Objective To evaluate the value of postoperative radiotherapy (PORT) in patients with stage ⅢA-N2 non-small cell lung cancer who received complete resection and chemotherapy. Methods Patients with stage ⅢA-N2 non-small cell lung cancer who received complete resection and chemotherapy were chosen from the SEER Research Plus Database [17 Registries, November 2012 Submission (2000-2019)]. The patients were divided into a PORT group and a non-PORT group according to whether the PORT was used. To balance baseline characteristics between non-PORT and PORT groups, R software was used to conduct a propensity score matching (PSM) with a ratio of 1 : 1 and a matching tolerance of 0.01. Both the Cox regression analysis and Kaplan-Meier survival analysis were conducted to evaluate the value of PORT in terms of overall survival (OS) and disease-specific survival (DSS). Results In total, 2468 patients with stage ⅢA-N2 non-small cell lung cancer were enrolled, including 1078 males and 1390 females with a median age of 65 (58-71) years. There were 1336 patients in the PORT group, and 1132 patients in the non-PORT group. Cox regression analysis showed that PORT was not significantly associated with OS (multivariate analysis: HR=1.051, 95%CI 0.949-1.164, P=0.338) and DSS (multivariate analysis: HR=1.094, 95%CI 0.976-1.225, P=0.123). No statistical difference was found in the OS or DSS between non-PORT group and PORT group after PSM analysis (P>0.05). Conclusion PORT does not have a survival benefit for patients with stage ⅢA-N2 non-small cell lung cancer who received complete resection and chemotherapy.

This study was aimed at establishing a method of ultra-fast quantitative PCR for Brucella detection. We used an exogenous recombinant plasmid as the internal reference and targeted the T4SS secretion system,an important Brucella viru-lence factor,to design specific primers and probes. The sensitivity, specificity, and repeatability of this method were evaluated, and a standard curve was constructed. The coincidence rate of detection findings with this method versus quantitative PCR was determined. This method markedly decreased the detection time to only 10 minutes. The standard curve demonstrated a good linear relationship(Y=-3.410 7x+38.357,R2=0.998 5)with a low minimum detection limit of 10 copies/μL.The method exhibited good specificity and did not specifically amplify several common clinical bacteria other than Brucella. The de-tection of three concentrations of positive plasmids yielded coefficients of variation(CVs)of 0.20%to 0.91%,thus demonstra-ting the method's excellent repeatability.Furthermore,140 clinical samples were analyzed concurrently with the fluorescence PCR method, which yielded a 100%compliance rate and consistent results. Our findings indicated that the Brucella ultra-fast quantitative PCR was ultrafast;had high sensitivity, high specificity, and good specificity;and can be used for the clinical de-tection of Brucella and emergency investigation of epidemics. Therefore, this method is valuable for the early diagnosis of Bru-cella.

OBJECTIVE To conduct qualitative analysis of chemical constituents in the crude extract of Modified Ermiao Formu-la,along with its blood-absorbed components and metabolites in rats post-administration employing ultra-performance liquid chroma-tography coupled with quadrupole-exactive orbitrap tandem mass spectrometry(UPLC-Q-Exactive Orbitrap-MS/MS).METHODS Separation was performed on a Waters HSS T3 column(100 mm×2.1 mm,1.8 μm)using gradient elution with 0.1%formic acid in water(mobile phase A)and 0.1%formic acid in acetonitrile(mobile phase B),under controlled conditions:column temperature maintained at 40℃,flow rate set to 0.3 mL·min-1,and injection volume fixed at 2 μL.Mass spectrometric data acquisition utilized an electrospray ionization(ESI)source with scanning in both positive and negative ion modes.RESULTS By analyzing precise mo-lecular weights, retention times, and MS/MS fragmentation patterns, and cross-referencing these against established databases and published literature,173 chemical constituents were definitively identified in the Modified Ermiao Formula crude extract. These prima-rily comprised flavonoids, organic acids, and alkaloids.Additionally,32 prototype components and 13 metabolites were characterized in the serum of dosed rats.Organic acids constituted the predominant class of serum prototype components, undergoing in vivo metabo-lism principally through demethylation and carboxyl glucuronidation.CONCLUSION This study provides the initial delineation of blood-absorbed prototype components derived from Modified Ermiao Formula, with concomitant identification of their metabolites, thereby establishing a foundational framework for elucidating the pharmacologically active constituents of this formula.

Objective:To analyze the characteristics and general patterns of adverse drug reactions associated with aripiprazole, and to identify potential risk signals, thereby providing references for the safe and rational use of medications in clinical practice. Methods:This is a descriptive study that collected and summarized reports of adverse reactions to aripiprazole submitted by medical institutions in Anhui Province to the National Center for Adverse Drug Reaction Monitoring, China from January 2019 to December 2024. The basic characteristics of patients, the treatment regimens used, the timing of adverse drug reaction occurrence, the clinical manifestations of the affected systems, and the outcomes of these adverse drug reactions were statistically analyzed to identify the clinical characteristics and risk factors of adverse drug reactions associated with aripiprazole. Results:A total of 1 106 adverse reaction case reports related to aripiprazole were collected, with a male-to-female ratio of 1:1.29 ( χ2 = 8.64, P = 0.003) and an average age of (35 ± 16) years. The primary diseases treated included schizophrenia (768 cases, 69.44%), bipolar affective disorder (96 cases, 8.68%), and depressive disorder (75 cases, 6.78%). Adverse reactions occurred within the first 2 weeks of medication in 689 cases (62.29%). All medications were oral formulations, with 978 cases (88.43%) falling within the recommended daily dose range of 10-30 mg/d, while 15 cases (1.36%) exceeding the maximum recommended dose of 30 mg/d. The most common clinical manifestations were related to the nervous system (696 cases, 62.93%), followed by cardiovascular system manifestations (146 cases, 13.20%). The majority of adverse reactions showed improvement or recovery (1 052 cases, 95.12%). Conclusions:Aripiprazole is widely used in clinical practice, and the occurrence of adverse reactions is correlated with patient characteristics and treatment regimens. It is advisable to enhance the rational clinical use and monitoring of aripiprazole. Such measures will aid in preventing or reducing the occurrence of severe adverse reactions.
