1.Study on Ecological Suitability Regionalization of Scutellaria barbata D.Don Based on MaxEnt Model and ArcGIS

Objective To predict the potential suitable habitats of Scutellaria barbata D.Don across China;To provide a theoretical basis for its artificial cultivation and standardized planting practices.Methods Distribution data were collected from 186 sample sites,combined with data from 90 environmental factors,and the MaxEnt model and ArcGIS 10.8.1 software were used to screen the main environmental factors affecting the distribution of Scutellaria barbata D.Don and to analyse and predict the distribution of Scutellaria barbata D.Don suitability zones in China.Results The dominant environmental factors affecting the distribution of Scutellaria barbata D.Don were the driest month precipitation,vegetation subclasses,minimum temperature in September,precipitation in October,and soil base saturation.The high-suitability zones for Scutellaria barbata D.Don are primarily concentrated in the northern Guangxi Zhuang Autonomous Region and its bordering areas with Hunan Province,the central of Guangdong Province,and the border regions between Jiangxi Province and Fujian Province.Conclusion The predicted results of this study can provide a reference for the selection of sites for artificial cultivation and standardized planting of Scutellaria barbata D.Don in southeast China and eastern southwest China(e.g.Zhejiang Province,Jiangxi Province,Sichuan Province,etc.),and provide a reference for the sustainable utilization of Scutellaria barbata D.Don medicinal resources.
2.Genetic analysis of a pedigree with hereditary coagulation factor Ⅺ deficiency caused by a homozygous p. Thr299Ser variant of F11 gene

Objective:To explore the phenotypic and genotypic characteristics of a Chinese pedigree affected with Hereditary coagulation factor Ⅺ (FⅪ) deficiency.Methods:A female patient with FⅪ deficiency and her family members (five individuals from three generations) who presented at Quanzhou First Hospital Affiliated to Fujian Medical University on September 19, 2024 due to diarrhea and fever were selected as study subjects. A retrospective study was conducted to collect the patients′ clinical data. Peripheral venous blood samples were collected from the patient and her family members. Genomic DNA was extracted, followed by sequencing of all exons and flanking sequences of the F11 gene. Candidate variants were validated by Sanger sequencing of the family members, and their pathogenicity was classified according to the guidelines of the American College of Medical Genetics and Genomics (ACMG). This study was approved by the Medical Ethics Committee of Quanzhou First Hospital [Approval No.: Quanyi Lun (2024) K281]. Results:The patient exhibited significantly prolonged activated partial thromboplastin time (APTT) of 80.9 seconds, while FⅪ activity (FⅪ: C) and FⅪ antigen (FⅪ: Ag) levels were extremely low (2% and 3%, respectively). Genetic analysis revealed that the proband harbored homozygous c. 896C>G (p.Thr299Ser) missense variant in exon 9 of the F11 gene, for which her son was heterozygous. The variant was located in a highly conserved domain. Although Mutation Taster predicted it as a polymorphism, SIFT, PolyPhen-2, and LRT analyses suggested it to be likely pathogenic. Protein modeling indicated that the p. Thr299Ser variant may alter the hydrogen bonds between amino acids, thereby affecting the structure and function of the FⅪ protein. According to the ACMG guidelines, c. 896C>G was rated as a likely pathogenic variant (PM1+ PM2_Supporting+ PP1_Strong+ PP3+ PP4). Conclusion:The c. 896C>G (p.Thr299Ser) missense variant of the F11 gene probably underlay the FⅪ deficiency in this pedigree. Above finding has enriched the mutational spectrum of the F11 gene and provided a basis for genetic counseling and prenatal diagnosis for this family.
3.Efficiency of targeted next-generation sequencing in diagnosis of pathogens causing acute respiratory tract infections:a meta-analysis

OBJECTIVE T o explore the value of targeted next-generation sequencing(t-NGS)in diagnosis of respir-atory tract pathogens through meta-analysis so as to provide reference for clinical application.METHODS PubMed database,Web of Science database,Wanfang database,CNKI database and Sinomed database were retrieved,and the time period of retrieval ranged from Jan.2010 to May 2024.The literatures were screened out based on the es-tablished standards.The quality was assessed by QU ADAS-2,the risk of bias graph was drawn by Revman 5.4,and the statistical analysis was performed by Stata 16.0.RESULTS A total of 9 literatures were included in the study.The result of meta-analysis showed that the heterogeneity test Q for sensitivity was 268.21,P<0.01,I2=97.02%,with the heterogeneity test Q for specificity 210.04,P<0.01,I2=96.19%,the combined sensitivity 0.88(95%CI:0.62 to 0.97),combined specificity 0.68(95%CI:0.41 to 0.86),combined positive likelihood ratio 2.72(95%CI:1.44 to 5.15),combined negative likelihood ratio 0.18(95%CI:0.06 to 0.53),combined diagno-sis score 2.74(95%CI:1.68 to 3.80),and combined diagnostic odds ratio 15.44(95%CI:5.34 to 44.66).The area under synthesize receiver operating characteristic(SROC)curve(AUC)was 0.85(95%CI:0.82 to 0.88).The result of Deeks funnel plot showed that P was 0.99,indicating that there was no obvious publication bias.CONCLUSIONS The sensitivity of tNGS is high in detection of the pathogens causing acute respiratory tract infection,the specificity needs to be improved,but its comprehensive ability is satisfactory.It has certain val-ue in early clinical diagnosis.
4.The mechanism of CD151 regulating vascular permeability through vesicle internalization and recycling

Objective :
To explore the effect and mechanism of CD151 on vascular permeability by regulating vesicle internalization and recycling.
Methods:
Wild-type mice and CD151 knockout mice were divided into WT-con group, WT-model group, KO-con group and KO-model group, with 6 mice in each group. WT-model group and KO-model group were intraperitoneally injected with LPS to prepare sepsis ALI model, and WT-con group and KO-con group were intraperitoneally injected with phosphate buffer saline(PBS) as a control. 24 h after modeling, pulmonary vascular permeability was measured by Miles test. The siRNA silencing CD151 expression(si-CD151) and negative control si-NC were transfected into EA.hy 926 cells. The permeability of endothelial cell layer to FITC-dextran at different time points was observed under basic conditions and vascular endothelial growth factor-A(VEGF-A) stimulation conditions. Transcriptome sequencing of endothelial cells in si-CD151 group and si-NC group; the distribution and internalization of CD151 in each group were measured using immunofluorescence. Western blot and real-time quantitative RT-qPCR were used to detect the expression of VE-cadherin in si-CD151 groupand other groups. The distribution and internalization of VE-cadherin in each group were measured using immunofluorescence.
Results :
Miles experiment results indicated that dye exudation in lung tissue of WT-model group was significantly higher than that of WT-con group(P<0.01). The dye exudation in the lung tissue of KO-model group increased compared with WT-model group(P<0.05). The results of endothelial cell layer permeability test showed that the permeability of FITC-dextran in si-CD151 group was significantly higher than that in control group after VEGF-A stimulation for 30, 60 and 120 min(P<0.05). Transcriptome sequencing results suggested that CD151 in endothelial cells was closely related to vesicle-mediated transport. Compared with other groups, protein and mRNA levels of VE-cadherin in CD151 knockdown endothelial cells was significantly lower(allP<0.01). The immunofluorescence assay demonstrated that after VEGF-A stimulation, the decrease of CD151 expression significantly impaired the expression of VE-cadherin at cell-cell contacts and reduced the CD151-VE-cadherin colocalization in the perinuclear region compared with other groups.
Conclusion
The absence of CD151 affects the internalization and recycling of endothelial cell vesicles, affects the expression and internalization of VE-cadherin, and then influences vascular permeability.
5.Study on rapid identification of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants with Fourier exchange infrared spectroscopy

Objective:To establish a Fourier transform infrared spectroscopy (FT-IR) method for rapid identification of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants.Methods:The Fourier transform infrared spectra of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants were established, and the second derivative spectral analysis, clustering analysis, principal component analysis, opls-da and cluster independent soft mode classification model were analyzed to explore the difference characteristic peaks of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants.Results:The first order infrared spectrum showed that the peak shape and peak intensity of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants were different. Clustering analysis, principal component analysis and OPLS-DA results showed that Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants showed good clustering characteristics. SIMCA method was used to construct the model, and the accuracy of the training set and the verification set were 100%, which further verified the feasibility of this method in identifying the authenticity of Cremastrae Pseudobulbus Pleiones Pseudobulbus.Conclusions:The second-order infrared spectroscopy can accurately distinguish the differences between Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants. The method is fast and accurate, and can be used for the authenticity identification of Cremastrae Pseudobulbus Pleiones Pseudobulbus.

Brain computer interface is faced with the problems of legal attribute,legal risk and legal liability.In terms of legal attributes,the user has the subject status,and the legal attributes of brain-computer interface should be judged according to integration degree between brain-computer interface and human body.In terms of legal risks,personal risks,confidentiality risks and exceptions to confidentiality must be analyzed.In terms of legal liabilities,whether the brain-computer interface equipment is faulty will affect the identification of civil liabilities.Legal interest is determined according to the legal attribute of brain-computer interface.When the brain-computer interface does not feedback the user's true will,the corresponding action does not constitute an act in criminal law.It is necessary for China to formulate"Human brain Computer Interface Management Regulations"to es-tablish the legislative purpose and basis,concept and classification,legal status,basic principles,informed consent rules,confi-dentiality rules and exception rules,and legal liability.
