1.The significance and advancements of circulating tumor DNA in the diagnosis and treatment of operable non-small cell lung cancer

Non-small cell lung cancer, characterized by high incidence and mortality rates, significantly threatens human health. Precisely assessing patient prognosis and implementing adaptive treatment strategies have emerged as pivotal issues in contemporary thoracic oncology. Postoperative minimal residual disease (MRD) detection through liquid biopsy has demonstrated substantial potential. Circulating tumor DNA (ctDNA) enables real-time dynamic monitoring of tumor alterations. With advancements in ctDNA detection technologies, ctDNA has become a core method for MRD assessment. In the context of postoperative surveillance, ctDNA detection facilitates more accurate prognostic stratification and early prediction of tumor recurrence. Regarding therapeutic interventions, ctDNA detection can predict the efficacy of neoadjuvant and adjuvant therapies. In the future, further elucidating the value of assessing ctDNA status in predicting patient prognosis and guiding drug therapies will contribute to the advancement of precision medicine and adaptive treatments.
2.Analysis of the success rate of CT-Guided 3D printed template-navigated radioactive seed implantation in the treatment of pancreatic cancer

Pancreatic cancer is highly malignant and difficult to treat. The implantation of radioactive seed has opened up a new treatment option for pancreatic cancer. Although the implantation of radioactive seed in the treatment of pancreatic cancer has achieved good results, due to the special anatomical location of pancreatic cancer and the dense distribution of important tissues and organs around the tumor, the operation of seed implantation for pancreatic cancer has become a clinical treatment challenge. We applied the method of 3D printing template navigation and constraining the direction of the puncture needle. The puncture needle followed the safe puncture path designed in the preoperative treatment planning system, effectively avoiding important tissues and organs, and implanted radioactive seeds in the tumor. The operation of seed implantation was simplified, homogenized and made safe. In this group of cases, the proportion of successful particle implantation using 3D printing template navigation was 89.5%. All cases met the preoperative treatment plan in postoperative dose verification and achieved the purpose of radioactive seed implantation treatment.

5.Values of combined detection of polygene methylation in stool for the diagnosis of colorectal cancer and precancerous lesions

Objective:To investigate the methylation status of SDC2, PPP2R5C and ADHFE1 genes in stool and their values in the screening of colorectal cancer and precancerous lesions.Methods:From August 2020 to March 2021, 64 patients with colorectal cancer, 72 patients with adenoma, 33 patients with hyperplastic polyps and 59 healthy people were recruited from Qingdao Central Hospital Affiliated to Qingdao University, and the morning stool samples were collected from the research subjects. The genomic DNA was extracted and modified with sulfite. The methylation status of SDC2, PPP2R5C and ADHFE1 genes were detected by methylation specific polymerase chain reaction (MSP), and the fecal occult blood test (FOBT) was performed. Taking the pathological results as the gold standard, receiver operating characteristic (ROC) curve and area under the curve (AUC) were used to compare the effect of combined detection of methylation of three genes and FOBT in predicting colorectal cancer and precancerous lesions. R-Studio software was used to construct a nomogram for the prediction of colorectal cancer with combined detection of gene methylation in stool and other clinical features, and the calibration and validation were performed.Results:The positive rates of combined detection of methylation of SDC2, PPP2R5C and ADHFE1 genes in stool were higher than those of FOBT in colorectal cancer+adenoma [74.3% (101/136) vs. 47.1% (64/136), χ2 = 23.20, P = 0.001], colorectal cancer [90.6% (58/64) vs. 70.3% (45/64), χ2 = 8.91, P = 0.003] and adenoma [59.7% (43/72) vs. 26.4% (19/72), χ2 = 14.43, P = 0.002]. There was no significant difference in the positive rates in hyperplastic polyps [21.2% (7/33) vs. 6.1% (2/33), χ2 = 0.12, P = 0.125] and healthy controls [10.2% (6/59) vs. 8.5% (5/59), χ2 = 4.01, P = 1.000]. The combined detection of gene methylation was better than FOBT in the prediction of colorectal cancer + adenoma [AUC: 0.85 (95% CI 0.80-0.91) vs. 0.71 (95% CI 0.64-0.78), P < 0.05], especially in the prediction of adenoma [AUC: 0.82 (95% CI 0.74-0.89) vs 0.64 (95% CI 0.57-0.69), P < 0.001]. The sensitivity and specificity of ADHFE1 gene methylation status in predicting colorectal cancer were high (90.6% and 96.6%). In colorectal cancer patients over 50 years old, the positive rate of combined detection of gene methylation was higher than that of FOBT [90.2% (55/61) vs. 68.9% (42/61), P < 0.05]. The nomogram calibration curve for predicting colorectal cancer constructed based on the combined detection of gene methylation and each clinical feature showed a high degree of concordance between the predicted and observed diagnostic performance of colorectal cancer. Conclusions:The methylation levels of SDC2, PPP2R5C AND ADHFE1 genes in stool are increased in patients with colorectal cancer or adenoma. The combined detection of gene methylation is expected to be a non-invasive method for the screening of colorectal cancer and precancerous lesions.
6.Study on the Quality Standards of Miao Medicine Oxalis corniculata

OBJECTIVE: To provide scientific basis for the utilization and development of Miao medicine Oxalis corniculata by promoting the quality standard of it. METHODS: Total of 12 batches of O. corniculata were collected from Guizhou, Anhui and Henan, etc. Microscopic characteristics of 12 batches of O. corniculata powder were observed. According to the corresponding methods in 2015 edition of Chinese Pharmacopoeia (part Ⅳ), TLC was used for qualitative identification [developing solvent was trichloromethane-methanol-formic acid (8 ∶ 1 ∶ 0.1, V/V/V)], and the contents of moisture, total ash, acid insoluble ash and alcohol soluble extractive from 12 batches of O. corniculata were determined. The content of isovitexin was determined by HPLC. The determination was performed on Venusil XBP C18 (L) with mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (15 ∶ 85, V/V) at the flow rate of 1 mL/min. The column temperature was 35 ℃, and the detection wavelength was set at 338 nm. The sample size was 10 μL. RESULTS: Microscopic observation showed that the powder was grayish brown to yellowish brown, with many non-glandular hairs and obvious fibrous pore. Results of TLC identification showed that the spots of the same color appeared in the corresponding positions of the test and the control chromatogram. The contents of moisture, total ash, acid insoluble ash and alcohol soluble extract from samples were 6.66%-12.13%, 9.16%-13.79%, 1.58%-4.63% and 5.22%-15.79%, respectively. Results of HPLC method showed that the concentration of isovitexin showed a good linear relationship in the range of 5.20-78.3 μg/mL (r=0.999 0); RSDs of reproducibility (n=9), intermediate precision (n=6) and stability (24 h, n=6) tests were all lower than 2.0%; and the recovery rates were 97.54%-99.52% (RSD=0.74%, n=6); the contents of isovitexin in 12 batches of O. corniculata were 0.036%-0.144% (n=3). CONCLUSIONS: Qualitative and quantitative identification methods of O. corniculate were established, which can be used as a reference for improving the quality standard of O. corniculata.
