1.Brusatol induces apoptosis in small cell lung cancer by inhibiting STAT3 phosphorylation

Aim To investigate the effect of Brusatol a-gainst small cell lung cancer(SCLC)and its potential mechanism.Methods CCK-8 assay and flow cytome-try were used to detect the cytotoxic effect of Brusatol on SCLC cells.Western blot was employed to measure the expression levels of apoptosis-related proteins,in-cluding cleaved poly(ADP-ribose)polymerase(cleaved-PARP),B-cell lymphoma 2(Bcl-2)and Bcl-2-associated X protein(Bax).Network pharma-cology databases were utilized to identify common tar-gets of Brusatol,SCLC,and apoptosis.Kyoto Encyclo-pedia of Genes and Genomes(KEGG)and Gene On-tology(GO)enrichment analyses were performed on the intersecting genes.Molecular docking simulations between Brusatol and core targets were conducted using the CB-DOCK2 online platform to calculate binding en-ergies and sites.Western blot was further applied to detect the expression levels of signal transducer and ac-tivator of transcription 3(STAT3)and phosphorylated-STAT3(p-STAT3).Results Brusatol inhibited SCLC cell growth and induced apoptosis,significantly downregulating Bcl-2 and cleaved-PARP while upregu-lating Bax expression(P<0.05).Network pharma-cology analysis revealed 108 common targets of Brusa-tol and SCLC,with the top three core targets being ep-idermal growth factor receptor(EGFR),STAT3,and tumor necrosis factor(TNF).Molecular docking re-sults indicated strong binding affinity between bruceine D and these core targets.Western blot validation con-firmed that bruceine D suppressed the expression of STAT3 and p-STAT3.Conclusion Brusatol exerts anti-SCLC effects by inhibiting STAT3 to induce apop-tosis in SCLC cells.

Chronic kidney disease(CKD)is a chronic disease characterized by renal structural damage and dysfunction.At present,there is still a lack of effective therapeutic drugs and prevention and treatment methods for CKD in clinical practice.More and more studies have shown that necroptosis,as a new type of programmed cell death,plays a vital role in the onset and progression of CKD.Targeting key molecules in the necroptosis pathway,such as RIPK1,RIPK3 and MLKL,the development of small molecule inhibitors has become an emerging strategy for the treatment of CKD,and has shown significant potential to pro-tect the kidneys and alleviate renal fibrosis in a variety of in vitro and in vivo models.Therefore,this article summarizes the re-search progress of the mechanism of necroptosis in recent years,and focuses on the potential role of necroptosis in the pathogene-sis of CKD and the therapeutic potential of targeting this path-way,providing a new perspective and research direction for the prevention and treatment of CKD in the future.

4.Clinical significance of postoperative recurrence pattern and prognosis of pancreatic ductal adenocarcinoma

Objective:This study aimed was to analyze the disease progression of postoperative recurrence/me-tastasis of pancreatic ductal adenocarcinoma(PDAC)and provide clinical evidence for evaluating the difference in prog-nosis between site and pattern of recurrence/metastasis.Methods:The clinical data of 167 patients with PDAC who underwent pancreatectomy in Panjin Liao-Oil Gem Flower Hospital from January 2011 to June 2019 were retrospec-tively analyzed.Kaplan-Meier curves and Cox-regression analyses were applied to assess survival outcomes.Results:In this study,96 patients(57.5%)recurred within 12 months after surgery,and the median recurrence time was 10.9 months.Compared to patients without recurrence,patients with recurrence/metastasis had higher baseline CA19-9>100 IU/mL and were treated more often with neoadjuvant chemotherapy(P<0.05).In the no recurrence group,patients had histopathological analyses yielded lower less poorly differentiated tumors,less positive margin,less pT3+4 stages and pN1+2 stages(P<0.05).In a multivariate Cox regression analysis,receipt of neoadjuvant chemotherapy,poor tumor differ-entiation and positive margin were independent predictors for recurrence/metastasis within 12 months after surgery(P<0.05).Neoadjuvant treated patients had less pT1+2 stages(P=0.015),a lower lymph node ratio,less perineural invasion,less positive margin,and were less likely to be treated with adjuvant chemotherapy compared to upfront surgery treated patients(P<0.05).Recurrence/metastasis rates were higher in the neoadjuvant treated cohort versus the upfront surgery cohort(P=0.014).There was no statistical difference in the recurrence/metastasis rate distribution between neoadjuvant therapy and upfront surgery,and neoadjuvant therapy exhibited no significant impact on recurrence/metastasis patterns in PDAC(P>0.05).The overall survival period of patients without recurrence/metastasis within 12 months was signifi-cantly longer than that of patients with recurrence/metastasis after PDAC resection(P<0.001).Conclusion:The varying site recurrence/metastasis rates and time to recurrence/metastasis,underline the inherent heterogeneity of PDAC and the specific interactions in the tumor microenvironments at the recurrence/metastasis sites.Further research is essential to establish optimal personalized treatment strategies for patients with recurrent PDAC.
5.Application of perfusion tube in the drying of digestive endoscope channels

Objective To compare the effects of perfusion tube and pressure air gun on endoscopic channels drying by using the stripped endoscope model,so as to explore the application value of perfusion tube in endoscopic channels drying.Methods After the standard manual cleaning and disinfection process,the stripped endoscope model was randomly divided into group A and group B.Each group was subjected to 30 tests.Group A was injected with a perfusion tube,and group B was injected with a pressure air gun.The liquid residues in the endoscope tubes were observed visually and with a magnifying lens,and the time of thorough drying of the endoscope channels and the physical fatigue of the operators were compared between the two groups.Results The average drying time of the group A was(366.47±75.74)s,the average drying time of the group B was(338.40±65.81)s,there was no significant difference between the two groups(t=-1.53,P=0.131).The fatigue degree of the group A was lower than group B(t=16.79,P<0.01).Conclusion There is no statistical difference in the air delivery time between the perfusion tube and the pressure air gun drying endoscopic tube,and the operator's physical fatigue is light when the perfusion tube is used,so it is recommended to use the perfusion tube drying endoscopic tube in clinic.
6.Investigation and recommendations on microbiological contamination of 115 endoscopes received before and after disinfection

To study the microbiological contamination of endoscopic forceps channel at the time of reception and after disinfection, and gain a preliminary understanding of their real disinfection quality, then analyse the problems and make recommendations. A total of 115 endoscopes received at Renmin Hospital of Wuhan University from June 2022 to December 2023 were chosen. Microbiological sampling and strain identification of the endoscopic forceps channel were carried out at the time of reception of the endoscopes and after cleaning and disinfection, respectively, then the qualified disinfection rate and microbial detection rate were compared. The overall qualified disinfection rate after cleaning and disinfection of endoscopes received (91.3%, 105/115) was higher than that at reception (57.4%, 66/115, χ 2=37.026, P<0.001). The overall microbiological detection after cleaning and disinfection of endoscopes received (13.0%, 15/115) was lower than that at reception (48.7%, 56/115, χ 2=41.000, P<0.001), the detection rate of high pathogenic organisms after cleaning and disinfection (7.0%, 8/115) was lower than that at reception (29.6%, 34/115, χ 2=24.039, P<0.001), low pathogenic organisms after cleaning and disinfection (6.1%, 7/115) was lower than that at reception (19.1%, 22/115, χ 2=13.067, P<0.001). At the time of reception, the qualified disinfection rate was low and the microbiological detection rate was high, and there may become a greater risk of cross-infection; after cleaning and disinfection, the qualified disinfection rate was increased and the microbiological detection rate was decreased, which may greatly reduce the risk of patient infection. Therefore, it is recommended that attention should be paid to the disinfection quality monitoring of endoscopes received before putting into clinical use.

9.Mechanism of dioscin inhibiting apoptosis in HT22 cells after OGD/R

Aim To investigate the neuroprotective effect of dioscin(DIO)on hippocampal neurons(HT22)after oxygen glucose deprivation/reoxygen-ation(OGD/R)and its possible mechanism.Methods HT22 cells were treated with 0,2.5,5,10,20,40,80,160,and 320 mg·L-1DIO for 24 h,and the cell proliferation rate was detected by CCK-8 method.The concentration that was non-toxic to HT 2 2 cells was se-lected for subsequent experiments.After OGD for 2 h,HT22 cells were randomly divided into the OGD/R group,1.25,2.5,and 5 mg·L-1DIO group,and posi-tive control group.HT22 cells were taken as the con-trol group.After drug intervention for 24 h,the cell proliferation rate was detected by CCK-8 method;the LDH release was detected by colorimetry;the cell ap-optosis rate was detected by TUNEL method;the ex-pression of proteins related to PARP-1/AIF pathway and caspase pathway was detected by Western blot.Results DIO intervention significantly upregulated the expression of AIF protein in mitochondria and PAR protein in nucleus of HT22 cells after OGD/R,and sig-nificantly downregulated the release of LDH,neuronal apoptosis rate,total protein expression of AIF and PAR,PARP-1,AIF in nucleus and protein expression of PAR protein in mitochondria,while the expression of Bax and caspase-3 proteins was not significantly differ-ent from that in the OGD/R group.Conclusion DIO can alleviate the apoptosis of HT22 cells induced by OGD/R by regulating the expression and translocation of proteins related to the PARP-1/AIF pathway,thus playing a neuroprotective role.
