Objective To explore and evaluate the association between mitotic spindle organizing protein 2A(MZT2A)and survival prognosis and immunotherapy response of skin cutaneous melanoma(SKCM),based on multiomics data mining.Methods The expression difference and survival prognosis of MZT2A in SKCM were analyzed by the Cancer Genome Atlas database,Genotype-tissue Expression Data,Human Protein Atlas,SUMMER database.The biological function of MZT2A and related enriched pathways were analyzed by David2.The differences of immune infiltrating cells,immune therapy response and immune escape ability between high-and low-MZT2A groups were analyzed and compared by using Tumor Immune Estimation Resource,Tumor Immune Single-cell Hub,Tumor-immune System Interactions database,the Cancer Imaging Archive and Tumor Immune Dysfunction and Exclusion databases.Results The expression of MZT2A gene elevated in a variety of cancers,especially in SKCM.SKCM samples with high expression of MZT2A had a poor overall survival.The expression of MZT2A was negatively correlated with the infiltration of CD8+T cells,CD4+T cells,macrophages,neutrophils and dendritic cells in SKCM tissues.In addition,the higher the expression of MZT2A,the therapeutic response of SKCM were worse to immune checkpoint inhibitors(ICI).Conclusion MZT2A is a novel survival prediction target of SKCM.SKCM patients with high expression of MZT2A have poor response to ICI treatment,which provides a theoretical basis for predicting the survival prognosis and guiding immune targeted therapy of SKCM.

T cell response plays an important role in anti-viral infection and anti-tumor immunity,and antigen-specific T cell detection is essential for study of T cell response.This article reviews progress of antigen-specific T cell detection technology,including enzyme-linked immunospot(ELISPOT)assay,intracellular cytokine staining(ICS)assay and activation-induced labeling(AIM)assay,which based on detection of cytokine secretion or activation phenotypes of specific T cells after stimulation and reactivation with antigen in vitro.Another class of methods include Tetramer technology based on known epitopes-human leukocyte antigen(HLA)restriction and recently developed single-cell transcriptomes and T-cell antigen receptor(TCR)sequencing technology.Application of the above methods has advanced our understanding of antigen specific T cell response:Strength and duration of the response,subpop-ulation information,epitopes and their associated HLA-restriction,TCR cloning information and transcriptome characteristic.

Objective:To investigate the anti-inflammatory activity of alcohol extract of Polyrhachis dives(PDAEs)against rheumatoid arthritis(RA)in vitro and in vivo.Methods:In vivo and in vitro anti-RA inflammatory response of PDAEs was investigated using a rat model of collagenous arthritis induced by bovine type Ⅱ collagen and an LPS-induced RAW264.7 cell inflammatory model.Results:PDAEs inhibited the polarization of M1 type macrophages in vivo and in vitro,reduced expressions of TNF-α,IL-1β,IL-6,iNOS,and promoted polarization of M2 type macrophages,enhanced expressions of anti-inflammatory cytokines,such as IL-10 and TGF-β,so as to achieve the anti-inflammatory effect.The experiments in vivo also showed that PDAEs had the immunomodulatory effect,the potential mechanism may be the regulation of Th17/Treg balance by regulating the expression of PD-1 and TGF-β,thus correcting the over-strong autoimmune response.Conclusion:PDAEs may reduce the inflammatory reaction of RA through anti-inflam-matory and immunomodulatory effects.

Combination of passive targeting with active targeting is a promising approach to improve the therapeutic efficacy of nanotherapy. However, most reported polymeric systems have sizes above 100 nm, which limits effective extravasation into tumors that are poorly vascularized and have dense stroma. This will, in turn, limit the overall effectiveness of the subsequent uptake by tumor cells via active targeting. In this study, we combined the passive targeting via ultra-small-sized gemcitabine (GEM)-based nanoparticles (NPs) with the active targeting provided by folic acid (FA) conjugation for enhanced dual targeted delivery to tumor cells and tumor-associated macrophages (TAMs). We developed an FA-modified prodrug carrier based on GEM (PGEM) to load doxorubicin (DOX), for co-delivery of GEM and DOX to tumors. The co-delivery system showed small particle size of ∼10 nm in diameter. The ligand-free and FA-targeted micelles showed comparable drug loading efficiency and a sustained DOX release profile. The FA-conjugated micelles effectively increased DOX uptake in cultured KB cancer cells that express a high level of folate receptor (FR), but no obvious increase was observed in 4T1.2 breast cancer cells that have a low-level expression of FR. Interestingly, in vivo, systemic delivery of FA-PGEM/DOX led to enhanced accumulation of the NPs in tumor and drastic reduction of tumor growth in a murine 4T1.2 breast cancer model. Mechanistic study showed that 4T1.2 tumor grown in mice expressed a significantly higher level of FOLR2, which was selectively expressed on TAMs. Thus, targeting of TAM may also contribute to the improved in vivo targeted delivery and therapeutic efficacy.

ObjectiveTo establish a rat model of pure sensory nerve injury or pure motor nerve injury combined with skin defect to examine the effect of highly selective nerve injury on skin wound healing.MethodsA total of 90 male Sprague-Dawley rats were randomly and equally divided into three different groups including Group A (posterior rhizotomy + cutaneous excision wounding group),Group B (anterior rhizotomy + cutaneous excision wounding group) and Group C ( sham operation + cutaneous excision wounding group).The wound healing rate was detected at days 2,7,14 and 21 after injury.The CGRP mRNA expression was determined by reverse transcription-polymerase chain reaction (RT-PCR) at days 1,3,7 and 14 after injury.The Bcl-2 protein expression was determined by immunohistochemistry at days 3,7,14 and 21 after injury.ResultsThe wound healing rate in the Groups A and C was significantly higher than that in the Group B at day 2 ( P ＜ 0.05 ),with no statistical difference between Group A and Group C (P ＞ 0.05 ).The wound healing rate in the Group C was higher than that in the Groups A and B at days 7 and 14 (P ＜0.05),with no statistical difference between Group A and Group B (P ＞0.05).The wound healing rate in the Groups B and C was higher than that in the Group A at day 21 ( P ＜ O.05 ),with no statistical difference between Group B and Group C ( P ＞ 0.05 ).The CGRP mRNA was expressed in three groups and the expression was up-regulated after wound.The CGRP mRNA expression in the Groups B and C was significantly higher than that in the Group A at days 1,3 and 7,with no statistical difference between Group B and Group C ( P ＞ 0.05 ).The CGRP mRNA expression was distributed as Group C ＞ Group B ＞ Group A at day 7 ( P ＜ 0.05 ) and Group B ＞ Group A ＞ Group C at day 14 (P ＜ 0.05 ).Immunohistochemistry indicated that there was no statistical difference in Bcl-2 protein expression at day 3 after injury in all groups; that the Bcl-2 protien expression in the Group C was significantly higher than that in the Groups A and B at day 7 ( P ＜ 0.05 ),with no statistical difference between Group A and Group B ( P ＞ 0.05 ) ; that the Bcl-2 protien expression was distributed as Group C ＞ Group B ＞ Group A at day 14 ; that the Bclo2 protien expression in the Group B was higher than that in the Groups A and C at day 21 (P ＜0.05),with no statistical difference between Group A and Group C (P ＞ 0.05).ConclusionsIn wound healing process,the sensory nerve plays a more important role than the motor nerve.Denervation of the nerves ( especially the sensory nerves) is not conducive to wound healing,and complete innervation is necessary for normal wound healing.

Objective: Evaluating ovarian carcinoma diagnosis,staging and resectability with EBCT. Methods: 25 cases of ovarian carcinoma were performed in total abdominal including pelvic with EBCT one week before operation.CT findings were compared and analysed with surgical and pathological results. Results: Of 25 patients,11 were serous cyst adenocarcinoma,six mucinous cyst adenocarcinoma,three malignant teratoma,two metastases,and each have only one for clear cell carcinoma,embryo carcinoma and undifferentiated carcinoma.Clinical staging of carcinoma were seven in stageⅠ,seven in stage Ⅱ,nine in stage Ⅲ,two in stage Ⅳ.Invaded vessels and nearby organs around ovarian carcinoma,and the status of peritoneum metastases,were main factors that influenced the difficulties of operation.Compared with conventional CT,EBCT has the virtues of ultrafast scanning,excellent quality of image,and displaying focus detail fine.Conclusion: EBCT scanning is very valuable in diagnosis,tumor staging and evaluating resectability of ovarian carcinoma.
