P<0.05 or 0.01).Multiple stepwise regression analysis showed that depression,type D personality,anxiety,coronary artery stenosis degree and degree of education were entered QOL regression equation(β =-16.31,-6.53,-7.69,-15.75,2.64,P < 0.05).Conclusion The type D personality of CHD exist high prevalence of anxiety and depression disorders,and the type D personality influence the QOL of patients with CHD.
