1.Clinicopathological characteristics of SEMA5A and PD-L1 expression and correla-tion of the two genes in gastric cancer

Purpose To investigate the expression of semaphorin 5 A(SEMA5A)and programmed death ligand-1(PD-L1)and their clinicopathological significance in gastric cancer.Methods Clinical data of 41 cases of gastric cancer tissues and paired adjacent tissues were collected.Immunohistochemical staining and RT-qPCR were used to de-tect the expression levels of SEMA5A and PD-L1,and analysed the correlation between SEMA5A and PD-L1 and clini-copathological features.In addition,we used lentivirus to construct SEMA5A stable low-expression cell lines.RT-qPCR and Western blot were used to analyse the correlation between the expression of SEMA5A and PD-L1 in gastric cancer tissues.Results The high expression rate of SEMA5A was 65.9%(27/41)in gastric cancer tissues and 39.0%(16/41)in paracancerous tissues,respectively.The positive rates of PD-L1 were 58.5%(24/41)and 14.6%(6/41),respectively.RT-qPCR showed that the relative expression levels of SEMA5A mRNA in gastric cancer and paracancerous tissues were 1.30±0.50 and 0.81±0.48,respectively,while the relative expression levels of PD-L1 mRNA were 0.70±0.42 and 0.12±0.09,respectively.SEMA5A expression was correlated with histological typ-ing of gastric cancer and lymph node metastasis(P<0.05).PD-L1 expression was correlated with tumour size,T stage,and pathological stage of gastric cancer(P<0.05).Pearson correlation analysis showed a positive correlation between the expression of SEMA5A and PD-L1 mRNA in gastric cancer tissues,and spearman correlation analysis showed that there was no correlation between the expression of the two in paracancerous tissues.Knockdown of SE-MA5A gene in human gastric adenocarcinoma cell lines resulted in down-regulation of PD-L1 expression.Conclusion Both SEMA5A and PD-L1 are highly expressed in gastric cancer tissues,and there is a correlation between the ex-pressions of SEMA5A and PD-L1.They can serve as potential molecular markers for prognostic evaluation and combi-nation therapy of gastric cancer.
2.Clinical characteristics and risk factors for surgical site infections of type Ⅰ incision in patients undergoing grade Ⅲ and Ⅳ surgeries

OBJECTIVE To explore the clinical characteristics and risk factors for surgical site infections(SSI)in the patients undergoing type Ⅰ incision surgeries of grade Ⅲ and Ⅳ so as to provide theoretical bases for effective prevention and control of SSI.METHODS The clinical data were prospectively collected from 280 patients with SSI who received major type Ⅰ incision surgeries of grade Ⅲ and Ⅳ in the key departments of the Second Affiliated Hospital of Jiaxing University from Jan.2021 to Jun.2024,and randomly from another 280 patients without infec-tions.The enrolled patients were assigned as the infection group and the no infection group,respectively.The clin-ical characteristics of the SSI were analyzed.The baseline data,including sex,age,hypertension,diabetes melli-tus,hyperlipidemia,heart disease,hepatorenal function,nutrition status,preoperative length of hospital stay,operation duration,implantation materials,emergency surgery,intraoperative warm maintenance,microscope,blood loss volume,blood transfusion and postoperative intensive care unit(ICU)stay as well as severe hypopro-teinemia,were collected from the enrolled patients.Univariate analysis and logistic regression analysis were per-formed for the influencing factors for the SSI.RESULTS A total of 16,706 patients underwent neurosurgery,or-thopedics and cardiothoracic surgery,280 of whom had SSI,the incidence of SSI was 1.68%.Totally 264 strains of bacteria were isolated,53.03%of which were gram-positive bacteria,46.97%were gram-negative bacteria;Staphylococcus aureus was the most common species of pathogenic bacteria.The result of multivariate logistic a-nalysis showed that the nutrition status,operation duration,microscope,postoperative ICU and severe hypopro-teinemia were the risk factors for the SSI(P<0.05).CONCLUSIONS The SSI is common among the patients un-dergoing type Ⅰ incision surgeries of grade Ⅲ and Ⅳ.It is necessary for the hospital to take targeted interven-tion measures in early stage based on the risk factors.

5.Study on the Quality Standards of Miao Medicine Oxalis corniculata

OBJECTIVE: To provide scientific basis for the utilization and development of Miao medicine Oxalis corniculata by promoting the quality standard of it. METHODS: Total of 12 batches of O. corniculata were collected from Guizhou, Anhui and Henan, etc. Microscopic characteristics of 12 batches of O. corniculata powder were observed. According to the corresponding methods in 2015 edition of Chinese Pharmacopoeia (part Ⅳ), TLC was used for qualitative identification [developing solvent was trichloromethane-methanol-formic acid (8 ∶ 1 ∶ 0.1, V/V/V)], and the contents of moisture, total ash, acid insoluble ash and alcohol soluble extractive from 12 batches of O. corniculata were determined. The content of isovitexin was determined by HPLC. The determination was performed on Venusil XBP C18 (L) with mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (15 ∶ 85, V/V) at the flow rate of 1 mL/min. The column temperature was 35 ℃, and the detection wavelength was set at 338 nm. The sample size was 10 μL. RESULTS: Microscopic observation showed that the powder was grayish brown to yellowish brown, with many non-glandular hairs and obvious fibrous pore. Results of TLC identification showed that the spots of the same color appeared in the corresponding positions of the test and the control chromatogram. The contents of moisture, total ash, acid insoluble ash and alcohol soluble extract from samples were 6.66%-12.13%, 9.16%-13.79%, 1.58%-4.63% and 5.22%-15.79%, respectively. Results of HPLC method showed that the concentration of isovitexin showed a good linear relationship in the range of 5.20-78.3 μg/mL (r=0.999 0); RSDs of reproducibility (n=9), intermediate precision (n=6) and stability (24 h, n=6) tests were all lower than 2.0%; and the recovery rates were 97.54%-99.52% (RSD=0.74%, n=6); the contents of isovitexin in 12 batches of O. corniculata were 0.036%-0.144% (n=3). CONCLUSIONS: Qualitative and quantitative identification methods of O. corniculate were established, which can be used as a reference for improving the quality standard of O. corniculata.
