1.Pharmacological effects and drug design research progress of fucoxanthin

Fucoxanthin is a pigment found in plants and animals such as algae, marine plankton and aquatic shellfish, and holds significant potential for development in the pharmaceutical field. This review introduces the anti-inflammatory, antioxidant, anticancer, anti-obesity, and other pharmacological effects of fucoxanthin, as well as recent advances in drug design research. It was found that fucoxanthin can exert anti-inflammatory and antioxidant effects through mechanisms such as activating AMP- activated protein kinase related signaling pathways, regulating the expression of inflammatory factors, altering microbial stability, thereby improving conditions such as metabolic associated fatty liver disease and colitis. It can exert selective antitumor effects through multi-target synergistic actions; and it was also found that it can exert anti-obesity effects by regulating the intestinal microbiota. Its characteristic functional groups (such as hydroxyl and epoxy groups) possess target specificity and reversible inhibitory properties, making it a suitable template for guiding the design and development of novel drugs, thereby providing new insights for breaking through the limitations of traditional drug design.
2.Tapping with plum-blossom needle combined with sulfur ointment and local irradiation for primary cutaneous amyloidosis: a case report.

The paper reports one case of primary cutaneous amyloidosis (PCA) treated by tapping with plum-blossom needle combined with sulfur ointment and local irradiation. PCA in this case was manifested as generalized erythema, papules, plaques, lichenification, and severe pruritus. In treatment, tapping with plum-blossom needle was delivered at typical lesions to induce local congestion, redness, and minimal bleeding. After cleaned with sterile gauze for 10 s, 25% sulfur ointment was evenly applied, followed by local irradiation with a TDP lamp for 15 min. This session was repeated twice a week. In 1 month of treatment, the lesions turned flat and the skin was soft as the normal, with pigmentation and mild pruritus left. In 3 months of follow-up, no papules recurred, and mild pruritus presented occasionally.

3.Celecoxib improves right heart function in mice after acute high-altitude hypoxia exposure by increasing 12,13-diHOME level

Objective To investigate the effect and mechanisms of celecoxib on right heart function in mice with acute high-altitude hypoxia exposure.Methods Male C57BL/6J mice(7 weeks old)were housed in a hypobaric chamber simulating an altitude of 5 800 m for 2 d to establish an animal model of acute hypobaric hypoxia.①Eighteen mice were randomly assigned to plain+saline(P+S),high-altitude hypoxia exposure+saline(H+S),and high-altitude hypoxia exposure+celecoxib(H+Cel).Body weight and routine blood indicators were measured,and cardiac ultrasound examination were performed for heart rate(HR),pulmonary artery acceleration time to ejection time ratio(AT/ET),tricuspid annular plane systolic excursion(TAPSE),tricuspid annular systolic velocity(S'),and left ventricular ejection fraction(LVEF)and fractional shortening(FS).Targeted metabolomic profiling was applied to detect the cardiac arachidonic acid(AA)metabolite levels.The contents of 12,13-dihydroxy-9Z-octadecenoic acid(12,13-diHOME)in the heart,liver,brown adipose tissue,and plasma were quantified by ELISA.② Eighteen mice were randomly assigned into plain+saline(P+S),high-altitude hypoxia exposure+saline(H+S)and high-altitude hypoxia exposure+12,13-diHOME(H+di)groups.Body weight,routine blood tests,and echocardiography were performed as above.③ Thirty-two mice were randomly divided into high-altitude hypoxia exposure+saline(H+S),high-altitude hypoxia exposure+celecoxib(H+Cel),high-altitude hypoxia exposure+soluble epoxide hydrolase inhibitor(sEHI)(H+sEHI),and high-altitude hypoxia exposure+sEHI+celecoxib(H+sEHI+Cel)groups.Body weight,routine blood tests,and echocardiography were performed as above.Cardiac and plasma contents of 12,13-diHOME and epoxyeicosatrienoic acids(EETs)were measured by ELISA.Results ① Compared to the P+S group,the H+S group exhibited significantly reduction of cardiac 12,13-diHOME level(P<0.001),increased counts of white blood cells(WBC)and neutrophils(P<0.01)and decreased TAPSE,S'and AT/ET both at resting state and under stress(P<0.01,P<0.001).Compared to the H+S group,the H+Cel group exhibited significantly increase of cardiac 12,13-diHOME level(P<0.05),reduced WBC and lymphocyte counts(P<0.01,P<0.05)and improved TAPSE and S'levels at resting state and under stress(P<0.01,P<0.001).② Compared to the H+S group,the H+di group demonstrated significantly improvement of TAPSE at basal and under stress(P<0.001)and a trend towards improved TAPSE at resting state(P=0.0532),but no obvious differences was observed in WBC and neutrophil counts between the H+di group and the H+S group.③ Compared to the H+Cel group,both the H+sEHI and H+sEHI+Cel groups exhibited significantly reduction of cardiac 12,13-diHOME level(P<0.01,P<0.05)though no statistical changes in cardiac function indicators.Compared to the H+S group,WBC counts and lymphocyte were decreased,and serum EETs level was incrased in the H+Cel group,H+sEHI group and H+sEHI+Cel group(P<0.01,P<0.001).Conclusion Celecoxib can elevate cardiac level of 12,13-diHOME and improves right heart function in mice after acute high-altitude hypoxia exposure through the CYP450-sEH metabolic pathway.
4.Muscone improves coronary microvascular disease by promoting VEGF-mediated microvascular angiogenesis

Objective To explore the impact of Shexiang Baoxin Pill(SXBXP,a traditional Chinese patent medicine,broadly applied for the treatment of cardiovascular diseases)on coronary microvascular disease(CMD)and investigate the role and underlying mechanisms of muscone,a key bioactive component of the pills,in the treatment.Methods A total of 16 ob/ob mice(8 weeks old)were randomly and equally divided into an ob/ob-SXBXP group and a ob/ob-Control group,receiving 10 mg/kg·d SXBXP or PBS via oral gavage,and another 8 wild-type mice with the same genetic background(WT group)were subjected as a negative control group.Cell model of CMD was established based on mouse coronary microvascular endothelial cells(MCMEC)under ischemia and hypoxic(HI)condition,and the cells were further treated with 20 μg/mL SXBXP(HI+SX)and 30 μmol/L muscone(HI+Muscone),respectively.Echocardiography was carried out for coronary flow reserve(CFR)and left ventricular function,and laser speckle imaging was applied to evaluate myocardial blood flow(MBF).Microvascular density in the heart was evaluated with CD31 immunofluorescence staining.The expression levels of vascular endothelial growth factor(VEGF)in cardiac microvascular endothelial cells of both mouse and cell models were detected by immunofluorescence staining and Western blot analysis.The proliferation and angiogenesis of MCMEC were observed by 5-ethynyl-2'-deoxyuridine staining and angiogenesis experiments.Results In the ob/ob-Control group of mice,the levels of CFR,MBF,and cardiac microvascular density were significantly lower than those in the WT group(P<0.05),and these indicators were significantly improved in the ob/ob-SXBXP group when compared with the ob/ob-Control group(P<0.05),which indicates that SXBXP improves the CMD phenotype.The expression level of VEGF in cardiac microvascular endothelial cells was significantly lower in the ob/ob-Control mice than the WT mice(P<0.05),while the level in the ob/ob-SXBXP group was significantly higher than that in the ob/ob-Control group(P<0.05),illustrating that SXBXP may ameliorate CMD through VEGF-mediated microvascular angiogenesis.In vitro experiments further revealed that the VEGF expression level and the proliferation and angiogenesis abilities in MCMEC were significantly lower in the HI group than the cells under the normoxia control condition(P<0.05).Both SXBXP and muscone treatment resulted in enhanced expression of VEGF and improved proliferative and angiogenesis abilities of MCMEC(P<0.05).These results suggest that muscone could improve CMD by VEGF-mediated microvascular angiogenesis.Conclusion SXBXP can improve CMD.Muscone,as a key component of SXBXP,promotes microvascular angiogenesis by inducing the expression of VEGF,then enhances myocardial perfusion,and consequently alleviates CMD.
5.Accuracy evaluation of a universal dental implant guide for simulating implantation in posterior area on dental molds

Objective This study aims to compare the accuracy of self-developed universal implant guide(SDG),3D printed digital guide(DG),and free hand(FH)simulated implantation in the posterior tooth area of dental models.Methods Ten junior dentists were selected to place three implants in the 35,37,and 46 tooth sites of the mandibular models(35,36,37,and 46 missing teeth)by using SDG,DG,and FH,and the process was repeated again to take the av-erage value.Cone beam computed tomography(CBCT)was used to evaluate the global coronal deviation,global apical deviation,depth deviation,and angular deviation between the actual position and preoperative planned position.Re-sults The coronal deviation and apical deviation of the three implant sites in the SDG group were not significantly dif-ferent from those in the two other groups(P>0.05).The depth deviation and angular deviation in the SDG group were smaller than those in the DG group(P<0.05)and FH group(P<0.05),respectively.All deviations at site 37 in the SDG group were not different from those at site 35(P>0.05),while the depth and angular deviation at site 37 in the DG group were higher than those at site 35(P<0.05).Conclusion The precision of the self-developed universal dental im-plant guide can meet the requirements of clinical posteri-or implantation.
6.Genomic characterization and cluster analysis of Carbapenem-resistant Klebsiella pneumoniae

To investigate the genomic features and perform cluster analysis of Carbapenem-resistant Klebsiella pneumoniae (CRKP) to provide an experimental basis for guiding the prevention and treatment of CRKP infections.A retrospective case-cohort study was conducted on 19 non-redundant CRKP strains isolated from the Tenth Affiliated Hospital of Southern Medical University between January and June 2023. Whole genome sequencing (WGS) and multilocus sequence typing (MLST) were performed to compare genomic features and analyze the resistance genes and homology of the strains.The results showed that the 19 CRKP strains were isolated from 8 different clinical departments, mainly from respiratory specimens. The whole genome sequencing revealed that the genomic lengths of CRKP ranged from 4.90 to 5.85 Mbp, with contigs N50 values>20 kb for each genome. The median overall GC content was 57.0% (50.4%-57.1%). Comparative genomic analysis identified three regions with high genomic variability. WGS detected 32 resistance genes across 11 categories. All 19 strains carried carbapenem resistance genes ( blaKPC-2 and blaOXA-48), blaTEM-1B extended-spectrum β-lactamase resistance genes, qnrS1 quinolone resistance gene, and fosA fosfomycin resistance gene, with each strain carrying only one carbapenemase gene. The detection rate of blaKPC-2 was 94.7% (18/19). MLST identified three sequence types: ST11, ST437 and ST147, with ST11 being predominant (89.5%, 17/19). Clustering analysis based on acquired resistance genes revealed three clonal transmission patterns among strains 72 and 90, and strains 88, 84, 66 and 79.In conclusion, CRKP strains carry multiple resistance genes, and clustering analysis indicating that nosocomial clonal transmission is closely related to acquired resistance genes. The ST11- blaKPC-2 type strain is the predominant clone. Strengthened surveillance and effective control strategies are necessary to reduce nosocomial transmission of CRKP.

8.Expression of serum macrophage migration inhibitory factor and Tau protein in patients with severe hypertensive intracerebral hemorrhage and their relationships with clinical outcome

Objective To explore the expression levels of serum macrophage migration inhibitory factor(MIF) and Tau protein in patients with severe hypertensive intracerebral hemorrhage(HICH),and analyze the relationships between their levels and patient outcome,and to provide a reference for clinical treatment of such patients. Methods We included 120 patients with severe HICH and 50 age-and sex-matched healthy controls from health examination in Ziyang Hospital of West China Hospital of Sichuan University from January 2019 to November 2022.The MIF and Tau protein levels of the two groups were compared. After a follow-up of more than half a year,the patients were divided into poor-prognosis group and good-prognosis group according to the defined criteria to compare their levels of MIF and Tau protein. The receiver operating characteristic(ROC) curve and Kaplan-Meier method were used to explore the critical values of MIF and Tau protein levels for patients with poor prognosis and the relationships with the clinical outcome of severe HICH. Results The levels of serum MIF and Tau protein in the patients with severe HICH were significantly higher than those in the control group(P<0.05). The poor-prognosis group showed significantly higher levels of serum MIF and Tau protein than the good-prognosis group(P<0.05). The ROC curve showed that the cutoff points of serum MIF and Tau protein levels for predicting poor prognosis were 64.43 ng/L and 216.25 pg/ml,respectively. The area under the curve for using MIF to predict poor prognosis was 0.815(95%CI=0.759-0.849),with sensitivity of 81.56% and specificity of 83.24%,and those values for using Tau protein to predict poor prognosis were 0.847(95%CI=0.764-0.831),82.26%,and 79.68%,respectively,all at high levels. Patients with serum MIF level <64.43 ng/L had significantly better survival than those with serum MIF level ≥ 64.43 ng/L(P<0.05 by log-rank test). Patients with serum Tau protein level<216.25 pg/ml had significantly better survival than those with serum Tau protein level ≥216.25 pg/ml(P<0.05 by log-rank test). Conclusion Patients with severe HICH had significantly higher serum MIF and Tau protein levels than healthy people. The patients with poor prognosis had significantly higher serum MIF and Tau protein levels than those with good prognosis. Serum MIF and Tau protein levels were closely related to the clinical outcome of the patients,which can be used as effective indicators to predict the prognosis of patients with HICH.
9.Optimal recognition algorithm for solid nuclear track images based on morphology and machine learning

Objective To propose a computer recognition algorithm for solid nuclear track images based on the machine learning method, and to realize the automatic, fast and accurate recognition of nuclear tracks and improve the efficiency of solid track image analysis. Methods Firstly, 143 images containing tracks were scanned by morphological method to determine the location of suspected tracks, and 1250 material images were captured. 50% of the material were selected as the training set and 30% as the validation set for training the machine learning model. Another 20% of the material were selected as the test set for testing the model recognition result. The algorithm code was written and trained based on the MATLAB software. Results The established solid track recognition algorithm had a strong recognition capability, and the recognition accuracy of the test set could reach 84.8%. The machine learning model program constructed by the algorithm could evolve continuously with the input of training data, further improving the accuracy. Conclusion Based on image morphology and machine learning, the track recognition algorithm was investigated, by which the automatic recognition of solid tracks was better realized. In the future, we will increase the data input of the model, optimize the algorithm, and improve the recognition accuracy, in order to provide a more accurate and efficient method for automatic image track recognition.
10.Logistic regression model-based analysis of influencing factors of initial testing qualified rate of condition monitoring of digital radiography devices in China

Objective:To analyze the influencing factors on initial testing qualified rate of condition monitoring of digital radiography (DR) devices in China and provide the scientific basis for key points of national medical radiation protection monitoring work.Methods:The data of quality control detection of DR devices, reported by 31 provinces in 2020, were collected from National Radiation Health Information Platform-National Medical Radiation Protection Monitoring System. Logistic regression was used to analyze the related influencing factors of initial testing qualified rate of condition monitoring of DR devices.Results:The result of logistic regression analysis showed that the nature of radiological health technical service institutions, the level of medical and health institution and the regions where they are located in China were the independent influencing factors of initial testing qualified rate of condition monitoring of DR devices ( χ2=15.41-21.18, P < 0.05). Private institution group ( OR=1.714, 95% CI: 1.310-2.243), tertiary hospital group ( OR=1.710, 95% CI: 1.310-2.232) and secondary hospital group ( OR=1.540, 95% CI: 1.203-1.971) had a higher qualified probability of DR devices, while western region group ( OR=0.670, 95% CI: 0.526-0.852) had a lower qualified probability of DR devices. Conclusions:Supervision on quality control of DR devices should be strengthened, such as strengthening the random inspection and double-check to the devices. And great attention should be paid to the quality control testing of DR devices in the western China and in the primary and ungraded hospitals.
