1.Selection of fecal antigen diagnostic markers for Echinococcus granulosus

This study aims to screen the diagnostic biomarkers for fecal antigen of Echinococcus granulosus(E.granulosus)in dogs with high specificity and sensitivity.The sheep-derived EgPSC artificially infected dogs were collected,and the negative and positive fecal samples of dogs with E.granulosus were prepared by arecoline hydrobromide leakage method.Polyclonal antibody,negative fecal antigen-polyclonal antibody conjugates and positive fecal antigen-polyclonal antibody conju-gates were purified by ammonium sulfate precipitation and affinity chromatography,three groups of samples were detected by ELISA and Western blot,LC-MS/MS and bioinformatics analysis were performed on the three groups of samples.The positive fecal antigen-polyclonal antibody con-jugate was used as the treatment group,the polyclonal antibody and the negative fecal antigen-polyclonal antibody conjugates were used as the control groups to screen the unique peptides of the treatment group.ELISA and Western blot showed that only the positive fecal antigen-polyclonal antibody conjugates were positive.According to LC-MS/MS and bioinformatics analysis,11 unique peptides were screened out only in the treatment group.Among them,3 proteins were related to E.granulosus,namely dysferlin,integrator complex 9 and diagnostic antigen gp50,which were mem-brane-associated proteins,INT complex components and diagnostic antigens.This study has pre-liminarily screened out three candidate canine E.granulosus fecal antigen diagnostic markers,pro-viding a reference for further exploration of diagnostic standards for E.granulosus,screening of echinococcosis target genes,and vaccine development.
2.Effects of Tiaoshu Anshen acupuncture on sleep quality and serum neurotransmitter levels in patients with chronic insomnia.

OBJECTIVE:
To observe the effects of Tiaoshu Anshen (regulating the hinge and calming the mind) acupuncture on sleep quality and serum levels of 5-hydroxytryptamine (5-HT) and dopamine (DA) in patients with chronic insomnia.
METHODS:
A total of 58 patients with chronic insomnia were randomly divided into an acupuncture group and a medication group, 29 cases in each group. Tiaoshu Anshen acupuncture was applied at Baihui (GV20) and bilateral Shenmen (HT7), Sanyinjiao (SP6), Benshen (GB13) in the acupuncture group, once a day, 1-day interval was taken after 6 consecutive days of treatment. Estazolam tablet was given orally before bed in the medication group, 1 mg each time. The 4-week treatment was required in both groups. Before and after treatment, the sleep quality was assessed by Pittsburgh sleep quality index (PSQI) and polysomnography (PSG), the serum levels of 5-HT and DA were detected by ELISA.
RESULTS:
After treatment, the item scores and total scores of PSQI were decreased compared with those before treatment in the two groups (P<0.05); in the acupuncture group, the scores of sleep quality, sleep latency, sleep time, sleep efficiency, sleep disorders and total score of PSQI were lower than those in the medication group (P<0.05). After treatment, the total sleep time (TST) was prolonged (P<0.05), the sleep latency (SL) and wake after sleep onset (WASO) were shortened (P<0.05), the sleep efficiency (SE%), percentage of non-rapid eye movement stage 3 (N3%), percentage of rapid eye movement stage (REM%) and serum levels of 5-HT were increased (P<0.05) compared with those before treatment; the percentage of non-rapid eye movement stage 1 (N1%), percentage of non-rapid eye movement stage 2 (N2%) and serum levels of DA were decreased (P<0.05) compared with those before treatment in the two groups. After treatment, in the acupuncture group, TST was longer, while SL and WASO were shorter than those in the medication group (P<0.05), SE%, N3%, REM% and serum level of 5-HT were higher, while N1%, N2% and serum level of DA were lower than those in the medication group (P<0.05).
CONCLUSION
Tiaoshu Anshen acupuncture may improve the sleep quality by regulating the serum neurotransmitter levels i.e. 5-HT and DA in patients with chronic insomnia.

4. Homeobox Gene Six3 is Required for the Differentiation of D2-Type Medium Spiny Neurons

Medium spiny neurons (MSNs) in the striatum, which can be divided into D1 and D2 MSNs, originate from the lateral ganglionic eminence (LGE). Previously, we reported that Six3 is a downstream target of Sp8/Sp9 in the transcriptional regulatory cascade of D2 MSN development and that conditionally knocking out Six3 leads to a severe loss of D2 MSNs. Here, we showed that Six3 mainly functions in D2 MSN precursor cells and gradually loses its function as D2 MSNs mature. Conditional deletion of Six3 had little effect on cell proliferation but blocked the differentiation of D2 MSN precursor cells. In addition, conditional overexpression of Six3 promoted the differentiation of precursor cells in the LGE. We measured an increase of apoptosis in the postnatal striatum of conditional Six3-knockout mice. This suggests that, in the absence of Six3, abnormally differentiated D2 MSNs are eliminated by programmed cell death. These results further identify Six3 as an important regulatory element during D2 MSN differentiation.

6.A Fluorescence Diffusion Optical Tomography System Based on Lattice Boltzmann Forward Model.

Fluorescence Diffuse Optical Tomography (FDOT) is significant for biomedical applications, such as medical diagnostics, drug research. The fluorescence probe distribution in biological tissues can be quantitatively and non-invasively obtained via FDOT, achieving targets positioning and detection. In order to reduce the cost of FDOT, this study designs a FDOT system based on Lattice Boltzmann forward model. The system is used to realize two functions of light propagation simulation and FDOT reconstruction, and is composed of a parameter module, an algorithm module, a result display module and a data interaction module. In order to verify the effectiveness of the platform, this study carries out the light propagation simulation experiment and the FDOT reconstruction experiment, respectively comparing the Monte Carlo (MC) light propagation simulation results and the real position of the light source to be reconstructed. Experiments show that the proposed FDOT system has good reliability and has a high promotion value.

7.A Method for Fluorescent Diffuse Optical Tomography Based on Lattice Boltzmann Forward Model on GPU Parallelization.

Fluorescent Diffuse Optical Tomography (FDOT) is an emerging imaging method with great prospects in fields of biology and medicine. However, the current solutions to the forward problem in FDOT are time consuming, which greatly limit the application. We proposed a method for FDOT based on Lattice Boltzmann forward model on GPU to greatly improve the computational efficiency. The Lattice Boltzmann Method (LBM) was used to construct the optical transmission model. This method separated the LBM into collision, streaming and boundary processing processes on GPUs to perform the LBM efficiently, which were local computational and inefficient on CPU. The feasibility of the proposed method was verified by the numerical phantom and the physical phantom experiments. The experimental results showed that the proposed method achieved the best performance of a 118-fold speed up under the precondition of simulation accuracy, comparing to the diffusion equation implemented by Finite Element Method (FEM) on CPU. Thus, the LBM on the GPU may efficiently solve the forward problem in FDOT.

8. Efficacy and safety of domestic dasatinib as second-line treatment for chronic myeloid leukemia patients in the chronic phase

Objective:
To investigate the efficiency and safety of domestic tyrosine kinase inhibitor (TKI) dasatinib (Yinishu) as second-line treatment for patients with chronic myeloid leukemia in chronic phase (CML-CP).
Methods:
A retrospective analysis of clinical data of CML-CP patients who received domestic dasatinib as second-line treatment in the CML collaborative group hospitals of Hubei province from March 2016 to July 2018 was performed. The optimal response rate, the cumulative complete cytogenetic response (CCyR), the cumulative major molecular responses (MMR), progression free survival (PFS), event free survival (EFS) and adverse effects (AEs) of the patients were assessed at 3, 6 and 12 months of treatment.
Results:
A total of 83 CML-CP patients were enrolled in this study. The median follow-up time was 23 months. The optimal response rates at 3, 6 and 12 months in 83 CML-CP patients treated with dasatinib were 77.5% (54/71), 72.6% (61/75) and 60.7% (51/69), respectively. By the end of follow-up, the cumulative CCyR and MMR rates were 65.5% (55/80) and 57.1% (48/73), respectively. The median time to achieving CCyR and MMR was 3 months. During follow-up time, the PFS rate was 94.0% (79/83) and the EFS rate was 77.4% (65/83). The most common non-hematological AEs of dasatinib were edema (32.5%), rash itching (18.1%) and fatigue (13.3%). The common hematological AEs of dasatinib were thrombocytopenia (31.3%), leukopenia (19.3%) and anemia (6.0%).
Conclusion
Domestic dasatinib was effective and safe as the second-line treatment of CML-CP patients and it can be used as an option for CML-CP patients.
9.Hippocampus MRI Parallel Segmentation Using Three Dimensions Lattice Boltzmann Model with Prior Information.

Getting volume change of hippocampus by segmenting on brain MRI is an important step in the diagnose of Alzheimer's disease and other brain disease. Three dimensional segmentation can make use of the correlation of image in gray and spatial position, so it has high accuracy. This paper proposes a novel three-dimensional lattice Boltzmann model combined with the surface evolution of deformable model and taking the prior information as an external force term to constrain the evolution of three dimensional surfaces. In order to solve the problem of high computational cost caused by 3D segmentation, the parallelization of the method is programmed on single GPU platform and dual GPU platform. Comparison experiments were set to test the accuracy of segmentation and computational efficiency between the novel LB method and another method by using 20 real AD patient's MRI from ADNI. In ensuring the accuracy of the segmentation, the time can be reduced to 12.76 s on single GPU platform, and 17.32 s on dual GPU platform, contrasting 132.43 s on CPU platform. It fully validates the characteristics of lattice Boltzmann method which can be highly parallelized.

10.Simulation Platform of Photoacoustic Imaging Based on Finite Element and -space Pseudospectral Method.

Numerical simulation is a powerful technology for photoacoustic imaging (PAI) in both theory studies and practical applications. In this paper, a simulation platform for PAI was designed and implemented based on Matlab. The simulation platform utilized finite element method (FEM) and -space pseudospectral method to calculate the forward and inverse problem of PAI. And a graphical user interface (GUI) was realized. Structural design, work process and other operating details of the platform was also provided. By compared with theoretical temporal waveform of photoacoustic signal and reconstruction results of COMSOL, the validity and reliability was verified. And a reliable simulation tool was proposed for PAI.
