1.Application of immunoprecipitation-mass spectrometry for screening of antibody-recognizing molecules against Echinococcus granulosus

We collected the positive serum of Echinococcus granulosus infection in sheep,an inter-mediate host with strong immune response,and used healthy serum as negative control,purified the serum and target protein to capture and enrich the corresponding antigen by immunoprecipita-tion,and obtained target protein-antibody-target protein complex.Mass spectrometry strategies were combined to screen and identify specific antigens associated with Echinococcus granulosus,and the proteins with the highest peptide coverage were analyzed bioinformatically using online prediction software.The results showed that 133 Echinococcus granulosus related proteins were i-dentified by IP-MS.Among them,one protein with peptide coverage≥70%was actin Ⅱ,and three proteins with peptide coverage between 30%to 40%were Ton B box domain containing protein,NADH dehydrogenase(ubiquinone)1 α-subcomplex 2(NADH dehydrogenase[ubiquinone])and lactic dehydrogenase.There were six proteins with 20%to 30%peptide coverage,namely,spli-cing factor 3B subunit 5,tumor protein D52,expressed conserved protein,NADH dehydrogenase(ubiquinone)1 alpha subcomplex 7,inosine-5'-monophosphate dehydrogenase,and aldo keto re-ductase family 1 member B4.Bioinformatics analysis revealed that actin protein has no signal pep-tide,it is probably a non-secretory protein and is subcellularly localized to the cytoskeleton,six op-timal potential antigenic epitopes are present,and the secondary and tertiary structures are consist-ently dominated by α-helices and irregular convolutions.The results indicate that immunoprecipita-tion-mass spectrometry is a high-throughput,simple,rapid and effective method for screening and identifying fine-grained Echinococcus granulosus antigens,which can provide a basis for screening specific molecules for serodiagnostic markers in intermediate host sheep and for the development of novel diagnostic techniques for hydatid diseases.

3.Effects of canthaxanthin in increasing reproductive hormone secretion through re-ducing ovarian oxidative stress injury on egg production rate of Huaixiang chickens

A total of 360 healthy 26 week old Huaixiang hens were randomly divided into 10 groups,of which,five groups were reared at normal temperature(NC,NT1,NT2,NT3,NT4)and other five groups were reared at high temperature(HC,HT1,HT2,HT3,HT4),all chickens were feed with 0,4,6,8,10 mg/kg canthaxanthin(CX)respectively.The pre-test period was 2 weeks and the main test period was 9 weeks.The results showed that high temperature reduced the egg pro-duction rate(P<0.05),led to damage and hemorrhage in the ovaries,decreased the activity of su-peroxide dismutase(SOD),glutathione peroxidase(GSH-Px)in the serum and the ovaries(P<0.05),decreased the levels of FSH,LH,P4,and E2(P<0.05),decreased the expression of FSHR,LHR,ER,and PR(P<0.05),and increased the levels of malonaldehyde(MDA)(P<0.05)in the ovaries.Addition of CX increased the egg production rate(P<0.05),increased the activity of serum and ovarian SOD and GSH-Px enzymes(P<0.05),decreased the content of serum and ovarian MDA(P<0.05)),increased the ovarian levels of FSH,LH,P4,E2(P<0.05),and increased the o-varian expression of FSHR,LHR,ER,and PR(P<0.05)in Huaixiang chickens at normal and high temperature environmental conditions,respectively.Among them,the highest egg production rate,the strongest antioxidant capacity of serum and ovarian tissues,the best integrity of ovarian tis-sues,ovarian reproductive hormone secretion and expression of reproductive hormone receptors were observed in Huaixiang chickens when 6 mg/kg of CX was added to the diet under normal temperature and when 8 mg/kg CX was added to the diet under high temperature conditions.The results showed that under normal and high temperature conditions,adding CX to the diet signifi-cantly increased the antioxidant capacity of serum and ovaries,alleviated the damage of ovary,pro-moted the secretion of reproductive hormone and the expression of reproductive hormone recep-tors,and increased egg production rate.Dietary supplemented with CX could not restore the dam-age and repair of ovarian tissue,the secretion of reproductive hormone and receptor to normal tem-perature level.The effects of CX on egg production rate,ovarian tissue structure,reproductive hor-mone and receptor expression in chickens were affected by the environmental temperature.When the egg production rate,antioxidant capacity of ovarian tissues,reproductive hormone levels and receptor expression of Huaixiang chickens were optimal,the amount of CX in high temperature en-vironment was more than that of the dose in normal temperature environment.

An essential step for cancer vaccination is to break the immunosuppression and elicit a tumor-specific immunity. A major hurdle against cancer therapeutic vaccination is the insufficient immune stimulation of the cancer vaccines and lack of a safe and efficient adjuvant for human use. We discovered a novel cancer immunostimulant, trichosanthin (TCS), that is a clinically used protein drug in China, and developed a well-adaptable protein-engineering method for making recombinant protein vaccines by fusion of an antigenic peptide, TCS, and a cell-penetrating peptide (CPP), termed an "all-in-one" vaccine, for transcutaneous cancer immunization. 5.Designed eye position of pilot′s helmet mounted display based on 3D scanning

Objective:To provide accurate eye position data for the system design of helmet mounted display by studying the eye position distribution and anthropometric items of pilots' head and face.Methods:The head and face data of 372 Chinese male fighter pilots were captured by a non-contact 3D scanning system to build cloud models. By noise cutting and mending processing each head and face model was built with same template mesh and unified anatomical coordinate system. The eye position design was defined according to monocular or binocular display. The positions (core of pupils) of left and right eyes and 12 anthropometric items were calculated from the 3D models.Results:The database of 372 fighter pilots′ head and face 3D models was established and it included 12 anthropometric items such as the coordinates of left and right eyes position, inter pupillary distance and vertex-pupil height.Conclusions:Based on 3D scanning technology, the distribution of eye locations (designed eye position and eye box) of pilots were built under the human head anatomical coordinate system and related anthropometric items of head and face are obtained, which could be used as the data foundation for designing pilot′s helmet mounted display system.

7.A Fluorescence Diffusion Optical Tomography System Based on Lattice Boltzmann Forward Model.

Fluorescence Diffuse Optical Tomography (FDOT) is significant for biomedical applications, such as medical diagnostics, drug research. The fluorescence probe distribution in biological tissues can be quantitatively and non-invasively obtained via FDOT, achieving targets positioning and detection. In order to reduce the cost of FDOT, this study designs a FDOT system based on Lattice Boltzmann forward model. The system is used to realize two functions of light propagation simulation and FDOT reconstruction, and is composed of a parameter module, an algorithm module, a result display module and a data interaction module. In order to verify the effectiveness of the platform, this study carries out the light propagation simulation experiment and the FDOT reconstruction experiment, respectively comparing the Monte Carlo (MC) light propagation simulation results and the real position of the light source to be reconstructed. Experiments show that the proposed FDOT system has good reliability and has a high promotion value.

8.Three-dimensional Reconstruction of Retinal Vessels Based on Binocular Vision.

In robot-assisted eye surgery, such as retinal vascular bypass surgery, precise positioning of operating points is required. In this study, a binocular vision-based 3D reconstruction method is proposed to locate the incision points on retinal vessels. Vessels in the image were extracted by CLAHE algorithm to remove the influence of background, then stereo matching was performed. Finally, the retinal vessel image was reconstructed by using the principle of parallax in binocular vision. Experimental results show that this method can accurately locate the incision points on retinal vessels and meet the requirements of ophthalmic surgery.

9.A Method for Fluorescent Diffuse Optical Tomography Based on Lattice Boltzmann Forward Model on GPU Parallelization.

Fluorescent Diffuse Optical Tomography (FDOT) is an emerging imaging method with great prospects in fields of biology and medicine. However, the current solutions to the forward problem in FDOT are time consuming, which greatly limit the application. We proposed a method for FDOT based on Lattice Boltzmann forward model on GPU to greatly improve the computational efficiency. The Lattice Boltzmann Method (LBM) was used to construct the optical transmission model. This method separated the LBM into collision, streaming and boundary processing processes on GPUs to perform the LBM efficiently, which were local computational and inefficient on CPU. The feasibility of the proposed method was verified by the numerical phantom and the physical phantom experiments. The experimental results showed that the proposed method achieved the best performance of a 118-fold speed up under the precondition of simulation accuracy, comparing to the diffusion equation implemented by Finite Element Method (FEM) on CPU. Thus, the LBM on the GPU may efficiently solve the forward problem in FDOT.

10.Clinical anatomical study on the development of pedicle of atlantoaxial pedicle in preschool children

Objective To discuss the characteristics,growth and development of atlantoaxial pedicle in preschool children,and to provide the basic theoretical basis for the design,production and clinical application of pedicle screw technique in the treatment of atlantoaxial disease in preschool children.Methods Excluded patients with atlantoaxial fractures,deformities,tuberculosis and tumor,the clinical data of 60 children aged from 3 to 6 years old with cervical spondylolisthesis were collected.The original CT images were reconstructed in DICOM format and the subjects were reconstructed with Mimics software.The subjects were grouped by age to measure and analyze the atlantoaxial pedicle correlation.Results The pedicle width(PW),pedicle height(PH) and total length of pedicle bone channel (PL) increased gradually with age.The PW/PH ratio of the vertebrae(C1) is greater than 1.0,and that is, the width of the pedicle of the C1 pedicle is greater than the height.The PW/PH ratio of the vertebral (C2) is less than 1.0,which means the width of the pedicle of the C1 pedicle is less than the height.Conclusion The growth and development of spine in preschool children are fast, and the pedicle of atlantoaxial calculus in different age groups has different developmental characteristics.In this study,the diameter of the pedicle of the atlantoaxial pedicle in the 3 to 6 year old children is increasing with age.The pedicle screw with the smallest diameter of 3.5 mm can be implanted into the atlantoaxial pedicle with feasibility in theory.
